굴 패각 구연산칼슘이 Papain으로 유도된 골관절염 C57BL/6J Mice에 미치는 영향 (Effects of Calcium Citrate Isolated from Oyster Shell on Papain-induced Osteoarthritis in C57BL/6J Mice)

  • 본 연구에서는 구연산칼슘 투여가 papain으로 유도된 골관절염 동물모델에 미치는 영향을 살펴보았다. 8주령의 C57 BL/6J 마우스의 우측 슬관절강에 papain $6{\mu}L$를 투여하여 골관절염을 유발하였다. 실험군은 각 군당 8마리씩 체중이 고르게 배정하여 총 5군으로 나누었다. 정상군은 우측 슬관절강에 생리식염수 $6{\mu}L$를 주사하였고, 대조군, 약물투여군, 구연산칼슘 투여군은 papain $6{\mu}L$로 골관절염을 유발하였다. 정상군과 대조군은 0.2 mL의 생리식염수를, 약물투여군은 diclofenac sodium 2 mg/kg/bw를, 구연산칼슘 투여군은 150, 300 mg/kg/bw를 28일간 1일 매일 정해진 시간에 1회 경구 투여하였다. 실험기간 동안 체중을 측정하였으며, 실험 종료일에는 부검하여 슬관절의 병리조직학적 관찰을 하였고 관절연골 내 proteoglycan 함유율, 골관절염 지수의 변화, 혈액 내 TNF-${\alpha}$, IL-$1{\beta}$, IL-6 함량을 측정하였다. 체중은 실험기간 동안 모든 개체 간의 유의적인 차이는 없었다. 혈액 내 TNF-${\alpha}$, IL-$1{\beta}$, IL-6 함량은 papain 투여 대조군이 정상군에 비해 유의적으로 증가하였으며 약물 및 구연산칼슘 투여군은 대조군보다 유의성 있게 낮았다. 관절연골 내 proteoglycan의 함유율은 약물 및 구연산칼슘 투여군이 papain 투여 대조군보다 유의성 있게 높았다. Hematoxylin과 eosin 염색 관찰 결과, 골관절염이 유발된 마우스에 구연산칼슘을 경구 투여하였더니 관절연골의 파괴와 골 침식 등 연골의 변성이 약물 투여군과 유사하게 나타났다. 이와 같은 결과는 구연산칼슘이 papain으로 유도된 골관절염에서 연골의 변성을 줄이고 염증을 억제함으로써 골관절염 치료에 효과가 있은 것으로 사료되며, 추후 더 자세한 연구가 필요하다.

  • 연구에서는 굴 패각 추출물이 papain으로 유도된 골관절염 동물 모델에 미치는 영향을 살펴보았다. 8주령의 C57BL/6J 마우스 우측 슬관절강에 papain $6{\mu}L$를 투여하여 골관절염을 유발하였다. 실험군은 각 군당 체중이 고르게 8마리씩 배정하여 총 다섯군으로 분류하였다. 정상군은 우측 슬관절강에 생리식염수 $6{\mu}L$ 주사하고 20일간 1일 1회 생리식염수 0.2 mL씩 경구투여 하였다. 대조군, 약물투여군, 굴 패각 추출물 투여군은 골관절염을 유발 후 각각 생리식염수, diclofenac sodium 2 mg/kg/bw, OSE 100, 200 mg/kg/bw를 20일 동안 매일 1회 정해진 시간에 경구투여 하였다. 실험기간 동안 체중을 측정하였으며, 실험 20일에 부검하여 슬관절의 병리조직학적 관찰, 관절연골 내 proteoglycan 함유율, 골관절염 지수의 변화, 혈액내 TNF-${\alpha}$, IL-$1{\beta}$, IL-6 함량을 측정하였다. 체중은 실험기간 동안 모든 개체 간의 유의적 차이가 없었다. 관절연골 내 proteoglycan 함유율은 DS 및 OSE 투여군이 papain 투여 대조군보다 유의성(P<0.05)있게 높았으며, 특히 OSE-200 투여군은 약물 투여군과 유사한 proteoglycan 함유율을 나타냈다. H&E 염색을 통하여 관찰한 결과, 골관절염이 유발된 마우스에 굴 패각추출물을 경구 투여하였을 때 관절연골의 파괴와 골 침식 등 연골의 변성이 약물 처리군과 유사한 것을 보아 굴 패각 추출물이 골관절염 치료에 효과가 있는 것을 확인할 수 있었다. 병리조직학적으로 골관절염 지수는 DS 및 OSE 투여군이 papain 투여 대조군보다 유의성(P<0.05) 있게 낮았으며, OSE-200 투여군이 OSE-100 투여군보다 낮았다. 혈액 내 TNF-${\alpha}$, IL-$1{\beta}$, IL-6 함량은 papain 투여 대조군이 정상군에 비해 유의적(P<0.05)으로 증가하였으며, DS 및 OSE 투여군은 대조군보다 유의성(P<0.05) 있게 낮았다. 특히 TNF-${\alpha}$ 및 IL-6 함량은 OSE-200 투여군이 약물 투여군과 유사한 감소 효과를 나타내었다. 이와 같은 결과는 굴 패각 추출물이 papain으로 유도된 골관절염에서 연골의 변성을 줄이고 염증을 억제함으로써 골관절염 치료에 효과가 있는 것으로 사료된다. 굴 패각은 탄산칼슘이 풍부하기 때문에 골관절염 개선에 효과가 있는 것으로 사료되나, 굴 패각 추출물 중 어떠한 기능성 물질이 골관절염 개선 효과를 나타내었는지에 대해 향후 심도 있는 연구가 필요하다.

어린이집 아동의 영양교육을 위한 사전조사 연구 - 식습관과 영양지식을 중심으로 - (A Preliminary Study on Nutrition Education for Preschool Children in Day-Care Center - Dietary Habit and Nutrition Knowledge -)

  • 본 연구에서는 어린이집에 다니는 미취학 아동의 식습관 및 영양지식상태를 파악하여 보건소의 영양 및 건강증진사업을 위한 영양교육 자료개발에 유용한 기초자료로 제공하고자 울산시 북구에 소재한 어린이집에 다니는 아동 1,200명을 대상으로 조사한 결과는 다음과 같다. 조사대상자의 성별은 남아 429명(52.6%), 여아 386명(47.4%)이었으며, 연령은 $3{\sim}7$세로 4세 240명(29.4%), 5세 294명(36.1%), 6세 163명(20.0%)으로 대부분을 차지하였고 3세와 7세는 각각 14.0%와 0.5%로 적었다. 부모의 직업으로는 지역의 특성상 생산직 근로자(38.7%)와 사무직(33.9%)이 높게 나타났고, 교육 정도에서 아버지는 전문대 졸업(51.6%)이 가장 많았고, 어머니는 고등학교 졸업이 58.0%로 가장 많았다. 월평균 소득은 $200{\sim}300$만원 미만이 42.0%로 가장 많이 나타났으며, 다음이 $100{\sim}200$만원 미만으로 20.6%로 나타났다. 조사대상 아동의 평균 신장과 체중은 108.0cm와 18.7kg으로 한국소아협회의 연령별 체위기준인 111.0cm, 19.0kg보다 약간 낮게 나타났다. PIBW에 근거한 저체중은 14.6%, 정상체중은 54.4%, 과체중 및 비만은 31.0%로 나타났다. 아동들의 일상적인 식습관을 나타내는 식습관점수는 전체 평균 22.0점(30점 만점)으로 남아 22.0점, 여아 21.9점으로 거의 비슷하게 나타났으며, 항목별 점수에서 가장 높은 점수는 '아침을 매일 규칙적으로 먹는다'였고, 가장 낮은 점수는 '당근, 시금치등 녹황색 채소의 섭취'로 나타났다. 식행동에서는 전체 아동의 69.1%가 편식을 하였으며, 편식식품으로는 채소류가 51.2%, 다음이 육류 및 생선류로 나타났다. 간식섭취 실태에서는 피자나 햄버거 등 패스트푸드의 섭취빈도가 가장 많았고 다음이 탄산음료, 튀김류의 순으로 나타났다. 영양지식점수는 교육실시 이전의 전체 평균은 7.7점(10점 만점)으로 남아가 여아보다 약간 높은 점수를 보였고 영양교육을 4주간 실시 후에는 전체 평균점수가 8.9점으로 향상되었으며 역시 남아가 여아보다 높게 나타났다(p<0.001). 이상의 연구결과를 종합해볼 때 미취학 아동들은 저체중과 과체중 및 비만의 문제를 동시에 가지고 있었고, 식습관에서 편식이 많았으며, 간식의 섭취비율이 클 뿐만 아니라 종류에서도 인스턴트 및 패스트푸드 식품을 선호하는 것으로 나타났다. 그러므로 건전한 식습관으로 아동의 영양상태 개선과 소아비만의 예방 및 올바른 식사예절의 정착을 위한 정기적인 영양교육이 필요하다고 하겠다.

다양한 식사섭취 조사방법을 활용한 비만 초등학생의 식생활 실태 분석 (Analysis of the diet of obese elementary school students using various dietary intake survey methods)

  • COVID-19로 인한 사회적 거리 두기 실천으로 소아 비만 증가하였고 비만 학생 대상 영양교육 및 상담의 필요성이 증가하고 있다. 이에 비만 초등학생의 식생활 문제점을 분석하여 비만 개선을 위한 영양교육 및 영양상담 시 활용할 수 있는 기초자료를 제공하고자 하였다. 본 연구는 2020년 부산시교육청 주관 영양캠프에 참여한 초등학생 4-6학년 88명을 대상으로 사진 활용 24시간 식사기록, 한 달 간의 식품섭취빈도 조사, DST 식습관 조사를 활용하여 식생활 실태를 분석하였다. 조사 대상자는 남학생 46.6%, 여학생 53.4%로 정상체중군 15.7%, 과체 중군 30.3%, 비만군 53.9%이다. 조사대상자의 평균 연령은 11.77 ± 0.77세, 평균 신장은 147.44 ± 8.08 cm, 평균 체중은 53.47 ± 1.56 kg, 평균 체질량지수는 23.96 ± 3.01 kg/m2 이었다. 사진활용 24시간 식사기록 조사 결과, 과체중군과 비만군이 정상체중군보다 녹색 채소류 (p < 0.001)와 흰색 채소류 (p < 0.01)의 섭취가 적었으며, 한 달 간의 식품섭취빈도 조사를 식품군으로 묶어 조사한 결과에서는 어패류 (p < 0.01)와 녹색 채소류 (p < 0.05)의 섭취빈도가 낮아 유의한 차이를 보였으며, 81가지 식품을 나누어 분석한 결과에서는 비만군이 정상체중군보다 라면 (p < 0.01)과 스넥과자류 (p < 0.05)의 섭취빈도가 높고 비만군이 정상체중군과 과체중군보다 탄산음료 (p < 0.01), 햄, 소시지 (p < 0.05)의 섭취빈도가 높은 반면, 과체중군과 비만군이 정상 체중군보다 멸치 (p < 0.05)와 브로콜리 (p < 0.05), 단호박 (p < 0.05)의 섭취가 낮아 유의한 차이를 보였다. DST 식습관 조사를 비교한 결과, 과체중군과 비만군이 정상체중군보다 채소류 (p < 0.01) 섭취가 낮은 반면, 유제품 (p < 0.05)과 단 간식 (p < 0.05) 섭취가 높게 나타나 유의한 차이를 보였다. 이상의 결과는 비만 아동의 문제를 해결하기 위해 다양한 방법을 활용하여 식생활 문제를 평가하고 개인 문제에 맞는 해결 방법으로 접근해야 함을 시사한다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo).   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. We use combination of MLP and fuzzy filter for pattern recognition to estimate the direction of a muscle and Hogan's method for the force estimation. EMG signals are acquired by using a amputation simulator and 2 dimensional joystick motion.
