Histological Development of the Digestive System in Artificially Produced Anguilla japonica Larvae (인공생산 뱀장어(Anguilla japonica) 자어의 소화기관 발달 특성)

  • Freshwater eel Anguilla japonica is a commercially important species in East Asia. However, given the continuously decreasing resources due to many problems as climate change, habitat destruction and overfishing, mass seed production is crucial. In particular, the eel larvae present a unique life history, spending prolonged periods in the Pacific Ocean. Therefore, finding suitable feed that lasts through the long larval stages is rather difficult. In the present study, we histologically observed the development of digestive organs for 50 days after hatching (DAH). The endogenous reserves were completely absorbed at 7 DAH, and the first exogenous feeding started thereafter. The teeth appeared at 4 DAH and dramatically changed from needle-like to broad-based types by 50 DAH. The intestine remained as straight gut until 50 DAH. The intestinal cilia appeared first at 6 DAH and their density continuously increased thereafter. Interestingly, goblet cells appeared in the intestine, but not in the esophagus, where these cells are commonly observed in other teleosts. In addition, high amounts of eosinophil substances, assumed to be zymogen granules, accumulated in the pancreas, suggesting a specific digestive strategy of this species. Overall, these observations largely update the previous reports and can be useful to improve A. japonica seed production.

Development of a High-Titer Culture Medium for the Production of Cholesterol by Engineered Saccharomyces cerevisiae and Its Fed-Batch Cultivation Strategy

  • Steroids are a class of compounds with cyclopentane polyhydrophenanthrene as the parent nucleus, and they usually have unique biological and pharmacological activities. Most of the biosynthesis of steroids is completed by a series of enzymatic reactions starting from cholesterol. Synthetic biology can be used to synthesize cholesterol in engineered microorganisms, but the production of cholesterol is too low to further produce other high-value steroids from cholesterol as the raw material and precursor. In this work, combinational strategies were established to increase the production of cholesterol in engineered Saccharomyces cerevisiae RH6829. The basic medium for high cholesterol production was selected by screening 8 kinds of culture media. Single-factor optimization of the carbon and nitrogen sources of the culture medium, and the addition of calcium ions, zinc ions and citric acid, further increased the cholesterol production to 192.53 mg/l. In the 5-L bioreactor, through the establishment of strategies for glucose and citric acid feeding and dissolved oxygen regulation, the cholesterol production was further increased to 339.87 mg/l, which was 734% higher than that in the original medium. This is the highest titer of cholesterol produced by microorganisms currently reported. The fermentation program has also been conducted in a 50-L bioreactor to prove its stability and feasibility.

Distribution and Bionomics of the Argentine Ant Linepithema humile (Mayr) (Hymenoptera: Formicidae: Dolichoderinae)

  • In this study, the distribution, bait preference, hosts, symbionts, habitat status, and cytochrome c oxidase subunit-I sequences of the ant Linepithema humile were analyzed. This species habitat comprised approximately 1.5 km along stone crevices, flower gardens, roadsides, and container yards in the vicinity of Busan station, Korea. Feeding activity of this species was confirmed in 9 species of 7 families of plants. Which was approximately 14% of total flora (63 species of 37 families) in the studied area. Particularly, it was observed that Dendranthema indicum and Camellia japonica were the most frequently visited. Further, we verified that they interacted with four species of aphids. We identified 22 species of ants (17 genera, 4 subfamilies) inhabiting the Busan station area, including L. humile, Tetramorium tsushimae, Nylanderia flavipes and the alien species Paratrechina longicornis. In areas where L. humile was the dominant species, the habitat of local native ants was reduced by approximately 30%. Bait preference was ranked in the following order: jelly, water with glucose (25%), and yogurt, with the highest preference for baits containing sugar and protein. The 630-bp cytochrome c oxidase subunit-I sequences of the local L. humile populations were 99% identical to those of L. humile in the National Center for Biotechnology Information database, and the Argentine ants that invaded Korea had a sequence identical to that of other invasive populations from China, Japan, and New Zealand. As L. humile rapidly increases due to domestic adaptation, continuous monitoring and control strategy for eradication are needed to protect domestic biodiversity.

Food Sources of the Ascidian Styela clava Cultured in Suspension in Jindong Bay of Korea as Determined by C and N Stable Isotopes (탄소 및 질소안정동위원소 조성에 의한 남해안 진동만 양식 미더덕의 먹이원 평가)

  • To examine the trophic ecology of the ascidian Styela clava in an aquaculture system of Korea, stable carbon and nitrogen isotopes were analyzed monthly in S. clava, coarse ($>20{\mu}m$, CPOM) and fine particulate organic matters ($0.7<<20{\mu}m$, FPOM). CPOM (means: $-18.5{\pm}1.2$‰, $9.3{\pm}0.7$‰) were significantly higher ${\delta}^{13}C$ and ${\delta}^{15}N$ values than those ($-20.5{\pm}1.5$‰, $8.4{\pm}0.5$‰) of FPOM. S. clava had mean ${\delta}^{13}C$ and ${\delta}^{15}N$ values of $-18.9({\pm}1.7)$‰ and $11.6({\pm}0.7)$‰, respectively. S. clava were more similar to seasonal variations in ${\delta}^{13}C$ and ${\delta}^{15}N$ values of FPOM than those of CPOM, suggesting that they rely largely on the FPOM as a dietary source. In addition, our results displayed that the relative importance between CPOM and FPOM as dietary source for the ascidians can be changed according to the availability of each component in ambient environment, probably reflecting their feeding plasticity due to non-selective feeding irrespective of particle size. Finally, our results suggest that dynamics of pico- and nano-size plankton (i.e., FPOM) as an available nutritional source to S. clava should be effectively assessed to maintain and manage their sustainable aquaculture production.

Studies on the Early Growth of Rockfish, Sebastes schlegeli (조피볼락, Sebastes schlegeli의 초기성장에 관한 연구)

  • The study has been conducted to know an appropriate feeding strategy and effects of the rearing density on larval growth of the rockfish, Sebastes schlegeli. The results obtained are as fellowed ; 1. Thirty-day-old larvae reached at $25.25{\pm}3.76$ mm in total length and $0.23{\pm}0.07$ g in body weight in experiment A, at which rotifer was provided from the beginning to the end of 30-day experiment, Anemia from 3th to 18th day, and artificial feed from 13th to 30th day after hatching. When rotifer was provided for 30 days, Artemia from 6th to 18th day, and artificial feed from 18th to 30th day after hatching (experiment B), these larvae grew up to $27.52{\pm}2.50$ mm in total length and $0.26{\pm}0.06$ g in body weight. On the other hand, when rotifer and artificial feed were supplied with the same time schedule as shown in experiment B, and Artemia was feed from 6th to 30th day after hatching (experiment C), the total length and body weight of those larvae were $23.22{\pm}3.44$ mm and $0.15{\pm}0.05$ g, respectively. The best result for larval growth was obtained from experiment B. The survival rates estimated were $57.6\%$ in experiment A, $66.4\%$ in experiment B, and $44.4\%$ in experiment C. 2. The growth in total length of the larvae according to their rearing days could be represented by the following equations : Experiment A : Y=4.350+0.116X+$1.887X^2$ (r=0.993) Experiment B : Y=4.500+8.931X+$2.221X^2$ (r=0.994) Experiment C : Y=4.478+5.734X+$1.881X^2$(r=0.990) The average number of Artemia nauplius intaken by the larvae was rapidly increased between 15th and 20th day afer hatching, and 9, 212, 242, 750, and 1,171 nauplius were found in the different sizes of larvae, whose total length were 5.65, 6.81, 9.45, 14.96, and 24.52 mm, respectively. 3. Larval growth in total length and body weight reared at four different densites (A: 1.8 $kg/m^3$, B; 4.0 $kg/m^3$, C; 5.0 $kg/m^3$, D; 6.2 $kg/m^3$) indicated that the best growth was found in experiment A, at which the larval were reared at the lower density and the final survival rates extimated were $92.9\%$ in exp. A, $99.5\%$ in exp. C, $89.0\%$ in exp. B, and $88.2\%$ in exp. D. The amount of production per cubic meter turned out to be 30.45 kg in exp. D, 25.89 kg in exp. C, 20.75 kg in exp. B and 10.48 kg in exp. A. therefore, considering both larval growth and survival rate, higher yields seemed to be attainable at the relatively high-rearing density.

A Literature Review of Fish Feeding Research in the Coast - Estuary Areas of Korean Peninsula (우리나라 연안-하구에 서식하는 어류의 식성 관련 연구 현황)

  • We carried out a literature review of fish food sources research in the coast - estuary areas of Korean peninsula for the last fifty years(1969~2018). A total of 101 research papers were reviewed, of which 81 were published in domestic journals, 7 were puliched in domestic reports and 14 were published in international journals. When divided into the decade periods, 21 papers (19.4%) between 1990 and 1999 and 26 papers (24.1%) between 2000 and 2009, and 42 papers (48.1%) were published in 2010 and 2018, respectively. When divided into the survey area by a bearing (East sea, South-East sea, South Sea, and West Sea), 65 papers(69.1%) were carried out in the South Sea. Followed by 13 papers (13.8%) in the East Sea and 8 papers (8.5%) in the West Sea and the South-East Sea, respectively. The surveyed area was dominant in Gwangyang Bay (18 papers; 24.3%), followed by Gadeok-do (8 papers; 10.8%). Of the diversity indices, 14 indices were used to calculate the ecological status. The dominant indices were IRI(Index of relative importance), Pi(Prey-specific abundance) followed by E (electivity index) and Bi (dietary breadth index). A total of 11 statistical methods were used to find feeding strategy. The correlation analysis and Bray-Curtis similarity matrix were applied most frequently. In terms of ecological research topics, the papers focused on seasonal or spatial differences before 2000. Since 2000, specifically, the international journal papers have been published based on competition, sympatric, niche overlap and other ecological topics.

Climate Change by Global Warming and Its Effects on Production Efficiency of Lactating Dairy Cows in Korea : a Simulation Modeling Approach (지구온난화에 따른 국내 기후변화와 젖소 착유우의 생산효율에 미치는 영향 평가 : 모델 시뮬레이션을 이용한 접근)

  • The objectives of this study were to access climate change by global warming in Korea, and to investigate its effects on production efficiency of lactating dairy cows. Two regions, Daegu and Daekwanryung, were selected to represent a warm and a cold area, respectively. Time-series analyses on meteorological records for 25 years (from January 1, 1988 to December 31, 2012) revealed significant and different climate changes in two regions. In the warm area there has been a significant (P<0.05) increase in low temperature during the summer, which can cause heat stress to the animal. On the other hand, a decrease in low temperature during the winter was observed in the cold region (P<0.01), and cold stress in winter can thus be an issue in this region. Simulations using a model integrated the Korean feeding standard for dairy cattle and the environmental effect module of Cornell Net Carbohydrate and Protein System, indicated that a reduction in feed efficiency can be a problem during the winter in the cold region while during the summer in the warm area. We conclude that the effect of climate change by global warming varies in different areas in Korea and a region-specific management strategy should be developed in order to maintain productivity, health and welfare of lactating dairy cows.

Biotypes of the Brown Planthopper, Nilaparvara lugens (Stal) (벼멸구의 생태형)

  • The brown planthopper, N. lugens (Stal), has become a serious pest of rice in tropical Asia during the last decade. At high pest density, its feeding damage causes 'hopperburn' or complete wilting and drying of the rice plant. It also transmits grassy and ragged stunt virus diseases. The estimated losses caused by the pest in tropical Asia exceed $US\$300$ millions. While cultivation of resistant rice varieties has proved to be highly effective against the pest, their long-term stability is threatened because of the evolution of prolific biotypes which can destroy these varieties. At present, identification of biotypes is based principally on the differential reactions of host rice varieties to the pest and on host-mediated behavioral and physiological responses of the pest. Recent findings of morphological differences in adult rostrum, legs, and antennae, body parts that possess receptors for host plant location and discrimination, and cytological differences in N. lugens populations maintained as stock cultures strongly complement other biotype studies. So far, three N. lugens biotypes have been identified in the Philippines. Biotype I can survive on and damage varieties that do not carry and genes for resistance, while Biotype 2 survives on resistant varieties carrying Bph 1 gene and Biotype 3 on varieties carrying gene bph 2. However, none of these biotypes can survive on varieties with genes Bph 3 or bph 4. Several varieties which are resistant in the Philippines are susceptible in India and Sri Lanka as the South Asian biotypes of N. lugens are more virulent than Southeast Asian biotypes. To monitor the pest biotypes in different geographical regions and to identify new sources of resistance, an International Brown Planthopper Nursery has been established in many cooperating countries. The evolution of biotypes is an exceedingly complex process which is governed by the interactions of genetic and biological factors of the pest populations and the genetic makeup of the cultivated varieties. While the strategy for sequential release of varieties with major resistance genes has been fairly successful so far, the monegenic resistance of these varieties makes them vulnerable to the development of the pest biotypes. Therefore, present breeding endeavors envisage utilizing both major and minor resistance genes for effective control of the pest.

Hypoglycemic effects of Welsh onion in an animal model of diabetes mellitus

  • Tight control of blood glucose is the most important strategy for the treatment of diabetes mellitus. Here, we investigated the beneficial effects of Welsh onion on fasting and postprandial hyperglycemia. Inhibitory activities of hot water extracts from the green stalk and white bulb, which are the edible portions of the Welsh onion, and the fibrous root extract against yeast ${\alpha}$-glucosidase were measured in vitro. To study the effects of Welsh onion on postprandial hyperglycemia, a starch solution (1 g/kg) with and without Welsh onion fibrous root extract (500 mg/kg) or acarbose (50 mg/kg) was administered to streptozotocin-induced diabetic rats after an overnight fast. Postprandial plasma glucose levels were measured and incremental areas under the response curve were calculated. To study the hypoglycemic effects of chronic feeding of Welsh onion, five-week-old db/db mice were fed an AIN-93G diet or a diet containing either Welsh onion fibrous root extract at 0.5% or acarbose at 0.05% for 7 weeks after 1 week of adaptation. Fasting plasma glucose and blood glycated hemoglobin were measured. Compared to the extract from the edible portions of Welsh onion, the fibrous root extract showed stronger inhibition against yeast ${\alpha}$-glucosidase, with an $IC_{50}$ of 239 ${\mu}g/mL$. Oral administration of Welsh onion fibrous root extract (500 mg/kg) and acarbose (50 mg/kg) significantly decreased incremental plasma glucose levels 30-120 min after oral ingestion of starch as well as the area under the postprandial glucose response curve, compared to the control group (P < 0.01). The plasma glucose and blood glycated hemoglobin levels of the Welsh onion group were significantly lower than those of the control group (P < 0.01), and were not significantly different from those fed acarbose. Thus, we conclude that the fibrous root of Welsh onion is effective in controlling hyperglycemia in animal models of diabetes mellitus.

Super-intensive Culture of Whiteleg Shrimp, Litopenaeus vannamei (Boone, 1931), in HDPE-lined Ponds with no Water Exchange (사육수 비교환방식을 이용한 포장 사육지에서의 흰다리새우, Litopenaeus vannamei (Boone, 1931)의 초고밀도양식)

  • Shrimp farming is the most important mariculture industry on the west coast of South Korea. However, it has suffered from mass mortality due to viral disease outbreaks and coastal pollution due to water discharge. This study developed an intensive shrimp culture method for outdoor ponds, without water exchange, which minimizes the chance of viral transmission from the environment, reduces coastal pollution by water discharge and enhances shrimp production. A culture trial was conducted in two high-density polyethylene (HDPE)-lined ponds with a $550\;m^2$ surface area. The ponds were stocked with postlarvae of Litopenaeus vannamei, the major farmed shrimp species in Korea, on July 10, 2007, and cultured for 90 days with no water exchange. The stocking density of the postlarvae (B.W. 0.0015 g) was $272\;ind./m^2$, which is eight times higher than in traditional pond culture in Korea. At harvest, the total production of ponds 1 and 2 was 1,362kg ($2.48\;kg/m^2$) and 1,282 kg ($2.33\;kg/m^2$), respectively. This is 20~22 times higher than the mean farmed shrimp production ($0.112\;kg/m^2$) in Korea and about eight times higher than in traditional ponds with a good harvest. Although there was no water exchange throughout the culture period, the mean concentrations of unionized ammonia and nitrite-nitrogen were as low as 0.038 and 6.0 mg/L, respectively. The feed conversion rate (FCR) was 1.38, which is 20~45% lower than that of traditional pond cultures. The high efficiency of the diet in this study is thought to be due to a well-managed feeding strategy and well-developed bioflocs used as diet additions for the shrimp. The final body weight of the shrimp at harvest was low (12.2~12.5 g), compared with that of traditional pond culture. This may have resulted from the combination of a short culture period, high density of shrimp, and low temperature. This study suggests that a super-intensive shrimp pond culture method using biofloc technology with no water exchange can minimize viral transmission via water exchange, reduce coastal pollution, and enhance shrimp production.
