본 연구에서는 2차원 형광 센서를 이용하여 ALA 생산 공정을 실시간 모니터링하고 오프라인 분석 데이터와 비교, 고찰하였다. 배양액의 pH나 균체의 성장 특성 등 생물공정내의 환경 변화에 따라 형광 특성에 차이를 보였으며 최소배지인 MS8 배지에 IPTG, LA 및 기질 (포도당, 숙신산) 등을 첨가한 후 형광 특성의 변화를 살펴본 결과 균체의 성장이나 세포내 대사현상의 변화를 형광 차이로부터 확인할 수 있었다 2차원 형광센서에 의해 실시간 모니터링 된 형광 스펙트럼데이터는 기질 및 생산물 그리고 세포내 효소 활성 등의 각종 공정 변수와 좋은 상관성을 보였다. 따라서 본 연구에서 사용한 2차원 형광센서는 ALA 대량 생산을 위해 주요한 공정변수를 실시간 모니터링 하는데 매우 효과적이라 할 수 있으며 향후 공정의 제어 및 최적화에도 이용될 수 있다.

The innermost structures of synovium consist of one to three layers of cells generally identified as synovial lining cells(SLC). The present studies were initiated to determine the protein expression patterns of fibroblast-like synovial(FLS) cells derived from the synovia of rheumatoid arthritis. Post-traumatic arthritis(PTA) is one of the most common causes of secondary osteoarthritis, and usually affects younger people. The proteins were separated by two-dimensional polyacrylamide gel electrophoresis and RNA expression investigated by RT-PCR Proteome analyses led to the identification of more than 1,500 protein spots and of 11 differently expressed protein spots among them. Six proteins were down-regulated, and five proteins were up-regulated in ACL-transected synovial tissue. Among these, spots 3 and 8 were identified as cofilin-1 and smooth muscle protein $22-\alpha$, respectively, Therefore, the proteome analysis of synovial tissue is a useful approach to investigate a joint after an injury and can be used to understand the pathogenesis of PTA.
BACKGROUND: Horticultural plant growth under field and/or greenhouse conditions is affected by the climate changes (e.g., temperature, humidity, and rainfall). Therefore investigation of hydroponics on field horticultural crops is necessary for year-round production of the plants regardless of external environment changes under plant factory system with artificial light sources. METHODS AND RESULTS: Common sage (Salvia plebeia), nasturtium (Tropaeolum majus), and hooker chive (Allium hookeri) plants were hydroponically culturing in the plant factory with blue-red-white LEDs (Light-Emitting Diodes) and fluorescent lights (FLs). Leaf numbers of common sage under mixture LED and FL treatments were 134% and 98% greater, respectively than those in the greenhouse condition. In hooker chives, unfolded leaf numbers were 35% greater under the artificial lights and leaf elongation was inhibited by the conventional sunlight compared to the artificial light treatments. Absorption pattern of NO3-N composition in hydroponic solution was not affected by the different light qualities. CONCLUSION(S): Plant factory system with different light qualities could be applied for fresh-leaf production of common sage, nasturtium, and hooker chive plants culturing under field and/or greenhouse. Controlled light qualities in the system resulted in significantly higher hydroponic growth of the plants comparing to conventional greenhouse condition in present.
When perceiving microbe-associated molecular patterns (MAMPs) or plant-derived damage-associated molecular patterns (DAMPs), plants alter their root growth and development by displaying a reduction in the root length and the formation of root hairs and lateral roots. The exogenous application of a MAMP peptide, flg22, was shown to affect root growth by suppressing meristem activity. In addition to MAMPs, the DAMP peptide PEP1 suppresses root growth while also promoting root hair formation. However, the question of whether and how these elicitor peptides affect the development of the vascular system in the root has not been explored. The cellular receptors of PEP1, PEPR1 and PEPR2 are highly expressed in the root vascular system, while the receptors of flg22 (FLS2) and elf18 (EFR) are not. Consistent with the expression patterns of PEP1 receptors, we found that exogenously applied PEP1 has a strong impact on the division of stele cells, leading to a reduction of these cells. We also observed the alteration in the number and organization of cells that differentiate into xylem vessels. These PEP1-mediated developmental changes appear to be linked to the blockage of symplastic connections triggered by PEP1. PEP1 dramatically disrupts the symplastic movement of free green fluorescence protein (GFP) from phloem sieve elements to neighboring cells in the root meristem, leading to the deposition of a high level of callose between cells. Taken together, our first survey of PEP1-mediated vascular tissue development provides new insights into the PEP1 function as a regulator of cellular reprogramming in the Arabidopsis root vascular system.
브라시노스테로이드는 식물의 생장과 발육 과정에 있어서 중요한 역할을 담당 할 뿐 아니라 생물학적/ 비 생물학적 스트레스에 대한 복합 저항성을 보인다고 알려져 있다. 따라서 본 연구에서는 브라시노스테로이드와 광범위스트레스 내성을 연결하는 중요한 생물학적 네트워크를 이해하기 위해, Agilent Arabidopsis $4{\times}44K$ oligo chip을 이용하여 브라시노스테로이드 신호가 강화된 bes1-D 계통의 전 전사체 비교분석을 수행하였다. 그 결과 bes1-D 계통에서 DEGs (Differentially Expressed Genes)를 1,091 (562 up-regulated, 529 down-regulated) 개 선발하였다. 또한 선발된 유전자들의 GO 와 단백질 상호작용 네트워크 분석을 통해 대사, 발달, 스트레스, 면역, 방어 반응에 관련된 주요 브라시노스테로이드 신호전달과 연결된 스트레스 관련 유전자군을 분리하였다. 선발된 유전자중 NB-ARC와 FLS2는 bes1-D 계통이 야생형 En-2 계통에 비해 약 6배 정도의 발현량이 증가되었으며, TIR1, TSA1, OCP3 유전자등은 bes1-D 계통이 야생형 En-2 계통에 비해 발현이 감소되었다. 또한 브라시노스테로이드 활성형 계통이 야생형 식물체 계통에 비해 가뭄 스트레스 및 병원균에 대해 저항력이 향상되었다. 따라서 microarray 분석을 통한 유전자 간 발현 네트워크와 유전체 정보를 결합하여 대단위 주요 기능 유전자들을 동정할 수 있는 방법을 고안하여 실험에 사용하였다. 이를 통해 기능 획득 돌연변이 bes1-D가 식물들이 다양한 스트레스 환경에 적응할 수 있는 반응을 조절한다는 사실을 보여주고 있다.
The age and growth of Pleurogrammus azonus in the coastal of Gangwon-do, East Sea were determined, from monthly samples of commercial catches, caught by the gill net and set net fishery from January to December in 2008. Gonadosomatic index (GSI) began to increase in September, and reached the maximum between November and December. After spawning GSI began to decrease from January. The annuli of P. azonus are formed once a year, with the boundary between opaque and translucent zones forming from December to January. The relationships between fork length (FL) and body weight (BW) were $BW=0.005FL^{3.240}$($R^2=0.963$) for females and $BW=0.006FL^{3.238}$($R^2=0.946$) for males. The FLs at annuli formation in otoliths were back-calculated from the otolith-length relationship and were adjusted to von Bertalanffy growth curves to $FL_t=70.54(1-{\exp}^{(-0.099(t+1.188))})$ for females and $FL_t=51.87(1-{\exp}^{(-0.135(t+1.697))})$ for males. Until the age of 3 years, males grew faster than females; however, from the age of 4 years, females grew faster than males. In the future, we want to study the relationship between early growth and water temperature changes in the East Sea.
1 keV $Ar^+$ 이온빔을 이용하여 polytetrafluoroethylene (PTFE)의 표면을 개질하고 그 위에 $5000\;{\AA}$의 구리 박막을 이온빔 스퍼터링법을 이용하여 증착하였다. 이온빔 조사에 의하여 PTFE의 표면에는 cone이 형성되며 cone의 높이는 이온 조사량이 증가함에 따라 점차로 증가함을 알 수 있었다. x-ray photoelectron spectroscopy (XPS) 분석을 통하여 조사된 PTFE의 표면에서는 이온 조사량이 증가함에 따라 Fls peak의 강도가 감소하며 이는 F 원자의 선택적인 스퍼터링에 기인한 것으로 생각된다. 또한 이온 조사에 의해 생성된 불안정한 사슬들은 crosslinking에 의하여 새로운 C-F 계열의 결합들을 생성하였다. 이온빔 스퍼터링법에 의하여 증착된 구리 박막은 PTFE 표면의 cone을 따라 균일하게 증착되며 PTFE의 표면 거칠기가 증가함에 따라 (111) 방향으로 우선 성장함을 알 수 있었다. 증착된 구리 박막의 비저항은 개질전 PTFE의 $2.7{\mu}{\Omega}cm$에서 $1{\tiems}10^{16}/\textrm{cm}^2$의 이온 조사량으로 개질된 PTFE의 $4.3{\mu}{\Omega}cm$까지 이온 조사량에 따라 점차로 증가하였다. $1{\tiems}10^{17}/\textrm{cm}^2$의 이온 조사량으로 개질된 PTFE 위에 증착된 구리 박막의 갑작스런 비저항 증가는 구리 박막의 단락에 의한 것으로 보인다.
Background: Calcineurin plays a crucial role in T cell activation, cell growth, apoptosis, and angiogenesis, and its over-expression has been implicated in the pathogenesis of cardiomyopathy and stroke. However, the expression and function of calcineurin in the pathologic lesion of chronic inflammatory diseases, like rheumatoid synovium, remain to be defined. This study was aimed to determine the role of calcineurin in inflammatory arthritis and investigate the expression and function of calcineurin in the rheumatoid synovium and synoviocytes, the actual site of chronic inflammation. Methods: Immuno-histochemical staining using specific antibody to calcineurin was perfomed in the synovium of rheumatoid arthritis (RA). Fibroblast-like synoviocytes (FLS) from RA and osteoarthritis (OA) patients were isolated from RA and OA patients, and cultured with IL-1${\beta}$ and TNF-${\alpha}$ in the presence or absence of cyclosporin A, a calcineurin inhibitor. The calcineurin expression was assessed by phosphatase assay and Western blotting analysis. IL-6, -10, -17, matrix metalloproteinase (MMP)-1, -2, -3, and -9 released into the culture supernatants were measured by ELISA. After transfection with GFP-Cabin 1 gene into synoviocytes, the levels of IL-6 and MMPs were measured by ELISA. Results: Calcineurin was highly expressed in the lining layer of synovium and cultured synoviocytes of RA patients. The elevated calcineurin activity in the rheumatoid synoviocytes was triggered by proin flammatory cytokines such as IL-1${\beta}$ and TNF-${\alpha}$. In contrast, IL-10, an anti-inflammatory cytokine, failed to increase the calcineurin activity. The targeted inhibition of calcineurin by the over-expression of Cabin 1, a natural calcineurin antagonist, inhibited the production of IL-6 and MMP-2 by rheumatoid synoviocytes in a similar manner to the calcineurin inhibitor, cyclosporin A. Conclusion: These data suggest that abnormal activation of calcineurin in the synoviocytes may contribute to the pathogenesis of chronic arthritis, and thus provide a potential target for controlling inflammatory arthritis.
정역교잡에 따른 보리의 겉보리와 쌀보리의 $F_1$세대에 대한 잡종강세의 정도와 이들의 유전적 작용을 평가할 수 있는 heterosis나 heterobeltiosis의 검정과 유전력 및 상관관계를 분석한 결과는 다음과 같다. 겉보리$\times$쌀보리, 쌀보리$\times$겉보리의 교배조합에 따른 heterosis는 간장, 수장, 망장, 엽장, 엽폭, 주당수수, 수당립수, 천립중 등의 모든 형질에서 정의 강세율을 보였다. 쌀보리$\times$겉보리의 교배조합이 겉보리$\times$쌀보리의 교배조합보다 heterosis나 heterobeltiosis가 간장, 엽장, 엽폭, 주당수수, 수당립수 등에서 강세의 정도가 다소 높았고, 피과성의 우성정도는 피성이 과성에 대하여 우성이었음을 확인하였다. 그리고 교배조합간의 유전력은 엽폭에서 94.5 %로 가장 높게 나타났으며, 각 조사형 질 대부분 76.7-94.5 %로 높은 수치를 나타내었다. 또한 겉보리와 쌀보리의 정역교배조합간 상관 관계는 정교배의 경우, 염장과 주당수수(0.970$^{**}$ ), 간장과 수당립수$(0.963^{**})$, 망장과 엽장$(0.862^{**})$이 정으로 높은 유전상관을, 역교배의 경우, 간장과 수장$(0.926^{**})$, 망장과 엽장$(0.922^{**})$, 주당수수와 수당립수$(0.713^{**})$가 높은 유전상관을 나타내었다.
