Antioxidant and Immune-Modulating Activities of Egg Yolk Protein Extracts

  • Egg yolk is widely used to extract lecithin, which is utilized in the food and cosmetics industry. After lecithin is removed, the rest of egg yolk is generated as a by-product. Thus, it is necessary to properly utilize it. In this study, egg yolk protein extracts were produced using ethanol (EYE-E) and water (EYE-W). Their antioxidant and immunomodulatory effects were then evaluated. Antioxidant activities of EYE-E and EYE-W were determined using cellular antioxidant capacity (CAC) assay and comet assay. EYE-E and EYE-W showed significant (p<0.05) scavenging effects on intracellular reactive oxygen species (ROS) in a dose dependent manner. At a concentration of 50 ㎍/mL, EYE-W showed higher (p<0.05) antioxidant activity than EYE-E. EYE-E and EYE-W also exhibited protective effects against DNA damage caused by oxidative stress. After treatment with EYE-E and EYE-W, DNA damage level of 48.7% due to oxidative stress was decreased to 36.2% and 31.8% levels, respectively. In addition, EYE-E and EYE-W showed immunomodulatory effects by regulating Th1 cytokines (TNF-α and IL-2) and Th2 cytokines (IL-10 and IL-4) in Balb/c mouse splenocytes. These data suggest that EYE-E and EYE-W could be used as functional food ingredients with excellent antioxidant and immunomodulatory activities in the food industry.

The adsorption pattern of protein to the soft contact lens and its effect on the visible light transmission and the contact angle (소프트콘택트렌즈의 단백질 부착 양상 및 가시광선투과도와 접촉각에 미치는 영향)

  • Protein deposit buildup on soft contact lenses(SCLs) is responsible for discomfort, optical defects and is even damaging to the eye. To better understand protein deposit formation on SCL, we investigated the pattern of protein attachment on patient-worn SCLs and the effect of the protein on the contact angle of SCLs and the transmission of visible light. This study used data from a clinical trial where subjects wore either daily-wear SCL(etafilcon A, N=40) for various period up to 14 days, or extended-wear SCL(balafilcon A, N=40) for 8 h day or night. The adsorption of protein in daily-wear SCLs was significantly increased in patient-worn time-dependent manner although the lenses were cleaned everyday by using the multi-purpose SCL solution. After 2 weeks wearing, the protein on the SCL influenced the contact angle of the lenses. Furthermore, the transmission of visible light of the lenses decreased up to almost 7%. Extended-wear SCLs worn during the closed eye condition were found to deposit significantly more protein than SCLs worn during the open eye condition. The amounts of protein adsorbed both after daily SCL wear and after overnight SCL wear were not affected the contact angle and the transmission of visible light of the lenses. The protein deposit buildup on SCL related to the corneal temperature and pH of tear fluid.

The Effects of Ultrasonic Cleaner for Eye Glasses on Protein Deposits and Parameters in Soft Contact Lens (안경용 초음파세척기에 의한 소프트콘택트렌즈의 단백질 침전물 세척효과)

  • Purpose: The cleaning effect of protein deposit and the change of contact lens parameters by ultrasonic cleaner for eye glasses on the soft contact lenses were investigated. Methods: Etafilcon A contact lenses contaminated with protein, was ultrasonicated by ultrasonic cleaner for eye glasses and for the control group, spoiled contact lenses were cleaned by multi-purpose solution. The remaining protein deposits on the contact lenses were determined after extraction and the changes of overall diameter, base curve, center thickness power, and water contents on contact lenses were measured and surfaces of contact lenses were observed by scanning electron microscope. Results: The cleaning efficacies of multi-purpose solution on protein deposited etafilcon A contact lenses were 6.08%, and 23.73~33.92% in the group of ultrasonic cleaner for eye glasses with multi-purpose solution and 0~12.99% in the group of ultrasonic clear for contact lens with multipurpose solution depending on the treatment time. The changes of parameters and surface on contact lenses by ultrasonication were not observed. Conclusions: Ultrasonic cleaner for eye glasses can be used to eliminate protein deposits for the diagnostic soft contact lens in the office since it was effective to eliminate protein deposits and not caused change of parameters on soft contact lenses.

  • The isolated soluble eye proteins from six species (Sepia esculenta, Sepiella japonica, Loligo chinensis, Todarodes pacificus, Loligo beka and Ocotopus minor) of class Cephaloopoda were compared by crossed immunoelectrophoresis methods using antibodies directed against total soluble eye proteins antigens. It showed a distinct antigen-antibody reaction between the species of order Sepioidea and various antiserum of class Cephalopoda. Our results suggested that the common precipitation lines of Sepia and Loligo species which were different from that of Octopus minor. By which obtained from elution of gel filtration chromatography and 12.5% SDS-polyacrylamide gel electrophoresis, the molecular weight of Todarodes pacificus eye protein (crystallin) was determined in abut 20-35 KDa.

  • A 14-year-old, male Shih-tzu was presented with glaucoma in the left eye. Menace response was negative in the left eye. On ophthalmic examination, exotropia, buphthalmos, bulging of upper lateral globe and episcleral injection was detected in the left eye. Also, there was corneal edema, corneal pigmentation and hyphema in the left eye. The pupil was difficult to observe due to corneal lesions in the left eye. On ultrasonographic examination, an amorphous large mass protruding from the region of the optic disc into the vitreous was observed in the left eye. Enucleation was performed in the left eye. Histologically, the left intraocular mass was diagnosed as glioma. Immunohistochemically, the neoplastic cells were positive to glial fibrillary acidic protein.

The effects of PEP-1-FK506BP on dry eye disease in a rat model

  • As FK506 binding proteins (FK506BPs) are known to play an important role in the regulation of a variety of biological processes related to cell survival, this study was designed to examined the protective effects of FK506 binding protein 12 (FK506BP) on low humidity air flow induced dry eye in a rat model using transduced PEP-1-FK506BP. After the topical application of PEP-1-FK506BP, tear volumes were markedly increased and significant prevention of cornea damage was observed compared with dry eye rats. Further, immunohistochemical analysis demonstrated that PEP-1-FK506BP markedly prevented damage to the cornea, the bulbar conjunctiva, and the palpebral conjunctiva epithelial lining compared with dry eye rats. In addition, caspase-3 and PARP expression levels were found to be decreased. These results demonstrated that topical application of PEP-1-FK506BP significantly ameliorates dry eye injury in an animal model. Thus, we suggest that PEP-1-FK506BP can be developed as a new ophthalmic drop to treat dry eye diseases.

Transduced PEP-1-FK506BP ameliorates corneal injury in Botulinum toxin A-induced dry eye mouse model

  • FK506 binding protein 12 (FK506BP) belongs to a family of immunophilins, and is involved in multiple biological processes. However, the function of FK506BP in corneal disease remains unclear. In this study, we examined the protective effects on dry eye disease in a Botulinum toxin A (BTX-A) induced mouse model, using a cell-permeable PEP-1-FK506BP protein. PEP-1-FK506BP efficiently transduced into human corneal epithelial cells in a time- and dose-dependent manner, and remained stable in the cells for 48 h. In addition, we demonstrated that topical application of PEP-1-FK506BP was transduced into mouse cornea and conjunctiva by immunohistochemistry. Furthermore, topical application of PEP-1-FK506BP to BTX-A-induced mouse model markedly inhibited expression levels of pro-inflammatory cytokines such as interleukin-$1{\beta}$ (IL-$1{\beta}$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and macrophage inhibitory factor (MIF) in corneal and conjunctival epithelium. These results suggest PEP-1-FK506BP as a potential therapeutic agent for dry eye diseases.

  • Olive flounder Paralichthys olivaceus is one of the most widely cultured fish species in Korea. Although olive flounder receive attention from aquaculture and fisheries and extensive research has been conducted eye morphological change in metamorphosis, but little information was known to molecular mechanism and gene expression of eye development- related genes during the early part of eye formation period. For the reason of eyesight is the most important sense in flounder larvae to search prey, the screening and identification of expressed genes in the eye will provide useful insight into the molecular regulation mechanism of eye development in olive flounder. Through the search of an olive flounder DNA database of expressed sequence tags (EST), we found a partial sequence that was similar to crystallin beta A1 and gamma S. Microscopic observation of retinal formation correspond with the time of expression of the crystallin beta A1 and gamma S gene in the developmental stage, these result suggesting that beta A1 and gamma S play a vital role in the remodeling of the retina during eye development. The expression of crystallin beta A1 and gamma S were obviously strong in eye at all tested developing stage, it is also hypothesized that crystallin acts as a molecular chaperone to prevent protein aggregation during maturation and aging in the eye.

Characterization of Beef Transcripts Correlated with Tenderness and Moisture

  • To identify transcriptional markers for beef traits related to meat tenderness and moisture, we measured the transcriptome of the Longissimus dorsi skeletal muscle in 10 Korean native cattle (KNC). We analyzed the correlation between the beef transcriptome and measurements of four different beef traits, shear force (SF), water holding capacity (WHC), cooking loss (CL), and loin eye area (LEA). We obtained non-overlapping and unique panels of genes showing strong correlations (${\mid}r{\mid}$ > 0.8) with SF, WHC, CL, and LEA, respectively. Functional studies of these genes indicated that SF was mainly related to energy metabolism, and LEA to rRNA processing. Interestingly, our data suggested that WHC is influenced by protein metabolism. Overall, the skeletal muscle transcriptome pointed to the importance of energy and protein metabolism in determining meat quality after the aging process. The panels of transcripts for beef traits may be useful for predicting meat tenderness and moisture.
