  • 임신초기 말의 태반으로부터 분비되는 융모성 성선자극 호르몬(eCG)은 황체형성(LH), 난포자극(FSH) 및 갑상선자극(TSH)호르몬과 같이 알파 및 베타 단체의 비공유결합으로 구성되어져 있는 당단백질 호르몬이다. 말의 융모성 성선자극 호르몬은 모체의 자궁내막에 침입한 태아 유래의 융모조직(자궁내막배)에서 합성ㆍ분비되어진다. 알파단체의 아미노산 배열은 동물종내에서 동일하지만, 베타단체는 호르몬에 따라 작용의 특이성을 가지고 있다고 알려져 있다. (중략)

Biosynthesis of a Biological Active Single Chain Equine Chorionic Gonado-tropin

  • The equine chorionic gonadotropin (eCG) subunits $\alpha$ and ${\beta}$ are transcribed from different genes and associate noncovalently to form the bioactive eCG heterodimer. Dimerization is rate limiting for eCG secretion, and dissociation leads to hormone inactivation. The correct conformation of the heterodimer is alto important for efficient secretion, hormone-specific post-translational modifications, receptor binding and signal transduction. To determine whether ${\alpha}$ and ${\beta}$ subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-eCG molecule by fusing the carboxyl terminus of the eCG ${\beta}$-subunit to the amino terminus of the af-subunit was construe-ted and transfected into chinese hamster ovary (CHO-Kl) cells. LH- and FSH-like activities were assayed in terms of testosterone production and aromatase activity in primary cultured rat Leydig cells and granulosa cells, respectively. The tethered-eCG was efficiently secreted and showed similar LH-like activity to the dimeric eCG ${\alpha}$/${\beta}$ and native eCG. FSH-like activity of the tethered-eCG was also shown similarly in comparison with the native and wild type eCG ${\alpha}$/${\beta}$. Our data for the first time suggest that the tethered-eCG can be expressed efficiently and the produced product by the CHO-K1 cells is fully LH- and FSH-like activities in rat in vitro bioassay system. Our results also suggest that this molecular can imply particular models of FSH-like activity not LH-like activity in the eCG. Taken together, these data indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion.

다낭성 난소증후군 치험 1례 (Clinical Study for the One Case that Diagnosed Polycystic Ovarian Disease)

  • Purpose: The purpose of this study is to report the effect of oriental treatments for PCOD(Polycystic Ovarian Disease) patient with amenorrhoea. Methods: A 24 years old women, who diagnosed PCOD(Polycystic Ovarian Disease), was enrolled in this study. She received oriental treatments such as herbal medicine, acupuncture for 5 months. And we proceeded to checkup female hormone regularly, such as Estrogen, Progesteron, LH, FSH, Prolactin, testosterone. We observed the menstruation period and figure out LH/FSH ratio. Results: 1. We treated PCOD patient with oriental herbal medicine(Changbudodamtang) and acupuncture. 2. PCOD patient restart menstruation regularly. 3. PCOD patient's LH/FSH ratio decreased. 4. PCOD patient's testosterone was within normal limit. Conclusion: 1. We treated PCOD patient with oriental herbal medicine(Changbudodamtang) and acupuncture, and then PCOD patient restart menstruation regularly. 2. We treated PCOD patient with oriental herbal medicine(Changbudodamtang) and acupuncture, and then we observed that LH/FSH ratio decreased.

Effects of GnRH on the Plasma FSH, LH and Estradiol Levels at Estrus Induced with Injection of PGF2α and eCG in Prepubertal Buffaloes (Bubalus bubalis)

  • The experiment was conducted to study the effect of GnRH administration at induced estrus on pituitary and ovarian response in buffalo heifers. Eight Murrah river buffaloes of 12 to 13 months of age were treated with $PGF_{{2}{\alpha}}$ and eCG combination. GnRH (Fertagyl) 200 ug was injected (iv) at estrus in four heifers (treated group) while saline (2 ml, iv) was injected in remaining four heifers (control group). Blood was collected through jugular catheter to estimate plasma FSH, LH and estradiol level. The pretreatment plasma FSH, LH and estradiol values ranged from $8.46{\pm}1.97ng/ml$ to $12.31{\pm}1.30ng/ml$, $0.87{\pm}0.21ng/ml$ to $1.19{\pm}0.29ng/ml$ and $19.09{\pm}2.38pg/ml$ to $20.24{\pm}1.00pg/ml$ respectively. The plasma estradiol concentration elevated significantly (p<0.05) within 24 hr after eCG administration and reached its peak levels of $154.09{\pm}17.28pg/ml$ and $181.95{\pm}31.82pg/ml$ at estrus in respectively treatment and control groups. The plasma FSH and LH concentrations did not increase during follicular development after eCG administration while initial significant (p<0.05) increases in both plasma FSH and LH concentrations occured within 5 and 10 min, reaching peak levels of respectively $110.06{\pm}23.56ng/ml$ and $13.15{\pm}3.13ng/ml$ within 90 min after GnRH injection was detected. A sharp and significant decline in plasma estradiol concentration ($59.27{\pm}8.78pg/ml$) associated with synchronized ovulation within 24 hours after GnRH injection was recorded. The observation suggest that the hypophysis of prepubertal buffaloes treated with eCG have gonadotrophins awaiting the releasing factor to evoke release of gonadotrophin during the follicular phase to induce synchronized ovulation.

한우와 샤로레 교잡종 빈우의 성성숙과 성호르몬 수준 변화에 관한 연구 (A Study on Puberty and Sex Hormone Levels In Korean Native and Charolais x Native Crossberd Heifers)

  • 한우와 Charolais와의 교잡종 빈우에 있어서 초발정일령과 체중 및 성장중의 혈중 LH, FSH prolactin, progesterone과 estradiol수준의 변화를 알고저 본 시험을 시도하였다. 공시된 빈우는 한우 4두와 교잡종$(F_1)$, 4두로서 NRC 사용표준에 따라 사료를 급여하였고 군사하였다. 체중은 출생일로부터 1개월 간격으로 16개월령까지 측정하였으며 13개월령까지 체중측정시마다 채혈한 다음 분리된 혈청내의 호르몬을 RIA로 분석하였다. 초발정일령은 유의성이 없었으나 교잡종이 한우보다 빨랐다($326.7{\pm}51.6$일과 $372.5{\pm}31.9$일) 초발정시 체중은 교잡종이 한우보다 월등히 (p<0.01) 무거웠다 ($223.0{\pm}16.6kg$$175.3{\pm}10.3kg$). 혈청내 LH와 FSH는 1두의 교잡종에서 출생시와 3개월령에서 높게 나타난 것을 제외하고는 전빈우의 전월령에서 본 시험에서의 호르몬분석한계수준(LH, $1.5mIU/m\ell$ : FSH, $1.2mIU/m\ell$) 이하였다. 혈청내 prolactin은 다른 월령에서 보다 $5\~8$개월령에서 더욱 높았고 $11\~12$개월령에서는 개체간 차이가 많았다. 혈청내 progesterone은 출생시는 낮았으며 그 후 $6\~8$개월령에서 높았고 8개월령 이후에서 $0.1\~0.2ng/m\ell$으로 다시 감소하였다. 혈청내 estradiol수준은 전빈우의 모든 월령에서 본 시험의 호르몬분석 한계수준$(10pg/m\ell)$ 이하였다. 성성숙동안에 LH, FSH와 estradiol의 수준이 낮았던 원인은 이들 호르몬의 상승을 측정하기에는 너무 채혈간격이 길었던 것으로 추측된다.

Gilt에 있어서 Difluoromethylornithine에 의한 LH분비 억제 (Suppression of LH Concentration by Difluoromethylornithine in Gilts)

  • Ornithine decarboxylase(ODC)가 polyamine 생합성에 주요 효소로 역할을 하지만 difluoromethylornithine(DFMO)은 polyamine 합성에 억제자로 작용하고 있다. Cycling crossbed gilt들을 무작위로 두 그룹으로 분배하였다(6/group). Indwelling silicone catheter를 모든 동물의 경정맥에 수술을 통해 이식하였다. DFMO는 생리식염수(20mg/ml)에 용해하여 매일 80mg/kg을 i.m.으로 주사하였으며 대조구에게는 같은 양의 생지식염수만을 주사하였다. DFMO는 estrous cycle day 16일부터 21일 혹은 발정때까지 하루 3번(08:00, 16:00, 24:00h) 주사하였다. Day 14일 부터 마지막 DFMO주사 이틀 후까지 하루 한번씩 10ml의 혈액을 채취하였다. Day 16일부터 21일까지 매일 다른 gilt로부터 8시간 동안(08:00∼16:00h) 15분 간격의 주기로 window 혈액을 채취하였다. Serum으로부터 progesteron(P4), Estradiol(E2), LH 및 FSH를 측정하였다. P4와 E2는 DFMO처리에 관계없이 follicular phase동안에 전형적인 profile을 보였다. DFMO처리는 발정직전의 LH농도를 저하시켰지만 (p<0.01), FSH에는 아무런 영향을 미치지 않았다. 결론적으로 gilt에 있어서 DFMO는 LH분비에 억제적인 영향을 미치는 한편, P4, E2 및 FSH에는 별로 영향을 미치지 않음이 나타났다.

호르몬 첨가가 돼지 난포란의 체외성숙과 체외수정에 미치는 영향 (Effect of Hormones on Pig Oocytes Maturation and Fertilization In Vitro)

  • This experiment was carried out to investigate in vitro maturation rate of pig follicular oocytes cultured from 30 to 48hr in TCM 199 supplemented with gonadotropins(FSH, LH) and estradiol-17$\beta$ and in vitro fertilization with ejaculated sperm preincubated in BO medium containing 2mM caffein and development of IVF oocytes. The results obtained in this experiments were as follows ; 1. In addition of hormones, in vitro maturation rate of follicular oocyte increased gradually from 36hr and 74.47% at 48hr in addition of hormones, but there was no differences among in vitro maturation rates after 36hr of culture. 2. Penetration rate of pig oocytes matured in FSH+LH+E2 and FSH+E2 was 71.8%, 71.0% and significantly increased by the addition of hormones. 3. Percentage of developed oocytes was 44.4% for oocytes matured in FSH+LH+E2-added medium and 48.7% for oocytes matured in FSH+E2-added medium, respectively. 4. Two to 16 cells stage embryos were obtained only when pig oocytes matuerd in vitro in hormones-added medium and 72hr after IVF. 5. From present results, it is concluded that gonadotropins and estradiol17$\beta$ can enhance in vitro fertilization and subsequent development as well as in vitro maturation pig follicular oocytes.

열목어 생식선자극호르몬의 cDNA cloning 및 CHO 세포를 이용한 발현검토

  • 경골어류의 뇌하수체에서는 두 종류의 생식선자극호르몬 (FSH, LH)이 생산되며, 이 호르몬들은 공통적인 α 쇄와 특이적인 β 쇄를 가진다. 연어과 어종들에서, FSH는 난황형성과 정자형성의 역할을 하며, LH는 배우자의 최종성숙을 조절한다. 냉수성 고유어종인 열목어 (Brachymystax lenok)의 멸종을 방지하고 종묘생산을 원활히 하기 위하여 먼저 열목어의 GTHα, FSHβ 및 LHβ 쇄를 cloning하여 염기서열을 결정하였다. 열목어 GTHα, FSHβ 및 LHβ의 cDNA는 산천어의 해당 cDNA와 높은 상동성 (각각 84, 95, 98%)을 보였다. 다음으로 기능적인 생식선자극호르몬을 제작하기 위해서 2개의 쇄를 single-strand로 연결하여 진핵세포를 숙주로하는 시스템에서 생식선자극호르몬을 생산할 수 있는 구조체인 FSHβ-GTHα (235 amino acids) 와 LHβ-GTHα (240 amino acids)를 각각 재조합하였다. 또한 각각의 융합단백질 생산용 구조체의 3'-말단에는 단백질추출이 용이하도록 histidine×6 구조를 첨가하였다. 이상의 단일쇄 FSH와 LH 유전자재조합 산물을 포유동물 유래의 세포 (CHO-K1)에 liposome chemical을 사용하여 유전자도입 후 세포에서 분비되는 단백질을 모니터링하였다. 배지를 부분정제한 후 SDS-PACE로 조사한 결과, LH 재조합 유전자를 도입한 후 48-60 시간째에 약 25 kDa의 단백질로서 관찰되었다. 현재 FSH 재조합 유전자에 대해서도 조사중이며, 향후 이를 재료로 하여 기능형 생식선자극호르몬을 생산하고 추출하기 위한 연구가 계속적으로 수행 될 것이다.

돼지 난포란의 체외성숙시 성선자극호르몬의 첨가가 체외성숙, 체외수정 및 배발생에 미치는 영향 (Effect of Gonadotropins added during Maturation of Porcine Oocytes on the In Vitro Maturation, In Vitro Fertilization and Development of Embryos)

  • This study was carried out to investigate the effects of gonadotropins added during maturation of porcine oocytes on the in vitro maturation(IVM), in vitro fertilization(IVF) and developmental potential of embryos. The follicular oocytes were cultured in TCM-199 medium containing different combination of gonadotropins(5$\mu$g /ml FSR or 1OIU /ml PMSG and 1O$\mu$g /ml LH or 1OIU /ml hCG), 10% FCS and 10% PFF for 36~48h in a incubator with 5% $CO_2$ in Air at 39$^{\circ}C$ and then matured oocytes were again cultured to 120h after IVF for 6~7h with heparin(100$\mu$g /m')-treated sperm. When the oocytes were matured for 42brs in the medium containing FSH+LH, FSH+hCG, PMSG+LH or PMSG+hCG, the JVF rate of each treatment was 50.0%, 52.9%, 66.7% and 70.0%, respectively. The highest CEI (cumulus cell expansion index) was obtained from PMSG+hCG-added medium and the highest polyspermic penetration resulted from FSH+LH-added medium. The cleavage of IVF oocytes derived from hormone added IVM was significantly(P<0.05) promoted by PMSG+hCG and the cleavage rate after 36-h, 42-h and 48-h maturation aws 53.0%, 56.7% and 45.6%, respectively. The highest developmental potential resulted from the oocytes derived from PMSG+LH -added IVM.

Intravenous Orexin Reduces LH Secretion in Castrated Camelus Dromedaries Fed a Sub-maintenance Diet

  • It has been shown that orexin has an inhibitory effect on gonadotropin secretions in non-ruminant animals. The goal of this study was to determine whether orexin affects LH, and FSH secretions in the camel, as a pseudo-ruminant animal, under different dietary energy content. Sixteen castrated camels were randomly divided into 4 groups. Animals in groups 1 and 2 were fed 100% and animals in groups 3 and 4 were fed 50% energy content in their diet for 20 days. After 20 days, animals in groups 1 and 3 received infusions of 1 $\mu{g}$ orexin and groups 2 and 4 received infusions of 2 $\mu{g}$ orexin into their jugular vein. Blood samples were collected from the jugular vein every 20 minutes from 4 h before the first infusion of orexin until 4 h after the last orexin infusion. Lower dietary energy intake and infusions of 2 $\mu{g}$ but not 1 $\mu{g}$ orexin significantly (p<0.01) decreased the mean plasma concentrations and pulse amplitudes of LH of the animals. Infusion of 1 and 2 $\mu{g}$ orexin did not change the secretions of LH of the animals fed NE. Different energy dietary intake and infusion of 1 and 2 $\mu{g}$ orexin did not change the mean plasma concentrations of FSH of the animals in all groups. Infusions of 1 and 2 $\mu{g}$ orexin significantly (p<0.01) decreased the glucose levels of animals fed LE but not in NE fed animals. Additionally, plasma glucose levels of the LE-fed animals in groups 3 and 4 were significantly (p<0.01) lower than those of the animals in groups 1 and 2 fed NE diet. The results of this experiment indicated that orexin may negatively affect LH and FSH in camels with negative energy balance, but not in those with positive energy balance.
