흰쥐의 성별과 체중에 따른 족삼리(足三里) 전침자극의 시간과 심도(深度)의 변화가 수장수송능(小腸輸送能)에 미치는 영향 (Study on Relation of Stimulated Duration and Depth of Electroacupuncture with Sex and Weight In Rats)

  • Acupuncture treatment cures disease by regulating the functional excessiveness or deficiency which occurs in the meridian and visceral organs, and by letting stagnated Qi and Blood flow through the whole body and then controlling Qi. Whether or not to have Qi feeling is the main factor needed to manifest the efficacy of acupuncture. So it needs the very stimulus to make Qi felt rather than the simple stimulus. In order to have Qi feeling, it needs to acupuncture a patient according to patient's thinness or fatness, constitution, richness or poverty and age. And also it needs to vary the way to acupuncture a patient according to meridian, kinds of pulse, degrees of disease, new or old diseases, seasons with a disease and local areas of disease. In order to ascertain that it is important for the quality and quantity of acupuncture stimulus and the state of feeling acupuncture to get the efficacy of acupuncture, the experiment was planned to confirm whether the form and Qi of subject has different result according to the stimulated duration and depth. On the basis of the report that acupuncture on derma, when using electroacupuncture at Zusanli(ST 36) which denotes small intestinal motility, is more efficient than full depth acupuncture, I got the following result, after I observed whether the stimulated duration and depth give different efficacy according to the sex and weights of rat in experiment. The increased effects of small intestinal motility by electroacupuncture on Zusanli(ST 36) appeared after stimulating full depth for thirty minutes without distinction of sex and weights. This significant change was observed only in the female experimental group when I distinguished the sex. The small interstinal motility in rats by electroacupuncture at zusanli(ST 36) decreased in the 10 minutes' stimulated group and 30 minutes' stimulated group by duration on the one hand, and derma-deep stimulated group and full depth stimulated group by the depth of stimulus on the other hand, as the weights increased. This result shows that the duration and depth of acupuncture depends on the differences of acupuncture points, sex of the experimental animals, ages, and, weights. And the further study on the experimental and clinical differences and sextual differences need to be continued on.

Does Vitex Agnus-Castus L. Have Deleterious Effect on Fertility and Pregnancy Outcome? An Experimental Study on Rats for Prediction of Its Safety

  • Objectives: Herbal medicine is a worldwide health topic. Vitex agnus-castus L. (VAC) is a popular plant used for gynecologic problems due to its hormonal effects. The aim of this study is to reveal VAC extract effect on fetus when this herb is used started from antenatal period or during pregnancy. Methods: Performed from starting day of January 2019 till February 2019, 48 rats were assigned in randomly divided eight-member six groups: control (C1), treated group with 365 mg/kg VAC from initiation of insemination (T1) and 30 days prior to pregnancy (T2), control that underwent caesarean section on 15th day of gestational age (C2) and treated group with 365 mg/kg VAC from initiation of insemination (T3) and 30 days prior to pregnancy (T4) that underwent caesarean section. Weight, sex and number of fetuses, abortion and still birth rate and estradiol level were evaluated using t-test by SPSS software. Results: We showed increased weight among T1 group considering totally and sex-dependent which is significant (all p-value < 0.05). We also detected significantly decreased weight in T2 in total (p-value < 0.0001) and when considering female fetuses (0.043) but not males (0.17). Although the results showed slightly non-significant increased weight among fetuses of T3 (totally or based on the fetus sex) compared to the control group (C2), T4 group had statistically decreased weight compared to control group. Pregnancy rate and pregnancy outcome were affected by VAC usage. The time of VAC initiation also affected live birth and abortion rates. Conclusion: VAC extract may affect pregnancy rate, live birth rate, abortion and stillbirth rates. Its effect on the weight and the sex showed dual pattern depends on the time of initiation and pregnancy trimester of evaluation. Prescribing this medicinal plant for patients being prone to pregnancy should be with caution. Further study is recommended.

The effect of local application of thymoquinone, Nigella sativa's bioactive component, on bone healing in experimental bone defects infected with Porphyromonas gingivalis

  • Purpose: This study was performed to evaluate the influence of local application of thymoquinone (TQ) on bone healing in experimental bone defects infected with Porphyromonas gingivalis (PG). Methods: Forty-two female rats were randomly divided into 6 groups. A bone defect was created on the right tibia of all animals. The PG, PG/collagen membrane (COL) and PG/TQ/COL groups were infected with PG. In the COL and PG/COL groups, the defects were covered with a COL; in the TQ/COL and PG/TQ/COL groups, the defects were covered with a TQ-containing COL. After 28 days, all animals were sacrificed. Quantitative measurements of new bone formation and osteoblast lining, as well as semiquantitative measurements of capillary density and tissue response, were analyzed. Furthermore, the presence of bacterial infections in defect areas was evaluated. Results: The new bone formation, osteoblast number, and capillary density were significantly higher in the TQ groups than in the control groups (P<0.001, P<0.001, and P<0.01, respectively). In a comparison between the TQ/COL group, with a TQ-containing COL (TQ/COL), and the PG-infected TQ-containing COL (PG/TQ/COL) group, the newly formed bone and capillary density were higher in the TQ/COL group (P<0.01). When the control group was compared to the PG, PG/COL, and PG/TQ/COL groups in terms of tissue response, the differences were statistically significant (P<0.001, P=0.02, and P=0.041, respectively). The intensity of the inflammatory cell reaction was higher in the PG, PG/COL, and PG/TQ/COL groups (P<0.05). Conclusions: Within the limitations of this study, the local application of a TQ-containing COL positively affected bone healing even if the bone defects were infected. The results suggest that TQ increased angiogenesis and showed promise for accelerating bone defect healing. Further research is warranted to support these findings and reach more definitive conclusions.

Acute oral toxicity and bioavailability of uranium and thorium in contaminated soil

  • A robust approach was conducted to determining the absolute oral bioavailable (fab) fractions of 238U and 232Th in rats exposed to contaminated soil along with their hematotoxicity and nephrotoxicity. The soil sample is the International Atomic Energy Agency-312 (IAEA-312) certified reference material, whereas blood, bones, and kidneys of in vivo female Sprague-Dawley (SD) rats estimate 238U- and 232Th-fab fractions post-exposure. We predict the bioavailable concentration (Cab) and fab values of 238U and 232Th after acute soil ingestion. The blood 238U (0.750%) and 232Th (0.028%) reach their maximum fab values after 48 h. The 238U (fab: 0.169-0.652%) accumulates mostly in the kidney, whereas the 232Th (fab: 0.004-0.021%) accumulates primarily in the bone. Additionally, 238U is more bioavailable than 232Th. Post 48 h acute ingestion demonstrates noticeable histopathological and hematological alterations, implying that intake of 238U in co-contaminated soil can lead to erythrocytes and proximal tubules damage, whereas, 232Th intake can harm erythrocytes. Our study provides new directions for future research into the health implications of acute oral exposures to 238U and 232Th in co-contaminated soils. The findings offer significant insight into the utilization of in vivo SD rat testing to estimate 238U and 232Th bioavailability and toxicity in exposure assessment.

출생 전.후 뇌의 성분화 결정시기에 DDT에 의한 KAP3 유전자 조절에 대한 연구 (Study on the Regulation of KAP3 Gene Involved in the Brain Sexual Differentiation by DDT during the Critical Period of Fetal and Neonatal Age)

  • 환경에 방출되어 있는 많은 내분비교란물질들은 사람과 동물의 내분비계에 교란을 일으킬 수 있는 잠재력을 가진다. 뇌의 성분화는 생식소 호르몬 영향하에 비가역적으로 진행되며 흰쥐의 경우 이 시기는 임신말기에서 생후 7∼10일 가량이다. 최근에 본 연구진은 횐쥐의 뇌 성 분화의 결정적인 시기에 발현되는 KAP3유전자를 클로닝하였다 (Choi & Lee, 1999). KAP3의 기능은 신경세포를 포함한 세포에서 aronal tansport를 조절하는 것으로 알려져 있다. 본 연구에서는 흰쥐 뇌 발생의 결정적인 시기에 내분비 교란물질인 Dichlorodiphenyl trichloroethane (DDT)가 KAP3유전자 발현과 성분화에 미치는 영향을 검토하였다. DDT에 노출된 임신 17일된 흰쥐 태아 암컷과 수컷의 뇌에서 KAP3 mRNA의 발현이 증가하였다. 그러나 출생후 DDT에 노출된 흰쥐 암컷과 수컷의 뇌에서는 KAP3 mRNA의 발현은 감소하였다. 또한 태어난 직후 DDT에 노출된 경우 체중이 현저히 감소하였으며 수정율도 DDT에 노출되지 않은 흰쥐에 비하여 크게 낮았다. 이러한 결과는 내분비 교란물질인 DDT가 뇌의 성 분화와 관련된 유전자인 KAP3의 전사에 영향을 미치며, 내분비 교란물질에 노출된 태아의 뇌 분화에서 독성을 보이는 것을 의미한다. 그리고 KAP3유전자는 동물의 신경세포의 발생에 미치는 내분비 교란 물질의 독성을 분자생물학적으로 연구하기 위한 유전자 지표로도 사용 가능하다고 생각된다.

난소 절제 흰쥐의 혈청 콜레스테롤 및 항산화효소에 미치는 비타민 E와 비타민 C의 효과 (The Effects of Vitamin E and C on Serum Cholesterol and Antioxidative enzyme in ovariectomized rat)

  • 난소를 절제한 female rat에 Vit C. Vit E, Vit C와 Vit E를 복합적으로 투여했을 때 rat의 간 조직 의 손상 정도 및 항산화 효소 활성을 관찰하기 위해 체중 150$\pm$10g이 되는 female rat에 100mg/kg 함 량의 각 실험 물질들을 이틀에 한 번 두달 간 복강내로 투여한 뒤 간 조직 중 과산화지질량 및 항상화 효소인 SOD, catalase, 총 glutathion 량 및 Glutathion peroxidase의 활성도를 측정함으로써 항산화계의 변화 및 생리 활성 변화를 살펴 보았다. 1. 간, 신장 조직 중의 과산화지질 함량은 노화군(Ovx)이 정상 군(Sham)에 비해 각각 1.78배, 1.61배 증가하였다. 그리고 각 비타민 투여군은 난소를 절제한 군에 비해 과산화질함량이 현저히 감소하였다. 2. 혈청 중 총 cholesterol 량은 노화군(Ovx)이 정상군(Sham)에 비 해 2.57배 높게 나타났다. 각 비타민 투여군은 노화군에 비해 수치가 현저히 감소하였는데, Ovx+Vit C 군의 경우는 94%정도의 억제율을 보였다. HDL-cholesterol은 노화군(Ovx)이 정상군(Sham)에 비해 36.7% 감소하였다. 각 비타민 투여로 HDL-cholesterol 함량이 좋은 수준으로 증가되었고, Ovx+Vit Erns은 노화군보다 2.52배 높게 나타났다. 동맥경화지수도 Ovx+Vit E 군이 아주 낮게 나타났다. 3. 간 조직 중 SOD 활성도는 노화군(Ovx)이 정상군(Sham)보다 5.3배 높게 나타났고, 각 비타민 투여로 free radical 생성이 억제되어 항산화효소인 SOD 활성도를 감소시켰다. 간 조직 중 catalase 활성도는 노화군 (Ovx)이 정상군(Sham)보다 1.58배 정도 높게 나타났는데, 각 비타민 투여로 catalase 활성도가 낮아졌 고, Ovx+Vit E 군은 탁월한 감소 효과를 보였다. 4. 간 조직 중 총 glutathion의 활성도는 정상군 (Sham)이 노화군(Ovx)에 비해 1.22배 정도 높게 나타났고, 각 비타민 투여군 중 Ovx+Vit Erns의 경우 간 조직 중 총 glutathionfid이 가장 높게 나타났다. 간조직 중 reduced-glutathion (GSH)함량과 총 glutathion 량에 대한 oxidized-glutathion (GSSG)의 비율, 또한 Ovx+Vit E 군의 수치가 가장 높게 나 타났다. Vit E가 항산화에 좋은 효과가 있음을 알 수 있었다. 그러나, GPx의 활성 수치는 그 다지 유의 한 변화를 나타내지는 않았다.

칠분도미(七分搗米)를 주식(主食)으로 한 한국인식단(韓國人食單)과 식량제한(食量制限)에 관(關)한 영양학적(營養學的) 연구(硏究) (Nutritional studies on the menu contained 70% polished rice and dietary restriction)

  • This study was designed to comparse the metabolic effects of Korean diet pattern which contained all-polished rice and 70% polished rice, and to compare dietary restriction. The results were as follows. 1. Food intake survey to catch the Korean diet pattern The calory intake showed women took much more than men and farm village was higher than city, However, protein intake showed men were higher than women and decrease gradually from city, fishing village, and farm village. Calcium intake showed Seoul was the first rank of all, but was merely 70% of recommended quantities. Fe intake was greatly short especially in women and vitamin C also showed same situation. In the view of food group intake, grains and potatoes were 70.06%, vegetables and fruits 13.05%, meats fishes and beans 11.99%, respectivelly. Oil and fat intake showed lowest percentage as 0.77%. 2. Nutritional experiments albino rats have been fed for 7 weeks with three different diet: all-polished rice diet, 70% polished rice diet, and the standard diet for contral group. Dietary restriction were at 4 levels: 5% , 10%, 15%, 20% in 70% polished rice det. Body weight of 70% polished rice group gained value compare to all polished rice group. In the final organ weight all-polished rice group and 70% polished rice group and 70% polished rice group revealed similar results, but organ weight almost decreased dued to dietary restriction and statistical data showed significant differences between dietary restriction group and none-restriction group. In the femur length, 70% polished rice group was longer than all polished rice group but there was no significant differences. The nitrogen content of liver and muscle of 70% polished rice group was higher than all-polished rice group. Furthermone there was significant differences in the female (P<0.1). In the 10% restriction group(female) and 15% restriction group(male), nitrogen retention was higher than 0% restriction group. The other biochemical analysis such as liver lipid, serum cholesterol, glucose of urine, feces and serum were not revealed any significant differences. As a result of studying, it seems clear that 70% polished rice intake influenced much nourshment to white rat growth than all-porished rice intake, and there is no significant influence to animal growth and metabolic effect even if it was restricted $5{\sim}10%$ of diet.

지방 함량에 따르는 흰쥐의 체내 대사 연구 (A study of metabolic effect in high and low fat diet on Albino Rat)

  • male female 80 마리의 흰쥐를 저지방식, 고지방식, 무지방식, 표준식이를 주어 16주간 동안 사육한 결과는 다음과 같다. 동물의 체중증가율 질소의 이용율, 간과 serum 내의 성분의 변화를 분석 시험한 결과를 요약하연 다음과 같다 지방의 식이내 함량에 따라서 체내 질소 대사에 미치는 영향으로 총 질소 보유율과 serum 내의 총단백질 albumin, globulin 의 함량, 각 장기 내의 질소 함유량들을 보았으나 대체적으로 현저한 변화를 가져오지 않는 것으로 본다. 그러나 각 장기에 있어서 총 질소 함유량을 보면 liver 와 spleen 이 식이 성분에 대해서 예민한 반응을 보였으며, brain 이 가장 적은 변화를 나타내었다. 한편 serum 내 glucose함량에는 식이 함유된 지방 함량에 따라서 큰 변화를 나타내지는 않았다. 각 군간의 유의적인 차이는 산발적으로 나타났지만 저지방군과 표준군을 male female에서 비교해 보면 다 같이 유의적인 차이를 나타내지는 않았다 . 이로 미루어 보면 2% 저지방군이 serum 내의 glucsoe 양에는 영향을 크게 미치지는 않는다고 볼 수 있다. serum 내 lipid 와 cholesterol 의 함량은 식이내 함유된 지방량에 따라서 현저한 차이를 나타냈다. 고지방군인 30% 지방 첨가로 serum 내의 lipid 와 cholesterol 의 함량이 증가되었으며 저지방군인 2% 의 식이내 지방 첨가는 표준군과 큰 차이를 나타내지 않는 것으로 보아 2%의 저지방군이 지방질 체내 축척에 있어서는 유리하다고 볼 수 있다 . 한국 영양 섭취 실정으로 미루어 보면 지방의 섭취량을 우선 일정량으로 올리는 것이 중요한 일이라고 보는데 본 실험의 결과에 따르면 식사내 2% 지방 첨가가 표준군과 모든 면에서 유사하였다. 반면 대부분의 구미 각국에서는 총열량에 대한 지방량의 비율이 $30{\sim}40%^{7)}$로서 상당히 많은 량의 지방을 섭취하고 있으나, 이들 각국은 지방 섭취량을 저하시키는 경향이므로 지방 섭취량의 대한 권장을 고려해야 할 필요가 있는 것으로 본다.

Suppression of Experimental Liver Tumors by Estradiol-3-Benzoate Treatment or Castration in Male Rats

  • Epidemiologically the incidence of liver cancer is markedly sex-differentiated, with a much higher frequency in men than in women. In experimental animals, it is also higher in male than in female irrespective of carcinogen-induced or spontaneous tumors. Therefore, we tried to investigate the modulating effects of sex hormones in experimental hepatocarcinogenesis. For induction of liver tumors, mini-osmotic pump containing diethylnitrosamine at a dose level of 47.5mg was implanted into the peritoneal cavity of the rat at 6 weeks old. To remove the effects of male sex hormones, the animals of group 2 were castrated one week prior to DEN treatment. To see the effects of estrogen, pellet containing 1g or 10g of estradiol-3-benzoate was infused subcutaneously to the animals of group 3 and 4 one week prior to DEN treatment. The pellets were exchanged every 4 weeks until sacrifice. All animals were sacrificed at 26 weeks after DEN treatment. The tumor incidences in group 1 (DEN alone), group 2 (DEN +castration), group 3(DEN +EB 1g) and group 4 (DEN +EB 10g) were 100% (15/15), 93.3% (14/15), 85.7% (12/14) and 66.7% (10/15), respectively, showing that the value of group 4 is significantly different from that of group 1. Tumor multiplicity data of group 1, 2, 3 and 4 were 5.470.73, 2.800.51, 2.070.41 and 1.670.46, respectively, showing castration or EB treatment reduced number of liver tumors significantly (P<0.001). With immunohistochemistry and Western blotting of ER the expressions were detected in normal adjacent liver cells but decreased or lost in tumor cells. From these results we conclude that female sex hormone, especially estrogen, may act as a liver tumor suppressor, and it seemed that the down regulation of ER may be associated with liver tumor development.

SD rats의 성별에 따른 형개 열수추출물의 방사선 방호효과 (Radiation Protection Effects of Schizonepeta Tenuifolia Water Extracts by Gender in SD Rats)

  • 본 연구는 형개 열수추출물이 흰 쥐의 성별에 따라 미치는 방사선 방호효과를 평가하는 것이다. 형개는 활성산소를 제거하는 대표적인 물질인 polyphenol과 flavonoid를 함유하고 있다. 이에 따라 방사선 방호효과를 알아보기 위해 형개 열수추출물을 SD rat 수컷과 암컷에 2 주 동안 경구투여 한 후 혈액학적 분석, 소장 융모 길이 변화 분석, SOD 활성 평가를 실시하였다. 수컷 SD rat ST+IR군은 IR군에 비해 혈소판의 회복능력이 조금 더 증가한 것으로 나타났다. 암컷 또한 ST+IR군이 IR군에 비해 혈소판 회복능력이 더 높게 나타났다. 림프구 수치도 혈소판 수치와 마찬가지로 수컷과 암컷 SD rat ST+IR군이 IR군보다 회복능력이 더 높게 나타났다. 방사선 조사 후 수컷 SD rat과 암컷 SD rat의 소장 융모 길이가 IR군보다 ST+IR군이 덜 감소한 것으로 보아 모든 성별의 SD rat 소장 융모가 손상이 적은 것을 확인하였다. 이 결과를 통해 형개 열수추출물이 방사선 방호효과가 있음을 확인 할 수 있었다.
