Fasudil Increases the Establishment of Somatic Cell Nuclear Transfer Embryonic Stem Cells in Mouse

  • Somatic cell nuclear transfer derived embryonic stem cells (NT-ESCs) have significant advantages in various fields such as genetics, embryology, stem cell science, and regenerative medicine. However, the poor establishment of NT-ESCs hinders various research. Here, we applied fasudil, a Rho-associated kinase (ROCK) inhibitor, to develop somatic cell nuclear transfer (SCNT) embryos and establish NT-ESCs. In the study, MII oocytes were isolated from female B6D2F1 mice and performed SCNT with mouse embryonic fibroblasts (MEFs). The reconstructed NT-oocytes were activated artificially, and cultured to blastocysts in KSOM supplemented with 10 μM fasudil. Further, the blastocysts were seeded on inactivated MEFs in embryonic stem cell medium supplemented with 10 μM fasudil. A total of 26% of embryos formed into blastocysts in the fasudil treated group, while this ratio was 44% in the fasudil free control group. On the other hand, 30% of blastocysts were established NT-ESCs after exposure of fasudil, which was significantly higher than the control group (10%). The results suggest that fasudil reduced blastocyst development after SCNT due to inhibition of 2 cell cleavage while improved the establishment of NT-ESCs through the anti-apoptotic pathway.

Can Narrow Band Chirp Stimulus Shake the Throne of 500 Hz Tone Burst Stimulus for Cervical Vestibular Myogenic Potentials?

  • Background and Objectives: The aim of the study was to compare effects of tone-burst (TB) and narrow-band (NB) Claus Elberling (CE)-chirp stimuli on amplitude, latency and interaural asymmetry ratio (IAR) of cervical vestibular evoked myogenic potentials (cVEMP) in healthy individuals. Subjects and Methods: The study included 50 healthy volunteers. cVEMP procedure was carried out using 500 Hz TB and NB-CE-chirp stimulus (360-720 Hz, up-chirp) in random order. cVEMP were recorded at 100 dB nHL. For each ear and each stimulus, P1 latency, N1 latency and P1N1 amplitude were measured. IAR was also calculated. Results: Mean age was 26.66±9.48 years. cVEMP's in response to both TB and NB CE-chirp stimuli were obtained in all subjects. No statistically significant difference in P1 latency, N1 latency, and P1N1 amplitude was found between the right and left ears for both TB and NB CE-chirp stimuli (p>0.05). In both sides, P1 and N1 latencies were significantly shorter in NB CE-chirp stimulation compared to TB stimulation (p=0.000). In both sides, no statistically significant difference was found in P1N1 amplitude between two types of stimuli (p>0.05). Conclusions: The chirp stimulus produces robust but earlier cVEMP than TB does. This largest series study on NB chirp cVEMP shows that NB chirp is a good and new reliable alternative.

딥러닝을 통한 하이엔드 패션 브랜드 감성 학습 (Deep Learning for Classification of High-End Fashion Brand Sensibility)

  • The fashion industry is creating innovative business models using artificial intelligence. To efficiently utilize artificial intelligence (AI), fashion data must be classified. Until now, such data have been classified focusing only on the objective properties of fashion products. Their subjective attributes, such as fashion brand sensibilities, are holistic and heuristic intuitions created by a combination of design elements. This study aims to improve the performance of collaborative filtering in the fashion industry by extracting fashion brand sensibility using computer vision technology. The image data set of fashion brand sensibility consists of high-end fashion brand photos that share sensibilities and communicate well in fashion. About 26,000 fashion photos of 11 high-end fashion brand sensibility labels have been collected from the 16FW to 21SS runway and 50 years of US Vogue magazines beginning from 1971. We use EfficientNet-B1 to establish the main architecture and fine-tune the network with ImageNet-ILSVRC. After training fashion brand sensibilities through deep learning, the proposed model achieved an F-1 score of 74% on accuracy tests. Furthermore, as a result of comparing AI machine and human experts, the proposed model is expected to be expanded to mass fashion brands.

The non-saponin fraction of Korean Red Ginseng ameliorates sarcopenia by regulating immune homeostasis in 22-26-month-old C57BL/6J mice

  • Background: The non-saponin fraction (NSF) of Korean Red Ginseng is a powder in which saponin is eliminated from red ginseng concentrate by fractionation. In this study, we examined the effect of NSF on age-associated sarcopenia in old mice. Methods: NSF (50 or 200 mg/kg/day) was administered orally daily to young (3-6-month-old) and old (20-24-month-old) C57BL/6 J mice for 6 weeks. Body weight and grip strength were assessed once a week during the oral administration period. The gastrocnemius and quadriceps muscle were excised, and the muscle fiber size was compared through hematoxylin and eosin staining. In addition, the effect of NSF on sarcopenia and inflammation/oxidative stress-related factors in hindlimb muscles was investigated by western blotting. Flow cytometry analysis was conducted to investigate the effect of NSF on immune homeostasis. Blood samples were collected by cardiac puncture, and the serum levels of insulin-like growth factor 1, pro-inflammatory cytokines, and glutathione were evaluated. Results: NSF significantly alleviated muscle strength, mass, and also fiber size in old mice. Age-associated impairment of immune homeostasis was recovered by NSF through retaining CD11b+F4/80+ macrophages and regulating inflammatory biomarkers. NSF also decreased the age-induced expression of oxidative stress factors. Taken together, NSF showed the effect of improving sarcopenia by inhibiting low-grade chronic inflammatory/oxidative stress factors. Conclusion: NSF exhibited anti-sarcopenia effects by regulating chronic inflammation and oxidative stress in old mice. Thus, we suggest that NSF is a promising restorative agent that can be used to improve sarcopenia in the elderly as well as maintain immune homeostasis.

COVID-19 Diagnosis from CXR images through pre-trained Deep Visual Embeddings

  • COVID-19 is an acute respiratory syndrome that affects the host's breathing and respiratory system. The novel disease's first case was reported in 2019 and has created a state of emergency in the whole world and declared a global pandemic within months after the first case. The disease created elements of socioeconomic crisis globally. The emergency has made it imperative for professionals to take the necessary measures to make early diagnoses of the disease. The conventional diagnosis for COVID-19 is through Polymerase Chain Reaction (PCR) testing. However, in a lot of rural societies, these tests are not available or take a lot of time to provide results. Hence, we propose a COVID-19 classification system by means of machine learning and transfer learning models. The proposed approach identifies individuals with COVID-19 and distinguishes them from those who are healthy with the help of Deep Visual Embeddings (DVE). Five state-of-the-art models: VGG-19, ResNet50, Inceptionv3, MobileNetv3, and EfficientNetB7, were used in this study along with five different pooling schemes to perform deep feature extraction. In addition, the features are normalized using standard scaling, and 4-fold cross-validation is used to validate the performance over multiple versions of the validation data. The best results of 88.86% UAR, 88.27% Specificity, 89.44% Sensitivity, 88.62% Accuracy, 89.06% Precision, and 87.52% F1-score were obtained using ResNet-50 with Average Pooling and Logistic regression with class weight as the classifier.

간호사의 병원윤리풍토와 의사소통 자기효능감이 미완료간호에 미치는 영향 (Effects of Hospital Ethical Climate and Communication Self-Efficacy on Nursing Cares Left Undone among Nurses)

  • Purpose: This study aimed to identify the effects of hospital ethical climate and communication self-efficacy on nursing care left undone. Methods: The participants were 142 nurses working in a general hospital. Data were collected from July 18 to August 30, 2021. Data were analyzed by t-test, ANOVA, Pearson correlation, and multiple regression analysis using the SPSS/WIN 25.0 program. Results: (a) The mean value of hospital ethical climate was 3.54±0.41, communication self-efficacy was 5.03±0.81, and the sum of nursing cares left undone was 3.68±3.14. (b) Nursing cares left undone had a negative correlation with hospital ethical climate(r=-.25, p=.003) but not with communication self-efficacy (r=-.13, p=.116). (c) Factors that affected nursing cares left undone included education (≥master) (β=.23, p=.005), marital status (single) (β=-.19, p=.018), age (26~27) (β=-.18, p=.022), and hospital ethical climate (β=-.18, p=.029); the explanation power was 18.0% (F=8.66, p<.001). Conclusion: Our study shows that hospital ethical climate plays a significant role in nursing cares left undone of nurses. It is important to strengthen hospital ethical climate to lower the incidence of nursing cares left undone. These results may serve as basic data to help develop strategies for reducing the incidence of nursing cares left undone.

대형 배양장치에서 기능그룹에 기초한 식물플랑크톤 천이 특성 (Characteristics of Phytoplankton Succession Based on the Functional Group in the Enclosed Culture System)

  • 2016년 8월 19일부터 12월 31일까지 현장원수와 퇴적물이 주입된 현장 배양장치 실험을 통해 환경요인의 영향에 따른 식물플랑크톤 기능그룹(phytoplankton functional group, PFG)의 천이 특성을 연구하였다. 본 연구에서 확인된 식물플랑크톤은 총 27속 50종이었으며, 분류군별로는 녹조류 30종, 규조류 11종, 남조류 7종, 편모조류 2종으로 나타났다. 실험구에서 총 25속 47종(녹조류 30종, 규조류 9종, 남조류 6종, 편모조류 2종), 대조구에서 총 23속 45종(녹조류 26종, 규조류 10종, 남조류 7종, 편모조류 2종)의 식물플랑크톤이 각각 확인되었다. 식물플랑크톤 기능그룹(PFG)은 총 19개(B, C, D, F, G, H1, J, K, Lo, M, MP, N, P, S1, $T_B$, $W_0$, X1, X2, Y)로 확인되었으며, 실험구와 대조구에서는 H1을 제외하고 18개의 PFG가 모두 출현하였다. 생체 부피(biovolume)에 기초했을 때 $W_0$, J 및 M 그룹은 원수만 주입된 대조구와 비교하여 원수와 퇴적물이 주입된 실험구에서 영양염의 내부순환(internal cycling)의 영향으로 더 높은 생체부피를 보이면서 뚜렷한 천이 양상을 나타내었다. PCA 분석 결과에 기초할 때 PFG의 천이 양상은 영양염, 수온 및 조도 등의 주요 환경인자들과의 상관성에 따라 뚜렷한 차이를 나타내었다. 결론적으로 본 연구에서 PFG를 적용하여 주어진 환경조건에서 식물플랑크톤 적응 및 천이(성장) 특성을 확인할 수 있었으며, PFG는 향후 국내 수계의 식물플랑크톤 천이를 이해하기 위한 새로운 방법이 될 수 있을 것으로 생각된다.

실외 실험적 온난화 처리에 따른 활엽수 4개 수종 묘목의생리적 반응 (Effects of Experimental Warming on Physiological Responses of Four Deciduous Tree Species Seedlings)

  • 본 연구는 실외 실험적 온난화 시스템 내에서 적외선등을 이용하여 온난화 처리를 한 다음 발아 당년의 활엽수 4개 수종(물푸레나무, 느티나무, 거제수나무, 굴참나무) 묘목의 생리적 반응을 알아보고자 수행되었다. 적외선등은 2015년 4월부터 가동되었으며, 온난화 처리구 내의 묘목들은 대조구에 비해 $2.7^{\circ}C$ 높은 대기온도 하에서 생육되었다. 기공전도도, 증산속도 및 순광합성률은 동년 7월과 9월에, 엽록소 함량은 7월과 10월에 각각 측정하였다. 온도 증가에 대한 묘목의 생리적 반응은 수종과 측정 시기에 따라 다른 결과를 나타내었다. 즉 기공전도도($mmol{\cdot}m^{-2}{\cdot}s^{-1}$)는 물푸레나무(대조구:$158.97{\pm}42.76$; 온난화 처리구:$42.07{\pm}8.24$), 느티나무(대조구:$170.53{\pm}27.22$; 온난화 처리구: $101.17{\pm}42.27$), 거제수나무(대조구:$249.93{\pm}47.39$; 온난화 처리구:$150.73{\pm}26.52$)에서 온난화 처리에 의해 감소하였다. 그리고 증산속도($mmol{\cdot}m^{-2}{\cdot}s^{-1}$)도 물푸레나무(대조구:$4.08{\pm}0.62$; 온난화 처리구:$1.74{\pm}0.39$), 느티나무(대조구:$4.32{\pm}0.44$; 온난화 처리구:$3.24{\pm}1.14$), 거제수나무(대조구:$6.21{\pm}0.38$; 온난화 처리구:$4.66{\pm}0.28$)에서 대조구에 비해 온난화 처리구에서 낮은 값을 나타내었다. 한편, 굴참나무의 경우 기공전도도와 증산속도의 처리 간 차이가 유의하지 않았다. 엽록소 함량은 대부분의 수종에서 온난화 처리에 의해 증가하였으나, 순광합성률은 모든 수종에서 처리 간의 차이가 통계적으로 유의하지 않았다. 이는 온난화 처리의 영향이 미미하여 순광합성률의 변화에까지 영향을 미치지 못하였기 때문인 것으로 판단된다.

꽈배기모자반의 발효를 통한 항염증 활성의 증진 (Enhancement of Anti-inflammatory Activity by Fermentation of Sargassum siliquanstrum)

  • 본 연구에서는 유산균으로 발효한 꽈배기모자반의 항염증 및 iNOS 발현저해 활성에 대하여 검증하였다. 항염증 활성은 lipopolysaccharide (LPS)로 자극시킨 RAW 264.7 세포에서 nitric oxide (NO)가 생성되는 양을 비교하여 나타내었으며, iNOS 발현저해 활성은 stable transfection시킨 RAW 264.7 세포에서 LPS에 의해 유도되는 inducible nitric oxide synthase (iNOS) 발현에 대한 저해효과를 reporter인 luciferase의 활성을 확인하여 나타내었다. NO 생성 억제능과 iNOS 발현에 대한 실험은 NO radical 소거 활성을 확인한 후 수행하였다. NO radical 소거 활성은 발효군이 비발효군에 비해 7.6~15.2% 증가되었으며, Lactobacillus sp. SH-1으로 접종한 군이 가장 높은 활성을 나타내었다. 그리고 해조류 침전물을 포함하여 발효한 군보다는 해조류 침전물을 포함하지 않고 발효한 군이 더 높은 NO radical 소거 활성을 나타내었다. Lactobacillus sp. SH-1을 접종하여 발효한 군은 LPS로 자극시킨 RAW 264.7 세포에서 NO 생성 억제능이 가장 높게 나타났다(64.1%). 또한 Lactobacillus sp. SH-1 접종군은 50, 100, 500, 1,000 μg/ml의 농도에서 LPS에 의해 유도된 iNOS 발현을 각각 28.6, 35.6, 49.4, 58.5% 감소시켰다. MTT법에 따르면, 발효 꽈배기모자반은 모든 농도에서 세포 생존율에 영향을 미치지 않았으므로, 세포독성을 나타내지 않는 것으로 사료된다. 본 연구에서는 NO radical 소거 활성, 항염증 및 iNOS 발현저해 활성 등의 꽈배기 모자반이 가지는 생리활성을 확인하였다. 따라서 발효에 의해 생리활성이 개선된 꽈배기모자반을 이용하여 기능성 식품을 개발할 수 있을 것으로 기대된다.
