Anti-Oxidative, Anti-Inflammatory, and Anti-Melanogenic Activities of Endlicheria Anomala Extract (Endlicheria anomala (Nees) Mez 추출물의 항산화, 항염증 및 미백 활성)

  • In this study, the anti-oxidative, anti-inflammatory, anti-melanogenic activities of Endlicheria anomala (Nees) Mez methanol extract (EAME) were evaluated by use of in vitro assays and cell culture model systems. The results revealed that EAME scavenges various radicals such as 1,1-diphenyl-2-picryl hydrazyl hydrogen peroxide induced reactive oxygen species, and lipopolysaccharide induced nitric oxide. Furthermore, EAME induced the expression of anti-oxidative enzymes such as heme oxygenase 1, thioredoxin reductase 1, NAD(P)H dehydrogenase 1, and their upstream transcription factor, nuclear factor-E2-related factor 2. Moreover, EAME inhibited in vitro DOPA oxidation and 3-isobutyl-1-methylxanthine induced melanogenesis in B16F10 cells. Its anti-melanogenic activity will have originated from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Taken together, these results provide the important new insight that E. anomala possesses various biological activities such as anti-oxidative, anti-inflammatory, and anti-melanogenic. Therefore, it might be utilized as a promising material in the fields of nutraceuticals and cosmetics.

Influence of the length and location of implants on distal extension removable partial dentures: finite element analysis (후방연장 가철성 국소의치에서 임플란트의 길이와 위치가 응력분산에 미치는 영향)

  • Purpose: To evaluate the effects of implant location and length on stress distribution and displacement in osseointegrated-implants that were associated with mandibular distal extension removable partial dentures (DERPD). Materials and Methods: A sagittally cut model with the #33, #34 teeth and a removable partial denture of the left mandible was used. Seven models were designed with NX 9.0. Models A, B, C had implants with lengths of 11, 6, 4 mm, respectively, under the denture base of the #37 artificial tooth. Models D, E, F had implants with lengths of 11, 6, 4 mm, respectively, under the denture base of the #36 artificial tooth. Model G did not have any implants. Axial force (250 N) was loaded on #36 central fossa. The finite element analysis was performed with MSC Nastran. Von Mises stress maps were plotted to visualize the results. Results: The models of #37 implant placement showed much lower stress concentration on the surrounding bone of the implant compared with #36. The #36 implant position tended to reduce displacement more than #37. Conclusion: When an IARPD is designed, the distal positioning of implant placement has more advantages in the edentulous bone of DERPD on the prognosis of short implants and the stress distribution of edentulous alveolar bone. Using implants with longer lengths are important for stress distribution. However, Additional studies are necessary of the effects of length on implant survival.

A Study on the Establishment and Application of Evaluation Criteria for Old Railway Station Considering the Level of Railway Service (철도 서비스수준을 고려한 노후철도역사 평가기준 마련 및 적용방안)

  • The total number of railroad stations managed in Korea is 322 (including general and wide-area railways), and a considerable number of stations are aging. In terms of the size of the existing railway station and the number of entrances, it has not been possible to secure adequate service capacity, and the demand for station improvement is increasing due to changes in surrounding conditions such as urban development. In the past, railroad stations were focused on the simple function of a connection passage in terms of maintenance or management, but in recent years, railroad stations are also changing to an atmosphere that they should be reborn as a user-centered comfortable, convenient, and safe service provision space. In this study, a case study related to the improvement of the old railway station was conducted to derive an improvement plan that meets the improvement standard of the old station, and the service level evaluation standard was developed. By introducing the concept of service level (LOS) in the development model, station congestion, station movement convenience, and station safety were selected as evaluation indicators. In addition, this development model applied an analytical stratification technique to divide various evaluation elements of each indicator into major and detailed elements and derive the relative importance of the elements by class. Priority for improvement was derived using the ratio of the number of E and F on the LOS for each facility. Based on this study, it is expected to be helpful in using it as an evaluation criterion for improving objective and equitable railway station.

Bayesian Network-based Probabilistic Safety Assessment for Multi-Hazard of Earthquake-Induced Fire and Explosion (베이지안 네트워크를 이용한 지진 유발 화재・폭발 복합재해 확률론적 안전성 평가)

  • Recently, seismic Probabilistic Safety Assessment (PSA) methods have been developed for process plants, such as gas plants, oil refineries, and chemical plants. The framework originated from the PSA of nuclear power plants, which aims to assess the risk of reactor core damage. The original PSA method was modified to adopt the characteristics of a process plant whose purpose is continuous operation without shutdown. Therefore, a fault tree, whose top event is shut down, was constructed and transformed into a Bayesian Network (BN), a probabilistic graph model, for efficient risk-informed decision-making. In this research, the fault tree-based BN from the previous research is further developed to consider the multi-hazard of earthquake-induced fire and explosion (EQ-induced F&E). For this purpose, an event tree describing the occurrence of fire and explosion from a release is first constructed and transformed into a BN. And then, this BN is connected to the previous BN model developed for seismic PSA. A virtual plot plan of a gas plant is introduced as a basis for the construction of the specific EQ-induced F&E BN to test the proposed BN framework. The paper demonstrates the method through two examples of risk-informed decision-making. In particular, the second example verifies how the proposed method can establish a repair and retrofit strategy when a shutdown occurs in a process plant.

Transformation of the Diatom Phaeodactylum tricornutum with its Endogenous (E)-4-Hydroxy-3-methylbut-2-enyl Diphosphate Reductase Gene (Phaeodactylum tricornutum의 (E)-4-Hydroxy-3-methylbut-2-enyl Diphosphate Reductase 유전자의 형질전환)

  • Phaeodactylum tricornutum is a model diatom that its genomic information and biological tools are well established. In this study, a gene encoding (E)-4-hydroxy-3-methylbut-2-enyl diphosphate reductase (PtHDR), a terminal enzyme of the methylerythritol phosphate pathway regulating chlorophyll and carotenoid biosynthesis, was isolated from P. tricornutum. The isolated gene was cloned into pPha-T1 vector containing fcpA promoter to prepare pPha-T1-HDR plasmid. As a positive control, pPha-T1-eGFP plasmid was constructed with egfp gene. Stable nuclear transformation was carried out with these plasmids by particle bombardment method and zeocin resistant colonies of P. tricornutum were selected on f/2 agar plate. In result, transformation efficiency was evaluated according to the amount of plasmid DNA coated with gold particles. Integration of introduced plasmids was confirmed with genomic DNA of each transformant by polymerase chain reaction. The eGFP fluorescence was visible in the cytoplasm, indicating that eGFP was successively expressed in P. tricornutum system. The transcript level of exogenous Pthdr gene was evaluated with the obtained transformants. The results presented here demonstrated that introduction of Pthdr gene into P. tricornutum chromosome succeeded and expression of PtHDR was enhanced under the fcpA promoter.

Establishment of a [18F]-FDG-PET/MRI Imaging Protocol for Gastric Cancer PDX as a Preclinical Research Tool

  • Purpose: The utility of 18-fluordesoxyglucose positron emission tomography ([18F]-FDG-PET) combined with computer tomography or magnetic resonance imaging (MRI) in gastric cancer remains controversial and a rationale for patient selection is desired. This study aims to establish a preclinical patient-derived xenograft (PDX) based [18F]-FDG-PET/MRI protocol for gastric cancer and compare different PDX models regarding tumor growth and FDG uptake. Materials and Methods: Female BALB/c nu/nu mice were implanted orthotopically and subcutaneously with gastric cancer PDX. [18F]-FDG-PET/MRI scanning protocol evaluation included different tumor sizes, FDG doses, scanning intervals, and organ-specific uptake. FDG avidity of similar PDX cases were compared between ortho- and heterotopic tumor implantation methods. Microscopic and immunohistochemical investigations were performed to confirm tumor growth and correlate the glycolysis markers glucose transporter 1 (GLUT1) and hexokinase 2 (HK2) with FDG uptake. Results: Organ-specific uptake analysis showed specific FDG avidity of the tumor tissue. Standard scanning protocol was determined to include 150 μCi FDG injection dose and scanning after one hour. Comparison of heterotopic and orthotopic implanted mice revealed a long growth interval for orthotopic models with a high uptake in similar PDX tissues. The H-score of GLUT1 and HK2 expression in tumor cells correlated with the measured maximal standardized uptake value values (GLUT1: Pearson r=0.743, P=0.009; HK2: Pearson r=0.605, P=0.049). Conclusions: This preclinical gastric cancer PDX based [18F]-FDG-PET/MRI protocol reveals tumor specific FDG uptake and shows correlation to glucose metabolic proteins. Our findings provide a PET/MRI PDX model that can be applicable for translational gastric cancer research.

MAGNETIC FIELD IN THE LOCAL UNIVERSE AND THE PROPAGATION OF UHECRS

  • We use simulations of large-scale structure formation to study the build-up of magnetic fields (MFs) in the intergalactic medium. Our basic assumption is that cosmological MFs grow in a magnetohy-drodynamical (MHD) amplification process driven by structure formation out of a magnetic seed field present at high redshift. This approach is motivated by previous simulations of the MFs in galaxy clusters which, under the same hypothesis that we adopt here, succeeded in reproducing Faraday rotation measurements (RMs) in clusters of galaxies. Our ACDM initial conditions for the dark matter density fluctuations have been statistically constrained by the observed large-scale density field within a sphere of 110 Mpc around the Milky Way, based on the IRAS 1.2-Jy all-sky redshift survey. As a result, the positions and masses of prominent galaxy clusters in our simulation coincide closely with their real counterparts in the Local Universe. We find excellent agreement between RMs of our simulated galaxy clusters and observational data. The improved numerical resolution of our simulations compared to previous work also allows us to study the MF in large-scale filaments, sheets and voids. By tracing the propagation of ultra high energy (UHE) protons in the simulated MF we construct full-sky maps of expected deflection angles of protons with arrival energies $E = 10^{20}\;eV$ and $4 {\times} 10^{19}\;eV$, respectively. Accounting only for the structures within 110 Mpc, we find that strong deflections are only produced if UHE protons cross galaxy clusters. The total area on the sky covered by these structures is however very small. Over still larger distances, multiple crossings of sheets and filaments may give rise to noticeable deflections over a significant fraction of the sky; the exact amount and angular distribution depends on the model adopted for the magnetic seed field. Based on our results we argue that over a large fraction of the sky the deflections are likely to remain smaller than the present experimental angular sensitivity. Therefore, we conclude that forthcoming air shower experiments should be able to locate sources of UHE protons and shed more light on the nature of cosmological MFs.

Health Risk Assessment and Evaluation of Asbestos Release from Asbestos-cement Slate Roofing Buildings in Busan (부산시내 석면슬레이트지붕 건축물로부터의 석면 노출 및 건강위해성 평가)

  • This study was performed to evaluate the asbestos exposure levels and to calculate excess lifetime cancer risk (ELCR) for the risk assessment of the asbestos fibers released from asbestos-cement slate roofing (ASR) building. Total number of ASR buildings was into 21,267 in Busan, and 82.03 percent of the buildings was residential houses, and 43.61 percent of the buildings was constructed in 1970s. For this study, ten buildings were selected randomly among the ASR buildings. The range of airborne asbestos concentration in the selected ten ASR buildings was from 0.0016 to 0.0067 f/mL, and the concentration around no-admitted ASR buildings was higher than that around admitted buildings. The ELCR based on US EPA IRIS (integrated risk information system) model is within 3.5E-05 ~ 1.5E-04 levels, and the ELCR of no-admitted ASR buildings was higher than 1.0E-04 (one person per million) level that is considered a more aggressive approach to mitigate risk. These results indicate that the cancer risk from ASR buildings is higher than other buildings, and systematic public management is required for control of no-admitted ASR buildings within near future.

  • With the continuous evolution of the numerical methods and the availability of advanced constitutive models, it became a common practice to use complex physical and geometrical nonlinear numerical analyses to estimate the structural behavior of reinforced concrete elements. Such simulations may yield the complete time history of the structural behavior, from the first moment the load is applied until the total collapse of the structure. However, the evolution of the cracking pattern in geometrical discontinuous zones of reinforced concrete elements and the associated failure modes are relatively complex phenomena and their numerical simulation is considerably challenging. The objective of the present paper is to assess the applicability of the Applied Element Method in simulating the development of distinct failure modes in reinforced concrete walls subjected to monotonic loading obtained in experimental tests. A pushover test was simulated numerically on three distinct RC shear walls, all presenting an opening that guarantee a geometrical discontinuity zone and, consequently, a relatively complex cracking pattern. The presence of different reinforcement solutions in each wall enables the assessment of the reliability of the computational model for distinct failure modes. Comparison with available experimental tests allows concluding on the advantages and the limitations of the Applied Element Method when used to estimate the behavior of reinforced concrete elements subjected to monotonic loading.

Clinical study on the maxillofacial prosthodontic treatment using dental implant: Part I-Color stability of maxillofacial elastomeric materials (치과용 임플란트를 이용한 악안면 보철치료에 관한 임상적 연구 1부-악안면보철용 실리콘의 색안정성)

  • 본 연구는 악안면보철용 실리콘의 색안정성을 연구하기 위하여 실리콘 재료로 MDX 4-4210 Silicone(Dow Coming. Midland, Mich.)과 A-2186(Factor II, Lakeside, Az)을 사용하고 Mars Violet. Golden Yellow, Red yellow, Raw Sienna등 4종의 색소(Factor II, Lakeside, Az)를 이용하여 시편을 제작한 후 1군은 섭씨5-55도에서 700회 thermocycling하였고, 2군은 10%의 비눗물(Ivory, Proctor & Gamble Inc,. Toronto, Ontario)에 담아 섭씨15-55도에서 700회 thermocycling하였다. 3군은 10%의 비눗물에 담아 75일간 실온에서 보관하였으며 4군은 실온의 암실에 75일간 보관하였다. 5군은 초여름인 6월 중순부터 75일간 햇볕이 잘드는 곳에 위치하였다. 이를 측색색차계(Model TC-6FX, Tokyo Denshoku Co., Japan)를 이용하여 측정하고 색변화를 연구한 결과 다음과 같은 결론을 얻었다. 1. $L^*$값의 경우 모든 실험군에서 대조군에 비해 유의하게 감소하였다. 2. $a^*$값의 경우 대부분의 실험군에서 증가하였으나 Raw sienna, Mars violet색소를 사용한 실험 3.4.5군의 MDX4-4210의 경우 감소하는 경향을 보였다. 3. $b^*$값은 실험1.2.3군의 경우 대부분 감소하였고 실험4.5.군의 경우에서 증가하였으나 3.4.5군의 경우에서 대부분 유의한 차이가 없었다. 4. ${\Delta}E$의 값은 실험군에서 대부분 감소하였으나 4군의 A-2186에서 증가하는 경향을 보였고 실험2군의 Red yellow, Mars violet을 사용한 MDX4-4210에서 비교적 큰 변화를 보였다. 5. $L^{\ast}$값과 a값의 변화량의 합은 A-2186의 경우에서 크게 나타났으나 ${\Delta}E$값의 변화량은 MDX4-4210에서 크게 나타났다. 6. $L^*$값의 변화량의 합은 실험4군에서 크게 나타났으며 $a^*$값은 1군에서, $b^*$값과 ${\Delta}E$값의 변화량의 합은 2군에서 크게 나타났다.
