Effect of the Dietary Protein Level on Plasma Glucose, Lipids and Hormones in Streptozotocin-Diabetic Rats

  • Atherosclerotic vascular disease is a major cause of the increased morbidity and mortality assciated with diabetes mellitus. The prominent role of nutrition in hypercholesteolemia and atherosclerosis is generally accepted. Diet is a key element in the management of diabetes (type I-IDDM), yet the appropriate diet for patient with diabetes mellitus is not well known. Dietary protein has been shown to have a significant effect on plasma cholesterol levels in both experimental animals and humans. The present experiment was designed to determine the effect of the dietary protein level(20% vs 60%) on plasma glucose concentration, lipids profile, insulin and glucagon levels from non-diabetic and streptozotocin-induced diabetic rats. Results showed that a high protein diet decreased triglyceride concentration in diabetic rats. Also diabetic rats fed a high protein diet were hypocholesterolemic than rats fed a control diet. There were no effects by level of protein on fasting blood glucose concentration and insulin/glucagon ratio. Results from the present study suggest that a high protein diet may be beneficial to control pasma lipids in chemically-induced diabetic rats.

A Study on the Glucose Level Control and Chronic Complication Rate in Diabetic Compliance Groups with Health Care Regimen (당뇨병 환자의 치료지시 이행군과 비이행군의 혈당과 만성 합병증 발생비교)

  • The purpose of this study is to examine to control of glucose level and the occurrence of chronic complications of diabetes by compliance groups with health care regimen The subjects were consisted of 300 out patients with type 2 diabetes mellitus from beginning of March through the end of April in 2001, who visited at the endocrinology department at Kangnam St. Mary's Hospital of Catholic University in Seoul. The patient's compliance level with health care regimen was assessed at questionnaire. However, the blood glucose level and the occurrence of chronic complications of diabetes were assessed at medical record review. The data were analyzed by SAS program for chi square test and t-test. The results were as follows. 1. Significant differences were found among the factors such as duration of diabetes, the number of participation of diabetes educational program, job, smoking, self monitoring of blood glucose and the methods of diabetes therapy between high and low compliance groups. High compliance group patients had a diabetes longer than low compliance group patients. High compliance group patients more frequently attended the educational program and checked themselves monitoring blood glucose than low compliance group patients. Also, they did not work recently, smoked less and got more insulin injection therapy compared to low compliance group patients. 2. No significant differences were found among the result of fasting blood glucose. 2-hour postprandial blood glucose, and $HbA_{l}c}$ between high and low compliance groups. 3. The occurrence rate of macrovascular complications of chronic complications of diabetes were lower, however, the occurrence rate of microvascular complications were higher in high compliance group than in low compliance group with health care regimen.

Effect of Fermented Soy Bean Extract Containing Herbal Medicines (Godjang) on Blood Glucose Levels and Histomorphology in Streptozotocin-induced Diabetic Rat (한약재 함유 대두발효 추출물이 streptozotocin으로 유발된 당뇨 흰쥐의 혈당 및 조직형태학적 변화에 미치는 영향)

  • Objective : This study was designed to investigate anti-diabetic effects of fermented soy bean extract with herbal medicines (Godjang) in diabetic rat models induced by streptozotocin (STZ) injection. Method : Changes in body weight, drinking water, and food intake were observed for 4 weeks before and after induction of diabetes mellitus in rats. The anti-diabetic capacity of Godjang was analyzed by fasting blood glucose (FBG) every week. Also, after 4 weeks of administration, the oral glucose tolerance test (OGTT) was performed, and then blood levels of insulin were checked. And serum levels of total cholesterol and triglycerides were determined. Histomorphological changes of liver, kidney and pancreatic tissues were also observed in STZ-induced diabetic rats and Godjang administered rats. Result : In Godjang administered group, body weight and water intake were more lower than that of STZ-induced diabetic rats. FBG was decreased in the Godjang administered group than STZ-induced diabetic group. According to OGTT, blood glucose levels at 30 minutes and 60 minutes significantly decreased in Godjang administered group than in STZ-induced diabetic control group. Administration of Godjang extract for 4W significantly decreased levels of serum glucose, total cholesterol (TC), triglycerides (TG) in diabetic rats. In histomorphological analysis of kidney, liver, Godjang administrated groups showed the inhibition of pathological damage. Conclusion : These results suggest that Godjang extract has an anti-diabetic action through decrease in serum glucose, TC, TG levels and recovery of the morphological changes in kidney and liver in STZ-induced diabetic rats.

The Effects of Dietary Patterns and Apolipoprotein E Phenotype on the Blood Lipid Profiles of Individuals from Cheju Area (제주지역 성인의 Apolipoprotein E Phenotype 분포와 식생활 및 혈청지질 농도의 관련인자 연구)

  • The purpose of this study was to determine the relation between serum lipid profiles, apolipoprotein E phenotype, and dietary patterns in a cross-section of healthy individuals from Cheju-Do. Age, gender, anthropometric measurements, blood pressure, dietary consumption, drinking / smoking habits and menopausal status were surveyed. Total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, fasting blood glucose, and insulin levels were measured from overnight fasting blood. The study involved a total of 286 individuals(147 men and 139 women) between the ages of 20 and 60 years old. All of the subjects were recruited from a population of healthy individuals living in Cheju-Do. The results of the study are as follows : 1) Among the males, those in their 20's had the maximum food intake, while those in their 40's had the minimum food intake. For the females, food intake was the highest for those in their 30's. Energy and nutrient intakes were directly proportional to the amount of food intake. Men in their 30's were heavier than other men and women in their 40's were heavier than other women. The activity index for men in their 20's and 30's appeared to be lower than that of men above 40. The activity index of women in their 20's appeared to be lowest among all aged groups, and the index appeared to increase from the age of 30 onwards. 2) In terms of changes In serum constituents with age, men in their 40's appeared to have the highest levels of serum constituents such as lipids, glucose, and insulin. Men in their 50's showed the highest levels of serum LDL-cholesterol and glucose. Men in their 30's showed peak levels of serum triglycerides. On the other hand, women in their 50's appeared to have peak levels of serum total cholesterol, LDL-cholesterol, and triglycerides. There was no ch:ange with age in HDL-cholesterol and insulin levels for men and women. The percentage of the subjects had the following apo E phenotypes : E3/3, 91.3% ; E3/2, 5.4% ; E4/3, 2.5% ; E4/2, 0.7%. Lee's reserch with Korean female college students showed that the percentage of ApoE3/3, E3/2, E 4/2, E4/3, and E4/4 were 84.8%, 6.7%, 6.7%, 0.9%, 0.9%, respectively. The number of samples with ApoE mutation was so small that there was no statistical significance in the relation between apolipoprotein E phenotype and se겨m lipids. 3) To investigate the relati onship between weight and serum constituents, the subjects of this study were divided into three groups by BMI underweight, normal weight, and overweight. The serum constituents of men and women below the age 40 in the overweight groups belonged to the normal domain. On the other hand, serum cholesterol levels of both men and women above the age 40 in the overweight groups remained in the borderline-high region(above 200mg/dl), and the mean value of LDL-cholesterol(above 130mg/dl) and triglycerides of men were above normal. Fasting blood glucose levels also remained in the borderline-high region. Total cholesterol levels of women above the age 40 in the overweight group was in the borderline-high region. Effects of Low Glycemic Index Nutrition Education on the Blood Glucose Control in Patients with Type 2 Diabetes Mellitus (제 2형 당뇨 환자의 저혈당지수 영양교육이 혈당관리에 미치는 영향)

  • This study was intended to assess the effects of low glycemic index (LGI) nutrition education on dietary management and glycemic control of patients with type 2 diabetes mellitus. The subjects were 48 sex-matched patients with type 2 diabetes mellitus, aged $66.5\;{\pm}\;6.2$ years, visiting a public health center. They were divided into two groups: the control group (males 10, females 14) and the educated group (males 10, females 14). The educated group was provided with a LGI nutrition education program for 7 weeks. The control group was educated only one time for general diabetic education. Anthropometric indices, knowledge and perception of efficacy of low glycemic index carbohydrates, dietary glycemic index (DGI) and glycemic load (DGL), fasting blood glucose, and HbA1c were assessed. In the educated group body weight, body mass index and systolic blood pressure (from $138.0\;{\pm}\;18.9\;mmHg$ to $130.6\;{\pm}\;15.0\;mmHg$) were significantly reduced after the nutrition education (p < 0.05). The scores of knowledge and perception of efficacy of low glycemic index carbohydrates increased significantly in the educated group. Dietary glycemic index and glycemic load of the educated group decreased significantly from $103.4\;{\pm}\;67.6$ to $45.4\;{\pm}\;27.1$ (p < 0.001), and from $173.3\;{\pm}\;135.9$ to $66.8\;{\pm}\;50.4$ (p < 0.001), respectively. Also fasting blood glucose and HbA1c levels of the educated group significantly decreased from $124.5\;{\pm}\;28.8\;mg/dL$ to $96.7\;{\pm}\;21.6\;mg/dL$ (p < 0.001) and from $7.1\;{\pm}\;1.3%$ to $6.4\;{\pm}\;1.2%$ (p < 0.05), respectively. The score of knowledge and perception of efficacy of low glycemic index significantly correlated with fasting blood glucose and HbA1c levels negatively. DGI, DGL and duration of diabetes significantly correlated with HbA1c level positively. From stepwise multiple linear regression analysis, DGI, DGL and the duration of diabetes were extracted as factors influencing HbA1c level of the subjects. The results of this study suggest that low glycemic index nutrition education programs is an effective intervention measure for the glycemic control in type 2 diabetic patients.

Hypoglycemic activity of diospyros peregrina fruits in diabetic rats

  • Diospyros peregrina Gurke. (Ebenaceae) is a small middle sized tree grows luxuriantly in the plains of costal West Bengal, India. The objective of the study was to explore the antidiabetic activity of methanol extract of matured fruits of Diospyros peregrina to substantiate the folklore claim of traditional practitioners. It was also aimed to establish correlation with reduction of oxidative state associated with diabetes. Methanol extract of matured fruits of Diospyros peregrina was administered orally at doses of 150 and 300 mg/kg body weight for 12 consecutive days to normal and streptozotocin induced diabetic rats. Fasting blood glucose level was estimated in both normal and diabetic rats while serum lipid profiles, liver glycogen level and pancreatic thiobarbituric acid reactive substances (TBARS) were evaluated for diabetic rats. Initial and final changes in body weight were also recorded. Oral glucose tolerance test was performed during the course of study. Experimental findings showed significant antidiabetic potential of extract in term of reduction of fasting blood glucose level of both normal and diabetic rats. It was found that extract at the dose of 300 mg/kg body weight is more effective and percentage reduction (55.64) of elevated blood glucose level is comparable to that of standard drug glibenclamide (60.60) at a dose of 10 mg/kg body weight. Observed data found statistically significant in reduction of serum lipid and pancreatic TBARS levels whilst improvement was observed in liver glycogen level and body weight profiles in extract treated diabetic rats.

Past and Present Obesity, Parameters of Body Fat and Blood Pressure in Korean Patients with Non-Insulin Dependent Diabetes Mellitus(NIDDM) (한국 인슐린 비의존형 당뇨병 환자의 체중 변화, 체지방, 혈압 및 C-peptide/혈당 비)

  • To determine the frequency of past and present obesity among patients with NIDDM and to identify the differences of body fat, blood pressure and C-peptide/glucose ratio according to obese diabetic patients (BMI$\geq$25 kg/$m^2$) and nonobese (BMI<25 kg/$m^2$). Also the final factor is to observe the anthrometric change patterns in the study. Method: The weight at 20 years-old, previous maximal body weight, and acute weight loss were queried. Current height, body weight, BMI, waist & hip circumferences, waist-hip ratio, skinfold thicknesses, blood pressure, fasting blood glucose, and fasting C-peptide were measured in one hundred sixty-seven NIDDM patients. The differences of the parameters ccording to obese and nonobese, and three anthropometric change patterns were analyzed. Result: Results were as follows: 1. 66.5 % of the NIDDM patients had a history of past obesity as assessed by their maximum weight, while only 33.2% of them were currently obese (p's < 0.001). 2. The waist & hip circumferences, skinfold thicknesses, systolic, diastolic & mean arterial blood pressure in obese patients were greater than those of nonobese patients (all p's < 0.001). 3. The waist and the hip circumferences, and skinfold thicknesses (subscapula & triceps) were highest among the obese-obese group. WHR and abdominal skinfold thickness in the obese-obese and obese-nonobese groups were higher than those in the nonobese- nonobese group. Systolic & diastolic and mean arterial blood pressures in the obese-obese group were higher than those of obese-nonobese and nonobese-nonobese groups(all p's < 0.005). 4. The abdominal and subscapular skinfold thicknesses in female diabetic patients were greater than those of male patients (all p's <0.0001). Conclusion: Although most Korean NIDDM patients were previously obese, many of them were not obese during the course of the study. Greater central and upper body adiposicity and higher blood pressure was shown in obese diabetic patients. Also, greater central and upper body adiposicity was demonstrated in female diabetic patients.

The Risk Factors Associated with Increased Body Mass Index in Heavy Industry Workers (중공업근로자의 체질량지수 상승요인)

  • This study was performed to determine the risk factors associated with increased body mass index (BMI) in 672 heavy industry men workers. Subjects were examined in March, 2010 to September, 2010 in Gyeongnam province. Height, weight, systolic blood pressure, and diastolic blood pressure were measured. And fasting blood glucose, total cholesterol, HDL-cholesterol, LDL-cholesterol, and triglyceride levels were measured by Olympus AU 680, and their BMIs calculated. Age, smoking statue, and alcohol drinking of the workers were surveyed by questionnairs. The mean total cholesterol levels were $190.06{\pm}36.62mg/dL$, HDL-cholesterol $53.65{\pm}11.92mg/dL$, LDL-cholesterol $104.28{\pm}30.31mg/dL$, triglyceride $151.11{\pm}99.53mg/dL$, and fasting blood glucose $96.92{\pm}19.53mg/dL$. Mean systolic blood pressure was $126.08{\pm}13.78mmHg$, and diastolic blood pressure $71.88{\pm}10.45mmHg$. Subjects were categorized into two BMI groups, BMI ${\geq}23$ and BMI ${\leq}22.9$. The subjects with BMI of 23 or above had significantly higher levels of LDL-cholesterol, systolic blood pressure, and triglyceride, with lower levels of HDL-cholesterol. BMI showed the highest level at the age of thirties, increasing with age. BMI of smokers was higher than BMI of non-smokers, not showing a link between alcoholics and non alcoholics. In conclusion, age, systolic blood pressure, and HDL-cholesterol are mostly relevant to the increase of BMI in this study.

The Effects of Dietary Fat on Survival Metabolism of Fasting Rat (단식중인 흰쥐에서 지방섭취가 생존 대사에 미치는 영향)

  • To investigate the effects of different type of dietary fat on survival metabolism of fasting rats, one group rats (FO) were fasted, another one group rats (BM) were fed normal diet and the others were fed only one of the following fat diets: beef tallow (FT), corn oil (FC), and perilla oil (FP) of 11.4g/kg respectively. Most FO group rats survived for 6 days and large part of the only-fat-diet groups rats survived for 16 days. Body weights of all rats in fasting and only-fat-groups, measured just one day prior to death owing to fasting or caloric malnutrition, decreased by 24.5%~25% only-fat to fasting rat somewhat extended the survival time but the specific properties of dietary fat types had no remarkably differential effect on survival time and body weight gain rate. The features of liver and kidney weight gain rate of all rats in fasting and only-fat-diet groups were similar to those of body weight gain rate. In FO groups blood levels of total-cholesterol, triglyceride, and glucose markedly reduced whereas GPT activities and BUN levels considerably increased as compared to BM group. However the types of dietary fat perse did not affect blood total cholesterol, triglyceride, glucose, BUN levels, and GPT activities in early stage of fasting in FC and FP group. GPT activities in rats of FP group just prior to death of starvation seemed to be affected by the dietary fat types. The results showed that only-fat-feeding to fasting rats somewhat extended survival time but the types of dietary fat had no remarkably differential effect on survival time and metabolism of fasting rats.

The Studies on Glycogenolytic Adrenergic Receptor in Rat (흰쥐에서의 해당작용(解糖作用) Adrenergic Receptor에 대(對)한 검토(檢討))

  • To evaluate the adrenergic receptors for glycogenolytic responses to catecholamine, the blood glucose level, liver glycogen content and muscle glycogen level in rats were studied with treatment of epinephrine, norepinephrine and isoproterenol. In addition, to study the possibility of interconversion of adrenergic receptors, the effects of catecholamines in feeding animal were compared with those in fasting animal. The results are summarized as follow; 1) Epinephrine and norepinephine showed dose dependent increase of blood glucose level but the effect of isproterenol was not significant. 2) The prandial states of animal did not influence on effects of catecholamines on blood glucose level. 3) Liver glycogen contents were lowered by epinephrine or by norepinephrins but effect of isoproterenol was not significant. 4) Glycogen content of skeletal muscle was significantly lowered by isoproterenol and. epinephrine shifted the dose-response curve to right, but the effect of norepinephrine was not significant. 5) The effects of epinephrine and norepinephrine on blood glucose were significantly blocked by ergotamine but not by propranolol. These results indicate that the glycogenolytic response to adrenergic agents in rat is mediated by an alpha-receptor in liver and by a beta-receptor in skeletal muscle.
