In this study, extracts from Caryopteris incana Miq. (C. incana) were investigated to assess anti-oxidation, skin-whitening and anti-wrinkle activity. The total phenolic compounds of C. incana extracts with water and 80 % ethanol showed 7.69 and 12.50 mg/g respectively. Antioxidation activity of C. incana extracts was measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS), protection factor (PF), and Thiobarbituric acid reactive substances (TBARs). At concentration of $200{\mu}g/mL$, the DPPH free radical scavenging activity of water and ethanol extracts were 84 and 92 %, respectively. ABTS radical scavenging activity of water and ethanol extracts were both at approximately 99 %. Antioxidant PF of water and ethanol extracts were 1.56 PF and 1.67 PF, respectively. The TBARs of water and ethanol extracts were 62 and 82 %, respectively. In anti-wrinkle and skin-whitening activity, 80 % ethanol extract had more outstanding effect than water extract at concentration of $200{\mu}g/mL$. The levels of elastase and collagenase inhibitory activity related with anti-wrinkle were 58 and 89 % in ethanol extract. The tyrosinase inhibitory activity related with skin-whitening was 13 % in ethanol extract. The astringent effect of ethanol extract was 50 %. Throughout the results, C. incana extracts showed an excellent effect on anti-oxidation, skin-whitening and anti-wrinkle activity. Therefore, C. incana extracts can be used as a new material for cosmetics.
Objectives : This study was conducted to evaluate the antioxidative and fibrinolytic activity of the water and ethanol extracts from medicinal plants. Methods : Five kinds of medicinal plants(Carthami Flos, Glycyrrhizae Radix, Schisandrae Fructus, Atractylodes Rhizoma, Shiitake mushiroom) were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and fibrilytic activities. Results : The highest polyphenol contents of the water and ethanol extracts from medicinal plants were 812.52 mg and 685.44 mg per 100 g of Carthamus tinctorius and Schizandra chinensis, respectively. The electron donating abilities (EDA) of the water extracts from all medicinal plants except Lentinus edodes were about 90% at 1,000 ppm and ethanol extracts were higher than those of water extracts. The highest SOD-like activity and nitrite scavenging abilities (NSA) were both of water and ethanol extracts from Schizandra chinensis. Five kinds of medicinal plants had fibrinolytoc activity and the highest activities were water and ethanol extracts from Glycyrrhiza uralensis. Conclusion : These results suggest that the medicinal plants can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidative and fibrinolytic activities.

Black ginseng was made by steaming raw white ginseng nine times at $100^{\circ}C$ for 2 h and drying. We then performed pilot experiments using the nine black ginseng extracts for different steaming and drying times to determine their anti-obesity effects. Two ginseng extracts, steaming and drying five times (FSFD) and steaming and drying nine times (NSND), prepared in water or ethanol solution decreased lipid accumulation of 3T3-L1 cells. FSFD in water and ethanol extracts showed higher levels of ginsenosides, in particular, Rh1, Rg2, and Rb1 than NSND, and levels of the three ginsenosides were higher in ethanol extracts than in water extracts. Treatment with FSFD and/or NSND in ethanol extracts significantly regulated $PPAR{\gamma}$, C/$EBP{\alpha}$ and AMPK phosphorylation in 3T3-L1 cells. We verified doubling time of stem cells from both abdominal fat and subcutaneous fat after FSFD and NSND in ethanol and water extracts were added. Although addition of FSFD and NSFD in water extracts had no effects on proliferation, ethanol extracts with FSFD and NSND increased doubling time of stem cells in subcutaneous fat. FSFD and NSND in ethanol extracts more effectively reduced adipogenesis compared to those in water extracts. FSFD in ethanol extracts promoted secretion of anti-inflammatory cytokine such as IL-10 and depressed MCP-1 infiltration in 3T3-L1 preadipocytes co-cultured with RAW264.7 cells. We concluded that FSFD and NSND ethanol extracts may be developed as a functional food for its anti-obesity effect, but anti-inflammatory effect was shown in ethanol extracted FSFD rather than in NSND.
This study was conducted to evaluate the antioxidative effect and antimutagenic capacity of ethanol extracts of Flammulina velutipes by employing biological and biochemical assays. The $IC_{50}$ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Flammulina velutipes was found to be 28.39 mg/assay, 9.33 mg/assay and 144.61 mg/assay respectively. Therefore, the most effective antioxidative capacity of ethanol extracts of Flammulina velutipes was $Fe^+$-induced linoleate peroxidation, among the method used this study. The indirect and direct antimutagenic effects of the ethanol extracts of Flammulina velutipes were examined by the Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-anthramine and on direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 and mediated by 2-nitrofluorene in Salmonella typimurium TA98 were 0%, respectively. These findings indicate that ethanol extracts of Flammulina velutipes have no effects on indirect and direct mutagenicity. Based on these results, it believed that the ethanol extracts of Flammulina velutipes has antioxidative capacities, and is a the candidate for the prevention and dietetic treatment of chronic diseases and the development of antioxidative functional food.
The present study was conducted to investigate extraction characteristics and antioxidative activity of Lycium chinense extracts. Lycium chinense were extracted by reflux extraction(RE) under different extraction conditions including solvent. The solid yield, turbidity, color value, titratable acidity, free sugar contents, electron donating ability(EDA) and superoxide dismutase(SOD)-like ability of Lycium chinense extracts were determined. The highest solid yield value were obtained with water of 10 fold. No significant difference in turbidity and color value were found among the extracts prepared with various extraction solvents, 75$\%$ ethanol, 50$\%$ ethanol and water. The highest titratable acidity were obtained with water of Lycium chinense. The free sugar contents of Lycium chinense extracted with water showed the highest value. Lycium chinense extracts with water included higher contents of free sugar compared with those of the other solvent extracts, 50$\%$ ethanol and 75$\%$ ethanol extracts. The total polyphenol compounds content of Lycium chinense extracted with 50% ethanol showed the highest value. Lycium chinense extracts with 50$\%$ ethanol included higher contents of total polyphenol compound compared with those of the other solvent extracts, water and 75$\%$ ethanol extracts.
The present study was conducted to investigate extraction characteristics and antioxidative activity of Cassia tora L. extracts. Cassia tora L. was extracted by reflux extraction under different extraction conditions including solvent. The solid yield, turbidity, color value, titratable acidity, free sugar contents, electron donating ability and superoxide dismutase-like ability of Cassia tora L. extracts were determined. The highest solid yield value was obtained with water of 10 fold. No significant difference in turbidity and color value were found among the extracts prepared with various extraction solvents, 75% ethanol, 50% ethanol and water. The highest titratable acidity was obtained with 50% ethanol of Cassia tora L.. The free sugar contents of Cassia tora L. extracted with water showed the highest value. Cassia tora L. extracts with water included higher contents of free sugar compared with then of the other solvent extracts, 50% ethanol and 75% ethanol extracts. The total polyphenol compound content of Cassia tora L. extracted with 50% ethanol showed the highest value. Cassia tora L. extracts with 50% ethanol included higher contents of total polyphenol compound compared with those of the other solvent extracts, water and 75% ethanol extracts.
The objective of this study was to evaluate the pre-heating treatment effects on the antioxidant properties of ethanolic garlic and onion extracts. Garlic and onion with or without heating ($100^{\circ}C$, 30 min) were extracted with ethanol, and the total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, iron chelating ability, reducing power, and antioxidant activity in a linoleic acid emulsion system were evaluated. Garlic (41%) had a higher drying yield than onion (11%). Regardless of pre-heating, ethanol extracts of onion resulted in an approximately 25-fold higher yield than those of garlic. Thermal treatment before extraction decreased the levels of ethanol-soluble phenolics for both garlic and onion. Regardless of pre-heating, the radical scavenging abilities of ethanol extracts from garlic were greater than the ethanol extracts from onion. The iron chelating abilities of ethanol extracts from fresh and heated garlic were 85 and 81% at 10 mg/mL, respectively, whereas those of onion extracts were 10 and 9% at the same concentration, respectively. However, no differences in reducing power between garlic and onion extracts were observed. Both garlic and onion inhibited the formation of hydroperoxide in linoleic acid emulsion systems when ethanol was used as a solvent. Overall, garlic extracts had greater antioxidant activity than onion extracts, and the antioxidant activity of garlic and onion extracts were not significantly affected by thermal treatment.
When SiHa cells were incubated for varying periods of time with extracts of PFF and PFM, the cytotoxicity of the ethanol extracts of PFF was higher than those of the other extracts. These results indicated that the extracts from fruiting bodies of p. ferulae contain antitumor Substances. When A549, SiHa and HeLa cells were incubated with different concentrations of PFF and PFM extracts, the ethanol extracts of PFF showed strong cytotoxicity against A549 tells at concentrations over $10{\mu}g/mL$ and against SiHa and HeLa cells at concentrations over $40{\mu}g/mL$. However, the differences in the cytotoxic effects of the hot water and ethanol extracts of PFM and the hot water extracts of PFF on all 3 cancer cells were not significant. Also, the PFF ethanol extracts induced synergistic effects on the TRAIL-induced apoptosis in A549 cells, which were strongly resistant to TRAIL. These results indicated that ethanol extracts of PFF were the most prominent antitumor agents toward lung cancer cells (A549).
This study was conducted to investigate the effect of Cassia tora ethanol extracts on carbon tetrachloride($CCl_4$)-induced hepatotoxicity in rats. Male Sprague-Dawley rats were divided into the following 4 groups: normal group, $CCl_4$, treated group, $CCl_4-0.25%$ Cassia tora ethanol extracts group and $CCl_4-0.5%$ Cassia tora ethanol extracts group. Rats were fed with each experimental diet and water for 5 weeks. Liver weights of rats treated only with $CCl_4$, were significantly increased compared to normal group, but not in rats fed diet containing Cassia tora ethanol extracts. Cholesterol and triglyceride contents in serum and liver were also not influenced by either $CCl_4$, treatment or the supplementation of Cassia tora ethanol extracts. $CCl_4$, treatment significantly increased ALP activities, however the supplementation of Cassia tora ethanol extracts significantly decreased the activities of serum ALT, AST, ${\gamma}-GTP$ in dose-dependent manner. Cassia tora ethanol extracts significantly reduced $CCl_4-induced$ elevation of liver TBARS contents. Activities of superoxide dismutase and catalase were decreased by $CCl_4$, treatment, however by the supplement of Cassia tora ethanol extracts slightly increased activities of SOD and catalase. The activity of glutathione peroxidase in groups fed diets containing Cassia tora ethanol extracts was significantly decreased compared to that of the control group. These results suggest that Cassia tora ethanol extracts may exert protective effect against $CCl_4-induced$ liver injury through the prevention of lipid peroxidation.
Red ginseng extracts were prepared with various concentrations of ethanol. Extract yields were examined and saponins in the extracts were identified and determined by TLC and HPLC, respectively. Yields of the extracts, $19.7{\sim]50.3%$, were the highest in water extract and showed significant decrease with the increase of ethanol concentration used for extraction. Contrary to the extract yields, saponin yields from red ginseng were conspicuously increased with the increase of ethanol concentration and were $3.47{\sim}5.13%$ of crude saponins and $1.28{\sim}1.93%$ of six major ginsenosides. Saponin contents in the red ginseng extracts were $6.9{\sim}24.2%$, of crude saponin and $2.57{\sim}9.22%$, of six major ginsenosides.
