Fifty mid-lactation Holstein cows were used in a six-week feeding trial to study effects of high-forage, high-fat diets on dry matter intake and production performance. Cows were divided into 10 replicates, each consisting of five cows. Each cow was assigned to a control (diet 1) or one of the four experimental diets (high-forage (75%), high-fat (7.5%) (diet 2); high-forage, medium-fat (5%) (diet 3); medium forage (65%), high-fat (diet 4); medium-forage, medium-fat (diet 5)), or a control diet containing about 50% forage and 2% fat. All diets were isonitrogenous (17.7% crude protein). The forage mixture consisted of 20% alfalfa hay, 40% alfalfa haylage, and 40% com silage. Supplemental fat included 80% rumen-protected fat and 20% yellow grease. Dry matter intake was decreased (p<0.01) in cows fed experimental diets (18.4, 20.9, 19.9, and 22.6 kg for cows fed diets 1-4, respectively vs. 27.5 kg for cows fed the control diet). Daily milk production was lower (p<0.05) for cows consuming experimental diets (30.5, 31.3, 31.0, and 32.5 kg for cows fed greater for cows consuming experimental diets (1.74, 1.55, 1.60, and 1.53 kg milk/kg dry matter intake for cows fed diets 1-4, respectively, vs. 1.26 kg milk/kg dry matter intake for cows fed the control diet).
BACKGROUND/OBJECTIVES: Obesity-associated insulin resistance is a strong risk factor for type 2 diabetes mellitus. The aim of this study was to investigate the effect of myricetin on adiposity, insulin resistance, and inflammatory markers in mice with diet-induced insulin resistance. MATERIALS/METHODS: Five-week-old male C57BL/6J mice were fed a basal diet, a high-fat, high-sucrose (HFHS) diet, or the HFHS diet containing 0.06% myricetin or 0.12% myricetin for 12 weeks after a 1-week adaptation, and body weight and food intake were monitored. After sacrifice, serum lipid profiles, glucose, insulin, adipocyte-derived hormones, and proinflammatory cytokines were measured. The homeostasis model assessment for insulin resistance (HOMA-IR) was determined. RESULTS: Myricetin given at 0.12% of the total diet significantly reduced body weight, weight gain, and epidydimal white adipose tissue weight, and improved hypertriglyceridemia and hypercholesterolemia without a significant influence on food intake in mice fed the HFHS diet. Serum glucose and insulin levels, as well as HOMA-IR values, decreased significantly by 0.12% myricetin supplementation in mice fed the HFHS diet. Myricetin given at 0.12% of the total diet significantly reduced serum levels of leptin, tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6) in mice fed the HFHS diet. CONCLUSIONS: These findings suggest that myricetin may have a protective effect against diet-induced obesity and insulin resistance in mice fed HFHS diet, and that alleviation of insulin resistance could partly occur by improving obesity and reducing serum proinflammatory cytokine levels.
$Dioscorea$$batatas$ Decne is a type of yam that eaten raw and used as a traditional oriental medicine in Asian countries. In this study, we evaluated the functional effects of yam water extracts on body weight levels and serum lipid concentrations in mice fed high fat diet. Mice were divided into four groups: normal diet control (ND), high fat diet control (HFD), HFD+yam extract 100 mg/kg (HFD-Y100), and HFD+yam extract 200 mg/kg (HFD-Y200). Yam extract was administrated orally to mice fed a high fat diet for 5 weeks. Treatment with yam extract significantly reduced body weight levels and energy efficiency in a dose-dependent manner in HFD-fed mice. Yam extract also attenuated serum levels of triglyceride, total cholesterol, and LDL-cholesterol, as well as organ weights of liver and abdominal adipose tissue in mice fed a high fat diet. Moreover, blood levels of glucose, insulin, and leptin significantly decreased in a dose-dependent manner upon administration of yam extract. Blood activities of GPT, GOT, and LDH were lower in the yam extract-treated groups compared to the HFD group. These results indicate that yam water extract may reduce elevated body weight and serum lipid concentrations in mice fed a high fat diet, suggesting its usefulness as a functional food for reducing body fat and hyperlipidemia.
This study investigated the effects of Wolbiga-banha-tang (WBBT) on high fat diet-induced rats. We divided the rats into 4 groups: normal (fed a general-fat diet), control (fed a high-fat diet and no treatment), $WBBT{\times}1$ (fed a high-fat diet and 250 mg/kg of Wolbiga-banha-tang extracts), and $WBBT{\times}2$ (fed a high-fat diet and 500 mg/kg of Wolbiga-banha-tang extracts). We measured the body weight, total cholesterol, HDL cholesterol, LDL cholesterol, triglycerides, free fatty acids, total lipids, AST, ALT, and CBC in the serum of the obese rats for 7 weeks. There was a significant decrease in the control group, $WBBT{\times}1$, and $WBBT{\times}2$ in triglycerides and free fatty acids. The results suggest that WBBT is effective for lowering triglycerides and free fatty acids. Therefore, WBBT may be applied to the treatment of lipid metabolism in obesity and further clinical studies are needed.
This study was performed to investigate the effects of liquid culture of Agaricus blazei Murill on the weight gains, food efficiency ratios, serum protein and mineral levels, and serum enzyme activities in growing male rats. Sprague-Dawley rats (7 weeks old) were given four different types of diets for a succeeding period of 6 weeks, respectively: a normal diet group, a control diet group (normal diet + 15% lard + 0.5% cholesterol), a 30% or 40% A. blazei diet groups (control diet + 30% or 40% A. blazei in water) according to the levels of A. blazei supplementation. The body weight gains, food efficiency ratios, and the liver of the rats fed control diet, 30% or 40% A. blazei diets were significantly increased compared to rats fed the normal diet, but those of rats fed the 30% and 40% A. blazei diets were similar to those of rats fed the control diet. The concentrations of total protein, albumin, glucose, and hematocrit value in serum of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. The concentrations of urea and creatinine in serum of rats fed the 30% or 40% A. blazei diets were similar to those of rats fed the control diet. but the urea of rats fed the 30% and 40% A. blazei diets were significantly decreased compared to rats fed the normal diet. The concentrations of Ca, P, Fe and Mg, and $Ca^{++}$, $K^+$, $Na^+$ and $Cl^-$ of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. There were no differences in the activities of aspartate aminotransferase, alanine aminotransferase, $\gamma$-glutamyltranspeptidase and alkaline phosphatase in the serum among the experimental groups. In conclusion, the rats fed the A. blazei maintained normal protein and mineral levels, and enzyme activities of serum. But the A. blazei feeding could not decrease the body and liver weights in the rats fed high cholesterol diets.
Electrocardiograms in rats fed diets with boiled eggs for 30 days was investigated. Amplitudes of P,Q and R waves were not significant differences among treatment groups. Amplitude of S wave in rats fed the diet with 95% boiled eggs was significantly tended to be increased compared with other groups(P<0.05). Amplitude of T wave in the rats fed the diet with 0% boiled eggs showed the highest values, and there were significant difference among treatment groups fed diets with 0% boiled eggs, 25% and 95% boiled eggs (P<0.05). Durations of P and PQ(PR) waves were high in the rats fed diets with 25% and 50% boiled eggs (P<0.05). Duration of QRS complex showed low in the rats fed diet 0% boiled eggs but not significant difference among treatment groups. Duration of QT was high in the rats fed diet with 0% boiled eggs(P<0.05). This result is assumed that electrocardiograms in the rats is not changed to intake the boiled eggs.
Biochemical studies such as growth rate, blood cholesterol and ascorbic acid contents in various organs of albino male rats were studied in the Garlic, Vitamin E, high and low protein diet fed groups. The results of this study were summarized as follows : 1) Either single 10% or 30% protein diet fed rats were not shown growth rate properly. In supplementation of Vitamin E or Garlic on 30% protein growth rate was a more increased than that of the 10% protein diet group. 2) The rate of food consumption of rats fed a 10% or 30% protein diet supplemented with Vitamin E and Garlic was more increased than that of the control Group receving 10% or 30% protein diet alone. 3) No essential difference was observed between the cholesterol level in blood of rats fed 10% protein diet supplemented with Vitamin E and Garlic and that of rats Receiving 10% protein diet alone. But the cholesterol level in blood of rats fed 30% protein diet supplemented with Vitamin E and Garlic was lower than that of control receiving 30% protein diet alone. 4) The Vitamin C contents in various organs of rats fed the diet supplemented with Vitamin E and Garlic seems to be a little higher than that of the control group fed the protein diet alone.
The adipose tissue hormone leptin has been proposed to be involved in the regulation of flood intake and energy expenditure via thermogenesis by uncoupling protein(UCP) in brown adipose tissue(BAT). The objective of the study was to examine the effects of high fat diet on the serum leptin levels, BAT UCPl expression and the body fat mass in rats after weaning. During experimental period of 12 weeks, 4 male Sprague-Dawley rats were killed for the baseline experiment at 4 weeks of age while the remaining rats were fed the two different diets: the control diet AIN-76A(n = 20), high fat(beef tallow) diet(n = 20) ad libitum, which provided 11.7% or 40% of calories as fat, respectively. At 16 weeks of age, the increase in the food efficiency ratio(FER) was related to fat mass in rats on high fat diet. Serum leptin level was increased by age and dietary high fat. There was no difference in serum insulin level between groups until 10 weeks of age, but rats fed high fat diet for 12 weeks showed hyperinsulinemia. The amount of body fat pads was increased significantly in high fat group compared to normal diet group. Visceral fat mass affected acutely by high fat diet, as a result, it was higher in rats fed high fat diet for 2 weeks than normal diet. At 16 weeks of age, BAT and visceral fat mass were significantly high in high fat group. Also, the serum leptin levels reflected the amount of body fat mass. BAT UCPI mRNA expression increased with age and dietary high fat. This study demonstrates that dietary high fat increased serum leptin levels, BAT UCPI expression and body fat mass. Futhermore, in rats fed high fat diets, the increases in leptin and UCPI expression counteracts only in part the excess adiposity and obesity.
This study was conducted to investigate the effects of shellfish hydrolysate on lipid metabolism in rats fed high fat diet. Male Sprague-Dawley rate weighting approximately 110g were fed basal control diet, high fat diet and high fat diet plus 4 different shellfish hydrolyates for 4 weeks. The shellfish hydrolysates from the different sources, were oyster, hard-shelled mussel, little neck clam and march clam. After 4 weeks, serum GOT, GPT, ${\gamma}$-GTP, triglyceride and total cholesterol was significantly decreased in shellfish hydrolysates supplementation with high fat diet compared to basal control and high-fat group(p<0.05). The total lipid and cholesterol content in liver showed significant decrease(p, 0.05). There were no different in serum GPT, HDL-cholesterol, liver total cholesterol and lipid of rats between basal control diet and high-fat diet. The unsaturated fatty acids, specific components of shellfishes were a little components in shellfish hydrolysate as they were a low and not different among the groups and were most well reflected in liver and plasma. Considering digestive and absorptive process of in human body, it was assumed that the hypolipidemic effect of shellfish was not under the influence of unsaturated fatty acids but the other components, peptides, taurine and betaine and so on was detected in the process of hyperlipidemia induced by high-fat diet.

BACKGROUND/OBJECTIVES: Doenjang, Korean traditional fermented soybean paste has been reported to have an anti-obesity effect. Because adipose tissue is considered a major source of inflammatory signals, we investigated the protective effects of Doenjang and steamed soybean on oxidative stress and inflammation in adipose tissue of diet-induced obese mice. MATERIALS/METHODS: Male C57BL/6J mice were fed a low fat diet (LF), a high-fat diet (HF), or a high-fat containing Doenjang diet (DJ) or a high-fat containing steamed soybean diet (SS) for 11 weeks. RESULTS: Mice fed a DJ diet showed significantly lower body and adipose tissue weights than those in the HF group. Although no significant differences in adipocyte size and number were observed among the HF diet-fed groups, consumption of Doenjang alleviated the incidence of crown-like structures in adipose tissue. Consistently, we observed significantly reduced mRNA levels of oxidative stress markers (heme oxygenase-1 and $p40^{phox}$), pro-inflammatory adipokines (tumor necrosis factor alpha and macrophage chemoattractant protein-1), macrophage markers (CD68 and CD11c), and a fibrosis marker (transforming growth factor beta 1) by Doenjang consumption. Gene expression of anti-inflammatory adipokine, adiponectin was significantly induced in the DJ group and the SS group compared to the HF group. The anti-oxidative stress and anti-inflammatory effects observed in mice fed an SS diet were not as effective as those in mice fed a DJ diet, suggesting that the bioactive compounds produced during fermentation and aging may be involved in the observed health-beneficial effects of Doenjang. CONCLUSIONS: Doenjang alleviated oxidative stress and restored the dysregulated expression of adipokine genes caused by excess adiposity. Therefore, Doenjang may ameliorate systemic inflammation and oxidative stress in obesity via inhibition of inflammatory signals of adipose tissue.
