Comparative effect of silkworm powder from 3 Bombyx mori varieties on ethanol-induced gastric injury in rat model

  • Gastric ulcer is a clinical symptom characterized by inflammation of the gastric mucosa. Stress and alcohol consumption have been identified as the major cause of gastric ulcer. However, the effects of silkworms on ethanol-induced gastric ulcer have not been studied yet. The mature silkworms that are difficult to eat have become easier to ingest due to recent technological development to make steaming and freeze-drying mature silkworm larval powder (SMSP). In this study, we investigated whether three silkworm varieties, Baekokjam, Golden-silk and Yeonnokjam could alleviate ethanol-induced gastric mucosal damage in vivo. Sprague-Dawley rats pretreated with 3 SMSPs (0.1 or 1 g/kg BW) or normal diet (AIN-76A) were exposed to absolute ethanol (3 g/kg BW, 3 h) by oral gavage. Morphological examination included ulcer index as a measurement of hemorrhages and hematoxylin and eosin staining was performed to analyze the severity of gastric ulcer. Results of macroscopic examination suggested that all 3 SMSPs pretreatment significantly protected gastric mucosa against ethanol-induced damage. Microscopic observations demonstrated significant mucosal erosion and inflammation in ethanol-treated rats, which was abrogated in rats pretreated with 3 SMSPs. In addition, pretreatment with all 3 SMSPs showed significant decreases the expression of pro-inflammatory mediators, IL-6 and cyclooxygenase-2. Among SMSP from 3 varieties of silkworm, preadministration of 1 g/kg Baekokjam SMSP showed the most effective protective effect against ethanol-induced gastric ulcer. These results suggest that Baekokjam SMSP can be a potential gastroprotective agent against ethanol-induced gastric ulcer.

THe Effect of Chronic Ehronic Treatment and Cold stress on Catecholaminergic Enzyme activity and mRNA in Rat Brain and Adrenals

  • Sprague-Dawley male rats (150 g) were chronically treated with 5 v/v % ethanol admixed with nutritionally complete liquid diet and fed ad libitum for 3 weeks. One half of each group was exposed to cold stress at 4 ^{\circ}C either for 24 h (for determination of mRNA by in situ hybridization) or for 48 h (for determination of enzyme activity). Chronic ethanol treatment (ethanol) did not affect tyrosine hydroxylase(TH) mRNA level in locus coeruleus(LC) of brain and adrenal medulla(AM) compared to controls. Cold stress showed strong increase of TH mRNA level in LC and AM compared to controls. Pretreated ethanol reduced the increased TH mRNA level by cold stress in LC and AM. Ethanol did not affect TH activity in LC and adenal glands(adrenals). Cold stress increased TH activity in LC but not in adrenals. Pretreated ethanol did not reduce the increased TH activity by cold stress in LC but this result was not shown in adrenals. Phenylethanolamine-N-methyltransferase(PNMT) activity in $C_{1}$$C_{2}$ and adrenals increased only in ethanol treated group. THese results suggest that ethanol does not affect TH mRNA level and activity in LC and adrenals, but increases PNMT activity in $C_{1}$$C_{2}$ and adrenals in normal rat. It is also suggested that pretreated ethanol reduces the magnitude of cold stress response, that is induction of TH mRNA in LC and AM, and does not reduce the protein activation of TH that is also cold stress response in LC.

구멍쇠 미역 에탄올 추출물의 섭취가 비만유도 흰쥐의 지질농도에 미치는 영향 (Effects of Dietary Intake of Agarum cribrosum Ethanol Extract on Lipid Level in Diet-induced Obese Rats)

  • 고지방식이로 비만 및 고지혈이 유발된 실험동물에서 체중, 혈중지질, 혈청생화학적 검사 및 장기무게를 측정하여 구멍쇠 미역 추출물 투여로 인한 혈중지질 개선효과와 구멍쇠 미역 추출물의 간과 신장의 기능 및 장기무게에 미치는 영향에 대해 조사하였다. 체중 및 사료이용효율은 고지방식이에 의해 고지방군에서 증가현상을 보였으며, 구멍쇠 미역 추출물 5 %와 10 % 투여군인 HF5S과 HF10S군에서 구멍쇠 미역 추출물 투여로 인한 체중증가량 감소와 식이효율 감소현상을 보였다. 혈청 생화학적 검사에서 고지방식이에 의해 혈중 ALT, AST의 증가가 관찰되었으나 구멍쇠 미역 추출물 투여에 의해 기본식이군과 비슷한 수준으로 유지되는 것을 볼 수 있었다. 혈중 creatinine은 고지방식이 및 구멍쇠 미역 추출물 투여에 영향을 받지 않았으나 g-GT의 경우 구멍쇠미역 추출물을 10 % 첨가하였을 때는 유의적으로 감소하여 기본식이군과 고지방식이군보다 낮아졌다. 혈중지질 변화는 총 콜레스테롤, LDL-콜레스테롤, triglyceride, 동맥경화지수(AI) 모두 고지방식이에 의해 유의적인 증가현상을 보였으며, 구멍쇠 미역추출물 투여한 경우 고지방식이군에 비하여 총콜레스테롤과 LDL-콜레스테롤은 감소하였으나 동맥경화지수는 변화가 없었다. 이상의 실험결과 구멍쇠 미역 추출물은 비만유도 흰쥐에서 체중과 간의 무게 감소와 더불어 혈청의 지질성상의 개선에 긍정적인 영향을 미치는 것으로 나타났다. 이러한 결과는 구멍쇠 미역에 함유된 다량의 식이섬유소와 미네랄 등의 생리활성 물질이 지질대사를 개선시키고 지방 축적을 억제하여 비만, 고지혈증, 이상지혈증의 예방 및 개선 효과를 나타낼 것으로 기대되며 나아가 심혈관계 질환의 예방 및 치료에 긍정적인 효과가 있을 것으로 사료된다.

미역 에탄올 추출물이 지방세포 형성과정에 미치는 영향 (Anti-adipogenic Effect of Undaria pinnatifida Extracts by Ethanol in 3T3-L1 Adipocytes)

  • 미역(Undaria pinnatifada)은 낮은 칼로리 및 요오드의 원료로써 천연체중조절식품으로 알려져 있다. 미역이 체중조절식품으로 알려져 있음에도 불구하고, 지방세포 분화 및 지방축적에 관한 저해 기작은 연구가 미비하다. 본 연구에서는 3T3-L1에서 지방세포로 분화가 일어나는 단계에서 미역에탄올추출물의 효과 및 기작을 확인하였다. 미역에탄올추출물의 독성과 지방축적저해효과는 MTT assay, Oil red O staining, RT-PCR과 western blot으로 분석하였다. 미역에탄올추출물은 50 ${\mu}g/ml$의 농도에서 독성을 띄지 않았다. 3T3-L1의 분화 및 지방세포에서 triglyceride축적과정동안 50 ${\mu}g/ml$의 미역에탄올추출물을 처리하였으며, 미역에탄올추출물은 지방세포에서 triglyceride의 축적을 40% 감소시켰다. 지방세포 특이적 단백질인 Peroxisome proliferator activated receptor ${\gamma}$ ($PPAR{\gamma}$), leptin과 Hormone sensitive lipase (HSL)의 발현은 RT-PCR과 western blot으로 확인하였다. $PPAR{\gamma}$의 과발현은 지방세포의 분화를 촉진시킨다. 또한 지방세포 크기의 증가와 세포 내 triglyceride의 함량에 따라 leptin은 세포 외로 분비된다. 그러므로 $PPAR{\gamma}$와 leptin은 비만의 지표로 사용된다. 첨가한 미역에탄올추출물의 농도가 높아질수록 $PPAR{\gamma}$와 leptin의 발현이 억제되었다. 이상의 결과를 통하여, 미역의 에탄올 추출물은 지방전구세포의 분화를 억제시키며, 지방세포 내 triglyceride의 축적을 저해하는 것으로 판단된다.

흰쥐에서 SAL5의 알코올성 지방간 형성에 미치는 영향 (Effect of SAL5 on chronic ethanol-induced fatty liver model)

  • Objective : In this study, we investigated the effect of SAL5(mixing extracts of Schisandra chinensis Baillon, Artemisia capillaris Thunb., and Aloe vera Linne) on chronic ethanol-induced fatty liver model. Methods : Sprague-Dawley male rats were fed Liber-DeCarli (normal), ethanol liquid diet (control), SAL5 (200 mg/kg). We administrated the SAL5 on chronic ethanol-induced fatty liver model for 5 weeks. We measured alkaline phosphtase (ALP), alanine transminase (ALT), aspartate transminase (AST) and ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG), superoxide dismutase (SOD), catalase, glutathione (GSH) and malondialdehyde (MDA) level in liver. Liver histopathology was examined by Hematoxylin-eosin and Oil red O staining of the fixed liver tissues. Real-time PCR was performed to measure the mRNA expression of inflammatory cytokines and MMP-2, MMP-9. Results : SAL5 administration resulted in significantly decreased liver marker enzymes activities of alanine transminase (ALT), ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG) activities in liver. The control group decreased the activities of superoxide dismutase (SOD), catalase (CAT) with the reduced level of glutathione (GSH) in liver. On the other hand, SAL5 group increased the activities of SOD, CAT and the level of GSH. SAL5 delayed the development of an alcoholic fatty liver by reversing fat accumulation in the liver, as evidenced in histological observations. The gene expression of mRNA were significantly decreased at the $IL-1{\beta}$, $TNF-{\alpha}$, NOS-II and MMP-2 by SAL5. Conclusions : These results indicate that SAL5 might have protective effect chronic ethanol-induced fatty liver models.

  • In order to verify the effect of Sineungyangjin-Dan on alopecia, I experimented the hair regrowth of the normal mice and of the high butter diet-pretreated mice by feeding each group of mice a certain amount of ethanol extract of Sineungyangjin-Dan and water extract and thus obtained the results as follow; 1. The ethanol extract of Sineungyangjin-Dan was proved to promote the hair regrowth of the hair-removed normal mice. 2. 360mg/kg ethanol extract of Sineungyangjin-Dan was proved to have the effect of promoting hair regrowth by restraining the increase of free cholesterol and triglyceride but by helping the increase of free fatty acid in skin. As a result of the above study Sineungyangjin-Dan was proved to have the effect of promoting hair regrowth.

흰쥐에서 양파추출물의 알코올성 지방간 개선 작용 (Onion Favorably Affects Serum Markers of Ethanol-induced Fatty Liver in Rats)

  • 양파추출물이 알코올성 지방간에 미치는 영향을 알아보고자 흰쥐를 meal-feeding 방법으로 모든 쥐가 매일 같은 양의 식이를 공급받는 실험조건 하에서 에탄올 없이 정상식이만 섭취한 대조군, 에탄올과 정상식이와 함께 투여한 에탄올군, 양파추출물과 에탄올을 정상식이와 함께 투여한 에탄올양파추출물군으로 5마리씩 3그룹으로 나누어 사육하였다. 양파 추출물을 28일간 흰쥐에게 먹여 체중의 변화, 장기 무게 변화, 혈청 중의 glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), alkaline phosphatase (ALP), gamma-glutamyl transpeptidase (GTP), total bilirubin, total protein, total cholesterol, triglyceride를 생화학적 검사를 통해 양파추출물이 에탄올로 유도된 지방간을 예방, 개선하는지 연구하였다. 대조군과 실험군 둘다 연구 시작부터 끝까지 지속적으로 체중 증가를 보였으나 그룹 간에 유의적인 차이를 보이지 않았다. 장기 무게는 각각의 그룹 간장, 신장, 심장, 비장, 고환 등의 무게에서 유의적인 차이점이 없었다. 혈중의 지방 변화를 보면 에탄올군에서는 혈중 중성지방이 유의적으로 증가하였고, 에탄올양파추출물군에서는 혈중 중성지방이 유의적으로 낮게 나타났다. 그러나 대조군과 에탄올양파추출물군 간에는 유의점이 없었다. GOT, GPT, ALP, GTP 의 4주간 실험한 혈중 농도에서 GOT와 GPT가 에탄올군에서 유의적으로 높았고, 에탄올양파추출물군에서는 유의적으로 낮게 나타났다. 이상의 실험 결과들을 종합해 볼 때, 양파추출물을 에탄올 식이와 함께 흰쥐에게 28일간 공급하였을 때, 지방간 관련 대표적인 혈액 지표인, 중성지방, GOT, GPT의 농도가 유의적으로 감소한다는 것을 이 실험을 통해서 확인할수 있었다.

알콜과 식이지방량이 흰쥐의 간 지질조성과 간조직형태에 미치는 영향 (Effects of Alcohol Consumption and Fat Content in Diet on Chemical Composition and Morphology of Liver in Rat)

  • Effects of alchohol and fat content in a balanced diet on chemical composition and morphology of liver were investigated in growing rats. Fourth eight male rats of Sprague-Dawley strain weighing about 160g were divided into 4 groups ; high fat diet group, alcohol-administered high fat diet group, low fat diet group and alcohol-administered high fat diet group, low fat diet group and alcohol-administered low fat diet group. High and low fat diets supplied 30% and 12%, respectively, of total calorie intake from fat, and alcohol was given by adding ethanol in drinking waster at 10%. Diets contained adequate amounts of all nutrients required for rats, including lipotrpoic agents(choline and methionine) to minimize effects of factors other than alcohol on liver damage. Ratios of liver weight to body weight were statistically different among groups. Liver/dody weight ratios alcohol-administered rats were significantly higher than those of non-alcohol groups after 6 weeks treatment. Although total lipid and triglyceride per gram liver were increased in alcohol-administered rats, especially low fat diet fed rats, the values were not significantly different. Opticmicroscopical observation revealed increase in cell size and no change in morphology of liver. Examination of hepatocytes by electron microscopy showed that fat droplets were observed in all groups but enlarged in the alcohol-administered low fat diet fed rat. Contents of protein, cholesterol and phospholipid were not affected by alcohol consumption. The level of lipid peroxide was significantly lower in the livers of alcohol-administered rats than in the livers of non-alcohol groups. The results of this study indicate that even moderate alcohol drinking and dietary fat content did not affect any significant change in composition and morphology of liver until 6 week treatment but that even moderate alcohol drinking caused some signs of steatosis of liver.

Mentha canadensis attenuates adiposity and hepatic steatosis in high-fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: Obesity is a major risk factor for metabolic syndrome, a global public health problem. Mentha canadensis (MA), a traditional phytomedicine and dietary herb used for centuries, was the focus of this study to investigate its effects on obesity. MATERIALS/METHODS: Thirty-five male C57BL/6J mice were randomly divided into 2 groups and fed either a normal diet (ND, n = 10) or a high-fat diet (HFD, n = 25) for 4 weeks to induce obesity. After the obesity induction period, the HFD-fed mice were randomly separated into 2 groups: one group continued to be fed HFD (n = 15, HFD group), while the other group was fed HFD with 1.5% (w/w) MA ethanol extract (n = 10, MA group) for 13 weeks. RESULTS: The results showed that body and white adipose tissue (WAT) weights were significantly decreased in the MA-supplemented group compared to the HFD group. Additionally, MA supplementation enhanced energy expenditure, leading to improvements in plasma lipids, cytokines, hepatic steatosis, and fecal lipids. Furthermore, MA supplementation regulated lipid-metabolism-related enzyme activity and gene expression, thereby suppressing lipid accumulation in the WAT and liver. CONCLUSIONS: These findings indicate that MA has the potential to improve diet-induced obesity and its associated complications, including adiposity, dyslipidemia, hepatic steatosis, and inflammation.

Alcohol Induced Hepatic Fibrosis in Pig

  • Hepatic disease has been noted and reported for involvement various detrimental factors. Among many detrimental injury factors, alcohol has been noted for hepatitis, fatty liver, fibrosis, and hepatic cirrhosis. The purpose of this study is to develop animal model for hepatic fibrosis in pig with ethanol, and to search new anti-fibrogenic agent. Twelve male Landrace pigs were divided into 3 groups of 4 animals each. Group 1, 2 and 3 were fed with ceramic water only, ceramic water + liquid diet containing 20% ethanol and normal tap water + containing 20% ethanol for 12 weeks, respectively. At week 12, all pigs were immediately sacrificed for collection each tissue and blood. Serologically, serum ALT and AST levels were significantly reversed in group 2, comparing to group 3. They were normal range in pigs of group 1. Microscopically, macrovesicular lipid droplets and moderate necrosis were evident in the tap water + ethanol fed group 3. However, ceramic water intake group 1 showed normal. Moreover, in group 3, little fatty changes and mild necrosis were observed. Collagen fibers were detected in the spaces of surrounding periportal and interlobular areas in the group 3 of tap water + ethanol, but collagen synthesis and its thickness of fibrotic septa connecting portal tracts was markedly reduced in the group 2 of ceramic water + ethanol. In immunohistochemistry, myofibroblasts were detected in the ethanol and tap water treated group 3. No or a few myofibroblasts were observed in groups 1 and 2. CYP 2E1 was rarely detected in group 1 fed ceramic water. However, group 2 showed slightly activation of CYP 2E1 in the area of pericentral, while CYP 2E1 was significantly activated in group 3 fed tap and ethanol. Taken together above, alcohol fibrosis model in pig was established. Furthermore, ceramic water had an inhibitory and protecting ability for alcohol-induced hepatic damages.
