Antioxidative activities of 50% ethanolic extracts from Du-zhong (Eucommia ulmoides Oliver) leaf and bark were investigated. Yields of the leaf and bark extract were 8.1±0.31 and 17.4±0.89%, respectively. Polyphenol contents of the leaf and bark extract were 64.1±3.35 and 42.4±2.38 ㎍ gallic acid equivalents/mg, respectively. Flavonoid contents of the leaf and bark extract were 24.0±3.15 and 36.7±3.18 ㎍ quercetin equivalents/mg, respectively. As concentration of the leaf and bark extract increased, their antioxidative activities proportionally increased. EC50 values of the leaf and bark extract for cation radical scavenging were 560.6±17.65 and 1,357.4±8.45 ㎍/mL, respectively. EC50 values of the leaf and bark extract for free radical scavenging were 574.2±14.70 and 2,103.1±108.59 ㎍/mL, respectively. EC50 values of the leaf and bark extract for ferric reducing antioxidant power were 319.9±13.42 and 705.9±26.08 ㎍/mL, respectively. EC50 values of the leaf and bark extract for nitrite scavenging were 2,329.2±35.11 and 5,467.6±243.92 ㎍/mL, respectively. In the presence of 74.8 ㎍/mL of the leaf extract and 177.2 ㎍/mL of the bark extract, linoleic acid peroxidation was inhibited by 70.0 and 79.1%, respectively. The Du-zhong leaf extract possessed higher antioxidative activities than its bark extract.
To evaluate a potential possibility of Eucommia ulmoides Oliver as a functional food, ACE (angiotensin converting enzyme) inhibitory activities of leaf, bark, stem and 4 compounds isolated from E. ulmoides were tested. The 4 compounds were isolated and purified by silica gel column chromatography, thin layer chromatography and reverse phase column chromatography. Compound I was pinoresinol-4,4'di-O-${\beta}$-D-glucoside (PG) and compound II was dehydrodiconiferyl alcohol 4,${\gamma}$'-di-O-${\beta}$-D-glucopyranoside (DAG) originating from Eucommial Cortex. The highest amount of PC was present at raw and roasted bark as 135.13 mg% and 163.67 mg%, and the highest amount of DAG was present at raw and roasted leaf as 117.93 mg% and 133.93 mg% respectively. In an ACE inhibition test, 10 mg/ml of roasted leaf, raw and roasted bark extracts of E. ulmoides Oliver were 77.49%, 75.72% and 75.36% respectively, and 10mg/ml of PC and DAG were shown to be 78.51 and 81.20% respectively. $IC_{50}$ values of PG and DAG were 0.6${\pm}$0.2 and 0.5${\pm}$0.2 mg/ml respectively.

Inhibitory effects of allergic contact dermatitis of hot water extract of Duchung (Eucommia ulmoides Oliver) leaf, bark and stem growing at Sancheong-gun were investigated for female BALB/c mouse induced by repeat elicitation of DNCB (2,4-dinitro-chlorobenzene). Skin reactions, consisting of increased ear thickness and the presence of ear inflammation, were observed in mice treated with DNCB and Duchung. Weight of lymph node, spleen and thymus in mice treated with Duchung extracts were lower than that of mouse treated with DNCB. Ear weight of mouse treated with Duchung extracts was decreased by increasing the concentration of sample as compared to control group and dropped as low as control level at 1,000 mg/kg. Ear thickness became thinner as test time on Duchung extract progressed. MDA (malondialdehyde) contents in liver tissue were not different in sample group with DNCB group, but were different in ear tissue. NO (nitric oxide) contents was decreased in Duchung extract groups at serum and ear tissues as compared to 1% DNCB group. In the present study, the results suggested that Duchung extract inhibits inflammatory response and oxidative damage induced DNCB allergen.

This study was performed to examine the neuroprotective effect of the ethyl acetate fraction from Eucommia ulmoides oliver leaf (EFEL) on PM2.5-induced cytotoxicity. EFEL had higher total phenolic and flavonoid contents than the other fractions. In ABTS and DPPH radical scavenging activities, the IC50 of EFEL was measured as 212.80 and 359.13 ㎍/mL, respectively. To investigate the neuroprotective effect of EFEL, MTT and DCF-DA assays were performed on HT22, MC-IXC, and BV-2 cells. EFEL effectively decreased PM2.5-induced intercellular reactive oxygen species (ROS) content and inhibited PM2.5-induced cell death. In the results of protein expression related to cellular cytotoxicity on microglial cells (BV-2), EFEL had an improvement effect on cell apoptosis and inflammatory pathways. Rutin and chlorogenic acid were identified as the main physiological compounds. Moreover, it was expected that EFEL, including rutin and chlorogenic acid, could be functional food substances with neuroprotective effects against PM2.5-induced oxidative stress.
Soybean sprouts presoaked to leaf extracts were on sale as taking a growing concern about wellbeing foods. The study was carried out to compare the effects of culture period after presoaking tea (Thea sinensis L.) and hard rubber tree (Eucommia ulmoides Oliver) leaf extracts on lateral root formation, growth and morphological characters of the sprouts. Seeds of three cultivars, Pungsannamulkong, Sowonkong and Junjery, were imbibed for 5 hours into 10% solutions immediately before 5, 6 and 7 day cultures. The cultured sprouts were classified into 4 categories to calculate their composition rates on the base of hypocotyllengths; >7 cm (A),4 to 7 cm (B),< 4 cm (C) and not germinated (D), and their morphological characters, fresh and dry weights were measured. Rate of A was the lowest in Junjery of the three cultivars, while that of C showed reverse result compared to A. Similar results to that of A were observed between the three cultivars in sprout length including hypocotyl and root, diameters in middle and hook parts of hypocotyl, total fresh and dry weights. Regardless of tea and hard rubber tree leaf extract treatments, longer culture periods increased rate of A due to decrements of B, C and D, and also did the growth and the morphological characters mentioned above. Although longer culture periods reduced the increment of lateral roots, the latter treatment was more delayed in their formation and less in the roots per sprout than the former one. The growth and the morphological characters above were less in the latter treatment than in the former one in general, meaning that hard rubber tree leaf extract could more repress growth of soybean sprouts than tea leaf extract.

Methanol extract (MeOH), n-hexane (Hx), chloroform ($CHCl_3$), ethyl acetate (EA), butanol (BuOH) and aqueous ($H_2O$) fractions of Eucommiae Cortex including geniposidic acid (GA), geniposide (GP) and aucubin (AU) were tested for their therapeutic efficacy on osteoporosis. The contents of GA, GP and AU in the cortex and leaf of Eucommia ulmoides Oliver were quantified by HPLC. The effect of Eucommiae Cortex on the induction of growth hormone (GH) release was studied by using rat pituitary cells. The proliferation of osteoblast-like cells increased by herbal extracts was assayed using a tetrazolium (MTT), alkaline phosphatase (ALP) activity, and [$^3H$]-proline incorporation assays. The inhibition of osteoclast was studied by using the coculture of mouse bone marrow cells and ST-2 cells. As a result, the GA, GP and AU were present in the cortex more than in the leaf of E. ulmoides Oliver. The MeOH (1mg/mL), Hx, $CHCl_3$ and EA fractions (each 20 $\mu$ g/mL) had potent induction of GH release. The $CHCl_3$ exhibited the potent proliferation of osteoblasts. The AU, GP and GA were increased proliferation of osteoblasts. In addition, GA ($IC_{50}: 4.43{\times}10^{-7}$M), AU and GP were significantly inhibited proliferation of osteoclast. In summary, it is thought that the components in a part of the fractions of Eucommiae Cortex participate in each step of mechanism for activating osteoblast to facilitate osteogenesis, and suppress osteoclast activity to inhibit osteolysis.
The effect of tu-chung (Eucommia ulmoides, Oliver) leaf meal on reducing lipid accumulation in chickens fed 1% cholesterol containing diet was studied. Forty male White Leghorn chickens aged 56 days were weighed and divided into four groups of ten chickens, and fed diets with or without 1% dietary cholesterol which were supplemented with 0 and 5% tu-chung. Tu-chung supplementation to the diet without cholesterol increased acetyl-CoA carboxylase (p<0.01) but decreased 3-hydroxy-3-methylglutaryl-CoA reductase activities (p<0.01) with no effect on fatty acid synthetase activities. However, its supplementation to the diet with cholesterol had no effect on these three enzyme activities as compared with the cholesterol containing diet without tu-chung. Tu-chung supplementation to the diet without cholesterol increased hepatic triglyceride (p<0.01), whereas its supplementation to the diet with cholesterol decreased it (p<0.01). Tu-chung supplementation to the diet with cholesterol decreased plasma cholesterol ester, free cholesterol, phospholipids (p<0.05) and triglyceride (p<0.01) as compared with the cholesterol containing diet without tu-chung. Supplementation of tu-chung to the diet without cholesterol decreased plasma free cholesterol (p<0.05). It is concluded that tu-chung leaf meal reduced an increase in lipid accumulation in chickens stimulated by 1% cholesterol feeding.
Natural products has been occasionally used in place of growth regulators to control lateral root formation and growth of soybean sprouts. The study was done to measure the effects of treatment concentration of hard rubber tree (Eucommia ulmoides Oliver) leaf extract on lateral root formation, growth and morphological characters of the sprouts. Seeds of three cultivars, Pungsannamulkong, Sowonkong and Junjery, were imbibed for 5 hours into water, its 5 and 10% solutions immediately before 6 day culture. The cultured sprouts were classified into 4 categories to calculate their composition rates on the base of hypocotyl lengths; >7 cm (A),4 to 7 cm (B),<4 cm (C) and not germinated (D), and their morphological characters, fresh and dry weights were measured. Rate of A was decreased with increased concentrations, while that of C showed reverse result compared to A. Regardless of concentrations, the former rate was less in cv. Junjery than in cv. Pungsannamulkong and Sowonkong than in cv. Junjery although the latter rate was truly opposite to the former one. In 10% treatment concentration, lateral roots were less formed in cv. Junjery although nearly formed in cv. Pungsannamulkong and Sowonkong. In all the 3 cultivars, lateral roots per sprouts were decreased with increased treatment concentration. shorter and thicker sprout was observed in cv. Junjery than in the other cultivars, and in increased concentrations compared to relatively lower ones. Total fresh weight was the greatest in cv. Sowonkong due to mainly increment of its hypocotyl, but was reduced with increased concentration due to mainly decrement of hypocotyl in all the cultivars.

The study was done to compare the effects of ocher, chitosan, corn kennel, tea (Thea sinensis L.) and hard rubber tree (Eucommia ulmoides Oliver) leaf extracts on lateral root formation, growth and morphological characters of the sprouts. Seeds of three cultivars, Pungsannamulkong, Sowonkong and Junjery, were imbibed for 5 hours into their 10% solutions and cultured for 6 days. The cultured sprouts were classified into 4 categories to calculate their composition rates on the base of hypocotyl lengths;>7 cm (A), 4 to 7 cm (B), < 4 cm (C) and not germinated (D), and their morphological characters, fresh and dry weights were measured. Composition rate of A was the lowest in Junjery of the three cultivars, while that of C showed reverse result compared to A. This results was the most distinct in hard rubber tree leaf extracts (HRTLE) of the five treatments. In HRTLE treatment, lateral root formation rate were formed in almost of Sowonkong although reduced in order of Pungsannamulkong and Junjery. However, there was no significant difference between the other treatments. Lateral roots per sprout were the lowest in HRTLE treatment of the 5 treatments. In all treatments except the chitosan treatment, the roots were most formed in Sowonkong but least in Junjery. Sprout length adding hypocotyl and root was the shortest in Junjery compared to the other two cultivars. and was the longest in tea leaf extract treatment but the shortest in HRTLE treatment. The result in total fresh weight of sprouts was similar to that of the sprout length.
