Effects of Synbiotics Containing Anaerobic Microbes and Prebiotics on In vitro Fermentation Characteristics and In situ Disappearance Rate of Fermented-TMR

  • This study was carried out to estimate effects of synbiotics containing anaerobic microorganisms and prebiotics on in vitro fermentation characteristics and in situ disappearance rate of fermented total mixed ration (F-TMR). For the in vitro trial, ninety vinyl bags were prepared to analyze temperature, pH, ammonia concentration, microbial growth rate and short chain fatty acid concentration. For the in situ trial, one hundred twenty nylon bags were prepared to analyze dry matter (DM), acid detergent fiber (ADF) and neutral detergent fiber (NDF) disappearance rate. Treatments consisted of a basal diet (US) with prebiotics and probiotics from anaerobic mold (MS), bacteria (BS), yeast (YS) or compound (CS). It was found that temperatures at 14 and 21 days were significantly higher (p<0.05) in the YS and CS than in the others. The pH at 21 days was lower in the CS than in the US. The synbiotic treatments had significantly increased (p<0.05) ammonia concentration at 21 days. The DM disappearance at 72 h was significantly higher (p<0.05) in the MS and CS than in the others. ADF and NDF disappearance rate tended to increase at a rate similar to the DM disappearance rate. Therefore, this study suggests that synbiotics (probiotics with prebiotics) may partially help the quality of fermentation and digestibility of TMR (MS and CS) as fiber disappearance.

Effects of Fermented Total Mixed Ration and Cracked Cottonseed on Milk Yield and Milk Composition in Dairy Cows

  • Four lactating Holstein Friesian crossbred cows, with an average initial weight of 450 kg, 48${\pm}$12 days in milk and initial milk yield of 18 kg/h/d, were randomly arranged according to a 2${\times}$2 factorial arrangement in a 4${\times}$4 in Latin square design with 21-d period to investigate the effects of type of total mixed ration (TMR) and type of whole cottonseed (WCS) on intake, digestibility and milk production. The dietary treatments were i) TMR and WCS supplementation at 0.5 kg/h/d, ii) TMR and cracked WCS (cWCS) supplementation at 0.5 kg/h/d, iii) fermented TMR (FTMR) and WCS supplementation at 0.5 kg/h/d, and iv) FTMR and cWCS supplementation at 0.5 kg/h/d. Voluntary feed intake was 15.9, 15.2, 15.4 and 15.6 kg DM/d in dietary treatment 1, 2, 3 and 4, respectively. Digestibility of DM, OM, CP, EE, NDF and ADF were not significantly different among dietary treatments. Ruminal pH, $NH_{3}-N$ and volatile fatty acids in the rumen were also not significantly different among type of TMR or type of WCS. Blood urea-N concentration was not significantly different among dietary treatments. Ruminal bacteria population tended to increase but ruminal protozoa population tended to decrease with supplementation of cWCS, but they were not affected by FTMR. Milk yield and 3.5% FCM were not statistically different among treatments (16.6, 16.2, 17.0, 16.3 kg/d and 18.0, 18.6, 19.9 and 19.0 kg/d, respectively). Milk composition was not significantly different among dietary treatments. However, unsaturated fatty acids in milk fat in cows fed FTMR were lower (p<0.05) than in cows fed TMR. In conclusion, fermentation is a conceivable method to improve the quality of TMR for long-time storage and the cracking method is suitable to release the fat from cottonseed for enhancing fatty acid deposition in milk. Thus, the combination of FTMR and cWCS supplementation would be an alternative strategy to improve performance of lactating cows.

Fermentation Characteristics and Lactic Acid Bacteria Succession of Total Mixed Ration Silages Formulated with Peach Pomace

  • The objective of this study was to assess the use of peach pomace in total mixed ration (TMR) silages and clarify the differences in aerobic stability between TMR and TMR silages caused by lactic acid bacteria (LAB). The TMR were prepared using peach pomace, alfalfa hay or Leymus chinensis hay, maize meal, soybean meal, cotton meal, limestone, a vitamin-mineral supplement, and salt in a ratio of 6.0:34.0:44.4:7.0:5.0:2.5:1.0:0.1 on a dry matter (DM) basis. Fermentation quality, microbial composition, and the predominant LAB were examined during ensiling and aerobic deterioration. The results indicated that the TMR silages with peach pomace were well fermented, with low pH and high lactic acid concentrations. The aerobic stability of TMR silages were significantly higher than that of TMR. Compared with TMR silages with alfalfa hay, TMR silage with Leymus chinensis hay was much more prone to deterioration. Although the dominant LAB were not identical in TMR, the same dominant species, Lactobacillus buchneri and Pediococcus acidilactici, were found in both types of TMR silages after 56 d of ensiling, and they may play an important role in the aerobic stability of TMR silages.

사료의 형태가 한우 거세우의 사료효율 및 도체성적에 미치는 영향 (Effect of Feed Type on Feed Efficiency and Carcass Characteristics of Hanwoo Steers)

  • 본 연구는 한우 거세우 95두 (평균체중 226 kg)를 이용하여 농후사료와 조사료 분리급여구, TMR구 그리고 TMR을 발효시킨 F-TMR구로 나누어 22개월동안 사육하여 도축하였을 때, 사료효율, 도체성적 및 경제성분석에 미치는 영향을 조사하기 위하여 실시되었다. 건물 및 TDN 섭취량은 육 비육 각 단계별로 F-TMR구에서 가장 많았다 (p<0.05). F-TMR구의 경우, 일일증체량은 육성기에는 처리구중 가장 적었으나, 비육후기에는 타 처리구에 비해 약 48%나 높았다 (p<0.05), 사료효율은 전 육성비육기간 동안 비슷한 수준으로 유지되었다. 도체중과 배최장근단면적에서는 F-TMR 급여구에서 가장 높았으나 (p<0.05), 등지방두께가 가장 얇았던 (p<0.05) 분리급여구에서 육량지수와 육량 A등급 비율이 높았다. F-TMR구의 근내지방도가 타구에 비해 현저히 (p<0.05) 증가되어 육질 1+등급이상 출현율이 96%에 달하였다. 경제성 분석에서 사료비는 F-TMR구가 분리급여구에 비해 40%가 증가되었으나 증체량과 육질등급의 향상으로 인하여 순수익은 29%나 증가되었다.

Evaluating fermentation quality, in vitro digestibility and aerobic stability of a total mixed ration ensiled with different additives on Tibet plateau

  • Objective: To investigate the improvement in utilization efficiency of total mixed ration (TMR) on Tibetan plateau, TMR were ensiled with different additives. Methods: A total of 150 experimental silos were prepared in a completely randomized design to evaluate the six treatments: i) control (without additive), ii) Lactobacillus buchneri (L. buchneri), iii) acetic acid, iv) propionic acid, v) 1,2-propanediol; and vi) 1-propanol. After 90 days of ensiling, silos were opened for fermentation quality and in vitro analysis, and then subjected to an aerobic stability test for 14 days. Results: Treating with L. buchneri, acetic acid, 1,2-propanediol and 1-propanol decreased propionic acid contents and yeast number, whereas increased (p<0.05) pH, acetic acid and ethanol contents in the fermented TMR. Despite increased dry matter (DM) loss in the TMRs treated with 1,2-propanediol and 1-pronanol, additives did not affect (p>0.05) all in vitro parameters including gas production at 24 h (GP24), GP rate constant, potential GP, in vitro DM digestibility and in vitro neutral detergent fibre digestibility. All additives improved the aerobic stability of ensiled TMR to different extents. Specially, aerobic stability of the ensiled TMR were substantially improved by L. buchneri, acetic acid, 1,2-propanediol, and 1-propanol, indicated by stable pH and lactic acid content during the aerobic stability test. Conclusion: L. buchneri, acetic acid, 1,2-propanediol, and 1-propanol had no adverse effect on in vitro digestibility, while ensiling TMR with the additives produced more acetic acid and ethanol, subsequently resulting in improvement of aerobic stability. There is a potential for some fermentation boosting additives to enhance aerobic stability of fermented TMR on Tibetan plateau.

Effect of Alcohol Fermented Feed on Lactating Performance, Blood Metabolites, Milk Fatty Acid Profile and Cholesterol Content in Holstein Lactating Cows

  • A feeding experiment with 40 lactating Holstein cows and 4 dietary treatments was conducted to investigate supplementation with different levels of alcohol fermented feed to the TMR on lactating performance, blood metabolites, milk fatty acid profile and cholesterol concentration of blood and milk. Forty Holstein lactating cows ($106{\pm}24$ d post-partum; mean${\pm}$SD) were distributed into four groups and randomly assigned to one of four treatments with each containing 10 cows per treatment. The treatment supplemented with TMR (DM basis) as the control (CON), and CON mixed with alcohol-fermented feeds (AFF) at a level of 5%, 10% and 15% of the TMR as T1, T2 and T3, respectively. Dry matter intake and milk yield were not affected by supplementation of AFF. An increased 4% FCM in the milk occurred in cows fed T3 diet compared with CON, while T1 and T2 diets decreased 4% FCM in a dose dependent manner. Supplementation of AFF increased the concentration of albumin, total protein (TP), ammonia, and high density lipoprotein-cholesterol in serum compared with CON. In contrast, supplementation with AFF clearly decreased concentration of blood urea nitrogen (BUN) and total cholesterol (TC) compare with CON. AFF supplementation increased the proportion of C18:1n9 and C18:2n6 compared to CON. A decrease in the concentration of saturated fatty acid (SFA) for T1, T2 and T3 resulted in an increased unsaturated fatty acid (USFA) to SFA ratio compared to CON. Concentration of cholesterol in milk fat was reduced in proportion to the supplemental level of AFF. Feeding a diet supplemented with a moderate level AFF to lactating cows could be a way to alter the feed efficiency and fatty acid profile of milk by increasing potentially human consumer healthy fatty acid without detrimental effects on feed intake and milk production. A substantially decreased cholesterol proportion in milk induced by supplementation AFF suggests that alcohol fermented feed may improve milk cholesterol levels without any negative effects in lactating cows.

Effect of lactic acid bacteria and yeast supplementation on anti-nutritional factors and chemical composition of fermented total mixed ration containing cottonseed meal or rapeseed meal

  • Objective: This study aimed to determine the appropriate supplementation level of lactic acid bacteria (LAB; Lactobacillus plantarum and Bacillus clausii), yeast (Saccharomyces cariocanus and Wickerhamomyces anomalus) for degrading free gossypol and glucosinolate in the fermented total mixed ration (TMR) containing cottonseed meal (CSM) or rapeseed meal (RSM), to improve the utilization efficiency of these protein sources. Methods: For LAB, L. plantarum or B. clausii was inoculated at 1.0×108, 1.0×109, 1.0×1010, and 1.0×1011 colony-forming unit (CFU)/kg dry matter (DM), respectively. For yeast, S. cariocanus or W. anomalus was inoculated at 5×106, 5×107, 5×108, and 5×109 CFU/kg DM, respectively. The TMR had 50% moisture and was incubated at 30℃ for 48 h. After fermentation, the chemical compositions, and the contents of free gossypol and glucosinolate were determined. Results: The results showed that the concentration of free gossypol content was reduced (p<0.05), while that of the crude protein content was increased (p<0.05) in the TMR containing CSM inoculated by B. clausii (1×109 CFU/kg DM) or S. cariocanus (5×109 CFU/kg DM). Similarly, the content of glucosinolate was lowered (p<0.05) and the crude protein content was increased (p<0.05) in TMR containing RSM inoculated with B. clausii (1×1010 CFU/kg DM) or S. cariocanus (5×109 CFU/g DM). Conclusion: This study confirmed that inclusion of B. clausii with 1.0×109 or 1.0×1010 CFU/kg DM, or S. cariocanus (5×109 CFU/kg DM) to TMR containing CSM/RSM improved the nutritional value and decreased the contents of anti-nutritional factors.

유형별 완전혼합사료 급여가 반추위내의 발효성상 및 영양소 소화율에 미치는 영향 (Effects of Types of TMR on Rumen Fermentation Characteristics and Nutrients Digestibility in Sheep)

  • 본 시험은 유형이 서로 다른 세 가지 형태의 TMR을 반추위에 누관이 장착된 면양을 이용하여 3${\times}$3 라틴 방각법으로 반추위내 발효성상, 효소역가 및 전장 소화율을 조사하였다. 시험 결과를 살펴보면, 전반적으로 반추위 내 pH, 암모니아태 질소 농도, 총 VFA, 각종 VFA생성량, A/P ratio 및 각종 효소(CMCase, Xylanase 및 Protease) 역가는 건TMR를 급여했을 때와 비교하여 습TMR과 발효TMR급여 시 높은 경향을 보였다. 또한 건물, 유기물, 조단백질, 조지방, NDF 및 ADF의 전장 소화율도 건TMR 급여 시 보다 습TMR과 발효TMR를 급여했을 때 높게 향상된 것을 볼 수 있다. 따라서 본 시험의 결과들을 종합해 보면 보다 안정적이고 활발한 반추위 발효과정과 영양소 이용효율 측면에서 건TMR보다 습TMR이나 발효TMR이 우수한 것으로 요약할 수 있다.

고초균을 이용한 조직대두단백의 고체 발효 기간에 따른 라디칼 소거 활성 및 물성 평가 (Evaluation of Radical Scavenging Activity and Physical Properties of Textured Vegetable Protein Fermented by Solid Culture with Bacillus subtilis HA According to Fermentation Time)

  • 조직대두단백을 이용하여 B. subtilis HA에 의한 장기간 동안 고체 발효를 통해 얻어진 발효물의 품질, 항산화 활성 및 물성 변화를 평가하였다. 발효시간에 따라 발효물의 색도는 L값은 감소하고 a, b 값은 증가하였으며 수용성 추출물의 갈색도는 발효 48시간까지는 서서히 증가하나 그 후에는 변화가 없는 것으로 나타났다. TVP 발효물의 물과 70% 에탄올 추출물의 DPPH radical 소거 활성 측정 결과 발효 24시간째 70% 에탄올 추출물의 $ IC_{50}$값이 0.99 mg/mL로 가장 활성이 우수하였고 발효시간이 길어져도 활성이 증가하지 않았다. 또한 ABTS radical 소거 활성도 70% 에탄올 추출물이 발효 72시간째 $ IC_{50}$값이 1.68 mg/g으로 가장 높았다. TVP 발효물의 점조도는 발효 48시간째 7.89 $Pa{\cdot}s^n$으로 가장 높았고 그 후에는 감소하는 것으로 나타났으며 점질물 함량은 발효 24시간째 26.49%로 가장 높은 함량을 보인 후 감소하는 것으로 나타났다. 발효물의 점탄성은 발효 48시간째 가장 높았으며 이후 감소하였고 탄성(G')보다 점성(G")이 높은 것으로 나타났다. $\gamma$-PGA 함량은 발효시간이 증가하면서 증가하는 경향을 보이면서 168시간에 37.72% 생산되었고, 분자량은 발효시간이 경과될수록 감소하였다. Levan 함량은 발효 초기인 12시간째 7.83%에서 발효시간이 경과할수록 감소하는 것으로 나타났다.
