Optimization of Cellulase Production in Batch Fermentation by Trichoderma reesei

  • Maximum cellulase production was sought by comparing the activities of the cellulases produced by different Trichoderma reesei strains and Aspergillus niger. Trichoderma reesei Rut-C30 showed higher cellulase activity than other Trichoderma reesei stains and Aspergillus niger that was isolated from soil. By optimizing the cultivation conditions during shake flask culture, higher cellulase production could be achieved. The FP(filter paper) activity of 3.7U/ml and CMCase (Carboxymethylcellulase) activity of 60U/ml were obtained from shake flask culture. When it was grown in 2.5L fermentor, where pH and DO levels are controlled, the enzyme activities were 133.35U/ml (CMCase) and 11.67U/ml(FP), respectively. Ammonium sulfate precipitation method was used to recover enzymes from fermentation broth. The dried cellulase powder showed 3074.9U/g of CMCase activity and 166.7U/g of FP activity with 83.5% CMCase recovery.

Quality characteristics of steamed rice cake with Schisandra chinensis powder or extract added prior to storage (오미자 분말 및 추출물 첨가 설기떡의 저장 중 품질특성)

  • The purpose of this study was to improve the quality of the steamed rice cakes added with powder and extract of Schisandra chinensis (SC). Steamed rice cakes were prepared by adding SC using 1% hot-air dried SC powder (DP), hot-air dried SC extract (DE), freeze dried SC powder (FP) and, frozen SC extract (FE). After packaging with nylon/polyethylene films, the cakes were stored at $25^{\circ}C$ for 5 days. The FE-treated steamed rice cake had the lowest pH value. The $L^*$ value of the control cakes was highest immediately after processing and decreased during storage. The $a^*$ values of FP- and FE-treated steamed rice cakes were higher than those of DP- and DE-treated cakes. The $b^*$ values of DP- and DE-treated steamed rice cakes were higher than those of FP- and FE-treated cakes. Total phenolic contents of DP and DE were lower than those of FP and FE. Anthocyanin contents of DP- and DE-treated steamed rice cakes were lower than those of FP- and FE-treated cakes. Schisandrin contents of DP- and FP-treated steamed rice cakes were lower than those of DE- and FE-treated cakes. Ferric reducing antioxidant power assays revealed that DP and DE had lower antioxidant powers than those of FP and FE. The results of this study indicate that, among the treatments methods used, the steamed rice cake treated with FE had the highest functional component and antioxidant activity levels.

Fluticasone Propionate and Beclomethasone Dipropionate in Asthmatic Patients (천식환자에서 Fluticasone propionate와 Beclomethasone dipropionate의 치료효과 비교)

  • Background: Corticosteroid is most potent and effective anti-inflammatory medication currently available and inhaled form has been used in the long-tenn control of asthma. Fluticasone propionate(Flixotide/Flovent: FP) is highly potent and topically active inhaled corticosteroid and has at least twice the potency of beclomethasone dipropionate(BDP) in the control of asthma. The aim of this study was to compare the efficacy of FP and BDP in several aspects. Method: Fifty patients with asthma were treated in a randomized, parallel group study of 4 weeks duration. During 2-week run-in period $\beta_2$-agonist was administered. After run-in period, FP $500{\mu}g/day$ was administered via Diskhaler or BDP $800{\mu}g/day$ via reservoir dry-power device. During the run-in and treatment period, morning and evening peak expiratory flow rate(PEFR) were measured daily. Daytime and nighttime asthma symptoms, daytime and night-time rescue bronchodilator use were checked daily. $FEV_{1.0}$ and FVC were measured biweekly in both groups. Results: Three patients treated with FP and seven patient treated with BDP were dropped out. Therefore forty patients completed the study. Morning and evening PEFR was increased and diurnal variation of PEFR decreased significantly in both groups. $FEV_{1.0}$ increased significantly in FP treatment group but not in BDP group. There were also improvements in daytime and night-time asthma symptoms, daytime and night-time rescue bronchodilator use in both groups after treatment There were no significant difference between groups in any of the efficacy parameters. Therapeutic effects were demonstrated earlier in patient treated with FP than BDP. Conclusion: In this study, $500{\mu}g/day$ fluticasone propionate was as effective as $800{\mu}g/day$ beclomethasone dipropionate in the control of asthma. Therapeutic effects were demonstrated earlier in patient treated with FP than BDP without adverse effect.

Effect of Coagulants on the Behavior of Ultra Fine Dust in a Coal Firing Boiler (석탄 화력 보일러에서의 응집제 이용에 따른 초미세먼지 거동)

  • Particulate matters of PM2.5, particularly focusing on 0.1~1 ㎛ decrease the efficiency of dust-collector due to the brownian-motion. This study is to verify the effect of coagulant on the particle size distributions of potassium and PM2.5. The activated coagulant was spayed to the coal fired fluidized bed combustion boiler by the weight ratio of 1,200 : 1 = coal : coagulant, and the size distributions of captured particles at both the cyclone (FP) and electrostatic precipitator (EP) were measured. As the result of XRP analysis, the potassium content of FP increased to 13.33% (averagely from 1.65% to 1.87%) and, in EP at 17.68% (averagely from 1.65% to 2.03%). And it was confirmed by the particle size distribution analyzer and SEM image analysis that the distribution rates of PM2.5 decreased at 89.53% on average in FP, and at 88.57% in EP. The total dust concentration (mg/㎥) confirmed by tele-monitering system (TMS) decreased during the primary test from 2.6 to 1.7~1.9 and also the secondary test from 2.9 to 1.7~1.9.

Evaluation for Optimal HUD Location on a Train Using EEG (뇌파를 이용한 열차 최적의 HUD위치 평가)

  • This paper suggested a new evaluation method for optimal HUD position through a correlation based on between biological signal as and statistical analysis which using (Electroencephalogram, EEG) and ANOVA. This experiment was conducted two kinds of method to evaluate the optimal HUD position. At first, visual stimulus suggested from six different positions(the top and the bottom of the left, the top and the bottom of the center and the top and the bottom of the right on the screen) in laboratory and an object image was shown for 30 seconds in a screen which has $235{\times}197cm2$ size. And second, HUD image was configured from three different positions and an object image was shown for 30 seconds in a screen. EEG, which used ${\alpha}$-wave and ${\beta}$-wave for evaluate an emotional stability, were measured from Fp1, Fp2, F7 and F8 channel based on ten to twenty electrode system. From the result in laboratory, F7 ${\beta}$-wave was shown statistically significant to significance probability of 0.006 and between ${\alpha}$-wave and ${\beta}$-wave were showed a negative correlation(r=-0.190). Also, Both the top of left and the bottom of center were showed lower ${\beta}$-wave than the bottom of right. From the result in railway simulator, Fp1 ${\beta}$-wave was appeared statistically significant as significance probability of 0.033 and it was showed lower ${\beta}$-wave than center. The outcome of this study will be helpful about evaluation of optimal HUD position through correlation between alpha wave and beta wave.

Robust Watermarking for Digital Images in Geometric Distortions Using FP-ICA of Secant Method (할선법의 FP-ICA를 이용한 기하학적 변형에 강건한 디지털영상 워터마킹)

  • This paper proposes a digital image watermarking which is robust to geometric distortions using an independent component analysis(ICA) of fixed-point(FP) algorithm based on secant method. The FP algorithm of secant method is applied for better performance in a separation time and rate, and ICA is applied to reject the prior knowledges for original image, key, and watermark such as locations and size, etc. The proposed method embeds the watermark into the spatial domain of original image The proposed watermarking technique has been applied to lena, key, and two watermarks(text and Gaussian noise) respectively. The simulation results show that the proposed method has higher speed and better rate for extracting the original images than the FP algorithm of Newton method. And the proposed method has a watermarking which is robust to geometric distortions such as resizing, rotation, and cropping. Especially, the watermark of images with Gaussian noise has better extraction performance than the watermark with text since Gaussian noise has lower correlation coefficient than the text to the original and key images. The watermarking of ICA doesn't require the prior knowledge for the original images.

Biotransformation of Fructose to Allose by a One-Pot Reaction Using Flavonifractor plautii ᴅ-Allulose 3-Epimerase and Clostridium thermocellum Ribose 5-Phosphate Isomerase

  • ${\text\tiny{D}}-Allose$ is a potential medical sugar because it has anticancer, antihypertensive, antiinflammatory, antioxidative, and immunosuppressant activities. Allose production from fructose as a cheap substrate was performed by a one-pot reaction using Flavonifractor plautii ${\text\tiny{D}}-allulose$ 3-epimerase (FP-DAE) and Clostridium thermocellum ribose 5-phosphate isomerase (CT-RPI). The optimal reaction conditions for allose production were pH 7.5, $60^{\circ}C$, 0.1 g/l FP-DAE, 12 g/l CT-RPI, and 600 g/l fructose in the presence of 1 mM $Co^{2+}$. Under these optimized conditions, FP-DAE and CT-RPI produced 79 g/l allose for 2 h, with a conversion yield of 13%. This is the first biotransformation of fructose to allose by a two-enzyme system. The production of allose by a one-pot reaction using FP-DAE and CT-RPI was 1.3-fold higher than that by a two-step reaction using the two enzymes.

Effect of Strarting Structures and Intercritical Annealing on Low Temperatures Mechanical Properties of a HSLA Steel (초기조직 및 이상역열처리가 저합금 고강도강의 저온기계적 성질에 미치는 영향)

  • Austenite formation and Low temperatures mechanical properties of HSLA steel of different starting structures have been studied by intercritical annealing(IA). The different starting structures are: ferrite+pearlite(FP1), martensite(M1), cold worked ferrite+pearlite(FP2) and cold worked martensite(M2). In most cases tensile strength and elongation was increased by decreasing the testing temperatures regardless of the IA time. Tensile strength of the cold worked starting structures was higher than that of the non-cold worked starting structures. However not any noticeable difference in elongation was found between two cases. Low temperatures impact properties were affected by the starting structures. Charpy V-notch impact transition temperatures of the M-starting structures were around $-40^{\circ}C$, and those of the FP starting structures were around $-10^{\circ}C$. Impact energy was lower in the cold worked specimens than in the non-cold worked specimens at the same starting structures. DP structure obtained from the M-starting structure has shown superior low temperatures mechanical properties than the DP structure obtained from the FP-starting structure.

Outcome of Locally Advanced Esophageal Cancer Treated with Concurrent Chemo-radiotherapy (국소진행된 식도암에서 동시적 항암화학방사선요법의 결과)

  • Purpose: We investigated the outcome and the prognostic factors of patients with locally advanced esophageal cancer who were treated with concurrent chemo-radiotherapy. Materials and Methods: Two hundred forty six patients with esophageal cancer that were treated by radiotherapy between January 1994 and July 2007. Of these, 78 patients who received radiotherapy of $\geq$45 Gy with concurrent chemotherapy were retrospectively enrolled in this study. We included patients stages IIA, IIB, III, IVA, and IVB with supraclavicular metastasis in the middle/lower esophageal cancer or celiac node metastasis in cervical or upper/middle thoracic esophageal cancer. The median radiation dose was 54 Gy and the combination chemotherapy with 5-FU and cisplatin (FP chemotherapy) was given concurrently with radiotherapy in most patients (88%). Results: The follow-up period ranged from 2 to 117 months (median 14 months). The treatment response of the 54 patients could be evaluated by computerized tomography or endoscopy. A complete response (CR) was observed in 17 patients, whereas a partial response was observed in 18 patients. In patients with a CR, the median recurrence time was 20 months and the first relapse sites constituted a locoregional failure in 3 patients and a distant failure in 7 patients. The 1-, 2-, and 5-year overall survival (OS) rates were 58.9%, 21.7%, and 12.2%, respectively. The median survival period was 14 months. A univariate analysis indicated that the treatment response and cycles of FP chemotherapy were significant prognostic factors for OS. Daily or weekly administration of cisplatin as a radiosensitizer showed a better treatment response than FP chemotherapy. Conclusion: This study has shown that results of concurrent chemo-radiotherapy in patients with locally advanced esophageal cancer is comparable to those of other studies. Daily or weekly cisplatin administration may be considered as an alternative treatment in patients that are medically unfit for FP chemotherapy.
