Assessment of Genetic Diversity, Relationships and Structure among Korean Native Cattle Breeds Using Microsatellite Markers

  • Four Korean native cattle (KNC) breeds-Hanwoo, Chikso, Heugu, and Jeju black-are entered in the Domestic Animal Diversity Information System of the United Nations Food and Agriculture Organization (FAO). The objective of this study was to assess the genetic diversity, phylogenetic relationships and population structure of these KNC breeds (n = 120) and exotic breeds (Holstein and Charolais, n = 56). Thirty microsatellite loci recommended by the International Society for Animal Genetics/FAO were genotyped. These genotypes were used to determine the allele frequencies, allelic richness, heterozygosity and polymorphism information content per locus and breed. Genetic diversity was lower in Heugu and Jeju black breeds. Phylogenetic analysis, Factorial Correspondence Analysis and genetic clustering grouped each breed in its own cluster, which supported the genetic uniqueness of the KNC breeds. These results will be useful for conservation and management of KNC breeds as animal genetic resources.

  • Samples of six varieties of soybeans (Bongeui, Danyup-kong, Dongpuktae, Hwangkeum-kong, Jangyup-kong, Kanglim) were analysed for their chemical composition and amino acid content by A.O. A. C. method (1980) and amino acid analyser in order to determine their nutritive value. 1) Chemical composition of soybean was affected by the varieties. The protein contents of 6 varieties of soybeans grown at Suwon were 48.84% in Kanglim, 45.57% in Bongeui, 43.73% in Dongputtae, 41.56% in Jangyup-kong, 40.86% in Hwangkeum-kong, and 40.05% in Danyup-kong. 2) Compared to FAO (1970) chemical score of soybean (A/E ratio and A/T ratio), three varieties of Korean soybeans (Bongeui, Danyup-kong, Hwangkeum-kong) showed higher levels of nutritive value in protein and three varieties of soybeans (Dongpuktae, Jangyup-kong, Kanglim) showed lower levels of nutritive value in protein. From protein content and chemical score, Bongeui showed the greatest potential for the best high protein source among Korean soybean varieties. 3) The determination of first limiting amino acid in six varieties of Korean soybeans by whole egg protein was methionine, but valine was the first limiting amino acid in Danyup-kong and methionine- cystine was other five varieties of Korean soybeans by FAO provisional amino acid pattern.

Intake of Food Additives in Foods by Total Diet (식이를 통한 식품첨가물의 섭취량)

  • This study has been carried out to measure the amount of the daily intake by Korean on preservatives (Dehydroacetic acid, Benzoic acid and ρ-Hydroxybenzoic acid asters) in foods. The amounts of preservatives were determined by HPLC in foods such as butter, cheese, margarine, aloe gel, carbonated beverages, mixed beverage, ginseng beverage, red ginseng drink, soy sauce, fruit and vegetable juices (except unheated fruit-vegetable juice), vineger, seasonings, fruit and vegetables only on the peel, fruit wine, rice wine and unrefined rice wine. The recovery ranges were found to be 99.2% for dehydroacetic Acid, 95.8% for benzoic acid and $94.2{\sim}97.2%$ for ${\rho}-hydroxybenzoic$ acid in foods, respectively. The data of average food intake for each food items per capita per day were obtained from the report of national nutrition survey carried by the Ministry of Health and Welfare in 1997. The detected number of samples and average intake range of preservatives have been figured cut to $ND{\sim}290.0{\;}ppm$ for dehydroacetic acid, $ND{\sim}400.0{\;}ppm$ for benzoic acid and $ND{\sim}93.9{\;}ppm$ for ${\rho}-hydroxybenzoic$ acid esters, respectively. Estimated daily intake (EDI) of each additives per capita per day were shown as follows; 1.56 mg for dehydroacetic acid, 2.25 mg for benzoic acid and 0.44 mg for ${\rho}-hydroxybenzoic$ acid esters in total respectively and these values were evaluated to be much lower than that of FAO/WHO's acceptable daily intake (ADI) $0{\sim}5{\;}mg/kg$ b.w./day for benzoic acid and $0{\sim}10{\;}mg/kg$ b.w./day for ${\rho}-hydroxybenzoic$ acid esters.

Residues of Endosulfan and its Metabolites in a Rice Field of Madurai, India

  • Thirty-day-old rice seedlings, IR-20 variety were transplanted into the experimental plots and were sprayed with endosulfan (35% EC) after 52 days at the rate of 0.64 kg a.i/ha. Residues of endosulfan in the plant, soil, and water were found to decrease steadily upto 15 days. A second application of the pesticide was made on the 31st day and the plant was harvested on 56th day after the first application. The residue level on hay and grains was 0.7 ${\mu}g/g$. This level is seven times higher than FAO/WHO-prescribed tolerance level of 0.1 ${\mu}g/g$. Metabolites of endosulfan were traced out in plants, soil, and water during the pre- and post-harvest period.

Evaluation of Energy Consumption in Heat Treatment of Pine Log (소나무 원목의 열처리 소요 에너지 평가)

  • The required energy for the heat treatment of pine log was evaluated in this study. A proper heat treatment of pine log infected by pinewood nematode (Bursaphelenchus xylophilus) can prevent spreading of the infection by pinewood nematode and make the infected pinewood valuable again. The FAO (Food and Agriculture Organization of the United Nations) heat treatment standard for various types of infected wood for which a heat treatment of the core part of the wood is necessary is 30 minutes at $56^{\circ}C$, taking into account the international standards for phytosanitary measures (ISPM No. 15). In this study, the energy consumption during the heat treatment was separated into two kinds of energy, initial energy for heating kiln drier and to reach set point temperature and relative humidity and the required energy supplementing heat loss. The initial required energy per unit time is greater than that during the treatment. The energy consumption per unit time varied little during the heat treatment. As a result, the set point relative humidity with set dry bulb temperature and density of wood dependent on moisture content are very important factors to change energy consumption in the experiment. The heat treatment at higher temperature and higher humidity levels requires more energy consumption but less treatment time. It is expected that a more effective energy program could be planed for the heat treatment of pine log through this study.

Residue Analysis to Establish an Index for the Safety Use of Propineb (Propineb의 안전사용(安全使用) 기준(基準) 설정(設定)을 위한 잔류분석(殘留分析))

  • Residue of Propineb in Sesame(Sesamum indicum L.) seed was determined to establish an index for the safety use of Propineb to Sesame. Evaluation was made on residual concentration of Propineb in Sesame seed as a function of application frequency and date when the mixed formulations of Propineb(56%), protectant fungicide, and Oxadixyl(8%), contact fungicide, were sprayed into Sesame leaves. Level of Propineb treatment was $0.028g/m^2$ with various combinations of application time from three to sixty days before harvest. Results are summarized as followings. 1. Recovery percentages of Propineb from Sesame seed were ranged from 84 to 96, and the minimum detectable limit of Propineb with the method employed in this experiment was 0.03mg/kg. 2. Residues of Propineb in Sesame seed were in the ranges of 0.14 to 1.38mg/kg, varying with frequency and date of Propineb application. 3. Residues of Propineb increased as increasing application frequency of Propineb or as being application date closer to harvest time. 4. Residue of Propineb in Sesame seed was decreased with time, showing to be fitted to the first-order kinetics. 5. Residues of Propineb in Sesame seen were, irresepective of treatments, lower than 2mg/kg, the Maximum Residue Limit(MRL) established by FAO/WHO. 6. Half-life of Propineb determined in this experiment was ranged from 12 to 16 days.

Trace Metal Contents in Fishes and Shellfishes and Their Safety Evaluatins (우리나라 어패류중 미량금속 함량 및 안전성 평가)

  • This study was carried out to estimate the contents of trace metals in the fishes and shellfishes available on Korean markets. The maples were digested with acids, then analyzed by ICP (INductively Coupled Plasma Spectrometer) and AAS (Atomic Absorption Spectrometer) for the contents of lead (Pb), cadmium (Cd), arsenic (As), copper (Cu), manganese (Mn) and zinc (Zn). The contents of mercury (Hg) were determined using a mercury analyzer. The contents of trace metals [minimum~ maximum (mean), mg/kg] in fishes were as follows; Hg;0.004~0.500(0.082), Pb;N.D.~ 1.87 (0.29), Cd;N.D. ~0.094 (0.020), As;0.01~5045 (0.84), Cu;0.04~3.74 (0.66), Mn; N.D. ~7.56 (0.57), Zn;0.92~18.33(6.03). Those contents in shellfishes were as follows (mg/kg); Hg;N.D.~0.221 (0.029), Pb;0.01~1.51 (0.38), Cd;0.02~1.93 (0.51), As;0.18~3.07 study showed that trace metal contents in both fishes and shellfishes on our markets of lead, cadmium and mercury from fishes and shellfishes takes about 1~13% of PTWI(Provisional Tolerable Weekly Intakes) that FAO/WHO Joint Food Additive and Contaminants Committee has set to evaluate their safeties.

Validation of Net Radiation Measured from Fluxtower Based on Eddy Covariance Method: Case Study in Seolmacheon and Cheongmicheon Watersheds (에디공분산 방법 기반의 플럭스 타워 순 복사에너지 검증: 설마천, 청미천 유역)

  • The necessity of clear understanding of water and energy cycles has been attracted recently due to the climate change. The micrometeorological flux tower networks play a role of cornerstone of the hydrological and ecological analyses. Although the eddy covariance techniques used for flux tower have been proven to be applicable for estimation of latent heat flux, the raw data are often underestimated and needs to be corrected. Among several methods, the Bowen ratio is recognized as the most useful method in which the net radiation and other flux data (Ground heat flux, Sensible heat flux) are used and needed to be validated. In this study, in order to validate the net radiation from flux tower in Seolmacheon and Cheongmicheon watersheds, we compare it with two version of calculated net radiation: (1) FAO 56 Daily net radiation proposed by Allen et al. (1998). (2) Instantaneous net radiation proposed by Bastiaanssen (1995). The results showed that the net radiation from the flux data had similar tendency with those calculated based on physical theory. In addition, after it was applied to Bowen ratio method, the corrected latent heat flux was considerably improved with making the energy balance much more closed.

Studies on the Nutritional Quality of Rapeseed Protein Isolates (평지(平地)씨 단백질(蛋白質)의 영양가(營養價)에 관(關)한 연구(硏究))

  • Protein efficiency ratio (PER), net protein utilization (NPU), nitrogen digestibility (ND), organ weights, and amino acid composition were investigated for rapeseed protein isolates (RPI) prepared by alkaline countercurrent extraction and isoelectric precipitation of defatted rapeseed meal (DRM). The PER values for 3 kinds of RPI obtained at pI of 6.7, 5.6 and 5.0 were 2.8, 3.1 and 2.9, and for DRM 2.5 while the NPU values for these RPI appeared to be 68, 73 and 71 %, and for DRM 56 %, respectively. Mean ND (85%), food intake (87), and weight gain (37.6) for RPI were significantly different from those of DRM (71 %, 77, and 28, respectively). There was no signicant difference in the PER among three RPI and casein (3.0), nor in the NPU among those proteins and casein (74%). These data attributed to the favorable amino acid patterns of proteins isolated which contained balanced essential amino acids in proportions which meet the requirements of human adult(FAO/WHO, 1973).

Development of Agricultural Groundwater Usage Model Considering Multipurpose Water in Jeju Island (다목적 용수를 고려한 제주도 농업용 지하수 이용량 모델 개발)

  • The estimation of groundwater usage in Jeju island is important to understand hydrologic cycle system and to plan management of water resource because large amounts of groundwater have been used for agricultural and domestic purpose. The model has been developed to estimate agricultural groundwater usage for garlic at uplands and citrus at orchards raising outdoors using the soil water balance model from FAO 56, respectively. The total amount of water supplied for the crop evapotranspiration and the multipurpose function such as sprout promotion can be simulated by the model. However, due to the discrepancy of water use in initial stage between calculated and observed, the model was calibrated and verified using actual groundwater usage monitoring data for 3.5 years (2011.6 to 2014.12) at three uplands for garlic and three orchards for citrus. Consequently, it would be concluded that the model simulated efficiently actual water usage in that root mean square (RMS) and normalized RMS of the validation stage were less than 8.99 mm and 2.43%, respectively, in two different conditions.
