Quality Characteristics and Content of Polysaccharides in Green Tea Fermented by Monascus pilosus

  • In this study, we designed a method to manufacture elevated fermented green tea by using Monascus pilosus, which is known as a functional microbe, and observe its antioxidant abilities and quality characteristics. The water-soluble substance (WSS) content of the fermented tea by M. pilosus (FTM) was lower than that of the non-fermented tea (NFT), although the alcohol-insoluble substance (AIS) content of the FTM was higher than that of NTM. On the other hand, the fractionated distilled water-soluble polysaccharide (DWSP), CDTA-soluble polysaccharides (CDSP), sodium carbonate-soluble polysaccharide (SCSP) and KOH soluble hemicellulose (HC) obtained from the AIS of the FTM was markedly higher than that of NFT. In the antioxidant parameters, the electron donating ability of all fractions, except HC, extracted from FTM was higher than that of NFT, and iron chelating ability of all fractions, except CDSP, extracted from FTM was higher than that of NFT. Whereas the DWSP and SCSP obtained from the FTM were higher than that of NFT, the activity of the HC fraction from both NFT and the FTM could not be detected. In addition, the xanthin oxidase (XO) inhibitory activities of the DWSP, CDSP and the SCSP obtained from the NFT were significantly higher than that of FTM, the aldehyde oxidase (AO) inhibitory activities of the DWSP and SCSP extracted from the FTM were markedly higher than that of the NFT. Meanwhile, the acceptance of NFT and FTM had no significant difference, while the quality of aroma, taste and mouthfeel of the FTM was higher than that of NFT. These results suggest that the post-fermented tea by Monascus microorganisms may be responsible for functional components as well as contribute to the improvement of the tea quality.

FTM을 이용한 파형격벽의 최적구조설계 (Optimum Structural Design of a Corrugated Bulkhead by using Flexible Tolerance Method)

  • In this paper, merits and demerits of Nelder and Mead Penalty Function Method(SUMTNM) and Flexible Tolerance Method(FTM) are investigated from the standpoint of generality, accuracy and efficiency. SUMTNM is combined with Nelder and Method and SUMT, but FTM improves the values of the objective function by using information provided by feasible points as well as certain nonfeasible points termed near-feasible points. Therefore, FTM uses more information than SUMTNM for minimizing object function. The structural analysis of a vertically corrugated bulkhead is performed by collapse mechanism and plate buckling analysis. Based on the results of this analysis, minimum structural weight design of a corrugated bulkhead by use of above two optimization techniques is carried out by investigating the effects of sizes of bulkhead on the structural weight.

FTM 튜브의 판넬과 마스크의 일치방법 개발 (Development of Registration Method of Panel and Mask for FTM Tube)

  • This paper presents a useful method of registration in manufacturing of shadow color mask for cathode ray tubes of the FTM (Flat Tension Mask) type, wherein the shadow mask and front panel are interchangeable when mask-panels are assembled, which is called ICM system. Theoretical analysis and alignment process are presented. The pattern of mask aperture is registered with a screen pattern of corresponding geometry of the panel in flat tension mask tube. Registration accuracy of panel and mask affects the purity of color cathode ray tube concerned with mislanding. It tries to minimize the misregistration caused by variances, which are mechanical error, mask stretching position error, restrictive number of fiducial point, etc.

Sodium polyanethol sulfonate의 혈액배양에서의 Salmonella typhi 분리에 대한 영향 (Effect of sodium polyanethol sulfonate on the isolation of Salmonella typhi from blood culture)

  • Blood is one of the most important clinical specimens for the isolation of bacteria. A rapid isolation and a high isolation rate of bacteria are very important in blood culture because bacteremic patients are mostly in grave condition. Various blood culture media which support growth of most fastidious bacteria are available commercially. However, growth of bacteria are frequently delayed because of antibacterial activity of blood. Sodium polyanethol sulfonate(Liquoid) has been reported to inactivate the antibacterial substance and disrupt phagocytic cells. The beneficial effect of SPS is well recognized in the isolation of gram-positive bacteria. However, the effect does not seem to be prominent for gram-negative bacilli isolation mainly due to the rapidity of their growth. It has been experienced with Sal. typhi that the growth is much slower than that of other gram-negative bacilli. For the rapid growth of the organism, use of bile broth has been recommended. Although Sal. typhi is the most frequently isolated organism at present, about one half of total isolates are other organisms and, in case bile broth is used, other media which support growth of these organisms should be used together. Fluid thioglycollate medium(FTM) which is always used in blood culture to isolate anaerobes is inferior to brain heart infusion(BHI) for the isolation of aerobes. This study was done to determine the effect of SPS on the isolation of Sal. typhi from blood. During the Sep. 1973 to Sep. 1974 study period, 2460 blood cultures were made from the Severance hospital patients: BHI and FTM sets 1431 specimens, BHI with SPS(0.05%) and FTM sets 396 specimens, BHI and FTM with SPS sets 359 specimens, BHI and BHI with SPS sets 274 specimens. Mean incubation time required for the macroscopic detection of growth of Sal. typhi were 3.5 days on BHI and 2.7 days on BHI with SPS. The 0.8 day difference was statistically significant. On FTM the mean incubation time was 3.8 days while it was 2.9 days on FTM with SPS. The 0.9 day difference was statistically significant. The result on BHI with and without SPS sets showed faster growth on BRI with SPS in 7 specimens and slower growth in one specimen and the remaining 12 showed growth at the same time. These specimens had mean incubation time of 3.2 days on BHI and 2.3 days on BHI with SPS. The 0.9 day difference was statistically significant. This study indicates beneficial effect of SPS for the rapid isolation of Sal. typhi from clinical blood specimens.

  • 시스템 아키텍쳐는 시스템의 가장 상위수준 개념이다. 이러한 아키텍쳐의 정립이 1997년 8월에 완료된 2수준 아키텍쳐에 이어 3, 4수준 아키텍쳐가 연구되었다. 본 연구는 그중 고속도로교통관리시스템(FTMS)에 관한 것을 제시한다. 제 3수준 아키텍쳐는 2수준 아키텍쳐를 서비스 구현단위로 분해한 서브시스템이며, 다양한 ITS 서비스를 '제품(Package)'화 하여 ITS 서비스를 선택적, 점진적으로 구현 가능케 하였다. 제 4수준 아키텍쳐는 물리아키텍쳐 개념을 도입하여 3수준에서 분해된 서브시스템을 구성하는 각종 물리적 요소를 정의하고, 이들 간의 또는 외부요소와의 정보흐름을 정의하였다. 이에 따라, FTMS는 고속도로 교통관리 서브시스템, 고속도로 교통류제어/연계 서브시스템, 고속도로 돌발상황관리 서브시스템으로 분해/정의되었다.

Report on the Occurrence of Perkinsus sp. in the Manila Clams, Ruditapes philippinarum in Korean

  • Five species of intertidal clams including Ruditapes philippinarum, Tegillarca granosa, Solen strictus, Heteromacoma irus, and Coecella chinensis were tested for the presence of the protozoan parasite, Perkinsus sp. using fluid thioglycollate medium (FTM) fortified with antibiotics and histological techniques. Each individual clam was placed in a test tube filled with 10ml FTM, placed in totally dark place, and incubated over a week. After incubation the clam tissues were stained with Lugol's iodine solution and examined under a light microscope to find out any hypnospores of Perkensus sp. in the tissues. Cross-sections of the clams were also embedded in paraffin, sliced to 3um, and stained with Harry's hematoxylene and Picro eosine to observe the presence of tomont or trophozoites. Perkinsus sp. were found in the presence of tomont or trophozoites. Perkinsus sp. were found in the tissues of R. philippinarum collected from Kangjin and Wando, along the south coast of Korea. However, Perkinsus sp. was not found in four other species of clams nor R. philippinaurm collected from Kimnyong and Waido in Cheju. A size-dependent Perkinsus sp. infection was found in R. philippinarum collected rom Kangjin and Wando the clams smaller than 15mm in shell width do not exhibit and Perkinsus sp. while other clams greater than 20mm in shell width exhibit almost 100% infection. To determine the number of Perkinsus sp. in the clams, FTM cultured clam tissues were digested with 2M NaOH solution and the number of hypnospores in the tube were counted. The number of hypnospores counted from the tissues indicated that each Manila clam contains 100,000 to 3,500,000 Perkinsus cells or 20,000 to 1,000,000 cells per gram tissue wet weight. The results of cell counts also suggests that such a high occurrence of Perkinsus sp. in the clam may cause mortality, as already reported from other studies of Perkinsus spp.

PCR 기법을 이용한 바지락포자충 Perkinsus 진단 기술개발 (Development of a PCR Assay for Detection of the Protozoan Parasite Perkinsus)

  • 이 연구에서는 해산 연체동물의 폐사를 유발하는 기생성 원생동물인 Perkinsus를 신속하고 특이적으로 검출하기 위하여 PCR 진단법을 개발하였다. 이를 위해 Perkinsus 속 4종을 공통적으로 증폭하거나 P. atlanticus만을 특이적으로 증폭할 수 있는 두 가지의 primer를 제작하였다. PCR분석은 기존 Perkinsus 진단법인 fluid thioglycollate medium (FTM) 방법과 2 M NaOH 기법을 병행하여 실시하였다. 실험구로서 전라남도 완도산, 제주도 김녕산, 제주도 서귀포산, 제주도 성산산 바지락과 in vitro 배양된 바지락포자충이 이용되었으며, 대조구로서 전라남도 강진에서 채집된 꼬막, T. granosa을 사용하였다. 실험 결과 Perkinsus특이성 DNA band가 전남 완도산과 제주도 성산산 바지락, in vitro 배양된 바지락 포자충에서 확인되었으나, 대조구였던 꼬막과 제주도 서귀포산, 제주도 김녕산 바지락에서는 나타나지 않았다. 이같은 진단 결과는 FTM과 2M NaOH 진단 결과와 일치하였다. 한편 P. atlanticus에 특이적인 primer에 의해 증폭된 band가 확인됨에 따라 국내산 바지락에서 검출되는 바지락포자충은 P. atlanticus와 동일 종임이 확인되었다. 결론적으로, 본 연구를 통하여 개발된 PCR을 이용한 진단법은 해산 연체동물내 Perkinsus 속 기생충과 P. atlanticus의 감염을 신속하고도 종 특이적으로 진단할 수 있어 수ㆍ출입 수산물의 검역과 바지락 포자충의 생태학적 특성을 규명하는데 효과적으로 이용될 수 있을 것으로 기대된다.

Construction of minimum time joint trajectory for an industrial manipulator using FTM

  • The path of an industrial manipulator in a crowded workspace generally consists of 8 set of Cartesian straight line path connecting a set of two adjacent points. To achieve the Cartesian straight line path is, however, a nontrivial task and an alternative approach is to place enough intermediate points along a desired path and linearly interpolate between these points in the joint space. A method is developed that determines the subtravelling- and the transition-time such that the total travelling time for this path is minimized subject to the maximum joint velocities and accelerations constraint. The method is based on the application of nonlinear programming technique, i.e., FTM (Flexible Tolerance Method). These results are simulated on a digital computer using a six-joint revolute manipulator to show their applications.

서해안 양식 바지락에 발생한 Perkinsus sp. 감염증과 신속검출 (Infection and Rapid Detection of Perkinsus sp. In Cultured Babyneck Clam, Ruditapes philippinarum from Western Coast of Korea)

  • 매년 폐사가 발생해 온 서해안 연안의 고창과 태안에서 채집한 양식 바지락에서 복합포자충류인 Perkinsus sp.가 관찰되었다. 기생충에 감염된 바지락은 육안적으로 아가미와 육질의 표면에서 유백색의 결절이 나타났다. 기생충의 영양체는 편재된 핵을 가지고 있으며, 아가미와 외투막, 간 췌장, 생식소 조직에서 이분열에 의해 증식하며, 육아종의 형성과 혈구의 침윤을 유발하였다. 감염된 바지락을 FTM에 배양한 결과 영양체의 크기가 커지면서 루골용액에 의해 검게 염색된 구형의 유주자낭들 형성하였다. Perkinsus sp.의 평균 감염율은 9개월의 조사 기간 동안 고창이 73.1%, 태안이 94.8%로 나타났으며, 감염율은 각장의 크기가 클수록 증가하는 경향을 나타내었다. Hemacolor kit는 바지락의 대량폐사와 밀접한 연관이 있는 Perkinsus sp.의 영양체의 신속진단에 유용한 것으로 나타났다.
