Comparison between Positive and Negative Ion Mode FAB CAD MS/MS Spectra of Linkage-Isomeric Oligosaccharides

  • Negative ion fast atom bombardment (FAB) mass spectra were found to allow the determination of the linkage positions in a series of underivatized linkage-isomeric oligosaccharides. A previous work (Laine et al., 1988) reported that ion patterns of linkage-isomeric trisaccharides could be distinguished by a positive ion. Negative ion FAB collison-activated dissociation (CAD) mass spectrometry (MS) spectra of trisaccharides exhibited better sensitivity than the positive ion mode and provided specific fragmentation patterns according to the linkage positions. Especially, the fragmentations, m/z 205 in F6 and m/z 221 in G6, not occuring in 1-3 or 1-4 linkage. were an indication of 1-6 linkage, by changing collision energies from + 10 eV to +60 eV. The survival ratios of molecular ions in each collision energy set gave support to previous results in which the order of bond stability was 1-6>1-4>1-3 linkage.

  • For the investigation of medicinal resources in Crataegus species, a study was carried out to clarify the constituents in the leaves of Crataegus Pinnatifida var. pubescens (Rosaceae), of which fruits have been used to stomachic, digestive, astringent, analgesic, and cardiotonic, etc. as a folk medicine in Korea. From the n-BuOH fraction of MeOH extract two flavone glycosides, vitexin(apigenin-8-C-$\beta$-D-glucopyranoside) and 2"-O-rhamnosyl vitexin(apigenin-8-C-$\alpha$-L-rhamnosyl-(1$\rightarrow$2)-$\beta$-D-glucopyrano side) were isolated and identified on the basis of their physico-chemical properties and spectroscopic evidences(UV, IR, NMR, FAB-Mass etc.) in comparision with authentic samples.amples.

Comparison of Self-living Ability, Obesity Indices and Nutrient Intake according to Physical Fitness among the Elderly in Rural Areas (농촌 지역 일부 노인에서 신체 기능 정도에 따른 활동 제한도, 비만도 및 영양소 섭취 수준의 변화 양상 비교)

  • Aging is associated with decreased energy expenditure, thermogenesis and energy requirements. Maintenance of physical fitness of the elderly has been reported to reduce the rate at which the basal metabolic rate, muscle strength, skeletal muscle mass and bone density deteriorate. Skeletal muscle disease is known to increase the risk of physical disability and psychological problems. This study was conducted to investigate changes in disability, emotional problems, body compositions, obesity indices and nutrient intake levels according to physical fitness with the elderly in rural areas. According to the results, physical fitness was negatively related with Activities of Daily Living(ADL, p<0.05), Instrumental Activities of Daily Living(IADL, p<0.001), Body Mass Index(BMI, p<0.001) and abdominal obesity(p<0.05), while it showed a positive correlation with the General Self Efficacy Scale(GSES)(p<0.001) and nutrient intake(p<0.05). When changes in these factors were compared according to the range of quartile of the Fullerton Advanced Balance Scale(FAB Scale), GSES(Q1=35.3, Q2=43.5, Q3=53.2, Q4=51.9, p<0.001), BMI(Q1=36.1%, Q2=34.7%, Q3=33.2%, Q4=28.6%, p<0.01), abdominal obesity(Q1=1.02, Q2=0.99, Q3=0.97, Q4=0.94, p<0.001) and nutrient intake(Q1=71.1%, Q2=75.4%, Q3=80.6%, Q4=80.2%, p<0.05) differed significantly. Taken together, these results suggested that better physical fitness would lead to a reduction in negative factors including physical disability and obesity indices, but to an increase in positive factors such as GSES and nutrient intake. The results of this study are expected to be used as basic data for the development of programs to promote the health of the elderly in a local society.

Isolation of 3-Galloylprocyanidin B3, a Glucosyltransferase Inhibitor from the Korean Green Tea Leaves

  • In the course of surveying the anti-plaque agents for dental caries prevention, the extract of Korean green tea leaves (KGTL) was tested for inhibitory activity against Streptococcus mutans adhering to glass surfaces in the presence of crude glucosyltransferase (GTase). The extracts of KGTL showed a positive inhibitory activity against GTase. The active compound was purified through Sephadex LH-20 and MCI gel CHP-20P columns. A positive reaction was shown in the anisaldehyde-$H_2SO_4$ test, which confirmed the condensed tannin. The inhibitory compound was identified as 3-galloylprocyanidin $B_3$ through IR, negative FAB-mass, and $^{1}H$-NMR spectroscopic analyses. Acetone extract and 3- galloylprocyanidin $B_3$ of KGTL showed inhibitory effect against GTase. The percent of inhibition was determinated to be 71.84% (P<0.01) with 10 mM 3-galloylprocyanidin B3. The 3-galloylprocyanidin $B_3$, which possessed a galloyl, showed a higher inhibitory activity against glucosyltransferase than monomeric (+)-catechin and procyanidine $B_3$ which had no galloyl group.

An Antifungal Antibiotic Purified from Bacillus megaterium KL39, a Biocontrol Agent of Red-Pepper Phytophthora-Blight Disease

  • Bacillus megaterium KL39, an antibiotic-producing plant growth promoting rhizobacterium (PGPR), was selected from soil. The antifungal antibiotic, denoted KL39, was purified from culture filtrate by column chromatography using Dion HP-20, Silica gel, Sephadex LH-20, and prep-HPLC. Thin layer chromatography, employing the solvent system of ethanol:ammonia:water=8:1:1, showed the $R_{f}$. value of 0.32. The antibiotic KL39 showed a negative reaction with ninhydrin solution, positive with iodine vapor, and also positive with Ehrlich reagent. It was soluble in methanol, ethanol, butanol, and acetonitrile, but insoluble in chloroform, toluene, hexane, ethyl ether, or acetone. Its UV spectrum had the maximum absorption at 208 nm. Amino acid composition, FAB-mass, $^{1}H-NMR,\;^{13}C-NMR$, and atomic analyses showed that the antibiotic KL39 (MW=1,071) has a structure very similar to iturin E. The antibiotic KL39 has a broad antifungal spectrum against a variety of plant pathogenic fungi including Rhizoctonia solani, Pyricularia oryzae, Monilinia froeticola, Botrytis cinenea, Altenaria kikuchiana, Fusarium oxysporum, and F. solani. An MIC value of $10\;{\mu}g/ml$ was determined for Phytophthora capsici. Macromolecular incorporation studies with P. capsici using radioactive [$^{3}H-adenine$] as the precursor, indicated that the antibiotic KL39 strongly inhibits the DNA biosynthesis of the fungal cell. Microscopic observation of the antifungal action showed abnormal hyphal swelling of P. capsici. The purified antibiotic KL39 was very effective for the biocontrol of in vivo Phytophthora-blight disease of pepper.

Isolation and Structural Determination of Anti-Helicobacter pylori Compound from Fungus 60686. (곰팡이에서 분리한 Helicobacter pylori 항균물질의 분리 및 구조규명)

  • Helicobacter pylori is a Gram-negative bacterium which causes chronic gastritis and is associated with gastric ulcer, duodenal ulcer and gastric carcinoma. In the process of screening of antibacterial activities against H. pylori from soil microorganisms, fungus No. 60686 was isolated. After fermentation of No.60686, the antibacterial compound was isolated, purified and followed by extraction of mycelium with organic solvents, acetone and ethyl acetate, through silica gel chromatography, LH-20 gel chromatography and HPLC. As a result of the structural analyses of the compound by IR, $^1$H- and $^{13}$C-NMR, FAB/Mass spectrophotometer, the compound having the antimicrobial activity was identified as chaetoglobosin A ($C_{32}H_{36}N_2O_5$), a cytochalasan derivative. The antimicrobial activity of chaetoglobosin A was tested against Gram-positive and negative bacteria by paper disk method. Among the test strains of 9 Gram-positive bacteria and 18 Gram-negative bacteria containing 4 H. pylori strains, the growth of 4 H. pylori strains and 3 S. aureus strains (SG 511, 285 and 503) was only inhibited by chaetoglobosin A. Also it was shown that its growth inhibition against H. pylori strains was stronger than that against S. aureus strains at the treatment of the same concentration. Therefore it was concluded that chaetoglobosin A has a specific growth inhibition against H. pylori of the tested bacteria.
