Effect of Fermentation Temperature on Quality of Doenjang (숙성온도가 된장의 품질에 미치는 영향)

  • There were four types of Doenjang fermentation as following conditions for investigation ; 1) low temperature fermentation at $13^{\circ}C$ for 180 days, 2) low temperature at $13^{\circ}C$ for 7 days to room temperature at $30^{\circ}C$ for 10 days, to low temperature at $13^{\circ}C$ for 163 days, and for 173 days, 3) low temperature at $13^{\circ}C$ 7 days to room temperature at $30^{\circ}C$, 4) room temperature at $30^{\circ}C$ for 180 days. There were no changes of moisture, NaCl and total nitrogen content during fermentation period of four types conditions, but pH and amino type nitrogen decreased in room temperature at $30^{\circ}C$ for 180 days. It required 3 times more fermentation period until same quantity of the amino type nitrogen. The low temperature fermentation sample was lower than room temperature fermentation sample in pH and amino type nitrogen. The yeast decreased in low temperature fermentation sample taken 15 to 30 days longer than room temperature sample. The yeast is increased up to 30 days, and decreased little by little. After 60 days, it remained a few without effectiveness on the Doenjang quality. The low temperature fermentation sample showed brighter than room temperature fermentation sample. Different fermentation condition affected Doenjnag quality, especially, low temperature fermentation sample showed bright color in Doenjnag. So low temperature fermentation must be expected as good method for getting high quality Doenjnag.

Changes of Chemical Components of Fermented Tea during Fermentation Period (미생물을 이용한 후발효차의 발효기간별 화학성분 변화)

  • To manufacture the fermented tea with hygienic quality, green tea was fermented using Bacillus subtilis, Saccharomyces cerevisiae and Lactobacillus bulgaricus and chemical composition and sensory changes were evaluated during fermentation period. The lightness of the fermented samples decreased; in contrast, redness and yellowness increased. Especially, the color change of the fermented tea using B. subtilis was higher than those of control and other samples with different microorganisms during fermentation period. Chlorophyll contents were decreased by similar level regardless of fermentation treatments. The fastest decrease of total catechins contents were found in the tea fermented with B. subtilis and significantly reduced by increase of fermentation period. However, total catechin contents of the tea fermented by L. bulgaricus were not decreased. The caffeine contents of the microbial fermented teas were more decreased than that of control, even though the decrease was slight. Sensory panelists preferred the tea fermented by B. subtilis to those of control or other fermentation treatment.

Physicochemical Properties of Dongchimi Added with Gatt (Brassica juncea)

  • To improve Dongchimi (watery radish kimchi) quality and preservation, 0, 3, 5, 10, and 15% of gatt (Brassica juncea; leaf mustard) per radish was added. Chemical characteristics were determined during fermentation at $10^{\circ}C$ for 45 days. Total acidity increased slowly by addition of gatt during initial fermentation period, and 15% treatment showed lowest total acidity. Total vitamin C content increased initially in all treatments depending on gatt content, and decreased thereafter. Period for reaching maximum value was delayed by addition of gatt. Highest total vitamin C was found in 15% treatments. In the case of reducing sugar, 5% treatment showed highest contents. Lactic, succinic, and tartaric acid contents consistently increased during fermentation, while those of malic and citric decreased. Turbidity and total solid contents of Dongchimi liquid increased in all treatments as fermentation proceeded, although the extent was rather suppressed by addition of gatt. Colorimetric lightness values decreased, while the initial increased and then decreased in redness and yellowness. Addition of gatt at above 15% weight level per radish accelerated fermentation at the later fermentation stage thus it should be avoided. Most changes in typical characteristics of fermentation were similar depending on treatments. More acceptable Dongchimi could be prepared by fermenting at 5% gatt concentration under given conditions.

Antioxidant Activity and Component Change of Steaming-Drying and Fermented Gastrodiae Rhizoma (증포 천마 발효물의 항산화 활성 및 성분변화)

  • Objectives : The objective of this study is to evaluate antioxidant activity and the main component content change of steaming-drying fermented Gastrodiae rhizoma extract. Methods : The antioxidant activities were determined for total polyphenol, flavonoid contents, SOD-like activity, electron donating activity, nitrite scavenging ability and major functional components(gastrodin and 4-hydroxybenzyl alcohol content) were also measured. Results : The polyphenol content of fermented Gastrodiae rhizoma by S. cerevisiae were higher than those of fermented Gastrodiae rhizoma by A. oryzae, and when the fermentation period is extended. SOD like activity of fermented Gastrodiae rhizoma showed to be increased by fermentation with S. cerevisiae than fermentation by A. oryzae. Electron donating activity of fermented Gastrodiae rhizoma were increased at almost parallel level as vitamin C, by fermentation. Notably, fermentation by A. oryzae was moderately better than fermentation by S. cerevisiae. Flavonoid content of Gastrodiae rhizoma showed to increasing by fermentation, particularly fermentation by S. cerevisiae was proven to be more effective than by A. oryzae. The more steaming-drying or increased period of fermentation would be resulted in more gastrodin contents but under the same conditions, 4-hydroxybenzyl alcohol content of fermentation by A. oryzae in case of steaming and drying 1 time and 3 time was higher than control. Conclusions : These results has strongly hint the possible applicability of fermentation might be effective to improve the diverse biological activities of Gastrodiae rhizoma and may further supports to develop a functional food materials.

Characteristic of Makgeolli and Pine (Pinus koraiensis) Extract Fermentation (소나무 (Pinus koraiensis) 추출물과 결합 된 막걸리 발효의 특징)

  • In this research, the improvement of Korean rice wine (makgeolli) with pine (Pinus koraiensis) extract addition was evaluated due to the increase in alcoholic Korean traditional beverage. Makgeolli fermentation was prepared using Korean rice and nuruk (traditional starter) supplemented by pine needle (MPN) and pine sprout (MPS) extract. The average of initial pH level for MPN was 3.95 and MPS was 4.55, the average of initial sugar content for MPN was 0.4% and MPS was 0.3%. The sugar content and pH level behavior were investigated every 24h during fermentation period. The observation of microbial colony was done at days 8 of fermentation period with three time sample dilution. Afterward, the physical appearance of fermentation solution and microbial development were investigated in the final of fermentation period. The number of yeast and LAB ($402{\times}103\;CFU/mL$) in MPN was greater than the yeast and LAB count in MPS ($224{\times}103\;CFU/mL$). The pH level obtained by addition pine sprout have value of R2 higher than addition of pine needles (leaf), the sugar content (%) behaviour was opposite with pH level behaviour.

The effect of Sodium chloride concentrations of Soaking-Water and Submaterials on Tongbaechu-Kimchi Taste and Fermentation (침지수의 염도 및 부재료가 통배추김치의 맛과 숙성에 미치는 영향)

  • This study was investigated to find effects of soaking-water salt concentration and submaterial on Kimchi taste and fermentation period. several parameters such as pH, acidity, reducing sugar, amino acid and sensory evaluation during fermentation at 2$0^{\circ}C$ were experimented. The results are as follows : As fermentation period increases, pH and content of reducing sugar of Kimchi have decreased. But acidity has increased. Especially, the pH of all samples rapidly decreased to a extent of pH 4.2. During fermentation the acidity and content of reducing sugar in 12% soaking-water were higher than those in 16% and 20%. Content of reducing sugar I Kimchi was observed to play an important role for the overall taste. As a result of sensory evaluation during fermentation, Kimchi in 16%, 2.6% and in 20%, 3.7%. Sensory evaluation revealed that leek-added Kimchi was highly evaluated in crispness, sour taste, flavor and overall acceptability. These results suggested that Kimchi fermented for 4days at 2$0^{\circ}C$ with 16% salt concentration of soaking water and leek as submaterials should taste most desirable.

Effect of Abiotic Factors on Fumosorinone Production from Cordyceps fumosorosea via Solid-State Fermentation

  • Cordyceps fumosorosea is an important species in the genus of Cordyceps, containing a variety of bioactive compounds, including fumosorinone (FU). This study was a ground-breaking assessment of FU levels in liquid and solid cultures. The present study focused on the impacts of solid-state fermentation (SSF) using solid substrates (wheat, oat, and rice), as well as the effects of fermentation parameters (pH, temperature, and incubation period), on the generation of FU. All the fermentation parameters had significant effects on the synthesis of FU. In a study of 25 ℃, 5.5 pH, and 21 days of incubation period combinations calculated -to give maximal FU production, it was found that the optimal values were 25 ℃, 5.5 pH, and 21 days, respectively. In a solid substrate medium culture, FU could be produced from SSF. At 30 days, a medium composed of rice yielded the most FU (798.50 mg/L), followed by a medium composed of wheat and oats (640.50 and 450.50 mg/L), respectively. An efficient method for increasing FU production on a large scale could be found in this approach. The results of this study might have multiple applications in different industrial fermentation processes.

Standardization for the preparation of traditional Korean whole cabbage Kimchi with salted shrimp (새우젓을 첨가한 전통적 통배추 김치의 최적 제조 조건 설정에 관한 연구)

  • This study was conducted to determine the optimum conditions for the preparation of traditional Korean whole cabbage Kimchi with salted shrimp. Sensory, physical and chemical characteristics of Kimchi with various salted shrimp level(1.8, 3.6%), fermentation time(12, 24, 36 hrs) and storage period(1, 4, 7 days) were measured. As the fermentation time and storage day were extended, pH of Kimchis decreased but titratable acidity of Kimchis increased. The more salted shrimp in Kimchi resulted in the higher titratable acidity. The longer fermentation time and the longer storage resulted in the more citric, malic and succinic acid, and the less lactic and acetic acid in Kimchi. The optimum conditions for the preparation of Korean whole cabbage Kimchi were 3.6% salted shrimp level, 12 hours fermentation time and 84 hours(3.5 days) storage period.

Effects of Red Peppers on the Its Pungency and Color during Kimchi Fermentation (고춧가루가 발효중 김치의 매운맛과 색도에 미치는 영향)

  • This study was carried out to investigate preparation of reconstructed red peppers, effects of pungency and redness of red peppers on the Kimchi quality using central composite design and response surfaces methodology. Capsaicinoids and ASTA (American Spice Trading Association) value put in X$_1$, X$_2$ of independent variable. The result of response surface regression analysis of reconstructed red peppers, correlation coefficient ($R^2$) of overall pungency intensity, persistence and degree of redness was 0.935, 0.935 and 0.821, respectively. After it was made Kimchi samples with reconstructed red peppers, it was examined pH, titratable acidity and lactic acid bacteria of its during fermentation. In the initial fermentation period of Kimchi, it showed pH of 5.46∼5.78, titratable acidity of 0.27∼0.31%, salt content of 2.26∼2.48% and lactic acid bacteria of 4.05${\times}$10$^{5}$ ∼6.23${\times}$10$^{5}$ , respectively. And it showed traditional fermentation pattern in the pH, titratable acidity and microbes of the middle (appropriate fermentation) and last (excessive) fermentation period. While capsaicinoids content in the Kimchi decreased a little according to extend fermentation period, ASTA value showed low correlation reconstructed red pepper and fermentation period. Also, it was analyzed correlation coefficient ($R^2$) of independent variables (capsaicinoids, X$_1$; ASTA value, X$_2$) between sensory attribute in the Kimchi during fermentation. The result of regression analysis, $R^2$ in the overall pungency intensity, persistence and degree of redness showed 0.515, 0.675, 0.784, respectively.

Fermentation Characteristics of Honey Wine by Saccharomyces bayanus (Saccharomyces bayanus를 이용한 벌꿀 발효주의 양조 특성)

  • Fermentation characteristics for the production of honey wine (mead and melomel) was investigated. Among the yeast strains tested, Saccharomyces bayanus showed higher alcohol production and better fermentability than the other strains at low temperature. Optimum pH and temperature for the production of honey wine by Saccharomyces bayanus were pH 4.0 and $20^{\circ}C$, respectively, and optimum soluble solid content of diluted honey solution for the fermentation was between 24 and $27^{\circ}Brix$. Total acidity and pH of honey wine (mead) did not change considerably during the whole period of fermentation, but those of Tangerine and Japanese apricot honey wine (melomel) changed during the fermentation. As the fermentation progressed, reducing sugar decreased continuously until the late period of fermentation, while alcohol content increased continuously during the same period. After fermentation of 21 days, honey wine consisted of about $8.5{\sim}9.1^{\circ}Brix$ of soluble solid, $1.90{\sim}2.32%$ of reducing sugar with the conversion rate of $90{\sim}92%$. After 21 days of fermentation, alcohol contents of mead fermented with polyflower and acasia flower were 13.3 and 13.7%, respectively. Final alcohol content was not affected significantly by the source of honey. While pH of the osmotically extracted fruit honey juice decreased rapidly to pH $2.92{\sim}2.97$ after 13 days of fermentation, total titratable acidity of Tangerine and Japanese apricot honey wine were 0.30 and 0.53%, respectively. After 13 days of fermentation, reducing sugar of fruit honey wines were reduced to $2.03{\sim}2.87%$, alcohol content were reached up to 13.1 and 12.5% for Tangerine and Japanese apricot honey wine, respectively. Generally, honey extracted fruit juices were fermented more efficiently than diluted honey juice.
