Directed evolution (DE) of desired locus by targeted random mutagenesis (TRM) tools is a powerful approach for generating genetic variations with novel or improved functions, particularly in complex genomes. TRM-based DE involves developing a mutant library of targeted DNA sequences and screening the variants for the desired properties. However, DE methods have for a long time been confined to bacteria and yeasts. Lately, CRISPR/Cas and DNA deaminase-based tools that circumvent enduring barriers such as longer life cycle, small library sizes, and low mutation rates have been developed to facilitate DE in native genetic environments of multicellular organisms. Notably, deaminase-based base editing-TRM (BE-TRM) tools have greatly expanded the scope and efficiency of DE schemes by enabling base substitutions and randomization of targeted DNA sequences. BE-TRM tools provide a robust platform for the continuous molecular evolution of desired proteins, metabolic pathway engineering, creation of a mutant library of desired locus to evolve novel functions, and other applications, such as predicting mutants conferring antibiotic resistance. This review provides timely updates on the recent advances in BE-TRM tools for DE, their applications in biology, and future directions for further improvements.
이 연구는 중학교 '진화' 단원을 디지털 교재로 개발하여 학생들의 진화 개념 형성과 학습에 대한 흥미에 효과가 있는지 알아보고자 하였다. 오랜 시간에 의한 생물의 변화를 설명하는 생물 진화에 대한 설명은, 많은 내용을 다양하게 제공할 수 있고, 영상을 제작하고 편집할 수 있으며, 어려운 개념을 재미있게 제공할 수 있는 디지털 교과서가 서책형 교과서를 통해 이루어질 때 효과적일 수 있다. 연구를 위해 먼저 5E 순환학습 모형을 기반으로 전자책 저작 도구인 iBooks Author를 활용하여, 4차시로 구성된 진화수업을 만들었다. 개발한 디지털 교재의 효과 검증을 위해 서책형 수업과 디지털 교과서를 활용한 수업을 비교했다. 서책형 교재를 통한 수업과 디지털 교재를 활용한 수업 모두 진화 개념 형성에 유의미한 효과를 보였으나 생명 과학 흥미도와 진화 흥미도는 디지털 교재 수업 집단에서만 유의미하게 증가하였다. 또한 학생을 유형별로 나누어 디지털 교과서 효과를 확인한 결과 디지털 리터러시에 따라 스마트 기기에 친숙한 그룹이 수업에 더 적극적이었고 흥미가 높았다. 개발된 디지털 교재의 만족도 역시 디지털 리터러시가 높은 그룹에서 긍정적인 점수를 나타냈다. 이 연구 결과는 진화 영역 디지털 교재 개발은 어려운 진화 개념을 쉽고 재미있게 접근할 수 있는 수업 도구가 될 수 있음을 알려준다.
We subcloned two chicken H3 histone genes and transfected them into Rat 3 cell line. One contains 300 bp 5' to its cap site and the other contains 130 bp 5' to its cap site when cloned into plasm ids. Both of them showed 5' phase specific expression of their mRNA about 8 fold higher (during 5' phase) than during Gl phase. This means that only 130 bp 5' to its cap site was enough to confer cell cycle regulated expression of the latter gene. The DNA sequences of their 5' flanking region did not reveal any particular homologies or subtype-specific sequences. The DNA sequence data also showed that even though the protein coding regions of the histone genes have been conserved exceptionally well throughout evolution, their 5' untranslated regions have not been conserved as well.
We examined the relations between CME speed and properties of magnetic helicity in the source region such as helicity injection rate and total unsigned magnetic flux, which reflect the magnetic energy in the active region. For this, we selected 22 CMEs occurred during the increasing phase of solar cycle 24, which shows extremely low activities and classified them into two groups according to evolution pattern of helicity injection rate. We then compared the relations with those from previous study based on the events in solar cycle 23. As the results, we found several properties as follows: (1) Both of CME speed and helicity parameters have very small values since we only considered increasing phase; (2) among 22 CMEs, only 6 events (27%) are classified as group B, which show sign reversal of helicity injection and they follow behind of appearance of group A events. This fact is well coincide with the trend of solar cycle 23 that only group A events was observed in the first 3 years of the period; (3) as the solar activity is increasing, the CME speed and helicity parameters are also increasing. Based on the observations of solar cycle 23, the helicity parameters was still increasing in spite of decreasing solar activity after maximum period.
오늘날 널리 개발되고 있는 생태계 모방형 시스템[1]은 대규모 네트워크 응용 서비스를 위한 효과적인 시스템 모델로 각광 받아왔지만, 이런 모형의 특징인 확장성과 적응성, 생존성 등을 응용에 맞추어서 구현하기 어렵다는 문제점을 가지고 있다. 본 논문에서는 이런 문제점을 해결하기 위하여 에코전트(Ecogent)라는 멀티 지능형 모바일 에이전트와 이런 에코전트가 세가지 생태계 특징을 위한 서비스를 제공하는 플랫폼을 설계하였다. 이 생태계 모델 플랫폼은 두 부분으로 나누어 설계 하였는데, Registration, Life Cycle, Migration, Communication, Location, Fault Tolerance와 같은 모바일 에이전트의 기본 기능들을 제공하는 ERS(Ecogent Runtime Services) 플랫폼과 Evolution & Stigmergy Control의 기능을 통해 진화력과 적응성을 제공하는 BIO 플랫폼으로 나누어 설계 및 구현하였다. 생존력과 자율성을 갖춘 에코전트와 융통성 및 확장성을 위해서 단순하게 모듈화된 플랫폼 구조는 생태계를 모방하는 다양한 시스템 모델 응용들의 개발을 쉽게 구현할 수 있도록 도와준다.
본 연구에서는 Cu와 Cu-Zn 합금의 저주기 피로 동안 발달한 전위 하부조직의 변화를 비파괴적으로 구분하고 평가하고자 하였다. 비파괴시험으로 초음파속도, 전기비저항 그리고 양성자소멸시간을 측정하였다. 서로 다른 적층결함 에너지를 갖는 Cu와 Cu-Zn에 대해 반복피로시험을 수행하고 이들 재료에서의 전위거동과 비파괴평가 파라미터와의 상관성을 연구하였다. Cu는 전위셀 하부구조를 형성하였지만, Cu-Zn 합금은 피로 사이클에 따라서 전위밀도는 증가하고 단지 평면배열의 전위구조를 형성하였다. 상온에서의 반복적인 피로에 의해 발달한 격자결함인 전위와 공공으로 인해 초음파속도의 감소, 전기비저항의 증가 그리고 양성자 소멸시간이 증가하였다. 비파괴평가파라미터의 지속적인 변화를 보이는 평면배열의 전위구조를 갖는 Cu-Zn에서와 달리, Cu에서는 전위셀구조가 발달하면서 더 이상의 큰 변화를 보이지 않았다.
The non-noble 1D nanofibers(NFs) prepared by electrospinning and calcination method were used as oxygen evolution reaction (OER) electrocatalyst for water electrolysis. The electrospinning process and rate of solution composition was optimized to prepare uniform and non-beaded PVP polymer electrospun NFs. The diameter and morphology of PVP NFs changed in accordance with the viscosity and ion conductivity. The clean metal precursor contained electrospun fibers were synthesized via the optimized electrospinning process and solution composition. The calcined $CuCo_2O_4$ NFs catalyst showed higher activity and long-term cycle stability for OER compared with other $Co_3O_4$, $NiCo_2O$ NF catalysts. Furthermore, the $CuCo_2O_4$ NFs maintained the OER activity during long-term cycle test compared with commercial $CuCo_2O_4$ nanoparticle catalyst due to unique physicochemical and electrochemical properties by1D nanostructure.
이 연구는 생명과학교사들의 진화개념 향상을 위한 논변활동 프로그램을 개발하고 이를 교사교육에 적용하여 진화개념에 대한 논변활동 특성을 분석함으로써 효과를 알아보고자 하는 것이었다. 이를 위해 먼저 진화를 이해하는데 필요한 주요 개념들을 문헌 조사를 통해 추출하여 '변이', '변이의 유전', '경쟁', '자연선택과 적응', '차등적 생식력', '개체군 내 개체분포의 변화', '대진화'를 주요개념으로 선정하였고, 각각의 개념에 대한 논변활동 질문지를 개발하였다. ADI모형을 변형한 7단계로 이루어진 순환적 논변활동 학습모형도 개발하였다. 총 7회에 대학원 교과교육 수업에 적용하였다. 현직 생명과학교사 4명과 예비 생명과학교사 2명을 대상으로 남, 여 3명씩 두 개의 소집단으로 구성하여 논변활동을 진행하였다. 프로그램이 진행되는 동안 녹음을 통한 담화와 논변글쓰기의 자료를 수집하여 분석하였다. 논변글쓰기는 '설명의 충분성', '설명의 개념적 수준', '증거의 수준', '정당화의 적절성'의 4가지 범주를 기준으로 논변의 질 수준을 분석하여 사전사후에 진화개념의 변화 정도를 확인하였고, 담화분석을 통해 진화주요개념에 대한 논변활동 특성을 분석하였다. 논변글쓰기와 담화를 분석한 결과 개발된 논변활동 모형과 프로그램은 생명과학교사들의 진화개념 이해 향상에 효과적이었다. 이 연구는 논변활동을 통한 교수학습 전략이 생명과학교사교육을 위한 효과적인 진화개념학습 전략이될 수 있음을 보여주었다.
Since the development of surface magnetic features should reflect the evolution of the solar magnetic field in the deep interior of the Sun, it is crucial to study properties of sunspots and sunspot groups to understand the physical processes working below the solar surface. Here, using the data set of sunspot groups observed at the ButterStar observatory for 3,364 days from 2002 October 16 to 2011 December 31, we investigate temporal change of sunspot groups depending on their Z$\ddot{u}$rich classification type. Our main findings are as follows: (1) There are more sunspot groups in the southern hemisphere in solar cycle 23, while more sunspot groups appear in the northern hemisphere in solar cycle 24. We also note that in the declining phase of solar cycle 23 the decreasing tendency is apparently steeper in the solar northern hemisphere than in the solar southern hemisphere. (2) Some of sunspot group types make a secondary peak in the distribution between the solar maximum and the solar minimum. More importantly, in this particular data set, sunspot groups which have appeared in the solar southern hemisphere make a secondary peak 1 year after a secondary peak occurs in the solar northern hemisphere. (3) The temporal variations of small and large sunspot group numbers are disparate. That is, the number of large sunspot group declines earlier and faster and that the number of small sunspot group begins to rise earlier and faster. (4) The total number of observed sunspot is found to behave more likewise as the small sunspot group does. Hence, according to our findings, behaviors and evolution of small magnetic flux tubes and large magnetic flux tubes seem to be different over solar cycles. Finally, we conclude by briefly pointing out its implication on the space weather forecast.
The microstructural evolution of AA1050/AA6061 complex aluminum alloy, which is fabricated using an accumulative roll-bonding (ARB) process, with the proceeding of ARB, was investigated by electron back scatter diffraction (EBSD) analysis. The specimen after one cycle exhibited a deformed structure in which the grains were elongated to the rolling direction for all regions in the thickness direction. With the proceeding of the ARB, the grain became finer; the average grain size of the as received material was $45{\mu}m$; however, it became $6.3{\mu}m$ after one cycle, $1.5{\mu}m$ after three cycles, and $0.95{\mu}m$ after five cycles. The deviation of the grain size distribution of the ARB processed specimens decreased with increasing number of ARB cycles. The volume fraction of the high angle grain boundary also increased with the number of ARB cycles; it was 43.7% after one cycle, 62.7% after three cycles, and 65.6% after five cycles. On the other hand, the texture development was different depending on the regions and the materials. A shear texture component {001}<110> mainly developed in the surface region, while the rolling texture components {011}<211> and {112}<111> developed in the other regions. The difference of the texture between AA1050 and AA6061 was most obvious in the surface region; {001}<110> component mainly developed in AA1050 and {111}<110> component in AA6061.
