Study on a 4-Week Recovery Test of Sweet Bee Venom after a 13-Week, Repeated, Intramuscular Dose Toxicity Test in Sprague-Dawley Rats

  • Objectives: This study was performed to check for reversibility in the changes induced by a 13-week, repeated, dose toxicity test of Sweet Bee Venom (SBV) in Sprague-Dawley (SD) rats. Methods: Fifteen male and 15 female SD rats were treated with 0.28 mg/kg of SBV (high-dosage group) and the same numbers of male and female SD rats were treated with 0.2 mL/kg of normal saline (control group) for 13 weeks. We selected five male and five female SD rats from the high-dosage group and the same numbers of male and female SD rats from the control group, and we observed these rats for four weeks. We conducted body-weight measurements, ophthalmic examinations, urinalyses and hematology, biochemistry, histology tests. Results: (1) Hyperemia and movement disorder were observed in the 13-week, repeated, dose toxicity test, but these symptoms were not observed during the recovery period. (2) The rats in the high-dose group showed no significant changes in weight compared to the control group. (3) No significant differences in the ophthalmic parameters, urine analyses, complete blood cell counts (CBCs), and biochemistry were observed among the recovery groups. (4) No changes in organ weights were observed during the recovery period. (5) Histological examination of the thigh muscle indicated cell infiltration, inflammation, degeneration, necrosis of muscle fiber, and fibrosis during the treatment period, but these changes were not observed during the recovery period. The fatty liver change that was observed during the toxicity test was not observed during the recovery period. No other organ abnormalities were observed. Conclusion: The changes that occurred during the 13-week, repeated, dose toxicity test are reversible, and SBV can be safely used as a treatment modality.

The Effects of Supplementing Methionine plus Cystine to a Low-protein Diet on the Growth Performance and Fat Accumulation of Growing Broiler Chicks

  • This experiment was conducted to determine the effects of a low-protein diet supplemented with DL-methionine plus L-cystine (Met + Cys) on the growth performance and fat accumulation of female broiler chicks during the growing period (3-6 wks old). A low-protein diet (17% CP; 3,200 ME kcal/kg) was supplemented with Met + Cys (1.1 : 1.0) at levels 0.75, 0.94, 1.25, 1.31 or 1.50% of diet, respectively. Another diet with 21% CP and 3,200 ME kcal/kg served as the control group. All essential amino acids were adjusted to meet the National Research Council (1984) requirement for chicks. Feed and water were given ad libitum. Body weight of the chicks fed the low-CP diets supplemented with Met + Cys were heavier than those of the control birds. Feed conversion ratio and feed intakes were not significantly different between and among the treatment groups. Similary, abdominal fat content was not significantly different among the various treatments except that of the chicks fed the low CP diet with 1.25% Met + Cys which was higher than that of the control group. Fatty acid synthetase (FAS), acetyl-CoA carboxylase (ACC) activities and carcass protein content were not influenced by dietary treatments. Carcass fat content was lowest in chicks fed low CP diet with 0.75% Met + Cys and highest in the group that received 1.50% Met + Cys supplementation. Liver triglyceride increased as Met + Cys supplementation level increased. Various lipid fraction concentrations (cholesterol ester, free cholesterol, and phospholipid) in the serum went up as Met + Cys increased up to 1.25% after which it levelled off. Results of this experiment suggest that it is possible to reduce dietary protein level from 21% to 17% for growing broiler chicks by the supplementation of Met + Cys when other EAA were sufficient.

Medium- and long-chain triglyceride propofol reduces the activity of acetyl-coenzyme A carboxylase in hepatic lipid metabolism in HepG2 and Huh7 cells

  • Medium- and long-chain triglyceride (MCT/LCT) propofol is widely used as an intravenous anesthetic, especially in the intensive care unit. The present study aimed to assess whether MCT/LCT propofol is safe in the hyperlipidemic population for long-term use. Free fatty acids (FFAs) were used to establish high-fat stimulation of HepG2 and Huh7 cells. Subsequently, these cells were treated with propofol at the concentration of 0, 4, or 8 ㎍/ml for 24 and 48 h. The results indicated that the cell viability was notably decreased when the cells were stimulated with 2 mmol/L FFAs and treated with 12 ㎍/ml MCT/LCT propofol. Accordingly, we chose 2 mmol/L FFAs along with 4 and 8 ㎍/ml MCT/LCT propofol for the subsequent experiments. Four and 8 ㎍/ml MCT/LCT propofol inhibited FFA-induced lipid accumulation in the cells and significantly reversed acetyl coenzyme A carboxylase (ACC) activity. In addition, MCT/LCT propofol not only significantly promoted the phosphorylation of AMPK and ACC, but also reversed the FFA-induced decreased phosphorylation of AMPK and ACC. In conclusion, MCT/LCT propofol reverses the negative effects caused by FFAs in HepG2 and Huh7 cells, indicating that MCT/LCT propofol might positively regulate lipid metabolism.

Effects of Yijung-tang (YJT) on Experimental Hypothyroidism in Mice (이중탕(理中湯)이 생쥐에 유발된 갑상선기능저하증에 미치는 영향)

  • Objectives: The present study was carried out to investigate the effects of YJT on experimental hypothyroidism induced by sodium perchlorate and methimazolein in mice. Methods: 30 one-month-old C57BL6 mice were decided into 4 groups: 1) normal (n=6), 2) sodium perchlorate and methimazolein-induced hypothyroidism control (n=8), 3) hypothyroidism mice treated with low YJT (n=8), 4) hypothyroidism mice treated with high YJT (n=8). Sodium perchlorate and methimazolein were administered for 4 weeks, YJT (low and high) was administered for 2 weeks after sodium perchlorate and methimazolein were initiated for a total duration of 2 week. The changes were observed : weight of body, T3, T4, TSH, follicular cells in the thyroid tissues, LDL cholesterol, HDL cholesterol, triglyceride and free fatty acid, FBG, AST, ALT and so on. Results: YJT did not affect body weight gain. YJT restored free T4 level decreased by sodium perchlorate and methimazolein and prevented shrinking of follicles and proliferation of follicular cells in the thyroid tissues. In addition, YJT lowered total and LDL cholesterol levels elevated by sodium perchlorate and methimazolein respectively and ameliorated distribution of fat in liver tissues. In addition, the effect on fasting blood glucose (FBG), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were also investigated. Conclusions: These data suggest that YJT can be used to treat woman patients which are accompanied with hypothyroidism relatively safely.

Differential Expression of Metabolism-related Genes in Liver of Diabetic Obese Rats

  • The Otsuka Long-Evans Tokushima Fatty (OLETF) rat, a model of spontaneous type 2 diabetes (T2D), develops hyperglycemic obesity with hyperinsulinemia and insulin resistance after the age of 25 weeks, similar to patients with noninsulin-dependent diabetes mellitus (DM). In the present study, we determined whether there are differences in the pattern of gene expression related to glucose and lipid metabolism between OLETF rats and their control counterparts, Long-Evans Tokushima (LETO) rats. The experiment was done using 35-week-old OLETF and LETO rats. At week 35 male OLETF rats showed overt T2D and increases in blood glucose, plasma insulin, plasma triglycerides (TG) and plasma total cholesterol (TC). Livers of diabetic OLETF and LETO rats also showed differences in expression of mRNA for glucose and lipid metabolism related genes. Among glucose metabolism related genes, GAPDH mRNA was significantly higher and FBPase and G6Pase mRNA were significantly lower in OLETF rats. For lipid metabolism related genes, HMGCR, SCD1 and HL mRNA were substantially higher in OLETF rats. These results indicate that gluconeogenesis in OLETF rats is lower and glycolysis is higher, which means that glucose metabolism might be compensated for by a lowering of the blood glucose level. However, lipid synthesis is increased in OLETF rats so diabetes may be aggravated. These differences between OLETF and LETO rats suggest mechanisms that could be targeted during the development of therapeutic agents for diabetes.

Effectiveness of Worksite Nutrition Counseling for Hyperlipidemic Employees in Kyung - buk Area (영양교육을 통한 사업장 근로자의 고지혈증 개선)

  • This study was aimed to implement and evaluate worksite nutrition counseling for industrial workers with hyperlipidemia in Kyung-buk area. Forty-six subjects consisted of 41 men and 5 women who were diagnosed hyperlipidemia though health examination had their average age, 40.3$\pm$ 6.0 and half of them were office workers and one third labor workers. Twenty-eight subjects had fatty liver with or without other complications and six had gastric problems. The subjects were interviewed using questionnaires on food habit, food frequency, smoking, drinking and exercise, and they were given nutrition counseling which was continued for 7 months. Desirable food selections were practiced using foods or food models and various written materials individually or as group during lunch times and breaks. After counseling, smoking and drinking were reduced and exercise and food habits were improved. Average levels of initial serum total cholesterol(T-Chol), HDL-cholesterol(HDL-Chol) and triglyceride(TG) were 214$\pm$ 44, 45.07$\pm$ 7.14 and 281$\pm$ 13mg/dl respectively and were not changed significantly except 2.30$\pm$ 7.04mg/dl increase of HDL-Chol(p<0.1) after nutrition counseling. However, subjects who improved their food habits and smoking, drinking and exercise habits had higher tendency to have reduced serum T-Chol and TG levels compared with those who drank less. Food habit score was negatively correlated with serum TG levels(r=-0.378, p<0.01). It is concluded that an efficient and regular nutrition counseling by dietitian at worksite is beneficial for health of industrial workers.

The Effect of Breakfast Regularity on Eating Habits, nutritional and Health Status in Adults (아침식사의 규칙성이 중년 남녀의 식습관 및 영양상태에 미치는 영향)

  • This study was performed to investigate the effect of breakfast regularity on eating habits, nutritional and health status in adult men and women. Also, eating habites and nutritional status expressed as percentage of recommended energy intake in IB(Irregular Breakfast group) were studied. Proportions of IB in men and women was 23.7% and 31.1%, respectively. In men, IB had irregular mealtime, overeating tendency. Also, they consumed high-fat meat and alcohol more frequently than RB (Regular Breakfast group). Total energy intake showed no significant difference between which two groups. Energy intakes from dinner and alcohol were higher in IB than in RB. Nutrient and food intakes were lower in IB than in RB. Percent body fat of IB was higher than that of RB. Weight, triglyceride and prevalence of fatty liver were higher in IB than in RB. In women, IB had irregular mealtime, and overeating tendency. Also, they had more frequent eating out and alcohol consumption. Total energy intake was lower in IB than in RB. In men, nutrient and food intakes were lower in IB than in RB. But health status showed no significant differences between two groups. In men, energy intakes from alchol and dinner were higher in +RDA group(percentage to recommended energy > 125%) than in other groups of IB. But energy intake from carbohydrate was lower in +RDA group. -RDA group (percentage to recommended energy < 75%) had Ca and vitamin A intake less than RDA. In women, +RDA group had overeating tendency. Also, they had more frequent eating out, high-fat meat, and alcohol consumption than other groups of IB. Energy intakes from dinner, snack and fat were higher in +RDA group. But -RDA group had protein and vitamin A intake less than RDA. From our study, it seemed that breakfast irregularity led to undesirable eating habits and had relation with increase of percent body fat. Breakfast regularity seemed to be very important to maintain a nutritional balance.

Expression of AGR-2 in Chicken Oviduct during Laying Period

  • The chicken oviduct is a dynamic organ that produces secretory proteins such as ovalbumin during the laying period. In this study, we identified oviduct-specific proteins in hens during the egg-laying period by proteomic analysis. Proteins extracted from the magnum of hens of different ages (5, 35, and 65 weeks) were analyzed by two-dimensional gel electrophoresis to compare the intensity of proteins among samples. Approximately 300 spots were detected on each gel. Based on the comparison of image gels, we found that the intensity of eight spots in 35-week magnums was increased at least by 2-fold compared with the others. Five of the eight spots were identified as calumenin, acidic ribosomal phosphoproteins (ARP), prohibitin, heart fatty acid-binding protein, and anterior gradient-2 (AGR-2). In particular, ARP and AGR-2 were highly expressed in 35- week magnums compared with 5- and 65-week magnums. In addition, the level of these proteins was consistent with their RNA levels. Expression of AGR-2 mRNA was detected in the mature magnum, whereas no signal was observed in premature tissue. Among various tissues, expression of AGR-2 mRNA was highest in the magnum, high in the isthmus, and five fold lower in muscle. It was undetectable in the liver and in other tissues (heart and kidney). However, the mRNA levels of other proteins were ubiquitous among tissues. In transcriptional activity of AGR-2, a 3.0 kb fragment of promoter region containing potential estrogen receptor binding sites had enhanced its activity strongly. In conclusion, these results suggest that AGR-2 has functional regulatory roles in the chicken oviduct during the egglaying period.

Effects of the Fermented Black Garlic Extract on Lipid Metabolism and Hepatoprotection in Mice (발효흑마늘 추출물이 흰쥐의 지질대사 및 간기능 개선에 미치는 영향)

  • This study was conducted to evaluate the functionality of fermented black garlic extracts under various conditions. Black garlic powder was prepared by aging for 0~72 hours at 80℃ depending on relative humidity (RH). It showed the highest antioxidant effects among the samples; the total antioxidant activity of black garlic powders at RH 75%, 84%, and 90% for 72 hours was increased 31.9 times, 28.2 times, and 22.6 times compared with that of the fresh garlic powder, respectively. Also, the alliin content was gradually decreased. S-ally-L-cysteine and S-ethyl-cysteine levels were increased; the highest values were 495.9 ㎍/g and 1,769.7 ㎍/g after aging for 72 hours at RH 75%. Aspartate transaminase (AST) and alanine transaminase (ALT) levels were increased following high fat diet feeding, but the rise was obviously reduced by administration of black garlic extract. The total cholesterol, LDL/VLDL-cholesterol, and triglyceride contents in serum were significantly lower in methionine and choline deficient (MCD) diet treatment groups than in the positive control group. The concentration was increased following the intake of black garlic and fermented black garlic extracts. Therefore, black garlic extracts could be an ideal material as a dietary supplement in healthy functional foods to improve the effects on fatty liver.

Effect of Gymnema sylvestre Extract on Body Weight, Adiposity, and Lipid Metabolism in ob/ob Mice (ob/ob 마우스에서 짐네마 추출물이 체중, 체지방 형성 및 지질대사에 미치는 영향)

  • This study investigated the effects of Gymnema sylvestre (GS) extract on body weight, adiposity, and lipid metabolism in leptin-deficient ob/ob mice. The experimental mice were divided into the following 4 groups: basal diet (AIN-93G diet) and 0.5%, 1%, and 1.5% GS supplemented groups. Each group was fed the experimental diet for 9 weeks. The final body weights, adipocyte sizes, and epididymal fat weights of the GS groups were significantly lower than those of the control group. There were no significant differences in food intake and food efficiency ratios among the treated groups. Serum triglyceride levels and the atherogenic index were significantly lower in the GS groups compared to the control group. Serum HDL cholesterol levels were significantly higher in the 1% and 1.5% GS groups compared to the control group. Serum glutamate oxaloacetate transaminase and glutamate pyruvate transaminase activities did not differ among the four experimental groups. The total hepatic lipid content was significantly lower in the GS groups, and hepatic cholesterol and triglyceride contents tended to be reduced in the GS groups compared to the control group. These results suggest that GS extract may be useful for ameliorating dyslipidemia and fatty liver.
