The investigation was aimed to study the effect of ginseng ethanol extract on the hepatic ethanol-metabolizing enzyme activity in vivo. The extract (100mg/kg/day) was administered orally to Sprague-Dawley rats for $7{\sim}10$ days and their microsomal ethanol oxidizing system(MEOS) and catalase activities were measured. The MEOS activity in the rat treated with the extract was not significantly different from that of the normal group. Microsomal fraction containing MEOS was separated and the MEOS activity was measured after preincubation for 5, 60 and 180 min, respectively. There were no significant differences in MEOS activities between the normal and treated groups preincubated for 5, 60 and 180 min. The activity in the rat treated with single i.p. injection of 95% $CCl_4$ (0.5ml/kg) was decreased by 48%, compared to the normal group and in the rat treated with the extract (100mg/kg) for $7{\sim}10$ days, the decrease of the MEOS activity was potentiated. Catalase activity in the rat treated with the extract (100mg/kg) was similar to that obtained from the normal group.
Streptococcus mutans (S. mutans) plays an important role in the information of dental plaque and it is being noticed as major causative bacteria of dental caries. In the present study, inhibitory effects of the ethanol extract of Dianthus superbus Linne (D. superbus) on the growth, acid production, adhesion and water-insoluble glucan synthesis of S. mutans were examined. The ethanol extract of D. superbus (0.5 - 4 mg/ml) significantly lowered the growth of S. mutans in a dose dependent manner. The acid production of S. mutans were inhibited by the presence of ethanol extract of D. superbus(1 - 4 mg/ml) significantly. The ethanol extract of D. superbus (0.25 - 4 mg/ml) also significantly lowered the adherence of S. mutans in a dose dependent manner. In water-insoluble glucan synthesis assay, 0.25 - 4 mg/ml of the ethanol extract of D. superbus significantly inhibited the formation of water-insoluble glucan. These results suggest that D. superbus may inhibit the caries-inducing properties of S. mutans. Further studies are necessary to clarify the active constituents of D. superbus responsible for such biomolecular activities.
Glutamate-induced oxidative stress results in neuro-degenerative disorders in many central nervous system (CNS) such as Alzheimer's disease, ischemia, Huntington's disease, and Parkinson's disease. Our study was performed to investigate neuroprotective effects of Allium hookeri extracts (leaf, root, and whole) on glutamate-induced HT22 cells. In this study, ethanol extract of A. hookeri showed the outstanding neuroprotective effect in HT22 cells. In addition, we found that ethanol extract of A. hookeri root increased heme oxygenase (HO)-1 in HT22 cells. Moreover, ethanol extract of A. hookeri root also upregulated nuclear accumulation of nuclear factor E2-related factor 2 (Nrf2) in HT22 cells. These results demonstrate that ethanol extract of A. hookeri root contributes neuroprotective effects against glutamate-induced oxidative stress in HT22 cells, via Nrf2-mediated HO-1 expression. Our study suggests that ethanol extract of A. hookeri root could be the potential agent for the treatment of many neuro-degenerative diseases.

Non-alcoholic fatty liver disease (NAFLD) is common in obesity. However, weight reduction alone does not prevent the progression of NAFLD to end-stage disease associated with the development of cirrhosis and liver disease. In a previous experiment, 50% ethanol extract of Acanthopanax senticosus stem bark (ASSB) was found to reduce body weight and insulin resistance in high fat diet-induced hyperglycemic and hyperlipidemic ICR mice. To evaluate the anti-steatosis action of ASSB, insulin-resistant ob/ob mice with fatty livers were treated with ASSB ethanol extract for an 8 week-period. ASSB ethanol extract reversed the hepatomegaly, as evident in reduction of % liver weight/body weight ratio. ASSB ethanol extract also specifically lowered circulating glucose and lipids, and enhanced insulin action in the liver. These changes culminated in inhibition of triglyceride synthesis in non-adipose tissues including liver and skeletal muscle. Gene expression studies confirmed reductions in glucose 6-phosphatase and lipogenic enzymes in the liver. These results demonstrate that ASSB ethanol extract is an effective treatment for insulin resistance and hepatic steatosis in ob/ob mice by decreasing hepatic lipid synthesis.
Objectives : This study evaluates neuroprotective effect of Gastrodiae Rhizoma on apoptosis in the cerebral infarct. Methods : Cerebral infarct was induced by the middle cerebral artery occlusion (MCAO) for 2 hours with intraluminal thread method in Sprague-Dawley rats. Then ethanol extract of Gastrodiae Rhizoma was administered orally for 3 days. Infarct area and volume were evaluated with TTC staining. Neuronal apoptosis was identified with TUNEL labeling. Apoptosis modulatory effect was observed with immunohistochemical Bax, Bcl-2, iNOS, and MMP-9 expressions in penumbra. Results : 1. Ethanol extract of Gastrodiae Rhizoma reduced infarct size partly and volume significantly of in the MCAO rat brain. 2. Ethanol extract of Gastrodiae Rhizoma reduced TUNEL positive cell ratio in the penumbra of MCAO rat brain significantly. 3. Ethanol extract of Gastrodiae Rhizoma suppressed Bax, iNOS and MMP-9 expression in the penumbra of MCAO rat brain significantly. 4. Ethanol extract of Gastrodiae Rhizoma did not change Bcl-2 expression in the penumbra of MCAO rat brain. But expression ratio of Bcl-2 against Bax was increased in the Gastrodiae Rhizoma group. Conclusions : These results suggest that Gastrodiae Rhizoma plays an anti-apoptotic neuroprotective effect through suppression of Bax, iNOS, and MMP-9 expressions and relative up-regulation of Bcl-2 in the ischemic brain tissue.
Background: A red seaweed, Callophyllis japonica has been traditionally eaten in the oriental area. In a recent study, it has been demonstrated that the ethanol extract of C. japonica have antioxidant activity. However, there are few studies about the effects of C. japonica on the function of immune cells. We investigated the immunomodulatory effects of C. japonica on the function of dendritic cells, the potent antigen-presenting cells. Methods: Bone marrow-derived dendritic cells (DCs) were used and the viability was measured by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay and trypan blue exclusion test. Cytokine and nitric oxide (NO) levels were determined by using ELISA and Griess reagent, respectively. The expression levels of DC surface markers were measured by flow cytometric analysis. Results: C. japonica ethanol extract did not significantly affect the DCs viability and the IL-12 production from DCs, irrespective of the presence of lipopolysaccharide (LPS). In addition, it did not significantly change the expression of DC surface markers. However, C. japonica ethanol extract significantly inhibited the LPS-induced NO production and also increased the proliferation of allogeneic lymphocytes activated by DCs. Conclusion: Our data suggests that C. japonica ethanol extract enhances the proliferation of allogeneic lymphocytes activated by DCs which is associated with inhibition of NO production from DCs induced by LPS.
The purpose of this study was to evaluate the color image of the natural-dyed silk fabrics. The dye was extracted from pine needle by boiling pine needle with ethanol at $78^{\circ}C$ for 3hours and distilled water at $100^{\circ}C$ for 2hours. The 100% silk fabric was dyed of extract in pH 5 at $90-100^{\circ}C$ for 1 hr. As mordants used were compounds of Al, Sn, Fe, and Cr, color image of pine-needle dyed silk fabrics was classified into 5 factors (pure, gentle, sophisticate, comfortable, pastorale) and the factor pure is most important one of those. Most cheerful image in pure factor was from the fabrics dyed with ethanol extract and then, none and Cr mordanting. Dignified image was from the fabrics dyed with ethanol extract and then, Cu or Fe mordanting. In production, products dyed with ethanol extracts was preferred to those dyed with distilled water extracts. Color image and preference of the silk fabrics dyed with pine needles extracted was affected by extraction solvents and mordants.
Rumex crispus (Curled Dock, Polygonaceae) is a perennial wild plant used in traditional medicine as a laxative, astringent, and to treat blood and skin disease. The ethanol extract of R. crispus was obtained and its carbohydrate contents were analyzed using high-performance anion-exchange chromatography with pulsed amperometric detection. The anabolic effects of R. crispus in human osteoblastic MG-63 cells were investigated using the WST-8 assay, alkalinephosphatase (ALP) assay, and mineralization assay. The ethanol extract increased the proliferation of MG-63 cells and stimulated ALP activity in a dose-dependent manner over a 72-hrs period. Additionally, the ethanol extract dose-dependently stimulated the formation of bone nodules in MG-63 cells treated for 12 days. The ethyl acetate fraction from the ethanol extract did not affect osteoblast viability but induced an increase in ALP activity. In conclusion, the ethanol extract of R. crispus increases the proliferation and bone-forming activity of osteoblasts, and hence it could be used in the development of bone-forming stimulatory nutraceuticals and osteoporosis-related medicines.
We examined dyeing properties using cotton, Tencel, general nylon 66 and hollow nylon 66 treated with aqueous and ethanol extracts without mordant. The antimicrobial properties of fabrics treated with Artemisia extracts against gram positive Staphylococcus aureus (S. aureus) and gram negative Klebsiella pneumonia (K. pneumonia) were also examined. The dying solution concentrations were determined from a calibration curve of the concentration and absorbance of Artemisia extracts. FTIR spectra confirmed that antimicrobial components and colorants (such as 1,8-cineol, thujone, caffeoylquinic acid and chlorophyll) were more present in ethanol extract than in aqueous extract. Nylons had higher $a^*$ and $b^*$, and lower $L^*$ values than cellulose fabrics dyed with aqueous solutions of Artemisia extracts; however, the dyed nylon fabrics were brown. Fabrics dyed with ethanol-extract added solutions were greener and had higher antimicrobial properties than those dyed with aqueous solutions; however, they faded and lost their antimicrobial properties after laundering. Fabrics regained their antimicrobial properties (especially against S. aureus) by the spraying of Artemisia ethanol extract; therefore, the application of Artemisia ethanol extract onto underwear is expected to relieve atopic dermatitis.
The study was accomplished to know the effects of garlic on body weight, and serum lipid, protein and glucose in male rats. The rats applied in this study were Sprague-Dawley strain of 42. In addition to basal diet, they were fed together raw garlic juice or ethanol garlic extract with 2.5% cholesterol solution solved by corn oil for 8 weeks respectively. These results were as follows. 1. The growth rate and efficiency ratio appeared to be more increased in ethanol garlic extract groups than in raw garlic juice groups. 2. The content of serum total cholesterol appeared to be the most decreased in 0.4ml ethanol garlic extract group. 3. The level of serum HDL-cholesterol had a tendency to be increased in all garlic groups. 4. Albumin/Globulin ratio appeared to be more decreased in raw garlic juice groups than in ethanol gar lie extract groups. 5. The level of serum glucose appeared to be more decreased in ethanol garlic extract groups than in raw garlic juice groups. Therefore we think that garlic is able to have an effect on atherosclerosis and diabetes.
