메밀 Flavonoids의 항돌연변이원성 및 지질대사 조절기능에 관한 연구 -메밀 잎 에탄올 추출물의 항돌연변이원성 연구- (Studies on Antimutagenic and Lipotropic Action of Flavonoids of Buckwheats -Desmutagenic Activity of Buckwheat Leaf Extracts)

  • In spore rec-assay using B. subtillus H17(rec) and M 45(rec) , the ethanol extract of buckwheat leaves showed antimutagenicity in condition of low concentrations, but its did comutagenicity in condition of high concentrations. In Ames test, the ethanol extract of buckwheat leaves reduced the mtabenicity of N-methyl-N' -nitro-N-nitrosoguaidine (MNNG), benzo (a) pyrene(B($\alpha$)P), 2-amino-fluorene(2AF), and 3-amino -1, 4-dime-thyl-5-H-pyrido(4, 3-b) indol(Trp-P-1) in Salmonella typhimurium TA98 and TA100. The ethanol extract was fractionated by hexane, ethylacetate, butanol and water. Among Them hexane fraction showed the highest inhibition rate on the mutagenicity of B($\alpha$)P, and so did chloroform fraction on the mutagenicity of MNNG in S. typhimurium Ta98 and TA100. To elucidate the antimutagenic mechanism of the ethanol extract, it was mixed and co-incubated with various metagens, S9 mix, and the bacteria with different experimental orders and different reaction times. The ethanol extract did not affect reversion rate of pre-mutated. S.typhimurium. However, when the ethanol extract was added to the mutagens before their interaction with S.typhimurium , it reduced the mutation rate to 152$\pm$12-273$\pm$18 colonies/plates in case of MNNG, and 135$\pm$13-195$\pm$10 colonies/ plates in case of B($\alpha$)P), showing strong desmutagenic activity.

Comparison of Immuno-stimulatory Activities by Purification Process of Schizandra chinensis Baillon Fruits

  • Two compounds from Gomisin N and Gomisin A were isolated from the fruits of Schizandra chinensis Baillon. The highest extraction yield as 21.36% was observed in the ethanol extract, compared to the yield obtained form the water extract. The extraction yields of the single compounds were measured to be 0.13 and 0.014 Gomisin A and Gomisin N, respectively. Approximately, 90% of the growth of human stomach adenocarcinoma cancer cells was inhibited after adding 1.0 g/l of the ethanol extract. The growth of the human normal lung cell was limited to 24% after adding the ethanol extract. The water extract lowered the specific secretion of $TNF-\acute{a}$ and IL-6 from T cells, $10.3{\times}10^{-4}\;pg/cell\;and\;12.1{\times}10^{-4}\;pg/cell$, respectively, compared to the ethanol extracts. On the other hand, a treatment with the ethanol extract increased the specific secretion of $TNF-\acute{a}$ and IL-6 from human T cells, to $11{\times}10^{-4}\;pg/cell\;and\;14.3{\times}10^{-4}\;pg/cell$, respectively. The crude ethanol extract had the highest effect on the differentiation of human promyelocytic leukemia cells compared to the other extracts and Gomisin A and N. In general, the biological activities of the extracts gradually decreased as the purification process proceeded, which suggests that higher immunostimulatory activities can be maintained by adding the crude extracts of the fruits rather than by adding a single compound.

어성초의 추출물에 따른 항산화력 연구 (Antioxidant Properties in Water and 70% Ethanol Extracts of Houttuynia Cordata Thunb)

  • 본 연구는 여러 생리활성 성분이 다량 함유되어 있다고 알려진 전통 약용 식물인 어성초의 추출 조건에 따른 항산화력을 관찰하였는데, 추출조건으로는 식용으로 활용가능한 물 추출과 70% 에탄올 추출을 통해 항산화력을 비교하였다. 항산화력은 총 페놀 함량, DPPH 라디칼 소거능, ABS 양이온 소거능, FRAP 방법으로 측정하였다. 결과적으로 총 페놀 함량은 어성초 물추출물이나 70% 에탄올 추출물 모두 77.8 mM Tannic acid/g extract로 나타나 유의적인 차이는 없었고 DPPH 라디칼 소거능은 물 추출물 2 mg/mL 농도에서 78.4%, 70% 에탄올 추출물은 91.8%로 유의적으로 소거능이 70% 에탄올 추출 시에 우수한 것으로 나타났다. 또한 ABTS 양이온 소거능에서도 물 추출물은 2 mg/mL 농도에서 47.7%, 70% 에탄올 추출물에서는 54.8%로 유의적으로 70% 에탄올 추출물이 저해능이 우수한 것으로 나타났으며 FRAP 방법으로 통한 환원 능도 70% 에탄올 추출물이 우수한 것으로 관찰되었다. 이와 같은 결과로 어성초 추출물 조건중 70% 에탄올 추출 시 항산화력을 높힐 수 있는 생리활성 성분이 추출되는 것으로 사료되며 식용 가능한 기능성 식품소재로써 어성초 에탄올 추출물을 사용할 수 있을 것으로 사료된다.

당귀약침액이 대식세포에서 산화질소(NO) 및 프로스타글란딘 (Prostaglandin) 생성에 미치는 영향 (Effects of Angelicae Gigantis Radix pharmacopuncture on Nitric Oxide and Prostaglandin E2 Production in Macrophage)

  • Objectives : Angelicae Gigantis Radix has been known traditional medicine with antimicrobial activities and it has been widely used for treatment of blood and inflammatory diseases. In the present study, some studies examined anti-inflammation effects of Angelicae Gigantis Radix but they usually were performed by ethanol extracted Angelicae Gigantis Radix pharmacopuncture. So We investigated the inhibitory effects of Angelicae Gigantis Radix pharmacopuncture by hot water and ethanol extract on Nitric oxide(NO) and Prostaglandin $E_2$($PGE_2$) production in lipopolysaccharide(LPS) induced macrophage cell. Methods : Angelicae Gigantis Radix was extracted by ethanol and hot water. Cell viability was determined by MTT assay. To evaluate anti-inflammation effects of Angelicae Gigantis Radix pharmacopuncture, we examined NO and $PGE_2$ production in LPS induced macrophages. The concentrations of NO and $PGE_2$ were measured by Griess assay and Enzyme Immuno-Assay. Results : 1) The MTT assay demonstrated that cytotoxic effect of Angelicae Gigantis Radix pharmacopuncture by hot water extract and ethanol extract in RAW 264.7 macrophage cells were not appeared. 2) Angelicae Gigantis Radix pharmacopuncture by ethanol extract and hot water extract inhibited NO production in LPS induced macrophages significantly. 3) Angelicae Gigantis Radix pharmacopuncture by ethanol extract tended to inhibiting $PGE_2$ production in LPS induced macrophages. And Angelicae Gigantis Radix pharmacopuncture by hot water extract inhibited LPS induced production of $PGE_2$ in RAW 264.7 macrophage cells significantly. Conclusions : This study suggests that Angelicae Gigantis Radix pharmacopuncture may have an anti-inflammatory property through the inhibition of NO and $PGE_2$ production in LPS induced macrophages. It may have a therapeutic potential for the treatment of various inflammatory diseases.

맥문동의 물추출액 및 에탄올추출액 이용 음료의 산화방지 활성 (Antioxidative Activity of Beverage with Water and Ethanol Extracts of Maegmundong (Liriope platyphylla))

  • 맥문동의 물추출액 및 70% 에탄올 추출액을 이용한 음료의 산화방지 활성과 유리아미노산, 무기질, 총사포닌, 폴리페놀, 플라보노이드 함량을 분석하였다. 산화방지 활성은 전자공여능과 ABTS 라디탈 소거능으로 측정하였다. 맥문동 에탄올추출액 이용 음료는 물추출액 이용 음료에 비해 총사포닌, 폴리페놀, 플라보노이드 함량이 높았으나 두 음료의 전자공여능과 ABTS 라디칼 소거활성에는 큰 차이가 없었다. 두 음료에는 6종의 필수 아미노산(이소루신, 루신, 리신, 메티오닌, 트레오닌, 발린)을 포함하여 20종의 아미노산 및 그 유도체가 함유되어 있었다. 맥문동 물추출액 이용 음료와 에탄올 추출액 이용 음료의 주 유리 아미노산은 아스파라긴(58.30, 60.68 mg%), 메티오닌(15.10, 13.95 mg%), 프롤린(12.31, 14.00 mg%), 알라닌(11.05, 11.46 mg%)이었으며 가장 많이 존재하는 무기질은 포타슘(238.68, 244.32 mg%)이었다.

우슬의 에탄올 추출물이 모발 성장에 미치는 영향 (Effect of ethanol extract from Achyranthis Radix on hair growth)

  • Objective : As more and more people are interested in appearance in modern society, the increasing number of hair loss population can have an important impact on psychological and social problems such as depression and inappropriate interpersonal symptoms. Therefore, much research is being done on treatments for alopecia using herbal extracts with relatively few side effects. This study was investigated about the effect of Achyranthis Radix (AR) extract with ethanol solvent on hair growth. Methods : We determined the promoting efficacy of AR-ethanol extract compared with minoxidil (MNXD) on the growth of human hair dermal papilla cells (HDPCs). Cell viability was measured by MTT assay and cell proliferation was confirmed by cell cycle analysis from flow cytometry in HDPCs. Also, we monitored the safe concentration range through MTT assay. And protein expression of hair growth-related genes (insulin-like growth factor 1 (IGF-1), Wnt3a, Protein kinase B (Akt), Extracellular signal-regulated kinase (Erk)) was monitored by western blot. Results : On cell cycle analysis, the G2/M phase was higher than that of the DW group in AR ethanol extract group at 0.05 and 0.1 mg/㎖. All protein expression levels of HDPCs were increased in AR ethanol extract groups and the MNXD group, compared to the DW group, respectively. Conclusion : As mentioned above, AR extract increased cell proliferation and the protein expression of IGF-1, Wnt3a, Akt, Erk in HDPCs. These results suggest that AR ethanol extract has promoted hair growth and it might be potential hair growth supplement.

자소 추출물의 기능성 성분과 자소 추출물을 함유하는 PVA 나노 섬유의 제조 (Functional Ingredients of Perilla Frutescens L. Britt Extracts and Preparation of PVA Nanoweb Containing Extracts)

  • The purpose of this study was to analyze the functional ingredients of Perilla Frutescens L. Britt extracts and to confirm the possibility of producing PVA nanofibers using extracts. Distilled water, 3% aqueous sodium hydroxide solution and ethanol were used as extraction solvents. The electrospinning was carried out at a PVA concentration of 12%, an applied voltage of 10 kV and a tip to collector distance of 15cm. The contents of volatile substances, essential oils, total polyphenols and flavonoids of the extracts were measured to examine the constituents of functional materials. Flavor components and esters were identified in 3% sodium hydroxide and ethanol extracts. The content of polyphenols and flavonoids in ethanol extracts was higher than that of medicinal plants. 1wt.% of Tween 20 was added to disperse the essential oil components of the ethanol extract. Addition of a dispersant made it possible to produce a homogeneous mixture by having some compatibility with the ethanol extracts and the PVA molecule. When the concentration of the ethanol extract was 0.25 and 0.5wt%, relatively uniform PVA nanofiber having an average diameter of 350 to 365nm could be produced. The results of FT-IR, XRD and DSC analysis confirmed that Perilla Frutescens L. Britt ethanol extract was well mixed with PVA molecules and was electrospun.

HPLC 및 HPLC/MS를 이용한 유자 씨앗 중 플라보노이드 및 리모노이드 화합물의 분석 (Determination of Flavonoid and Limonoid Compounds in Citron (Citrus junos Sieb. et Tanaka) Seeds by HPLC and HPLC/MS)

  • 유자씨앗 중의 플라보노이드 및 리모노이드 화합물을 methanol 및 ethanol로 추출 HPLC로 정량한 결과 ethanol추출의 경우가 거의 모든 화합물에서 높게 나타났고 화합물의 종류도 더 많이 검출되었다. Limonin이 methanol 추출의 경우 140.34 mg/100g, ethanol 추출의 경우 170.98 mg/100g으로 개별화합물 중 가장 높은 함량을 나타내었고 그다음이 caffeic acid, naringin, lutin, nomilin 등의 순으로 높았다. HPLC/MS에 의하여 약42가지 화합물의 분자량 측정이 가능하였으며 mass spectrum과 extracted current ion chromatogram으로 분자량 측정이 가능하였다.

블루베리 듀크 추출물의 예쁜꼬마선충 내의 항산화 효과 (Anti-oxidative Effect of Blueberry Duke Extract in Caenorhabditis elegans)

  • We investigated the anti-oxidative effect of the blueberry duke (Vaccinium corymbosum L., Ericaceae) ethanol extract in Caenorhabditis elegans model. The ethanol extract of blueberry duke showed relatively significant DPPH radical scavenging and superoxide quenching activities. To prove antioxidant activity of the extract, we checked the activities of superoxide dismutase (SOD), catalase, intracellular ROS, and oxidative stress tolerance in C. elegans. In addition, to verify if the increased stress tolerance of C. elegans by treating with the extract was due to regulation of stress-response genes, we checked SOD-3 expression using a transgenic strain. As a consequence, the blueberry duke ethanol extract increased SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Besides, blueberry duke ethanol extract-treated CF1553 worms showed higher SOD-3::GFP intensity.

백서(白鼠)에 인삼(人蔘) 투여시(投與時) 간(肝)의 에탄올 대사(代謝) 효소(酵素) 활성(活性)에 미치는 효과(效果) (The Effect of Ginseng on the Hepatic Ethanol-Metabolizing Enzyme Activity in Rat Liver)

  • The investigation was aimed to study the effect of ginseng ethanol extract on the hepatic ethanol-metabolizing enzyme activity in vivo. The extract (100mg/kg/day) was administered orally to Sprague-Dawley rats for $7{\sim}10$ days and their microsomal ethanol oxidizing system(MEOS) and catalase activities were measured. The MEOS activity in the rat treated with the extract was not significantly different from that of the normal group. Microsomal fraction containing MEOS was separated and the MEOS activity was measured after preincubation for 5, 60 and 180 min, respectively. There were no significant differences in MEOS activities between the normal and treated groups preincubated for 5, 60 and 180 min. The activity in the rat treated with single i.p. injection of 95% $CCl_4$ (0.5ml/kg) was decreased by 48%, compared to the normal group and in the rat treated with the extract (100mg/kg) for $7{\sim}10$ days, the decrease of the MEOS activity was potentiated. Catalase activity in the rat treated with the extract (100mg/kg) was similar to that obtained from the normal group.
