Pre-extraction of Hemicelluloses from Korean Mixed Hardwood Chips Using Hot Water and Alkali Solution and its Characteristics (열수 및 알칼리 용액을 이용한 국산 목재 칩으로부터 헤미셀룰로오스의 선추출 및 특성 구명)

  • Hemicelluloses are useful materials for papermaking and other biotechnological applications. To recover and further utilize the hemicelluloses, this study carried out the pre-extraction of the hemicelluloses from Korean mixed hardwoods chips using hot water and sodium hydroxide solution at various temperature and time conditions. Through the hot water pre-extraction, the wood components of 22% was extracted and the 35% of the hemicelluloses which were contained in the original wood chips were dissolved into the pre-extractives. The yield of the pre-extractives was more significantly influenced by the pre-extraction temperature than the time. The pre-extractives with the yield of 30% could be obtained by alkali pre-extraction and alkali concentration was the most important factor. It had lower hemicellulose content but high molecular weight, compared with the hot water pre-extractives.

  • The process of extracting active ingredients from wheat sprout using ultrasound assisted method was optimized with a central composite design model. The response value of the central composite design model established the extraction yield and the total flavonoids content, main effects and interactive effects were analyzed depending on independent variables such as the extraction time, volume ratio of ethanol to ultrapure water, and ultrasonic irradiation power. The volume ratio of ethanol to ultrapure water and ultrasonic irradiation power were relatively large for the extraction yield and the extraction time was most significantly affected the total flavonoids, Considering both the extraction yield and total flavonoids content, the optimal extraction conditions were as follows: the extraction time of 17.00 min, volume ratio of ethanol to ultrapure water of 50.25 vol%, ultrasonic irradiation power of 551.70 W. In this case, the extraction yield and total flavonoids content were 28.43 wt% and $29.99{\mu}g\;QE/mL\;dw$, respectively. The actual experimental extraction yield and total flavonoids content under this condition were 8.73 wt% and $29.65{\mu}g\;QE/mL\;dw$, respectively with respective error rates of 1.05 and 1.13%.

  • Effects of modifier and soaking on extraction of triterpenoid saponin (glycyrrhizin) from liquorice were examined using supercritical $CO_2(SC-CO_2)$ at 50 MPa, $60^{\circ}C$, and flow rate of 3 mL/min, and glycyrrhizin content was analyzed by HPLC. Additon of undiluted methanol, ethanol or isopropanol as modifier to $SC-CO_2$ had little influence on extraction yield of glycyrrhizin. Soaking process using water increased the extraction yield as the sample to solvent ratio was increased. Addition of 70% methanol, ethanol or isopropanol to $SC-CO_2$ significantly increased the extraction yields, with 70% methanol resulting in the highest yield. When water at 90% (w/w) of sample weight was used for soaking, the extraction yield and rate increased, 70% ethanol-modified $SC-CO_2$ was almost equal to that obtained using 70% methanol.

  • In the semiconductor manufacturing, yield enhancement is an urgent issue. It is ideal to prevent all the failures. However, when a failure occurs, it is important to quickly specify the cause stage and take countermeasure. Reviewing wafer level and composite lot level yield patterns has always been an effective way of identifying yield inhibitors and driving process improvement. This process is very time consuming and as such generally occurs only when the overall yield of a device has dropped significantly enough to warrant investigation. The automatic method of failure pattern extraction from fail bit map provides reduced time to analysis and facilitates yield enhancement.   • The chemical constitution of a drug has been accepted as an important basis for pharmacological action in Unani medicine. Various dosage forms have been developed on this concept, such as decoctions (Joshanda), infusions (Khesanda), extract (Rub / Usara), and syrup. Zarawand Mudaharaj (ZM.) / Aristolochia rotunda L. root was subjected to extraction process using Soxhlet's apparatus by using Response Surface Methodology (RSM) to design the number of random runs of the extracts with variation in the factors of temperature, the concentration of ethanol in water, time for extraction, for optimizing and maximizing the yield concentration. The data obtained, was analyzed with regression equation and ANOVA two-way summary to interpret the interaction of the factors for yield maximization. Minitab version 18 was used to design and analyze data. Validation of the optimum conditions for maximum yield of the whole extract of ZM. Root was carried out by re-run of the extract using the optimized conditions. The maximum yield percentage thus obtained using RSM was 20.87% whereas using these optimum conditions 21.35 % yield was obtained thereby validating the method. The association between the response functions and the process variables was identified by a three-factor recorded Box-Behnken design. In the present study RSM is used because itis a cheap and affordable method to optimize maximum yield percentage which may be reliably used by researchers. The study set in the surface conditions for ZM. root extraction by the Soxhlet apparatus for maximizing the yield percentage.

Study on Extraction of Mucopolysaccharide-protein Containing Chondroitin Sulfate from Chicken Keel Cartilage

  • The objective of this study was to investigate technical methods for extraction of mucopolysachharide-protein containing chondroitin sulfate from keel cartilage of chickens. The chemical composition of chicken keel cartilage was determined. For the preparation of mucopolysaccharide-protein from lyophilized chicken keel cartilage, hot water extraction and alcalase hydrolysis methods were examined. Results showed that the optimum condition of hot water extraction was incubation for 120 min with a yield of 40.09% and chondroitin sulfate content of 28.46%. For alcalase hydrolysis, the most effective condition was 2% alcalase in 10 volumes of distilled water for 120 min. The yield of hydrolysate was 75.87%, and chondroitin sulfate content was 26.61%. For further separation of chondroitin sulfate from the alcalase hydrolysate, which has a higher yield than that of hot water, 60% ethanol precipitation was performed. The yield of the ethanol precipitate was 21.41% and its chondroitin sulfate content was 46.31%. The hot water extract, alcalase hydrolysate and ethanol precipitate showed similar electrophoretic migration with standard chondroitin sulfate (chondroitin sulfate A), using cellulose acetate membrane electrophoresis. These results indicated that a significant amount of mucopolysaccharide-protein containing chondroitin sulfate could be acquired form chicken keel cartilage. Therefore, keel cartilage in chicken may provide an inexpensive source of chondroitin sulfate for commercial purposes.

Monitoring on Extraction Conditions of Old Pumpkin Using Response Surface Methodology (반응표면분석법에 의한 늙은 호박 추출조건의 모니터링)

  • Response surface methodology was used for monitoring extraction conditions, based on quality properties of old pumpkin extracts. Hunter's color L value of extracts was maximized at 101℃, 2.6 hr and decreased gradually after maximum point. The polynomial equation for Hunter's color L value showed 10% of significance level and 0.8799 of R². Hunter's color a value was minimized at 117℃, 3.9 hr and R² of polynomial equation was 0.9852 within 1% significance level. Hunter's color b value and ΔE value increased as the extracting temperature and time increased. Extraction yield of old pumpkin was maximized at 110℃, 4 hr and increased in proportional to the extracting temperature and time, but decreased after 113℃ and 2 hr. Viscosity of pumpkin extracts were maximized at 120℃, nearly 3 hr. R² of polynomial equations for yield, viscosity and sugar content were 0.9532, 0.9812 and 0.8869, respectively. Optimum ranges of extraction conditions for quality properties of old pumpkin were 102∼109℃, 2.5∼3.5 hr, respectively. Predicted values at the optimum extraction condition agreed with experimental values.

Optimization of Saponin Extraction Conditions in Ginseng Milk using Response Surface Methodology (반응표면분석 법을 이용한 인삼우유 중 사포닌 추출조건의 최적화)

  • To develop the methodology of the quantitative analysis of saponin in ginseng milk, conditions of the saponin extraction were optimized using the fractional factorial design with 3 variables and 3 levels by a RSM computer program. The extraction of saponin increased with an increase in extraction temperature up to $90^{\circ}C$ and then decreased significantly at $100^{\circ}C$. Extraction time affected the saponin yield in a similar trend. On the other hand, decreasing cooling temperature increased the amount of the saponin extracted. Recovery yield of the saponin from ginseng milk varied from 70.0% to 92.9%. The optimum extraction temperature, time and cooling temperature determined by partial differentiation of the model equation were $86^{\circ}C$, 2.83 hrs and $4^{\circ}C$, respectively.   • This study was concentrated to investigate the dependency of recovery yield on the extraction conditions. From fruiting body, the extracted amount of polysaccharide was studied with various solvent. In order to maximize the yield, the optimum extraction conditions were elucidated with respect to solvent, temperature and time. In addition, the dialysis was applied to obtain the higher purity of polysaccharide with varying the exposure time.

  • Several solvents or combinations of solvents were tested for the extraction of wet or dried biomass at different extraction mode from plant cell cultures. Methanol gave the highest paclitaxel recovery with the least amount of solvent usage. before extraction drying of biomass wass helpful to decrease solvent usage in extraction step./ in this case drying method was very important to obtain high yield from dried biomass. In thid mode of operation counter-current extraction process can be able to decrease solvent usage but paclitaxel recovery was almost same with both batch and counter-current mode of operation. The number of extraction times was at least four to obtain high yield(>99%) from cell and one to obtain highyield(>96%) from cell debris in batch mode. Equilibrium (i.e. the ratio of paclitaxel in biomass to paclitaxel in the extraction solvent) was reached within 5 minutes. The minimum methodal concentration (90%) and solvent amount(biomass : solvent=1 Kg : 1L) are enough to obtain high yield(>98%) for extraction from biomass.
