Fed-batch cultures of Hansenula polymorpha were studied to develop an efficient biosystem to produce recombinant human serum albumin (HSA). To comply with this purpose, we used a high-purity oxygen-supplying strategy to increase the viable cell density in a bioreactor and enhance the production of target protein. A mutant strain, H. polymorpha GOT7, was utilized in this study as a host strain in both 5-l and 30-l scale fermentors. To supply high-purity oxygen into a bioreactor, nearly 100% high-purity oxygen from a commercial bomb or higher than 93% oxygen available in situ from a pressure swing adsorption (PSA) oxygen generator was employed. Under the optimal fermentation of H. polymorpha with highpurity oxygen, the final cell densities and produced HSA concentrations were 24.6 g/l and 5.1 g/l in the 5-l fermentor, and 24.8 g/l and 4.5 g/l in the 30-l fermentor, respectively. These were about 2-10 times higher than those obtained in air-based fed-batch fermentations. The discrepancies between the 5-l and 30-l fermentors with air supply were presumably due to the higher contribution of surface aeration over submerged aeration in the 5-l fermentor. This study, therefore, proved the positive effect of high-purity oxygen in enhancing viable cell density as well as target recombinant protein production in microbial fermentations.
The potential of algal-bacterial culture was investigated for advanced treatment of animal wastewater. Fed-batch experiments were carried out to examine treatability of nitrogen and phosphorus in different microbial consortium: Chlorella vulgaris, activated sludge, three microalgae strains (Scenedesmus, Microcystis, Chlorella) and Bacillus consortium, and three microalgae strains and sludge consortium. Single culture of C. vugaris showed the better efficiency for nitrogen removal but was not good at organic matter and phosphorus removal compared with activated sludge. Three microalgae and Bacillus consortium was best culture among the culture and consortium for pollutants removal tested in this experiment. Effect of $CO_2$ addition was studied by using three microalgae and Bacillus consortium. $CO_2$ addition enhanced T-P removal efficiency up to 60%. However, removal efficiencies of T-N and ammonia nitrogen reduced on the contrary.
New problems have been recently posed on the abnormal foaming (Scum) in an aeration tank and the sludge flotation in a final sedimentation tank during the activated sludge process. However, the activities of the causing bacteria, Nocardia-amarae in an aeration tank have not been searched out at all. Therefore, in this article the activities of Nocardia-amarae in an aeration tank have been closely examined by means of the changes of (F/M) ratio, SRT and inflowing substrate using continuous type and fed-batch type. Summarized results of experiments are as follows. 1. Regrading continuous culture when synthetic wastewater was used substrate neither the increase in the number of Nocardia-amarae in the aeration tank nor the Occurrence of Scum was observed. 2. In the case of fed-batch culture, Nocardia-amarae in the aeration tank increased due to the partial change in substrate and the effect of SRT was significant. 3. Once the scum was formed and the quantity of added Nocardia-amarae and substrate were not changed, the effect of STR was not significant.
An accurate and efficient method for measuring the mass of hairy roots using conductometry is established. A conductivity equation expressed in terms of the concentration of the ion species in the medium is suggested. By using this equation, the effect of the individual ions on the total conductivity can be quantitatively analyzed. An equation for the in situ estimation of the cell growth coefficient for determining the mass of hairy roots is established based on measurements of the nitrogen concentration and conductivity during cultivation. The proposed equation does not require preliminary experiments to determine the cell growth coefficient. Instead, the physiological characteristics of the plant species are reflected by introducing the cellular nitrogen content. Since the cell growth coefficient is determined by measuring the major ionic nutrient concentrations, it is more effective to express the dynamics of an actual culture system. This improved method for determining the mass of hairy roots was successfully utilized in a fed-batch culture system.
According to the controllability of pulse times and the amount of jumps in the states at these times in the process of fed-batch culture producing 1,3-propanediol, this paper proposes a terminal optimal control model, whose constraint condition is the nonlinear impulsive delay system. The existence of optimal control is discussed and an optimization algorithm which is applied to each subinternal over one cycle for this optimal control problem is constructed. Finally, the numerical simulations show that the terminal intensity of producing 1,3-propanediol has been increased obviously.
The optimal glucose concentration for the high-density culture of recombinant yeasts was obtained using dynamic simulation. An adaptive and predictive algoritilm complimented by the rule base was proposed for the control of the fed-batch fermentation process. The measurement of process variables has relatively long sampling period and relatively long time delay characteristics. As one of the solution on these problems, prediction techniques and rule bases were added to a classical recursive identification and control algorithm. Rule bases were used in the determination of control input considering the difference between the predicted value and the measured value. A mathelnatical model was used in the estimation and interpretation of the changes of state variables and parameters. Better performances were obtained by employing the control algorithm proposed in the present study compared to the conventional adaptive control method.

For the coexpression of endoxylanase and endoglucanase genes in yeast Saccharomyces cerevisiae, the genes were separately inserted downstream of the yeast ADH1 promoters, resulting the plasmid pAGX3 (9.83 kb). In the batch culture on YPD medium of the yeast transformant, S. cerevisiae SEY2102/pAGX3, the total activities of the enzymes reached about 7.91 units/ml for endoxylanase and 0.43 units/ml for endoglucanase. In the fed-batch culture with intermittent feeding of yeast extract and glucose, the total activities of 24.9 units/ml for endoxylanase and 0.84 units/ml for endoglucanase were produced which were about 3.1-fold and 2.0-fold increased levels, respectively, compared to those of the batch culture. Most of endoxylanase and endoglucanase activities were found in the extracellular media. This recombinant yeast could be useful for the development of simultaneous saccharification bioprocess of the cellulose and xylan mixture.
In order to maximize the cell growth and the biosurfactant production by the Pseudomonas aeruginosa BYK-2(KCTC 18012P), in the fed-batch fermentation processes were performed varying the feeding medium concentrations and the feeding rate. Feel-batch culture was performed with the optimal agitation speed of 200rpm and the aeration rate of 0.67vvm in a 7L Jar fermentor containing 3L of modified medium and 2.0-2.5%(v/v) fish oil as a carbon source. Addition of fish oil(2.5mL/l00mL modified medium), when fish oil was depleted, the cell and biosurfactant concentration were 6.1g/L and 22.7g/L, respectively.
Utilization of yeast extract and formation of byproduct metabolite were investigated for hyperthermophilic archaeon Sulfolobus solfataricus (DSH 1617). In both batch and fed-batch cultivations of S. solfataricus, maximal cell density, {{{{ { NH}`_{4 } ^{ +} }}} ion production and pH change were highly dependent on the ratio of yeast extract to glucose in the medium. Variation of {{{{ { NH}`_{4 } ^{ +} }}}} ion level was identified as a major cause of pH change during cultivation, and acidification of culture broth was attributed to consumption of {{{{ { NH}`_{4 } ^{ +} }}}} ions rather than formation of acid byproducts. It was also observed that increase of {{{{ { NH}`_{4 } ^{ +} }}}} ion concentrations in the medium resulted in greater degree of growth inhibition.

Controlling the dissolved oxygen (DO) in the fed-batch culture of the medicinal mushroom Ganoderma lucidum led to a 2-fold increase of the maximum biomass productivity compared with uncontrolled DO conditions. By contrast, extracellular polysaccharide (EPS) production was two times higher under oxygen limitation (uncontrolled DO) than under increased oxygen availability (controlled DO). Morphologically, dispersed mycelium was predominant under controlled DO conditions, with highly branched hyphae, consistent with the enhanced culture growth noted under these conditions, whereas in the uncontrolled DO process mycelial clumps were the most common morphology throughout the culture. However, in both cultures, clamp connections were found. This is an exciting new finding, which widens the applicability of this basidiomycete in submerged fermentation. In rheological terms, broths demonstrated shear-thinning behavior with a yield stress under both DO conditions. The flow curves were best described by the Herschel-Bulkley model: flow index down to 0.6 and consistency coefficient up to 0.2 and 0.6 Pa $s^n$ in uncontrolled and controlled cultures DO, respectively. The pseudoplastic behavior was entirely due to the fungal biomass, and not to the presence of EPS (rheological analysis of the filtered broth showed Newtonian behavior). It is clear from this study that dissolved oxygen tension is a critical process parameter that distinctly influences G. lucidum morphology and rheology, affecting the overall performance of the process. This study contributes to an improved understanding of the process physiology of submerged fermentation of G. lucidum.
