Protoplast Fusion of phaffia rhodozyma (Phaffia rhodozyma의 원형질체 융합)

  • Cell fusion between complementary mutants isolated from astaxanthin-producing yeast, Phaffia rhodozyma, was carried out to obtain astaxanthin-overproducing strains by protoplast fusion technique. The frequency of protoplast fusion was ranged from 2.3$\times$10-5 to 6.0$\times$10-5, and nuclear fusion in the cells of hybrids was demonstrated by several techniques such as isolation of recombinants after mitotic segregation of parental genetic markers, estimation of DNA content, direct observation of nuclei with nuclear staining, and comparison of survival rate to UV exposure. One of several hybrids, Fl, showed approximately 3-fold increase in astaxanthin content when compared with wild parent.

Sterilization of Food-Borne Pathogenic Bacteria by Atmospheric Pressure Dielectric Barrier Discharge Plasma (대기압 유전체장벽방전 플라즈마에 의한 식품유해 미생물 살균)

  • This study aimed to explore the potential for food-industry application of atmospheric pressure dielectric barrier discharge plasma (atmospheric pressure DBD plasma) as a non-thermal sterilization technology for microorganism. The effects of the key parameters such as power, oxygen ratio, exposure time and distance on Escherichia coli KCCM 21052 sterilization by the atmospheric pressure DBD plasma treatment were investigated. The experimental results revealed that increasing the power, exposure time or oxygen ratio and decreasing the exposure distance led to an improvement in the sterilization efficiency of E. coli. Furthermore, the atmospheric pressure DBD plasma (1.0 kW power, 1.0% (v/v) $O_2$, 5 min exposure time and 20 mm exposure distance) treatment was very effective for the sterilization of food-borne pathogenic bacteria. The sterilization rate of E. coli, Bacillus cereus KCCM 40935, Bacillus subtilis KCCM 12027, Bacillus thuringiensis KCCM 11429 and Bacillus atrophaeus KCCM 11314 were 72.3%, 74.6%, 88.5%, 84.7% and 91.3%, respectively.

Sterilization and quality variation of dried red pepper by atmospheric pressure dielectric barrier discharge plasma (대기압 유전체장벽방전 플라즈마에 의한 건고추의 식중독균 살균효과 및 품질변화)

  • This study was conducted to explore the potential for use of atmospheric pressure dielectric barrier discharge plasma (atmospheric pressure DBD plasma) as a non-thermal sterilization technology for microorganisms in dried red pepper. The effects of key parameters such as power, exposure time and distance on the sterilization efficiency and the quality of red dried pepper by the atmospheric pressure DBD plasma treatment were investigated. The results revealed that the plasma treatment was very effective for sterilization of Staphylococcus aureus, with 15 min of treatment at 1.0 kW and 20 mm sterilizing 82.6% of the S. aureus. Increasing the power or exposure time and decreasing the exposure distance led to improved sterilization efficiency. The atmospheric pressure DBD plasma treatment showed no effect on the ASTA (American spice trade association) value or hardness of dried red pepper. Furthermore, no effects of atmospheric pressure DBD plasma treatment were observed on the sensory properties of dried red pepper. To assess the storage stability, the dried red pepper was treated with atmospheric pressure DBD plasma (1.5 kW power, 15 min exposure time and 10 mm exposure distance), then stored for 12 weeks at $25^{\circ}C$. Consequently, the ASTA value, hardness and capsaicin concentration of dried red pepper were maintained.

  • In this article, the properties of ${CO_2}^+(CO_2)_n$ clusters in a Paul ion trap have been investigated using mass-selective instability mode which conducted by chosen precursor ions, mainly $Ar^+$ and ${CO_2}^+$ produced by a mixture of Ar and $CO_2$. Exposure of ${CO_2}^+$ ions to $CO_2$ molecules, lead to the formation of ${CO_2}^+(CO_2)_n$ clusters. Here, Ar gas react as a buffer gas and lead to form ${CO_2}^+(CO_2)_n$ cluster by collisional effect.

  • The conditions for the protoplast fusion of auxotrophic mutants of Penicillium verruculosum were determined. A preparation of commercial enzyme Novozym 234 was used to successfully isolate protoplast from the 20hr old mycelium of P. verruculosum. Under optimal condition, the protoplast yield ranged from 2.4$\times$10$^7$ to 3.0$\times$10$^7$ protoplasts from 400mg of damp mycelia of various auxotrophic mutant strains. The regeneration frequency ranged from 26.6 to 42.4% and the spontaneous reversion frequency of the protoplasts on the regeneration minimal medium was less than 10$^7$. The optimal concentration of PEG 6000 was 20%, and exposure of protoplasts to PEG for 10 min was found to be sufficient for protoplast fusion. Optimal pH of fusion mixture was deter-mined as 5.5 and l0mM of calcium chloride in fusion mixture effectively enhanced the protoplast fusion frequency. Under optimal condition, the fusion frequency between various auxotrophs ranged from 1.8$\times$10$^{-3}$ to 3.5$\times$0$^{-3}$.

Change of Fixation Disparity and Accommodation when the Fusion Contrast Varied (융합대비에 따른 주시시차와 조절의 변화)

  • Purpose: To study the change of fixation disparity and accommodation as fusion contrast is deteriorated. Methods: 16 subjects who had above 20/20 and stereopsis took part. Monocular and binocular refraction were done with Zeiss Polatest Classic whereas the critical angle for stereopsis was done with TNO. A computer programmed with Random-Dot stereogram and vernier test managed a precise change of the fusion contrast and exposure time. Results: The fixation disparity was influenced by reduction of fusion contrast and had tendancy to exophoria (p=0.0004), especially it is considerably higher when uncrossed disparity was shown to exophoric subjects. Although accommodation was not influenced by a change of fusion contrast (p=0.803), vernier acuity was influenced (p=0.0000). Conclusions: Exophoric trend arose as the fusion contrast was reduced, nevertheless there was no accommadative change.

  • This study intends to analyze T-shirt designs, particularly in connection with pop art. 444 pieces of T-shirts which conveyed the pop art spirits of well-known designers were selected for five years from 2001 to 2005, through fashion magazines(Collection: Dong-ah TV, Collezioni: Italy). The findings are as follows. As for formative characteristics of pop art in T-shirts, previous studies were analyzed to set classifications criteria such as popularization of images, eroticism, lettering and graffiti, assemblage. The design factor of the image popularization includes everyday images, cartoon, celebrities, and caricature. Everyday images ranked first followed by cartoon, caricature and celebrities. The design factor of eroticism ranges from the see-through look, partial exposure of human body, symbolization of underwear, and to printing of sexy images. The see-through look was ranked first, followed by partial exposure of human body, symbolization of underwear, and printing of sexy images. The design factor of lettering and graffiti encompasses letters, numbers, symbols and logos. Adoption of letters, numbers or symbols was ranked first, followed by brand logos and graffiti. The design factor of Assemblage is closely related to a three-dimensional effect. Varied expressions are possible: combination of two different textiles and fusion of textile and non-textile. Combination of two different textiles are higher than fusion of textile and non-textile.

Facile preparation of self-assembled wool-based graphene hydrogels by electron beam irradiation

  • Three dimensional self-assembled graphene hydrogels were easily fabricated by electron beam irradiation (EBI) using an aqueous solution of wool/poly(vinyl alcohol) and graphene oxide (GO). After exposure to various levels of EBI radiation, the highly porous, self-assembled, wool-based graphene hydrogels were characterized using scanning electron microscopy and Fourier-transform infrared spectroscopy; to determine the gel fraction, degree of swelling, gel strength, kinetics-of-swelling analyses and removal of hexavalent chromium (Cr(VI)) from the aqueous solution. X-ray diffraction results confirmed that EBI played a significantly important role in reducing GO to graphene. The adsorption equilibrium of Cr(VI) was reached within 80 min and the adsorption capacity was dramatically increased as the acidity of the initial solution was decreased from pH 5 to 2. Changes in ionic strength did not exert much effect on the adsorption behavior.

  • To evaluate the radioprotective effects of Bujeongsaengjintang studies were done experimentally. The results were obtained as follows: 1. WBC, Platelet and RBC were significantly increased in Bujeongsaengjintang treated group as compared with control group after exposure to radiation by Liniac. 2. By FACS analysis of splenic leukocyte after exposure to radiation by Liniac, T cell, T-helper cell and macrophase were significantly increased in Bujeongsaengjintang treated group. 3. In histological changes of ileum and jejunum of Balb/C mice after exposure to radiation by Liniac, exclusion and fusion of villi were decreased in Bujeongsaengjintang treated group as compared with control group. From above results, it is suggested that Bujeongsaengjintang is available to a clinic for the protection from damage by radiotherapy to cancer.

Increase in the Contents of Ginsenosides in Raw Ginseng Roots in Response to Exposure to 450 and 470 nm Light from Light-Emitting Diodes

  • An light-emitting diode (LED)-based light source was used as a monochromatic light source to determine the responses of raw ginseng roots (Panax ginseng Meyer) to specific emission spectra with respect to the production of ginsenosides. The ginsenoside content in the ginseng roots changed in response to the LED light treatments at $25^{\circ}C$ relative to the levels in the control roots that were treated in the dark or at $4^{\circ}C$ for 7 d. Ginseng roots were exposed to LEDs with four different peak emission wavelengths, 380, 450, 470, and 660 nm, in closed compartments. Compared with the control $4^{\circ}C$-treated roots, roots that were treated with 450 and 470 nm light showed a significantly increased production of ginsenosides (p<0.05), with increases of 64.9% and 74.1%, respectively. The contents of the ginsenosides $Rb_2$, Rc, and $Rg_1$ were significantly higher (p<0.05) in the 450 and 470 nm-treated root samples. The ratio of protopanaxadiol ginsenosides ($Rb_1$, $Rb_2$, Rc, and Rd) to protopanaxatriol ginsenosides ($Rb_1$, $Rb_2$, Re, and Rf) was significantly higher (p<0.05) in the 450 and 470 nm-treated root samples than in the control $4^{\circ}C$-treated roots. This is the first report that demonstrates the increase and conversion of ginsenosides in raw ginseng roots in response to exposure to LED light.
