단백식이 조건을 달리하여 성장시킨 흰쥐에 Bromobenzene 투여가 간손상에 미치는 영향 (An Effect of Bromobenzene Treatment on the Liver Damage of Rats Previously Fed Low or High Protein Diet)

  • To evaluate an effect of dietary protein on the liver damage, the bromobenzene was intraperitoneally injected to the rats fed a low or high protein diet and then the liver weight per body weight and serum levels of alanine aminotransferase (ALT) activities were determined to demonstrate the differences in liver damage between the groups fed low or high protein diet. Hepatic aniline hydroxylase (AH), glutthione (GSH) content and glutathione s-transferase(GST) activity were also determined to clarify causes of liver damage between the two groups. Increases of liver weight per body weight and serum ALT activities were higher in brombenzene treated rats fed low protein diet than those fed high protein diet. The increasing rate of hepatic AH activity was higher in bromobenzne-treated rats fed low protein diet than that in those fed high protein diet. Furthermore , hepatic glutathione contents and GST activities in bromobenzene-treated rats were higher in rats fed high protein diet than those fed low protein diet. In case of control group, the heaptic glutathione content and GST activity were also higher in rats fed high protein diet than those fed low protein diet.

  • 식이성단백이 methanethiol에 의한 간기능에 어떠한 영향을 미치는지를 검토코자 흰쥐를 저단백식이(LP : casein 7%) 및 고단백식이 (HP : casein 20%)로 1개월간 성장시킨 뒤 methanethiol을 투여한 다음, 간무게, 혈청 ALT 활성을 측정하여 간기능 정도를 두 군간에 비교하였다. 동시에 이에 대한 원인을 규명하는 일환으로 간조직 중 glutahione함량과 glutathione 포함효소의 일종인 glutathione S-transferase (GST)활성을 측정하여 다음과 같은 실험 결과를 얻었다. Methanethiol 투여로 인한 대조군에 대한 간무게와 혈청 ALT활성 증가율은 LP군이 HP군보다 높았으며, 간 glutathione함량과 간 GST활성이 HP군 보다 LP군이 낮게 나타났다. 이상 성적을 종합해 볼 때 methanethiol에 의한 간 독성은 식이 중 단백함량감소에 비례해서 증가되며 이는 LP군이 HP군 보다 methanethiol해독에 관여하는 간 glutathione함량과 GST활성의 감소에 기인되기 때문인 것으로 생각된다.

칼슘과 철의 과다섭가 성장기 흰쥐의 체내 무기질 이용성에 미치는 영향 (Effect of Calcium and Iron Loading on Bioavailability of Minerals in Normal and Ca/Fe-deficient Rats)

  • This study examined the effect of excess loading of calcium (Ca)and iron(Fe) on the bioavailability of minerals in both normal and Ca-and Fe-deficient rats. Three-week-old male rats were divided into four groups and fed experimental diets for six weeks, containing either normal (0.5%) or high(1.5%) Ca and normal (35ppm) or high (350ppm)Fe. Likewise, three-week-old male rats were first fed a Ca-and Fe-deficient diet for three weeks, and then fed one of four experimental diets for additional three weeks. In both normal and Ca-and Fe-deficient rats, ca contents of serum, liver, kidney and femur were not significantly affected by dietary Ca and Fe levels. Apparent Ca absorption(%) decreased in rats fed a high Ca diet regardless of dietary Fe levels. Magnesium(Mg) contents of serum, liver and femur significantly decreased in rats fed a high Ca diet. Fe contents of serum and liver significantly increased in rats fed a high-Fe diet, but decreased in rats fed a high Ca diet. Fe content of serum and liver significantly increased in rate fed a high-Fe diet, decreased in rats fed a high-Ca diet. Apparent Fe absorption increased in rats fed a high-Fe diet, and decreased in rats fed a high-Ca diet in Ca-and Fe-deficient rats, but dietary Ca did not seem to affect Fe absorption in normal rats. Phosphorus(P) contents of serum and femur were not significantly affected by dietary Ca and Fe levels in both normal and Ca-and Fe-deficient rats. Serum copper(Cu) decreased in rats fed a high-Fe diet, while serum zinc(Zn) decreased in rats fed a high-Ca diet in normal rats. Cu contents of liver, and Zn contents of serum and liver decreased in rats fed a high-Fe diet in Ca-and Fe-deficient rats. There results suggest that a dietary overload of Ca and Fe in both normal and Ca-and Fe-deficient rats may decrease mineral bioavailability leading to potential health problems.

고 트립토판 식이를 섭취한 마우스에서 Immobilization 스트레스로 인한 면역변조와 Serotonin 대사의 변화에 대한 연구 (A Study on Immunomodulation and Serotonin Metabolism Changes by Immobilization Stress in Mice Fed Tryptophan Supplemented Diet.)

  • We fed high trypotophan diet(3.5% tryptophan/diet(w/w) to mice for 7 days and treated then with 3 hour immobilization(IMMB) stress to investigate tryptophan metabolism and immunomodulation. The levels of serum tryptophan, brain tryptophan, serotonin(5HT) and 5-hydroxyindoleacetic acid(5HIAA) in the tryptophan diet fed animals were higher than those of the normal diet fed animals. Feeding tryptophan supplemented diet to stressed animal significantly decreased the levels of serum and brain tryptophan and 5HT levels. However, the amount of 5HIAA which is the metabolite of serotonin was increased in brain. Plasma corticosterone level was increased by the stress in both groups but the degree of this increase was smaller in high tryptophan fed animals. The relative numbers of CD8+ T cells, CD4+ T cells and B cells in spleen were decreased in high tryptophan diet fed and stressed animals compared to control diet fed and no stressed animals. CD8+ T cells decreased more than CD4+ T cells. The decrease of CD8+ T cells in high tryptophan fed and stressed animals was similar to that in high tryptophan fed animals or normal diet fed and stressed animals. Stress and tryptophan supplement acted synergistically to decrease the number of B cells. This study suggests that stress and tryptophan supplement could modify the number of lymphocyte cells, and indicates that the interaction of stress and tryptophan supplement on immune fuction depends on the types of immune cells.

신령버섯이 고지방 식이를 급여한 흰쥐의 지질대사에 미치는 영향 (Effects of Agaricus blazei Murill on Lipid Metabolism in Rats Fed High Fat Diet)

  • 신령버섯이 고지방식이를 섭취한 흰쥐의 지질대사에 미치는 영향을 조사하고자, 생후 21주링의 音쥐 에 표준식이를 급여한 정상군, 표준식이에 20% 돈지를 첨가한 식이를 급여한 고지방군, 고지방 식이에 신령버섯 자실체 분말을 3% 및 5%씩 첨가한 식이를 급여한 군(3% 및 5% 신령 버섯군) 등 4군으로 나누어 10주간 사육한 결과는 다음과 같다. 실험동물의 체중증가량 및 식이효율은 고지방군에 비해 3% 신령버섯군이 다소 감소되었으나, 5% 신령버섯군은 유의하게 감소되었다. 간과 신장의 무게는 고지방군과 신령버섯군들이 비슷하였으나, 부고환지방은 고지방군에 비해 신령버섯군들이 유의하게 감소되었다. 간의 콜레스데롤 농도는 고지방군과 신령버섯군들이 비슷한 수준으로 감소효과가 나타나지 않았다. 혈청 및 간의 중성지질 농도는 고지방군에 비해 5% 신령버섯군이 유의하게 감소되었다. 혈청의 총 콜레스테롤과 LDL-콜레스테롤 농도 및 동맥경화지수는 고지방군에 비해 3% 및 5% 신령버섯군이 유의하게 감소되었다. 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율은 고지방군에 비해 신령버섯군들이 유의하게 증가되었다. HDL-콜레스테롤 및 인지질 농도는 고지방군과 신령버섯군들이 비슷한 수준으로 신령버섯 섭취에 따른 영향은 나타나지 않았다. 이상의 결과로 보아 고지방식이를 급여한 흰쥐에 신령버섯을 5% 급여시 혈청과 간의 중성지질 농도, 혈청의 총 콜레스테롤, LDL-콜레스테롤 농도 및 동백경화지수를 낮추고, 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율을 증가시키는 것으로 나타났다.

고콜레스테롤 식이를 급여한 흰쥐에서 Xylooligo당의 혈중 지질개선 효과 (Effects of Dietary Xylooligosaccharide on Lipid Levels of Serum in Rats Fed High Cholesterol Diet)

  • The purpose of this study was to investigate the effects of dietary xylooligosaccharide on lipid levels of serum in rats fed high cholesterol diet. Male Sprage-Dawley rats weiging 100$\pm$10g were randomly assigned to groups of two normal(N, N+10X) and four high cholesterol diets which contained 1%(w/w) cholesterol. High cholesterol diet groups were classified to xylooligosaccharide free diet (C group), 5% xylooligosaccharide diet(C+5X group), 10% xylooligosaccharide diet(C+10X group) and 15% xylooligosaccharide diet(C+15X group) according to the level of dietary xylooligosaccharide supplementation. These experimental diets were fed ad libidum for 4 weeks. The weight gain of high cholesterol diet group were significantly increased more than that of normal group, but those of 10% and 15% dietary xylooligosaccharide groups were significantly decreased more than that of high cholesterol diet(C)group. The higher content of xylooligosaccharide, the more food intake was increased. The food efficiencies of 10%, 15% cholesterol diet groups were lower than that of high cholesterol diet(C)group. The levels of serum triglyceride(TG) and total cholestoral were significantly high in cholesterol diet groups compared with normal diet group but were decreased in groups fed 5% and 10% dietary xylooligosaccharide. Especially, the lowest level showed in group fed high cholesterol diet containing 10% xylooligosacchride. High cholesterol diet group containing 10% xylooligosaccharide increased HDL-cholesterol level and then decreased LDL-cholesterol level and atherogenic index compared with other groups. The level of TBARS(thiobarbituric acid reactive substances) in serum was decreased in rat group fed xylooligosaccharide in high cholesterol diet. The higher content of xylooligosacchride, the more gastrointestinal transit time was decreased. The results indicate that dietary xylooligosaccharide can improve status of TG and total cholesterol and repress lipid peroxidation in serum lipid at hypercholesterolemia induced by high cholesterol diet.

신령버섯 균사체 액체배양액이 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향 (Effects of Liquid Culture of Agaricus blazei Murill on Lipid Metabolism in Rats Fed Cholesterol Diet.)

  • 신령버섯 균사체 액체배양액이 고콜레스테롤 식이를 섭취한 흰쥐의 지질대사에 미치는 영향을 조사하고자, 생후 7주령의 숫쥐에 표준식이를 급여한 정상군, 표준식이에 15% 돈지와 0.5% 콜레스테롤을 첨가한 식이를 급여한 대조군, 대조식이에 신령버섯 균사체 액체배양액을 음료수에 30% 및 40%로 혼합 급여한 군(30% 및 40% 신령버섯군) 등 4군으로 나누어 6주간 사육한 결과는 다음과 같다. 실험동물의 체중 증가량, 식이섭취량 및 식이효율은 대조군과 30%와 40% 신령버섯군이 유사하였다. 간, 신장 및 부고환지방의 무게는 대조군과 신령버섯군들이 비슷하였다 간의 콜레스테롤 농도, 간과 혈청의 중성지질 농도는 대조군과 신령버섯군들이 비슷한 수준으로 감소효과가 나타나지 않았다. 혈청의 총 콜레스테롤과 LDL-콜레스테롤 농도 및 동맥경화지수는 대조군에 비해 30% 및 40% 신령버섯군이 유의하게 감소되었다. 총콜레스테롤에 대한 HDL-콜레스테롤의 비율은 대조군에 비해 신령버섯군들이 유의하게 증가되었다. HDL-콜레스테롤 농도는 대조군과 신령 버섯군들이 비슷한 수준으로 신령 버섯섭취에 따른 영향은 나타나지 않았다. 분으로 배설된 지방은 대조군에 비해 40% 신령버섯군이 유의하게 증가되었다. 이상의 결과로 보아 고콜레스테롤 식이를 급여한 흰쥐에 신령버섯 균사체 액체배양액을 30%나 40% 급여시 혈청의 총 콜레스테롤, LDL-콜레스테롤 농도 및 동맥경화지수를 낮추고, 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율을 증가시키는 것으로 나타났다.

고지혈증 모델 흰쥐에서 칼슘과 소디움 섭취수준이 체내 지질대사에 미치는 영향 (Effects of Dietary Calcium and Sodium Levels on Lipid Metabolism in Hyperlipidemic/Hypercholesterolemic Rats)

  • The effects of dietary Ca and Na levels on lipid metabolism in hyper lipidemic/hypercholesterolemic rats were examined. In Expt. 1, normal rats were divided into six groups and fed high fat(15%, w/w)/cholesterol(1%, w/w) diet containing two levels of Na, low (0.05) or high(1.5%) and three levels of Ca, low(0.1%), normal (0.5%), or high(1.5%) for 8 weeks. In Expt. 2, hyperlipidemia / hypercholesterolemia rats were induced by feeding high fat / cholesterol diet for 4 weeks. They were divided into four groups and fed the high fat / cholesterol diet, containing two levels of Na, low or high and two levels of Ca, low or high for 4 weeks. In Expt. 1, total lipid and total cholesterol contents in serum and liver were significantly lower in rats fed high Ca diet than in rats fed normal or low Ca diet regardless of dietary Na levels. Serum TG was the highest in rats fed low Ca and low Na diet. In Expt. 2, Serum total lipid, TG, and total cholesterol levels decreased by 24, 35, 26% respectively in rats fed high Ca diet regardless of dietary Na levels. Serum total lipid level tended to increased in rats fed low Na diet. The total lipid and TG contents in liver slightly decreased in rats fed high Ca diet. Another observation was that high Ca intake significantly faciliated the fecal lipid and cholesterol excretion regardless of dietary Na levels. There results suggest that the hypolipidemidc/hypocholesterolemic effects of high Ca diet could be partly due to increase in lipid and cholesterol excretion and these effects may be independent of dietary Na levels.

식이 지방의 수준과 종류가 조기이유한 흰쥐의 체내 지질대사와 세포성장에 미치는 영향 (Effects of Dietary Fat Sources and Levels on Lipid Metabolism and Cellular Growth in Early Weaned Rats)

  • This study was performed to observe the effects of dietary fat levels and sources on lipids contents and cellularities of liver, brain, and adipose tissue of early weaned rats. Male Sprague-Dawley rats were prematurely weaned from postnatal 17th day with the experimental diets differ in fat levels : low(5%), medium(10%), high(20%) and fat sources : butter, soybean oil, butter+ soybean oil. On the postnatal 29th day, contents of total lipid, triglyceride, cholesterol and phospholipid of serum, liver, brain and adipose tissue were determined, and DNA was determined to assess the cell growth. Rats early weaned fed high fat diet showed lower total lipid and triglyceride levels in serum and liver than those fed medium or low fat diet Rats early weaned fed high fat diet had adipocytes of fewer number, but larger size than those of rats fed low or medium fat diets. Rats early weaned fed soybean oil diet had more adipocytes thu those fed butter diet. Rats normally weaned to commercial chow diet showed lower total lipid, triglyceride, and cholesterol levels in serum and liver, had fewer adipocytes than all early weaned rats except for rats fed high fat-butter diet. These results suggest that high fat-butter diet is ideal weaning diet at early weaning.

Gene Expression Profile in the Liver Tissue of High Fat Diet-Induced Obese Mice

  • The purpose of this study was to investigate the gene profiles that were up- or down-regulated in the livers of high-fat diet-induced obese mice and $db_-/db_-$ mice with deficient leptin receptor. C57/BL6 normal mice and $db_-/db_-$ mice, respectively, were divided into two groups and fed a standard or high-fat diet for four weeks. Liver weight was unchanged in the normal mice but the high-fat diet led to a 10% weight increase in the $db_-/db_-$mice. Adipose tissue mass increased by about 88% in the normal mice that were fed a high-fat diet and by about 17% in the $db_-/db_-$mice on the high-fat diet. In terms of serum lipids, total cholesterol significantly increased in mice on the high-fat diet. Microarray analysis was carried out using total RNA isolated from the livers of standard or high-fat diet-fed mice of the normal and $db_-/db_-$ strains. The change of gene expression was confirmed by RT-PCR. About 1.6% and 6.8% of total genes, respectively, showed different expression patterns in the normal mice fed the high-fat diet and $db_-/db_-$ mice. As a result of microarray, many genes involved in metabolism and signal pathways were shown to have different expression patterns. Expression of Mgst3 gene increased in the livers of normal and $db_-/db_-$ mice that were fed a high-fat diet. Wnt7b and Ptk9l were down-regulated in the livers of the normal mice and $db_-/db_-$ mice that were fed a high-fat diet. In conclusion, a high-fat diet induced obesity and affected gene expression involved in metabolism and signal pathway.
