A study of apoptosis induction of Euonymus alatus (Thunb.) Sieb via mitochondrial pathway prooxidant in leiomyomal smooth muscle cells (귀전우(鬼箭羽)의 인간 자궁근종 세포에서 미토콘드리아 경로를 통한 산화제로서 apoptosis 유도작용에 관한 연구)

  • Purpose : 귀전우(Euonymus alatus, EA)는 현재까지 항종양활성을 나타낸다고 보고되었지만 그 작용 메커니즘에 대해서는 아직 밝혀지지 않은 채 남아 있다. 본 연구에서는, 자궁근종세포(ULSMC)에서 EA의 분자적 수준에서의 작용메커니즘을 연구${\cdot}$검토하고자 하였다. Methods : EA의 열수추출액이 자궁근종세포(ULSMC)와 caspase-3 pretense의 활성도에 미치는 영향을 측정하였다. Results : 우리는 자궁근종에서 EA 유도 세포독성의 메커니즘을 검토하였는바, 근종 세포들은 20-200g/ml 농도의 EA추출물에 6시간 배양될 때, caspase-3가 활성화되고, 그때 세포들은 apoptosis를 유발하게 되었다. EA에 의한 apoptosis의 유도가 진행되었으며, cytochrome- c의 세포질분획에서 양적증가가 caspase-3의 활성보다도 우세하였다. GSH합성의 저해제인 5mM buthionine용액에 전처리는 EA유도 apoptosis를 용이하게 하지만 pan-caspase inhibitor인 Z-VAD-fmk용액 전 처리는 부분적으로 apoptosis유도를 억제하였다. 한편, EA는 건강한 지원자들로 부터 채취한 말초혈액 단핵세포들에 있어서는 독성의 효과는 없었다. Conclusion : 이들 결과들은 EA가 prooxidant로 작용을 하고 그리고 caspase-3 activation과 mitochondrial pathway를 경유하는 apoptosis를 유발한다는 것을 나타낸다. EA의 탕제약제로서 열수추출액이 항산화활성뿐만 아니라, 종양세포에 대한 세포독성효과를 나타낸다고 보고된 바, 이에 향후 근종치료에 대한 임상연구가 필요할 것으로 보인다.

Antidiabetic Activity of Formula Containing Euonymus Alatus (Thunb.) Sieb. and Mori Folium in Multiple Low Dose Streptozotocin-induced Diabetic Rats (저용량 스트렙토조토신 유도 당뇨 흰쥐에서 화살나물.상엽 복합 처방의 항당뇨 활성)

  • We investigated the hypoglycemic effect of formula containing Euonymus alatus (EA) and Mori Folium (MF) in multiple low dose (MLD) streptozotocin (STZ)-induced diabetic rats. In order to iduce hyperglycemic state 25 mg/kg of STZ was injected intraperitoneally for 5 consecutive days. SD rats were randomly divided into diabetic control and treatment groups. Treatment groups were administered with either 250 mg/kg of EA and 250 mg/kg of MF (E1Ml), or 500 mg/kg of EA mixed with same dose of MF (E2M2) for 3 weeks. Blood glucose levels and body weights were measured every 5th or 6th day. E1Ml and E2M2 both significantly reduced food intake, water intake, and fasting blood and urine glucose levels as compared to those in diabetic control group in a dose dependent manner. Body weight in diabetic control group was increased slightly after 3 weeks. Treatment group, however, showed gradual increase in body weights during 3 week-period. While plasma insulin levels of the diabetic control group were decreased to the level of 387$\pm$14 pg/ml from 534$\pm$36 pg/ml, those levels in E1Ml and E2M2-treated groups were both markedly increased by 13% and 26%, respectively. Urine glucose levels in E1Ml and E2M2-treated groups were also remarkably reduced by 17 and 26% compared to the levels of diabetic control group. While expression of membrane-bound glucose transporter-4 (GLUT-4) protein in skeletal muscle was reduced by 45% in diabetic control compared to the normal control, GLUT-4 protein expressions in E1Ml and E2M2-treated groups were augmented by 2 and 3.5 times compared to the diabetic control, respectively. Pancreatic HE staining experiments showed that E2M2-treated group revealed much less infiltrated mononuclear cells, indicating that E2M2 efficiently blocked insulitis induced by multiple low dose streptozotocin. Taken together, we conclude that formula containing EA and MF may prevent or delay the development of hyperglycemia through overexpression of GLUT-4 protein in skeletal muscle and prevention of insulitis.

Antioxidant Activity and Biological Properties in Extracts of Euonymus alatus (Thnub.) Sieb. (화살나무 추출물의 항산화 활성 및 생물학적 특성)

  • Antioxidant activity and biological properties in the MeOH extracts from different plant parts of Euonymus alatus (THNUB.) were measured by DPPH free radical scavenging ability and inhibition ability against xanthine/xanthine oxidase and proliferation in HL-60 cells. DPPH free radical scavenging activities in extracts of plant parts were high such as leaf, wing, root, seed and stem, respectively. The EtOAc fractions of plant parts were purified through LH-20 column chromatography and identified by GC/MS. LH-4 fraction and LH-5 fraction of leaf, stem and root showed stronger activities than other fractions in the inhibitor activity of DPPH and xanthine/ xanthine oxidase. $IC_{50}$ values of LH-4 fraction eluted from stem extracts showed such as 2.38 and 5.32 in DPPH and xanthine/ xanthine oxidase assay. Polyphenolic compounds were identified in purified LH-20 fractions showed highest $IC_{50}$ value in DPPH and xanthine/xanthine oxidase assay The activity of POD according to sampling time was high in root harvested in May and leaf harvested in September, respectively. The activity of SOD showed only in the extracts of stem in plant parts. SOD and POD in leaf were similar in the patterns of isozyme to those of stem. The purified extracts from Euonymus alatus (THNUB.) exerted inhibition ability of proliferation in HL-60 cells.

Inhibition of Cyclooxygenase and Prostaglandin E2 Synthesis by Crude Methanolic Extract from Euonymus Alatus (Thunb.) Sieb in SKBR3 Human Breast Cancer Cell Line

  • In the present study, we examined the effect of crude methanolic extract (CME) from Euonymus alatus (Thunb.) Sieb on arachidonic acid (AA) cascade in SKBR3 human breast cancer cell line. CME had a potent inhibitory activity of prostaglandin E2 (PGE2) release induced by A23187, a $Ca^{2+}$ ionophore. The inhibition was concentration-dependent, with the 50 value of about 5 M. CME had no inhibitory effect on A23187-induced phosphorylation of p42/p44 extracellular signal regulated kinase/mitogen-activated protein kinase or on the liberation of [14C]-AA from the cells labeled with [14C]-AA. However, CME concentration-dependently inhibited the conversion of AA to $PGE_2$ in microsomal preparations, showing its possible inhibition of cyclooxygenase (COX). In enzyme assay in vitro, CME inhibited the activities of both constitutive COX (COX­I) and inducible COX (COX-2) in a concentration-dependent manner, with the 50 values of about 0.8 and 2M, respectively. Lineweaver-Burk plot analysis indicated that CME competitively inhibited the activities of both COX-l and -2. This study is a first demonstration that CME directly inhibits COX activity.

Growth inhibitory and antioxidative effects of crude methanolic extract from Euonymus alatus (Thunb.) Sieb on SKBR3 human breast cancer cell line (SKBR3 유방암세포주에 대한 귀전우 메탄올 추출물의 성장억제 및 항산화효과)

  • 이 연구는 SKBR3 인간 유방암세포주에 대한 귀전우 메탄올추출물(CME)의 증식억제 효과, 세포사 유발 효과 및 항산화 활성을 확인하기 위해 이루어졌다. SKBR3 유방암세포주는 48시간동안 다양한 농도($0{\sim}20{\mu}g/m{\ell}$)의 CME가 제공된 곳에서 배양되었고, MMT 측정법을 이용하여 세포생존율을 평가하였다. CME의 50%에서 효과를 나타내게 하는 약물농도인 $ED_{50}$ (effective dose 50%)은 $6.5{\pm}0.3{\mu}g/m{\ell}$) 이며, 투여량이 증가함에 따라 농도에 의존하여 세포증식이 억제되는 것으로 나타났다. 또한 CME의 증식억제 효과는 유방암세포주의 세포사와 관련됨의 세포의 형태학적 변화와 올리고뉴클레오솜 DNA 파편의 확인을 통해 알 수 있었다. 또한 다양한 농도와 배양시간에서 CME가 ROS의 생산을 억제한다는 것을 확인할 수 있었다. 이런 결과들은 귀전우의 메탄올추출물이 SKBR3 인간 유방암세포주에 대해 강력한 증식억제 효과와 강한 항산화 효과를 나타낼 뿐만 아니라 세포사를 유도하는 효능을 가지고 있음을 시사한다. 이러한 효능은 약물에 대한 노출시간과 투여량에 의존하였다. 따라서 귀전우는 다양한 기전에 의해 유방암 세포에 대한 억제효과를 가질 수 있을 것으로 인식할 수 있다.

Physiological Activity and Effects on Lipid Peroxidation of Hot Water-Extract Obtained from Euonymus alatus in Cultured Rat Hepatocyte (Rat의 hepatocyte에서 $amyloid-{\beta$}$로 유발된 세포사, 지질과산화 및 세포산화에 대한 귀전우 열수 추출물의 보호효과)

  • This study was carried out to investigate the physiological activityof the water extract from EA. The present study was done to investigate the effects of EA on cultured hepatocyte cell system and lipid peroxidation in $A{\beta}$ treatment conditions. Pretreatment of EA attenuated in cell cytotoxicity enhanced by increasing concentrations of $A{\beta}$. MDA level induced by $A{\beta}$ treatment was significantly increased and the level was slightly reduced by pretreatment of EA. The ability of EA to reduce cell death and MDA level induced by $A{\beta}$ suggest that EA may be a protective agent against free radical generating compounds such as $A{\beta}$. EA exhibited anti oxidative activity at all concentration tested.The extract was as good as antioxidative activity of the synthetic antioxidants, butylated hydroxy toluene and ascorbic acid. Furthermore, this was superior to that of natural antioxidant, a-tocopherol. In the presence of heavy metal ions ($Fe^{2+},{\;}Zn^{2+}$), EA showed strong antioxidative activity. The extracts showed about 3075% in the nitrite scavenging effect under pH 1.2 and $37^{\circ}C$ for 1 hr. There was significant difference among concentration of extracts.

Screening of Stat3 inhibitory effects of Korean herbal medicines in the A549 human lung cancer cell line

  • Background: The transcription factor signal transducer and activator of transcription 3 (Stat3)is constitutively activated in many human cancers. It promotes tumor cell proliferation,inhibits apoptosis, induces angiogenesis and metastasis, and suppresses antitumor hostimmune responses. Therefore, Stat3 has emerged as a promising molecular target for cancertherapies. In this study, we evaluated the Stat3-suppressive activity of 38 herbal medicinestraditionally used in Korea.Methods: Medicinal herb extracts in 70% ethanol were screened for their ability to suppressStat3 in the A549 human lung cancer cell line. A Stat3-responsive reporter assay system wasused to detect intracellular Stat3 activity in extract-treated cells, and Western blot analyseswere performed to measure the expression profiles of Stat3-regulated proteins.Results: Fifty percent of the 38 extracts possessed at least mild Stat3-suppressive activities(i.e., activity less than 75% of the vehicle control). Ethanol extracts of Bupleurum falcatumL., Taraxacum officinale Weber, Solanum nigrum L., Ulmus macrocarpa Hance, Euonymus alatusSieb., Artemisia capillaris Thunb., and Saururus chinensis (Lour.) Baill inhibited up to 75% of thevehicle control Stat3 activity level. A549 cells treated with these extracts also had reducedBcl-xL, Survivin, c-Myc, and Mcl-1 expression.Conclusion: Many medicinal herbs traditionally used in Korea contain Stat3 activity-suppressing substances. Because of the therapeutic impact of Stat3 inhibition, these resultscould be useful when developing novel cancer therapeutics from medicinal herbs.
