Influence of Wheat Straw Pelletizing and Inclusion Rate in Dry Rolled or Steam-flaked Corn-based Finishing Diets on Characteristics of Digestion for Feedlot Cattle

  • Eight Holstein steers ($216{\pm}48kg$ body weight) fitted with ruminal and duodenal cannulas were used to evaluate effects of wheat straw processing (ground vs pelleted) at two straw inclusion rates (7% and 14%; dry matter basis) in dry rolled or steam-flaked corn-based finishing diets on characteristics of digestion. The experimental design was a split plot consisting of two simultaneous $4{\times}4$ Latin squares. Increasing straw level reduced ruminal (p<0.01) and total tract (p = 0.03) organic matter (OM) digestion. As expected, increasing wheat straw level from 7% to 14% decreased (p<0.05) ruminal and total tract digestion of OM. Digestion of neutral detergent fiber (NDF) and starch, per se, were not affected (p>0.10) by wheat straw level. Likewise, straw level did not influence ruminal acetate and propionate molar proportions or estimated methane production (p>0.10). Pelleting straw did not affect ($p{\geq}0.48$) ruminal digestion of OM, NDF, and starch, or microbial efficiency. Ruminal feed N digestion was greater (7.4%; p = 0.02) for ground than for pelleted wheat straw diets. Although ruminal starch digestion was not affected by straw processing, post-ruminal (p<0.01), and total-tract starch (p = 0.05) digestion were greater for ground than for pelleted wheat straw diets, resulting in a tendency for increased post-ruminal (p = 0.06) and total tract (p = 0.07) OM digestion. Pelleting wheat straw decreased (p<0.01) ruminal pH, although ruminal volatile fatty acids (VFA) concentration and estimated methane were not affected ($p{\geq}0.27$). Ruminal digestion of OM and starch, and post-ruminal and total tract digestion of OM, starch and N were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal NDF digestion was greater (p = 0.02) for dry rolled than for steam-flaked corn, although total tract NDF digestion was unaffected (p = 0.94). Ruminal microbial efficiency and ruminal degradation of feed N were not affected (p>0.14) by corn processing. However, microbial N flow to the small intestine and ruminal N efficiency (non-ammonia N flow to the small intestine/N intake) were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal pH and total VFA concentration were not affected ($p{\geq}0.16$) by corn processing method. Compared with dry rolled corn, steam-flaked corn-based diets resulted in decreased acetate:propionate molar ratio (p = 0.02). It is concluded that at 7% or 14% straw inclusion rate, changes in physical characteristics of wheat straw brought about by pelleting negatively impact OM digestion of both steam-flaked and dry-rolled corn-based finishing diets. This effect is due to decreased post-ruminal starch digestion. Replacement of ground straw with pelleted straw also may decrease ruminal pH.

근관치료시 Nd:YAG Laser 사용과 세척액에 따른 치근단 폐쇄효과의 비교 (THE EFFECTS OF ND:YAG LASER AND IRRIGANTS ON CANAL SEALING ABILITY)

  • The application of Nd:YAG laser and irrigants to the root surface can change its surface configurations. The purpose of this study was to investigate the effects of Nd:YAG laser and irrigants on the apical seal of obturated canals. In this study, 66 single rooted teeth were randomly assigned to 4 group of 14 teeth each. 8 teeth were served us positive and negative controls. The teeth were divided into 6 groups as follows. Group A: Nd:YAG laser, 5% NaOCl + Rc-prep Group B: Nd:YAG laser, Saline Group C: 5% NaOCl + Rc-prep Group D: Saline Group E: Positive control Group F: Negative control 66 teeth were instrumented using Maillefer ProFile$^{\circledR}$ (Orifice Shapers, .04 taper, .06 taper Dentsply, Switzerland). Two of each group were selected at random, and the canal wall surfaces were examined under a SEM. 12 teeth of each group were obturated using by lateral condensation technique. Specimens were immersed in india ink for 7days, decalcified by 10% nitric acid, dehydrated by 75. 80. 85, 90, 95 and 100% alcohol in order cleared by methyl salicylate and then measured of dye penetration with stereomicroscope($\times$15 magnification) and Image Pro plus. The data were analyzed statistically by one-way ANOVA test and Duncan's Multiple Range test. The results were as follows : 1. The mean leakage was 0.128$\pm$0.376 for group A, 0.237$\pm$0.325 for group B, 0.397$\pm$0.468 for group C, 0.586$\pm$0.402 for group D, and there were statistically significant differences between group A and group D, group B and group D. (p<0.05). 2. Group A had better sealing ability than Group C, but there was statistically no significant differences. (p>0.05). 3. Group B had better sealing ability than Group D and there was statistically significant difference. (p<0.05). 4 Group A had better sealing ability than Group B, but there was statistically no significant difference. (p>0.05). 5. Group C had better sealing ability than Group D, but there was statistically no significant difference. (p>0.05). 6. As a result of observation under SEM, Smear layers were removed in Group A, B. but Smear layers were partially removed and smear plugs were remained in Group C, Smear layers were not removed in Group D. To be specially, Melting of smear layer were showed in Group C. 7. These results suggests that the laser has a potential in reducing the apical microleakage of obturated canals.

GC 및 GC/MS를 이용한 벌꿀 및 시판 음료 중의 Fructose, Glucose 및 Sucrose의 동시분석 (Simultaneous Determination of Fructose, Glucose, and Sucrose in Honey and Commercial Drinks by GC and GC/MS)

  • 본 연구는 GC 및 GC/MS에 의한 벌꿀과 시판음료 중에 함유되어 있는 fructose, glucose, sucrose의 TMS 및 TMSO 유도체화를 통한 동시분석방법을 확립하고자 하였다. 3종 당류를 분석하기 위한 동시분석방법으로 당류의 TMSO 유도체화가 TMS 유도체화 방법보다 정성 및 정량성이 우수한 것으로 나타났으며, TMSO화된 당류의 GC 최적분석조건으로 초기 oven온도 $175^{\circ}C$에서 5분간 머무르게 하다가 $10^{\circ}C/min$$280^{\circ}C$까지 승온시키는 조건으로 3종의 당류를 19분 이내에 분석할 수 있었다. GC 분석에서 fructose와 glucose 그리고 sucrose의 직선성은 50-5000 ${\mu}g/mL$ 농도 범위 내에서 $r^2$값이 모두 0.9999 이상이었으며, 회수율은 각각 100.5, 101.0, 99.7%이였으며, 검출한계와 정량한계는 0.68, 0.47, 0.53 ${\mu}g/mL$와 2.27, 1.58, 1.77 ${\mu}g/mL$이었다. 일내 정확도와 정밀성은 fructose, glucose, sucrose에서 각각 99.0-100.0%와 0.47-1.06%, 일간 정확도와 정밀성은 100.1-101.2%와 1.09-1.79%로 나타났다. 위와 같은 우수한 분석 값은 GC/MS에 의한 분석에서도 유사하게 나타났다. 확립된 GC 분석방법으로 아카시아 벌꿀 시료 12종의 당 함량을 조사한 결과, 아카시아 벌꿀의 fructose와 glucose 평균함량은 각각 $42.58{\pm}1.97%$, $27.74{\pm}1.16%$이었으며, F/G ratio는 $1.53{\pm}0.07$로 HPLC로 분석한 값과 거의 동일하였다. 그러나 sucrose 함량은 GC분석에서는 평균함량이 $0.79{\pm}0.52%$였으나, HPLC $NH_2$ column을 사용 시 $2.24{\pm}4.62%$를 나타내었고, carbohydrate column으로 분석했을 때는 분리도의 저하로 몇 개의 시료에서는 정확한 함량분석에 어려움이 있었다. 따라서 본 연구에서 실시한 GC, GC/MS에 의한 fructose, glucose, sucrose의 TMSO 유도체 동시분석방법은 감도, 분리도, 회수율, 직선성, 정밀성, 정확도면에서 우수한 정량분석방법이라고 판단된다.

중학교 남학생들의 건강관리 실천과 이에 영향을 미치는 요인 조사 연구 (A Study OR Investigation of the Factors having Affect on Junior Highschool Boys관 Practice of Health Care)

  • Accepting the health as the fundamental human right, the nation and society came to admit the duty to give it to all the people. Korean government is expanding the Primary Health Care as one of the policies for developing the people's health by the lead of community. Like this current situation the School Health Service which is the center of community Health Service schould be active to keep, promote and maintain the health of students and teachers. This investigation was attempted to help to establish the basis of the Health Education Program which would perform the health education efficiently and bring the reforming of student's health control, by measuring the degree of junior high school student's practice concerning health care and con-firming the relationship of knowledge, concern and level of health. The subjects for this study were made up 296 boys at the third grade in a school, located in Seoul. The tool for this study was questionaire. Data were collected for five days, September 22∼26, 1983. The investigators explained the students how to answer the questions of the survey questionaire and then collected the survey cards immediately. The data were analyzed by means of percentages, Pearson Correlation Coefficient and Stepwise Multiple Regression. The results of this study are as follows. 1. The subjects' reaction to the practice concerning Health Care; According to the average proportion of practice concerning health care, subjects' practice in the aspect of Infectious Disease Care is the highest 82.4% and they showed their practice in the aspect of the accident prevention by 77.5% and in the aspect of mental health by 74.8%. Their practice in the aspect of personal hygiene and daily lifehabit is the lowest 71.2%. 2. Health Knowledge; The whole mark distribution of health knowledge is ranged from the lowest 4 point to the highest, 30 point, therefore point range is 26.0. The Average point is 16.58. 11.2% of students solved more than two thirds of personal knowledge levels. 81.4% of students did more than one third of them. 7.415 of students did less than one third of them. 3. Health Concern; The Students of the average 3.99 point respond positively to the question about the health concern, ‘They think health is more important than money’, this is the highest rate. The students of the average 2.78 respond“when they are sick, they enter the appointed hospital where they own choose”, while it is the lowest. 4. Subjects' response to the health level are at follow: Very healthy 26.0%, healthy 47.0%, less healthy 10.5%, 34.9% of them have ever been sick within two weeks, the number of symptoms they reported amount to 114 and the number of cases poi one person is 0.35. 5. The hypotheses test about the practice concerning health care and the factor which effect on it. 1) The main first hypothesis:“The more knowledge of health the subjects have, the better they practice health care.”was accepted. (r=0.1582, p <0.05) 2) The minor first hypothesis:“The more interest in health subjects have, the better they practice”was accepted. (r=0.4354, p <0.001) 3) The minor second hypothesis;“The healthier subjects are, the better they practice health care”was accepted. (r=0.1069, p<0.05) As other test, partical correlation test is performed in other refine whether health knowledge, a fact influencing the students practice concerning health care, is associated with the practice after controlling the third variables. First, after controlling health concern, the correlation of health knowledge and practice concerning health care was kept. (r=0.1347, p <0.005) Second, after controlling health level, the correlation of health knowledge and practice concerning health care was kept. (r=0.1526, p <0.005) And finally, after controlling economic state, the correlation of health knowledge and practice concern-ing health care was kept. (r=0.1413, p <0.05) Additionally Stepwise Multiple Regression between practice concerning health care and variables. 1.6591 of compliance was explained with the know-ledge (F=5.584, p <0.05), 20.0% of compliance was explained with the health concern added to knowledge. (F=63,213, p <0.005) As the above, health knowledge obviously have effects on the practice about the health care. But, contrary to researcher's expectation, health concern has more affects than health knowledge. Therefore, we must grope the plan to enhance health concern through the regular curriculum and systematic health education for students. Besides, we must study further on, to find the factors which have affect on the practice concerning health care.

골 성숙도 결정 시 혈청 IGF-1, IGFBP-3 농도와 수완부 방사선 사진의 상관관계 (Correlation between Serum IGF-1, IGFBP-3 Levels, and Hand-Wrist Radiographs in Determining Skeletal Maturity)

  • 이 연구의 목적은 Skeletal maturity indicator (SMI) 및 Middle phalanx of the third finger (MP3)와 혈청 Insulin-like growth factor-1 (IGF-1) 및 Insulin-like growth factor binding protein-3 (IGFBP-3)의 상관 관계를 통해 골격적 성숙도의 예측 지표로 사용될 수 있는지 알아보는 것이다. 2018년 1월부터 2022년 12월까지 아주대학교 치과병원 및 소아청소년과에 내원한 만 7세 이상 17세 이하 환자 205명의 의무기록을 후향적으로 분석하였다. 혈청 IGF-1 농도는 SMI 6 - 8, MP3 - G에서 가장 높았고(p < 0.0001) 혈청 IGFBP-3 농도는 SMI 9 - 10, MP3 - I에서 가장 높았다(p = 0.010, 0.030). SMI, MP3와 혈청 IGF-1 농도 간 비교적 높은 Pearson 상관계수를 나타냈다(r = 0.698, 0.622, p < 0.0001). 혈청 IGF-1는 소아청소년의 골격적 성숙도를 평가하는 보조적 수단으로 활용될 수 있을 것이며 임상적으로는 472 ㎍/L에서 510.63 ㎍/L의 범위를 최대성장기로 고려할 수 있다.

Prediction of Digestible and Metabolizable Energy Content and Standardized Ileal Amino Acid Digestibility in Wheat Shorts and Red Dog for Growing Pigs

  • Two experiments were conducted to evaluate the effects of chemical composition of wheat shorts and red dog on energy and amino acid digestibility in growing pigs and to establish prediction models to estimate their digestible (DE) and metabolizable (ME) energy content and as well as their standardized ileal digestible (SID) amino acid content. For Exp. 1, sixteen diets were fed to thirty-two growing pigs according to a completely randomized design during three successive periods. The basal diet was based on corn and soybean meal while the other fifteen diets contained 28.8% wheat shorts (N = 7) or red dog (N = 8), added at the expense of corn and soybean meal. Over the three periods, each diet was fed to six pigs with each diet being fed to two pigs during each period. The apparent total tract digestibility (ATTD) of energy in wheat shorts and red dog averaged 75.1 and 87.9%. The DE values of wheat shorts and red dog averaged 13.8 MJ/kg (range 13.1 to 15.0 MJ/kg) and 15.1 MJ/kg (range 13.3 to 16.6 MJ/kg) of dry matter, respectively. For Exp. 2, twelve growing pigs were allotted to two $6{\times}6$ Latin Square Designs with six periods. Ten of the diets were formulated based on 60% wheat shorts or red dog and the remaining two diets were nitrogen-free diets based on cornstarch and sucrose. Chromic oxide (0.3%) was used as an indigestible marker in all diets. There were no differences (p>0.05) in SID values for the amino acids in wheat shorts and red dog except for lysine and methionine. Apparent ileal digestibility (AID) and SID values for lysine in different sources of wheat shorts or red dog, which averaged 78.1 and 87.8%, showed more variation than either methionine or tryptophan. A stepwise regression was performed to establish DE, ME and amino acid digestibility prediction models. Data indicated that fiber content and amino acid concentrations were good indicators to predict energy values and amino acid digestibility, respectively. The present study confirms the large variation in the energy content and amino acid digestibility in wheat shorts and red dog, and describes the factors that influence this variation and presents equations based on chemical composition that could probably be used to predict the DE and ME values as well as the amino acid digestibility of wheat shorts and red dog.

냉장실의 온도 정온화가 냉장 식품의 품질과 미생물의 생육에 미치는 영향 (Effects of Temperature and Fluctuation Range on Microbial Growth and Quality of Foods Stored in Domestic Refrigerator)

  • 가정용 냉장고의 냉장실 온도는 통상 $4^{\circ}C$를 적용하고 있으나, 냉장온도를 $2^{\circ}C$로 낮추고, 온도 편차를 줄일 경우 저장효과의 향상 정도를 비교하기 위하여, 각각 다른 조건에 저장한 과일과 어패류의 품질을 비교 측정하고 아울러 냉장온도에서 생육이 가능한 미생물의 생육정도를 비교하였다. 냉장 온도를 $2^{\circ}C$로 낮추고 온도 편차를 ${\pm}0.3^{\circ}C$ 정도로 낮춘 경우 사과의 경도는 저장 14일째까지 크게 변화하지 않았으나, $4^{\circ}C$의 냉장고에서는 7일 이후 경도가 서서히 감소하는 것으로 나타났으며, 도미의 근단백질 분해 정도 또한 $2^{\circ}C$에서는 느리지만, 저장온도가 높거나 온도편차가 심할 경우 근단백질의 분해가 빨리 나타나 도미의 신선도가 빨리 저하됨을 보여 주었다. 굴의 저장 중 미생물수의 변화는 $2^{\circ}C$에서는 저장 9일째까지 균수의 증가를 보이지 않은 반면, 저장온도가 $4^{\circ}C$인 경우 저장 5일 이후 급격한 증가를 보였고, pH 저하 또한 $2^{\circ}C$에서는 비교적 완만하였으나 $4^{\circ}C$에서는 급격히 저하되었다. Listeria monocytogenes는 우유 중에서는 $2^{\circ}C$에서도 성장하였으나, 햄에서는 냉장온도가 $2{\pm}0.3^{\circ}C$인 경우 증식이 억제되어 생존 균수가 감소하였고, $4{\pm}1.2^{\circ}C$인 경우 약간 성장하였고, 온도 편차가 보다 심한 $4{\pm}4^{\circ}C$에서는 성장이 지속되어 생존균수의 증가를 보여 주었다. 따라서 냉장온도를 $2^{\circ}C$로 낮추고 온도의 편차를 줄임으로써 과일이나 어패류의 저장기간을 보다 연장시키고 유해 미생물의 증식이 상당히 억제될 수 있으리라 기대된다.

생물방제균 Pseudomonas sp. 3098이 생산하는 Chitinase의 정제 및 특성 (Purification and Characterization of Chitinase from Antagonistic Bacteria Pseudomonas sp. 3098.)

  • 길항미생물로 분리.동정된 Pseudomonas sp. 3098이 생산하는 chitinase를 황산암모늄 침전, DEAE-cellulose column chromatography, Bio-Gel P-100에 의한 겔여과, 1차 및 2차 hydroxyapatite column chromatography 과정을 거쳐 회수율 5.8%,정제도 15.7배의 정제효소를 얻었고, 효소의 순도는 SDS-PAGE로 확인하였으며, 분자량은 45kDa으로 추정되었다. 정제된 chitinase의 최적 온도와 pH는 45$^{\circ}C$와 5.0이었고, 정제효소는 pH 5.0~9.0 사이에서 안정하였고, 5$0^{\circ}C$, 3시간 및 6$0^{\circ}C$, 30분까지는 비교적 안정하였다. 금속염 및 화학물질의 영향을 조사한 결과 Fe$^{2+}$, Ag$^{1+}$ 및 단백질변성제인 Hg$^{2+}$ 이온에 의해 효소활성이 크게 저해되었고, p-CMB, iodoacetic acid, urea, 2,4-DNP 및 EDTA에 의해 효소활성이 약간 저해되었다. 기질특이성을 조사한 결과 colloidal chitin 및 shrimp shell 유래의 chitin은 분해가능하였으나 crab shell 유래의 chitin, chitosan등은 분해하지 못하였다. Colloidal chitin에 대한 본 효소의 Km값은 0.11%였고, 분해율은 24시간 반응시 34%였다.

Effects of Levels of Feed Intake and Inclusion of Corn on Rumen Environment, Nutrient Digestibility, Methane Emission and Energy and Protein Utilization by Goats Fed Alfalfa Pellets

  • The effect of high and low level of feed intakes on nutrient digestibility, nutrient losses through methane, energy and protein utilization by goats fed on alfalfa (Medicago sativa L.) pellets based diets was investigated in this study. Twelve castrated Japanese goats were employed in two subsequent digestion and metabolism trials. The goats were divided into three groups, offered three diets. Diet 1 consisted of 100% alfalfa pellet, Diet 2 was 70% alfalfa pellet and 30% corn, and Diet 3 was 40% alfalfa pellet and 60% corn. The two intake levels were high (1.6 times) and low (0.9 times) the maintenance requirement of total digestible nutrients (TON). Rumen ammonia nitrogen ($NH_3$-N) level of Diet 1 was lower (p<0.001) compared to Diets 2 and 3, but the values were always above the critical level (I50 mg/liter), The pH values of rumen liquor ranged from 6.02 to 7.30. Apparent digestibility of nutrient components did not show differences (p>0.05) between the two intake levels but inclusion of corn significantly altered the nutrient digestibility. Diet 3 had highest (p<0.001) dry matter (DM), organic matter (OM), ether extract (EE) and nitrogen fee extract (NFE) digestibility followed by the Diet 2 and Diet 1. The crude protein (CP) digestibility values among the three diets were in a narrow range (70.1 to 70.8%). Crude fiber (CF) digestibility for Diet 3 was slight higher (p>0.05) than that for other two diets. When alfalfa was replaced by corn, there were highly significant (p<0.001) increases in DM, OM, EE and NFE apparent digestibility and a slight increase in the CF digestibility (p>0.05). There were no differences (p>0.05) in energy losses as methane ($CH_4$) and heat production among the diets but energy loss through urine was higher for the Diet 1. The total energy loss as $CH_4$ and heat production were higher for the high intake level but the energy loss as $CH_4$ per gram DM intake were same (0.305 kcal/g) between the high and low intake level. Retained energy (RE) was higher for Diet 3 and Diet 2. Nitrogen (N) losses through feces and urine were higher (p<0.001) for Diet 1. Consequently, N retention was lower (p>0.05) for Diet 1 and higher in Diets 3 and 2. It is concluded that inclusion of corn with alfalfa increased the metabolizable energy (ME) and RE, and retained N through reducing the energy and N losses. The high level of intake reduced the rate of nutrient losses through feces and urine.

강우시 도로유출수 수질특성 및 입경분포 (Water Quality and Particle Size Distributions of Road Runoff in Storm Event)

  • 도시지역은 불투수층 면적비율이 높아 작은 강우에도 초기유출(first flush)이 강하게 발생하고 유출되는 오염물질 농도도 높다. 도로유출수에는 미세입자물질, 중금속, 유기물질 성분을 다량 함유하고 있다. 도로유출수의 수질과 입자의 유출특성을 파악하고자 교통량이 비교적 많은 도로 2개 지점을 대상으로 총 6회의 강우시료를 채수하여 분석하였다. 2개 조사 지점에서 분석된 도로유출수 수질농도범위는 SS $45{\sim}2,396\;mg/L$, $COD_{Cr}\;40{\sim}931\;mg/L$, TKN $0.1{\sim}19.6\;mg/L$, TP $0.2{\sim}25.1\;mg/L$, Fe $0.33{\sim}8.15\;mg/L$, Cr $0.06{\sim}0.50\;mg/L$, Pb $0.06{\sim}0.97\;mg/L$, Cr $0.01{\sim}0.12\;mg/L$으로 조사되었다. 강우초기의 농도는 극단적으로 높은 특성을 나타내었다. 도로유출수 SS와 상관계수가 높은 수질항목으로는 $COD_{Cr}$, TP, Cu, Pb이고, 상관계수가 가장 낮은 항목으로는 TKN으로 분석되었다. 도로유출수의 입경을 분석한 결과 평균 f3경간의 범위는 $6.7{\sim}23.4\;{\mu}m$이고 90% 입경의 범위는 $36.2{\sim}105.2\;{\mu}m$로 분석되었다. 또한, 입경은 유출량에 비례적으로 증가하는 경향을 나타내었다. 균등계수를 분석한 결과 평균범위는 $6.4{\sim}10.2$로 입경 분포범위가 크게 나타났다.
