Net Analyte Signal-based Quantitative Determination of Fusel Oil in Korean Alcoholic Beverage Using FT-NIR Spectroscopy

  • Purpose: Fusel oil is a potent volatile aroma compound found in many alcoholic beverages. At low concentrations, it makes an essential contribution to the flavor and aroma of fermented alcoholic beverages, while at high concentrations, it induced an off-flavor and is thought to cause undesirable side effects. In this work, we introduce Fourier transform near-infrared (FT-NIR) spectroscopy as a rapid and nondestructive technique for the quantitative determination of fusel oil in the Korean alcoholic beverage "soju". Methods: FT-NIR transmittance spectra in the 1000-2500 nm region were collected for 120 soju samples with fusel oil concentrations ranging from 0 to 1400 ppm. The calibration and validation data sets were designed using data from 75 and 45 samples, respectively. The net analyte signal (NAS) was used as a preprocessing method before the application of the partial least-square regression (PLSR) and principal component regression (PCR) methods for predicting fusel oil concentration. A novel variable selection method was adopted to determine the most informative spectral variables to minimize the effect of nonmodeled interferences. Finally, the efficiency of the developed technique was evaluated with two different validation sets. Results: The results revealed that the NAS-PLSR model with selected variables ($R^2_{\upsilon}=0.95$, RMSEV = 100ppm) did not outperform the NAS-PCR model (($R^2_{\upsilon}=0.97$, RMSEV = 7 8.9ppm). In addition, the NAS-PCR shows a better recovery for validation set 2 and a lower relative error for validation set 3 than the NAS-PLSR model. Conclusion: The experimental results indicate that the proposed technique could be an alternative to conventional methods for the quantitative determination of fusel oil in alcoholic beverages and has the potential for use in in-line process control.

Nondestructive Evaluation for the Viability of Watermelon (Citrullus lanatus) Seeds Using Fourier Transform Near Infrared Spectroscopy

  • Purpose: Conventional methods used to evaluate seeds viability are destructive, time consuming, and require the use of chemicals, which are not feasible to implement to process plant in seed industry. In this study, the effectiveness of Fourier transform near infrared (FT-NIR) spectroscopy to differentiate between viable and nonviable watermelon seeds was investigated. Methods: FT-NIR reflectance spectra of both viable and non-viable (aging) seeds were collected in the range of 4,000 - 10,000 $cm^{-1}$ (1,000 - 2,500 nm). To differentiate between viable and non-viable seeds, a multivariate classification model was developed with partial least square discrimination analysis (PLS-DA). Results: The calibration and validation set derived from the PLS-DA model classified viable and non-viable seeds with 100% accuracy. The beta coefficient of PLS-DA, which represented spectral difference between viable and non-viable seeds, showed that change in the chemical component of the seed membrane (such as lipids and proteins) might be responsible for the germination ability of the seeds. Conclusions: The results demonstrate the possibility of using FT-NIR spectroscopy to separate seeds based on viability, which could be used in the development of an online sorting technique.

  • In 1989, at the 2nd ICNIRS Meeting in Tsukuba, I projected certain directions for NIR technology would take in the future. Among those projections were: (1) An inrush of companies producing FT-NIR instruments. (2) Hybrid NIR Systems (3) Hand-held NIR Technology. All three thrusts have resulted in numerous commercial offerings over the last 12 years Hand-held technology for all fields is growing at an astonishing rate. To date, NIR work at North Carolina State University has produced four (4) hand-held NIR units for: (1) Nicotine and Moisture in tobacco, (2) Vanillin and Moisture in Vanilla Beans, (3) Protein, Moisture and Nitrogen in plant tissue, (4) Chlorophyll and Moisture in Growing Plants: A NIR Spectrometer for Developing Countries. This paper will discuss these developments, including design and performance data.

  • In this study, a FT-NIR spectroscopy was used to determine skin moisture. NIR diffuse reflectance spectra were collected from separated dorsal and abdominal hairless mouse skin. Partial least squares regression (PLSR) was applied to develop calibration models that determine the water content. The seven spectra regions, such as 833-2500, 1100-2250, 1100-1750, 1750-2250, 1200-1600, 1800-2200, and 1200-2200 except 1600-1800 nm, were investigated for the best model by PLSR. The developed model predicted skin moisture for validation set with a standard errors of prediction (SEP) of 4.43%, when used 833-2500 nm. (omitted)

Rapid Prediction of Amylose Content of Polished Rice by Fourier Transform Near-Infrared Spectroscopy

  • Fourier transform near-infrared (FT-NIR) spectroscopy and partial least squares (PLS) regression were used to predict the amylose content of polished rice. Spectral reflectance data in a wavelength range of 1,000 to 2,500 nm were obtained with a commercial spectrophotometer for 60 different varieties of Korean rice. For a comparison of this spectroscopic method to a standard chemical analysis, the amylose contents of the tested rice samples were determined by the iodine-blue colorimetric method. The highest correlation for the rice amylose ($R^2=0.94$, standard error of prediction=0.20% amylose content) was obtained when using the FT-NIR spectrum data pre-treated with normalization, the first derivative, smoothing, and scattering correction.

  • 국내에서는 건물의 화재 방지를 위하여 인증된 내화뿜칠재를 사용하도록 규정되어 있다. 하지만 일부 현장에서는 성능이 없는 흡음뿜칠재을 사용함으로 해서 내화구조 부실시공의 원인이 되고 있다. 따라서 성능이 인정된 내화 뿜칠재와 일반 흡음뿜칠재를 현장에서 분석하여 적정 시공 여부를 확인하는 방법을 제시하고자 한다. 이에 본 연구에서는 성능이 인정된 내화 뿜칠재 9종과 정상 내화 뿜칠재 및 내화성능이 없는 일반 흡음뿜칠재 3종을 비교 측정하였다. 측정하기 전에 분석시료를 대상으로 mill을 사용하여 powder로 전처리를 하였으며 현장에 접근성이 용이한 NIR을 사용하였다. 측정은 NIR중에서 FTNIR을 사용하였으며, 측정 모듈은 적분구(integrating sphere)를 사용하여 측정하였다. 이 측정된 흡수 스펙트럼은 통계 처리 방법 중에서 주성분 분석법인 PCA 기법으로 분석함으로 해서 정상과 비정상 내화뿜칠재를 판별이 가능 함을 확인하였다.

  • Near infrared (NIR) spectroscopy is a powerful technique for non-destructive analysis that can be obtained in a wide range of environments. Recently, NIR measurements have been utilized as probe for quantitative analysis in agricultural, industrial, and medical sciences. In addition, it is also possible to make practical application on NIR for molecular structural analysis. In this work, Fourier transform near infrared (FT-NIR) measurements were carried out to utilize extensively in the relative amounts of different secondary structures were employed, such as Iysozyme, concanavalin A, silk fibroin and so on. Several broad NIR bands due to the protein absorption were observed between 4000 and $5000\;^{-1}$. In order to obtain more structural information from these featureless bands, second derivative and Fourier-self-deconvolution procedures were performed. Significant band separation was observed near the feature at $4610\;^{-1}$ ,. Particularly the peak intensity at $4525\;^{-1}$ shows a characteristic change with thermal denaturation of fibroin. The structural information can be also obtained by mid-IR and CD spectral. Correlation of NIR spectra with protein structure is discussed.

농약 제품의 동일성 판별을 위한 FT-NIR 분석 사례 연구 (Study of Feasibility Test: FT-NIR Spectrometer for Discrimination Analysis of Agrochemical Products)

  • 본 연구는 농약 제조수입회사에서 등록한 제조처방에 따라 농약을 유통판매하고 있는지의 동일성을 확인함에 있어서 FT-NIR의 적용 가능성을 알아보고자 하였다. 농약 등록용으로 제출된 시료와 유통 중인 농약 83점의 스펙트럼 간 상관관계를 조사하였다. 그리고 유효성분과 제형이 동일한 유통농약 23점의 스펙트럼 간 상관관계를 조사하였다. 또한, 유효성분이 다른 유통농약 68점의 스펙트럼 간 상관관계도 조사하였다. 등록용으로 제출된 시료와 유통 중인 농약 83점의 1차 미분 스펙트럼 간 상관관계 값은 95.86~100%로 나타났다. 상관관계 값 한계치를 Standard Deviation of Population에서 주로 적용하는 95%에는 모두 적합으로 판정될 수 있을 것이다. 한편, 농약의 유효성분과 제형은 동일한 유통농약 23점의 1차 미분 스펙트럼 간 상관관계 값은 29.09~99.83%로 나타났다. 99.0%이상 높이 나온 3점은 제조사는 다르지만 유효성분과 제형 및 사용된 부자재까지 같은 농약임을 확인할 수 있었다. 3점을 제외한 나머지는 95% 미만으로 나타났다. 농약의 유효성분이 다른 68점의 스펙트럼 간 상관관계 값이 2.00~93.70%까지 다양하게 나왔다. 상관관계 값이 93.70%처럼 높게 나타난 것은 농약의 유효성분은 다르지만 사용된 부자재의 유사성에 기인한 것으로 추정된다. 동일성 판별 상관관계 값의 한계치 95%를 적용하면 모두 부적합으로 판정될 수 있을 것이다. 따라서 본 결과를 토대로 등록 농약과 유통 중인 농약의 동일성을 정성적으로 판별하는데 있어서 FT-NIR의 활용 가능성이 확인되었다.

비파괴 분석 방법인 푸리에 변환 근적외선 분광 분석을 이용한 한지 기록물의 산성도 및 함수율 정량 분석 연구 (The study of quantitative analytical method for pH and moisture of Hanji record paper using non-destructive FT-NIR spectroscopy)

  • 한지기록물의 상태를 평가하는 데 있어 기록물의 훼손을 방지하는 것은 필수적인 요소이다. 사용된 공시재료는 1900년대 생산된 한지기록물을 사용하였다. 이 종이 기록물의 상태를 평가하기 위하여 비파괴적 분석기인 12,500~4,000 $cm^{-1}$ 파장영역을 가진 푸리에변환(FT) 근적외선 분광 분석기를 이용하였다. 푸리에변환 방식의 간섭계는 뛰어난 정밀도와 정확도를 가지며, 신호-대-잡음비가 가장 우수 하여 정량 분석에서 가장 많이 사용되고 있다. 한지기록물의 산성도와 함수율을 적분구(integrating sphere)의 확산반사로 사용하여 측정하였다. 산성도(pH)의 경우에 전처리를 하지 않았을 때의 상관계수($R^2$)는 0.92, 표준예측오차(SEP)는 0.317이었고, 전처리를 하였을 때의 $R^2$는 0.98, SEP는 0.208 이었다. 그리고 함수율의 경우 산성도와 달리 전처리를 하지 않았을 경우에도 $R^2$와 SEP는 각각 0.99와 0.458로 확인 되었다. 전처리 중 다산란 보정방법(MSC)의 경우에는 $R^2$가 0.97, SEP가 0.558로 증가하는 것을 확인 할 수 있어 함수율은 전처리를 하지 않았을 때가 좋은 결과가 나오는 것으로 확인 되었다. 이러한 결과를 통해 비파괴적인 분석방법으로 한지기록물을 적분구와 FT 근적외선 분광 분석기를 이용하여 한지의 상태를 정확히 평가할 수 있을 것으로 판단되었다.

천연과 CVD 합성 다이아몬드의 감별을 위한 물성 연구 (Properties of the Natural and CVD Synthetic Diamonds for Identification)

  • Recently, Chemical Vapor Deposition (CVD) synthetic diamonds have been introduced to the jewelry gem market, as CVD technology has been making considerable advances. Unfortunately, CVD diamonds are not distinguishable from natural diamonds when using the conventional gemological characterization method. Therefore, we need to develop a new identification method that is non-destructive, fast, and inexpensive. In our study, we employed optical microscopy and spectroscopy techniques, including Fourier transform infra-red (FT-IR), UV-VIS-NIR, photoluminescence (PL), micro Raman, and cathodoluminescent (CL) spectroscopy, to determine the differences between a natural diamond (0.30 cts) and a CVD diamond (0.43 cts). The identification of a CVD diamond was difficult when using standard gemological techniques, UV-VIS-NIR, or micro-Raman spectroscopy. However, a CVD diamond could be identified using a FT-IR by the Type II peaks. In addition, we identified a CVD diamond conclusively with the uneven UV fluorescent local bands, additional satellite PL peaks, longer phosphorescence life time, and uneven streaks in the CL images. Our results suggest that using FT-IR combined with UV fluorescent images, PL, and CL analysis might be an appropriate method for identifying CVD diamonds.
