While nitric oxide (NO) produced by inducible nitric oxide synthase (iNOS) is beneficial for host survival, it is also detrimental to the host. Thus, regulation of iNOS gene expression may be an effective therapeutic strategy for the prevention of unwanted reactions at various pathologic conditions. During the screening process for the possible iNOS regulators, we observed that esculetin is a potent inhibitor of cytokine-induced iNOS expression. The treatment of 3T3-L1 adipocytes with the tumor necrosis factor-${\alpha}$ (TNF) induced iNOS expression, leading to enhanced NO production. TNF-induced NO production was inhibited by esculetin in a dose-dependent manner. Esculetin inhibited the TNF-induced NO production at the transcriptional level through suppression of iNOS mRNA and subsequent iNOS protein expression. These results suggest esculetin, a component of natural products, as a naturally occurring, nontoxic means to attenuate iNOS expression and NO-mediated cytotoxicity.

The potential utility of injection media (sucrose, PEG, and liposome) was demonstrated for direct gene transfer into olive flounder (Paralichthys olivaceus) muscles. Based on the use of sucrose (final cone. 20%), PEG 8,000 (final cone. 10%) or liposome (twice us of DNA injected), the present injection strategy significantly improved the level of transgene expression as well as persistent duration of expression. The increased amounts of expression in DNA injection with sucrose, PEG, and liposome were as high as from 2.1 to 4.9-folds of conventional TE-based DNA injection. The best result was obtained from injections of liposome-encapsulated DNA in which the expression was detectable at least 32 days after injection when compared to only 8-16 days from TE-based injections.

This study tries to find how architects interpret and express fashion brand images. First, it examines the concept of brand images and its architectural expression of them in contemporary commercial architecture. Second, it investigates the brand strategies and architectural expression strategies of world-famous fashion brand companies such as Prada and Louis Vuitton. Third, it finds out architects' expression of the brand strategies focusing on Rem Koolhaas, Herzog & do Meuron, Jun Aoki, Barthelemy-Grino, Kumiko Inui. In conclusion, architects have their own expression tools for fashion brand images but the extent of expression vanes according to each brand strategy.

본 연구는 IPTV 광고효과에 영향을 미치는 요인이 무엇인가를 알아보기 위해 이루어졌다. 연구결과 첫째, 양방향성 메시지를 담고 있는 IPTV 특성반영 광고가 일반 TV 광고보다 광고주목율과 광고 태도가 긍정적으로 나타났다. 둘째, 양방향성 메시지를 담고 있는 IPTV형 광고의 경우 제품 상세 정보를 제시하는 조건이 효과적인 것으로 나타났다. 셋째, 양방향성 메시지 표현 중 플로팅 표현전략 효과가 가장 높게 나타났다. 넷째, 매체 수용자의 혁신성 특성은 IPTV에 관한 지식수준과 추가 정보 탐색의도에 영향을 미치는 것으로 나타났다. 본 연구결과는 IPTV 사업자들에게 양방향성 메시지를 탑재하는 것이 효과적이라는 이론적 근거를 제시해 줄 뿐 아니라 플로팅 표현전략이 광고의 클릭의도를 높일 수 있는 방안이라는 것을 시사한다.

Preproenkephalin (PPE) is a precursor molecule for multiple endogenous opioid peptides Leu-enkephalin (ENK) and Met-ENK, which are involved in a wide variety of modulatory functions in the nervous system. Despite the functional importance of ENK in the brain, the effect of brain-derived factor(s) on PPE expression is unknown. We report the dual effect of neural epidermal growth factor (EGF)-like-like 2 (NELL2) on PPE gene expression. In cultured NIH3T3 cells, transfection of NELL2 expression vectors induced an inhibition of PPE transcription intracellularly, in parallel with downregulation of protein kinase C signaling pathways and extracellular signal-regulated kinase. Interestingly, these phenomena were reversed when synthetic NELL2 was administered extracellularly. The in vivo disruption of NELL2 synthesis resulted in an increase in PPE mRNA level in the rat brain, suggesting that the inhibitory action of intracellular NELL2 predominates the activation effect of extracellular NELL2 on PPE gene expression in the brain. Biochemical and molecular studies with mutant NELL2 structures further demonstrated the critical role of EGF-like repeat domains in NELL2 for regulation of PPE transcription. These are the first results to reveal the spatio-specific role of NELL2 in the homeostatic regulation of PPE gene expression.

This study investigated the moderating effects of conflict-coping strategies between marital conflict and satisfaction according to gender and marital period. The subjects of this study were 178 couples from Cheongju who have been married less than fourteen years. For husbands and wives who have been married less than 7 years, nothing moderated the interaction between marital conflict and satisfaction. However, for husbands who have been married between 8 and 14 years, their conflict withdrawal moderated the interaction between marital conflict and satisfaction. For their wives, their conflict-expression strategy and their husbands' conflict-expression strategy moderated the interaction between marital conflict and satisfaction.

Telomerase reverse transcriptase (TERT) is an enzymatic ribonucleoprotein that prolongs the replicative life span of cells by maintaining protective structures at the ends of eukaryotic chromosomes. Telomerase activity is highly up-regulated in 85-90% of human cancers, and is predominately regulated by hTERT expression. In contrast, most normal somatic tissues in humans express low or undetectable levels of telomerase activity. This expression profile identifies TERT as a potential anticancer target. By using an RNA mapping strategy based on a trans-splicing ribozyme library, we identified the regions of mouse TERT (mTERT) RNA that were accessible to ribozymes. We found that particularly accessible sites were present downstream of the AUG start codon. This mTERTspecific ribozyme will be useful for validation of the RNA replacement as cancer gene therapy approach in mouse model with syngeneic tumors.

Liver fibrosis is caused by metabolic problems such as cholestasis, genetic problems, or viral infections. Inhibiting hepatic stellate cell (HSC) activation or inducing selective apoptosis of activated HSCs is used as a treatment strategy for liver fibrosis. It has been reported that when HSCs are activated, their apoptosis sensitivity to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is enhanced because the expression of death receptor 5 is elevated. Finding a natural compound that can enhance the apoptotic effect of TRAIL on HSCs is a necessary strategy for liver fibrosis treatment. It was confirmed here that mangosteen-derived gartanin increased the effect of TRAIL-induced apoptosis by increasing the expression of DR5 in a p38-dependent manner in the hepatic stellate cell line LX-2. Combined treatment with gartanin and TRAIL accelerated DNA cleavage through caspase-3 activation and enhanced antifibrotic effects in LX-2 cells.

Human heavy chain (H-) and light chain (L-) ferritins were amplified from a human cDNA library. Each ferritin gene was inserted downstream of the T7 promoter of bacterial expression vectors, and two types of coexpression vectors were constructed. The expression levels of recombinant ferritins ranged about 26-36% of whole-cell protein. H-ferritin exhibited a lower expression ratio compared with L-ferritin, by a coexpression system. However, the coexpression of HL-ferritins was significantly increased above the expression ratio of H-ferritin by cultivation without IPTG induction overnight. Purified recombinant H-, L-, HL-, and LH-ferritins were shown to be homo- and heteropolymeric high molecular complexes and it was indicated that their assembled subunits would be able to work functionally in the cell. Thus, these results indicate an improvement in the expression strategy of H-ferritin for heteropolymeric production and studies of ferritin assembly in Escherichia coli.

In this paper, a strategy for utilizing the Korean tradi-tional spaces and interior design elements in inner space in order to express Korean-style interior design has been studied. Characteristics of Korean tradition was gathered from literatures in two categories; space and el-ements. Space again has been studied in detail in the area of split, continuity, hierarchy, elasticity, dynamics. And study of elements includes floor, wall, ceiling, dan-chung, lattice, laytiles on a roof, rafter, extended eaves, the line of eaves. 30 Korean restaurants were selected, analysed and compared with literature review. Based on the compari-son, a strategy for proper expression and utilization of Korean tradition is suggested. In the process of compari-son, current status of implementation and problems were found. Traditional elements are used in about 50% of Korean restaurants located in hotels, and 25% of those located in department stores. With the survey and other professional's opinions, an implementation plan is suggest-ed as follows; 1. Succession design method of tradition should use main-ly amelioration method and use copy and partly abstraction method. 2. Expression of tradition has to include all of space, ele-ments, and decoration. In space structure, Korean tra-dition space structure must be applied. 3. In order to design with feeling of Korean tradition, various different Korean elements have to be used. 4. In order to express high quality design, high-quality elements has to be used.
