The Effects of Temperature and Nutritional Conditions on Mycelium Growth of Two Oyster Mushrooms (Pleurotus ostreatus and Pleurotus cystidiosus)

  • The influences of temperature and nutritional conditions on the mycelium growth of oyster mushroom Pleurotus ostreatus (PO) and Pleurotus cystidiosus (PC) were investigated in laboratory experiment during the summer season of 2014. The results of the experiment indicated that potato dextrose agar (PDA) and yam dextrose agar (YDA) were the most suitable media for the mycelium growth of oyster mushroom PO while four media (PDA, YDA, sweet potato dextrose agar, and malt extract agar medium) were not significantly different in supporting mycelium growth of oyster mushroom PC. The optimal temperature for mycelium growth of both oyster mushroom species was obtained at $28^{\circ}C$. Mycelium growth of oyster mushroom PO was improved by carbon sources such as glucose, molasses, and at 1~5% sucrose concentration, mycelium colony diameter of mushroom PO was achieved the highest value. Whereas glucose, dextrose, and sucrose as carbon sources gave the good mycelium growth of oyster mushroom PC, and at 1~3% sucrose concentration, mycelium colony diameter of PC was achieved the maximum value. Ammonium chloride concentrations at 0.03~0.09% and 0.03~0.05% also gave the greatest values in mycelium colony diameter of mushroom PO and PC. Brown rice was found to be the most favourable for mycelium growth of two oyster mushroom species. In addition, sugarcane residue, acasia sawdust and corn cob were selected as favourable lignocellulosic substrate sources for mycelium growth of both oyster mushrooms.

A Study on the Classification and Prediction of the Chip Type under the Specified Cutting Conditions in Turning (선삭가공시 절삭조건에 의한 Chip형태의 분류와 예측에 관한 연구)

  • In recent years, the rapid development of the machine tool and tough insert has made metal removal rates increase, and automatic system without human supervision requires a higher degree reliability of machining process. Therefore the control of chips is one of the important topics which deserves much attention. The chip classification was made based upon standard deviation of the mean cutting force measured by a tool dynamometer. STS304was chosen as the workpiece which is known as the difficult-to-cut material and mainly saw-toothed chip produced, and the chip type according to the standard deviation of mean cutting force was classified into five categories in this experiment. Long continuous type chip which interrupts the normal cutting process, and damages the operator, tool and workpiece has low standard deviation value, while short broken type chip, which is favourable chip for disposal, has relatively large standard deviation value. In addition, we investigated the possibility that the chip type can be predicted analyzing the relationship between chip type and cutting condition by the trained neural network, and obtained favourable results by which the chip type can be predicted with cutting conditon before cutting process.

Interaction of Escherichia coli K1 and K5 with Acanthamoeba casfellanii Trophozoites and Cysts

  • The existence of symbiotic relationships between Acanthamoeba and a variety of bacteria is well-documented. However, the ability of Acanthamoeba interacting with host bacterial pathogens has gained particular attention. Here, to understand the interactions of Escherichia coli K1 and E. coli K5 strains with Acanthamoeba castellanii trophozoites and cysts, association assay, invasion assay, survival assay, and the measurement of bacterial numbers from cysts were performed, and nonpathogenic E. coli K12 was also applied. The association ratio of E. coli K1 with A. castellanii was 4.3 cfu per amoeba for 1 hr but E. coli K5 with A. castellanii was 1 cfu per amoeba for 1 hr. By invasion and survival assays, E. coli K5 was recovered less than E. coli K1 but still alive inside A. castellanii. E. coli K1 and K5 survived and multiplied intracellularly in A. castellanii. The survival assay was performed under a favourable condition for 22 hr and 43 hr with the encystment of A. castellanii. Under the favourable condition for the transformation of trophozoites into cysts, E. coli K5 multiplied significantly. Moreover, the pathogenic potential of E. coli K1 from A. castellanii cysts exhibited no changes as compared with E. coli K1 from A. castellanii trophozoites. E. coli K5 was multiplied in A. castellanii trophozoites and survived in A. castellanii cysts. Therefore, this study suggests that E. coli K5 can use A. castellanii as a reservoir host or a vector for the bacterial transmission.

An Experimental Study for Dryer (건조기 고안 제작에 관한 연구)

  • A newly devised dryer with heated air for the farm products, especially suited for high water content materials such as red pepper, Beer ground, each Vegetables, and Low water content materials such as Rough rice was tested for its thermal efficiency and drying mechanism, and the optimum conditions for each sample were established. In order to improve the present rural situation of drying farm products which entirely dependent upon natural solar radiation, a study upon an economic multi-parpose dryer was conducted. A series of drying tests were run first with red pepper which is one of the important cash crop in Korean farm. And successive series of tests were also run with such proaucts as garlic, sweet potatoes, green onion, radish, Beer ground and Rough rice. The results from the above experiment in drying system with heat dryer can be summarized as follows. 1. Drying duration could be shortened by the tempering effect in high water content crop such as red pepper and beer ground. 2. The color changes occured in around 20% water content in red pepper. The degree of color change was heavily affected by high temperature and short drying duration. 3. The drying condition of red pepper was most favourable at the temperature of 85$^{\circ}C$ in early stage and 80$^{\circ}C$ in middle stage and 75$^{\circ}C$ at the final stage, and with the air rate of 0.81㎥/sec and with sample amount of 200kg. 4. The drying condition of Rough rice(I.R.667) was most favourable at the templature of 40$^{\circ}C$ in early stage and 35$^{\circ}C$ in middle stage and final stage and with the air rate of 0.2㎥/sec and with sample amount of 75kg. 5. In order to prevent the color change of red pepper and to assure high efficiency in drying mechanism, it was necessary to lower the temperature as the time passes in drying process. 6. For vege tables, the drying rate were short in early stage and there was also tempering effect. However, for garlics, Constant drying rates through the early and final stages were observed and there were no tempering effects. 7.   • Dynamic behaviour of a package of a microwave oven under the drop-impact conditions is evaluated by the finite element analysis and tests. PAM-CRASH software is used for the finite element analysis and the tests are performed according to the ISTA(International Safe Transit Association) specification. Results of the analyses are compared with those of the tests and accuracy is shown to be favourable. Under the drop-impact condition of the original design, severe deformation occurs and an improved design is proposed to reduce it. The approach presented in this research can be successfully applied to reduce costs and time required to develop new models of the microwave oven.

Suitable Conditions for Mycelial Growth of Phellinus spp.

  • The fungus Phellinus is a mushroom that is widely used medicinally. The optimal conditions for mycelial growth of 13 strains of the fungus were investigated. Mycelial growth was optimal at 25$^{\circ}C$ and was uniformly minimal at 15$^{\circ}C$ and 35$^{\circ}C$. Growth was optimal at pH 6$\sim$7. The mycelial phenotype was best promoted by growth using Potato Dextrose agar, Hamada, Glucose peptone, and Yeast-Malt media, whereas Czapek Dox, Hennerberg, and Lilly media were the most unfavorable for the mycelial growth of Phellinus spp. Glucose, sucrose, fructose, and dextrin were the most suitable carbon sources for mycelial growth, while lactose, maltose, and galactose were unsuitable. Among tested nitrogen sources, ammonium phosphate, potassium nitrate, and arginine best promoted mycelial growth, while alanine, urea, and histidine least promoted mycelial growth.

  • Recently, WC-Co have some excellent properities as the material for the mechanical component such as metallic moulding parts, ball dies parts, and punch parts. This paper describes the surface roughness and grinding force caused by experimental study on the surface grinding of WC-Co with ultra-precision like a mirror shape using diamond wheel. Also, some investigations are carried out using WA grinding wheel to increase improved ground surface roughness such as polishing, lapping effect. Some important results obtained here are summarized as follow. 1) Within this experimental grinding condition, we can be obtained $R_{max}.\;2\mu\textrm{m}\;R_a\;0.3\mu\textrm{m}$ whichare the most favourable ground surface roughness using #140 diamond wheel, and improved surface roughness values about 20 .approx. 25% throughout 5 times sparkout grinding 2) The value of surface roughness is Rmax. $0.49\mu\textrm{m},\;R_a\;0.06\mu\textrm{m}$ using #600 diamond wheel. 3) The area of no rack zone is less than $F_{n}$ 0.27N/mm, Ft 0.009N/mm

Current Level of Reproductive Performances in Japanese Black Cows

  • A set of 313,169 reproduction records of Japanese Black cows calving between 1987 and 1996 were collected in nation widely Japan to investigate the current level of reproductive performances and the factors influencing the performances such as calving interval, first calving age, services per conception, calving difficulty, non-gestation period and gestation length. All the records of reproductive performances are outcome of artificial insemination. The means of first calving age, calving interval, non-gestation period and gestation length were 25.0 months, 389.9, 101.9 and 287.5 days, respectively. Services per conception were 1.41 and degree of calving difficulty was 1.07, which suggests most of the calving, did not need assistance. There were chronological tendencies that first calving age became younger while calving interval became longer. Differences in reproductive performance were found for seasons and calving numbers. Calving interval became shorter towards the fourth calving but became longer afterwards. The cows calving in May had the shortest calving interval followed by those in April and June and first calving age had a similar tendency. The cows with the standard body condition score showed more favourable reproductive performances compare with those with rather fatty or thin body condition scores.

Plywood Properties by PEG Treatment Conditions on Veneer (단판(單板)의 PEG 처리조건(處理條件)에 따른 합판(合板)의 성질(性質))

  • This study was carried out in order to investigate the treatment effect of PEG soln which is a common dimensional stabilizer to green log. sawing panel etc, on bonding product including plywood widely-used in secondary processing unit. The 30% concentration of aqueous PEG soln. with molecular weight of 400, 1.000 and 4,000 were prepared respectively, and also dipping the veneer in the PEG soln. spreading the PEG soln. on veneer and mixing the PEG soln. in the adhesive were allowed. Then the ratio of PEG impregnation on veneer, the adhesive strength of plywood were epitomized as follows: The ratio of impregnation by PEG 4,000 at dipping condition was highest. while that by PEG 400 at same condition was lowest. However, the effect of PEG molecular weight on the ratio of impregnation at spreading condition did not occur. 2. The adhesive strength was great in the order of 4,000>400>1,000 in molecular weight of PEG at dipping and spreading conditions. In case of mixing the PEG soln. in the adhesive, the adhesive strength was great in the order of 400>1,000>4,000 in molecular weight of PEG. Throughout three treatment conditions, PEG 400 was relatively favourable with about 10kg/$cm^2$ dry strength. 3. The adhesive strength was great 10 the order of spreading >dipping >mixing condition. 4. Although adhesive strength with the 30% concentration of aqueous PEG soln. was decreased by 35% and over, compared to control (non-treatment) adhesive strength, all types of PEG treatment except mixing the PEG soln. in the adhesive exceeded the standard dry strength for common use panel. 7.5kg/$cm^2$. 5. In warm water-proof test, the adhesive strengths by all PEG treatment conditions were less than the standard wet strength, 7.5kg cot, and also delamination of glue line occured mostly in mixing in the PEG soln. in the adhesive condition.

Extraction of Athabasca Oil Sand with Sub- and Supercritical Water (아임계 및 초임계수를 이용한 Athabasca 오일샌드의 추출)

  • Bitumen extraction and sulfur removal from Athabasca oil sand were conducted using water in sub- and supercritical condition. Bitumen yield in micro reactor was investigated in the pressure range of 15~30 MPa, the temperature of 360 and $380^{\circ}C$ and water density $0.074{\sim}0.61g/cm^3$ for 0~120 min. Bitumen yield increased with reaction pressure irrespective of temperature and dramatically increased in especially supercritical region due to hydrogen formed from water gas shift reaction. Total amount of gas product decreased with reaction pressure but the portion of sulfur and hydrogen increased a little with increasing pressure to 25 and 30 MPa. It is seen that supercritical condition was favourable to the hydrogen formation and sulfur removal. Bitumen yield and sulfur removal from original oil sand reached a maximum 22% and 40% respectively in supercritical condition(the reaction time of 60 min at $380^{\circ}C$ and 25 or 30 MPa).
