다낭성 난소 유발 rat에 단미 한약제가 미치는 영향에 관한 연구분석 (Effects of the Korean Medicinal Herbs for Treatment of Polycystic Ovary induced Rat Models: a Review)

  • Objectives: This study is to review the effect of Korean medicinal herbs on treatment of polycystic ovary induced rat models reported in domestic & foreign journals. Methods: 7 electronic databases (Pubmed, CNKI, EMBASE, Oasis, RISS, CENTRAL, Koreankt) were searched with term as polycystic ovary, and study reports on polycystic ovary impairment with Korean medicinal herbs. After selecting several studies, the analysis was focused on items reflected in the Rotterdam criteria for diagnosis of polycystic ovary syndrome such as hyperandrogenism (HA), ovarian weight, the number of follicle cysts, estrous cycles, and etc. Results: 15 studies were reviewed. As a polycystic ovary induction material, estradiol valerate (EV) was used in 9 studies, testosterone propionate (TP) in 2 studies and dihydrotestosterone (DHT), dehydroepiandrosterone (DHEA), Poretsky's method and letrozole (LE) was used in rest 4 studies, respectively. Tribulus terrestris (TT), Nardostachys jatamansi (NJ), Lycii cortex (LC), Cinnamomum cassia kees (CCK) and Atractylodis macrocephalae koidz (AMK) produced statistically significant results related to hyperandrogenism. In ovarian weight, Silybum marianum (SM), AMK and Alllium sativum (AS) were statistically significant. In the the number of follicle cysts, SM and TT were statistically significant. In estrous cycles, AMK, NJ, TT and Coix lacryma-jobi (CL) were statistically significant. Conclusions: Based on the diagnosis criteria of Rotterdam, TT, NJ, AMK, SM, AS, and CL produced statistically significant results in rat model.

Leptin Is a Metabolic Signal for GnRH-LH/FSH Axis in Feed-restricted Ewes

  • Two experiments were conducted to determine whether leptin is a metabolic signal for gonadotropin secretion in ewes. In the first experiment, twenty-eight cyclic Chal ewes were assigned randomly to an energy restricted, no leptin group (ERNL) (60% of maintenance; n = 14) and an energy normal, no leptin group (ENNL) (100% of maintenance; n = 14) for 71 days (6 estrous cycles). Estrus was synchronized with seven consecutive injections of $PGF_{2{\alpha}}$ Biweekly, body weight (BW) and body condition score (BCS) were determined and blood samples were collected to measure plasma leptin concentration. Blood samples were also taken to determine plasma progesterone concentration twice weekly. After each PG injection from the second injection to the end of experiment, four ewes were selected and blood samples were collected at 20 minutes and at hourly intervals for 3 h to detect plasma LH and FSH concentration. In the second experiment, after the ceasing of the estrous cycle caused by energy restriction, six acyclic ewes were selected and randomly allotted to two groups (n = 3) and received the following treatment for four days. Ewes in an energy restricted, leptin group (ERL) were fed with a ration which provided 60% of maintenance energy requirements and intravenously injected with $4{\mu}g$ leptin/kg BW daily. Ewes in an energy excess, no leptin group (EENL) were fed with a ration that provided 180% (120%+60%) of maintenance energy requirements and intravenously injected with 1 ml saline daily. In both groups, blood samples were collected at 20 minutes and at hourly intervals for 3 h before feeding on d 0 and d 5, and for 3 h before and after injections as above on d 2 and d 4 to detect plasma LH and FSH concentration. In the first experiment, BW and BCS from the $2^{nd}$ estrous cycle, and leptin from the $3^{rd}$ estrous cycle to the end of the experiment significantly (p<0.05) decreased. In ERNL ewes, mean plasma concentrations of FSH significantly (p<0.01) decreased from the $4^{th}$ estrous cycle to d 71 and LH pulsatile secretion was suppressed on d 71, so that, mean plasma concentrations of LH (p<0.05), LH pulse frequency (p<0.01) and LH pulse amplitude (p<0.05) significantly decreased. In the second experiment, injection of leptin significantly increased mean circulating concentrations of LH (p<0.05), LH pulse frequency (p<0.01), LH pulse amplitude (p<0.05) and mean circulating concentrations of FSH (p<0.01) and leptin (p<0.01). High energy intake significantly (p<0.05) stimulated pulsatile secretion of LH and leptin secretion (p<0.01), but non-significantly increased plasma FSH concentration. The results of this study indicate that leptin is a metabolic signal for the GnRH-LH/FSH axis in feed-restricted fat-tailed ewes.

Miniatur Schnauzer견에서 발정 주기 동안 질세포 검사의 유용성 (The Usefulness of Vaginal Cytology during the Estrous Cycle in Miniature Schnauzer Dogs)

  • Vaginal cytology with behavioral observation was performed in 34 estrous cycles in 16 Miniature Schnauzer dogs to evaluate the its usefulness. The mean duration of proestrus and estrus in Miniature Schnauzer based on behavioral observation and vaginal cytology was $7.58{\pm}1.50$ (Mean ${\pm}$S.D., range: $5{\sim}10$) and $8.46{\pm}2.82\;(6{\sim}17)$ days for proestrus, and $6.97{\pm}1.66\;(4{\sim}10)$ and $10.29{\pm}2.61\;(6{\sim}19)$ days for estrus, respectively. The duration of each phase of the estrous cycle was not significantly different based on between behavioral observation and vaginal cytology. The gestational length from the first day of male acceptance was $66.55{\pm}2.91\;(64{\sim}76)$ days, $57.70{\pm}1.92\;(54{\sim}62)$ days from the first male refusal, and $56.90{\pm}1.62\;(54{\sim}60)$ days from the onset of cytologic diestrus, respectively. Vaginal cytology during the estrous cycle were significantly characteristic of large intermediate cells in proestrus, anuclear cells in estrus, small intermediate cells in diestrus, and parabasal cells and small intermediate cells in anestrus (p<0.001), respectively. Cornification index (CI) by vaginal cytology was higher in proestrus ($69.90{\pm}1.44%$) and estrus ($91.35{\pm}1.09%$), then it was decreased in diestrus ($13.53{\pm}1.28%$) and anestrus ($1.16{\pm}1.26%$).

The effect of Equilume light masks on the timing of seasonal ovulation of Thoroughbred mares in South Korea

  • Advancing the estrous cycle of mares is an essential breeding strategy that is routinely conducted by Thoroughbred breeders to improve economic outcomes. For this purpose, Equilume light masks have been developed as an alternative to existing technologies such as artificial lighting or hormonal treatments because they are considered as valid as existing methods with additional animal welfare advantages. For example, with the Equilume light masks, horses can be let out into the pasture, whereas they have to be kept indoors during lighting treatment. Because the function of Equilume light mask on the estrous cycle of mares is influenced by environmental factors such as nutrition condition and temperature, Equilume light mask should be studied in various environments. The objective of the present study was thus to verify the effect of Equilume light masks on the onset of the estrous cycle in Thoroughbred mares in South Korea. Mares were randomly selected and separated into two groups at two Thoroughbred horse breeding farms. The mares in the treatment group were equipped with Equilume light masks from November 18 to February 10 the following year. The body condition, the number and size (> 35 mm) of uterine follicles, and the uterine horn score of the mares were assessed on January 6 and February 10. The body condition scores were not different between the two groups. The treatment with the Equilume light mask had no positive effects on developing follicles and the reproductive tract. In conclusion, the use of Equilume light masks did not influence the seasonal resumption of the estrus cycles in Thoroughbred mares in South Korea.

한국재래빈산양의 번식과정에 따른 혈청내 Progesterone과 Estradiol-$17\beta$수준변화에 관한 연구 (Studies on the Changes of Progesterone and Estradiol-$17\beta$ Levels in Serum of Female Korean Native Goats during the Reproductive Stages)

  • This study was conducted to find out the changes of progesterone and estradiol-17$\beta$ levels in the serum of female Korean native goats during the reproductive stages such as the estrous cycle, pregnancy and periparturient period. Nine heads of female Korean native goats of 3 year old in average and weighing 35.7$\pm$1.4 kg were offered for the experiment. Blood samples were taken at 0, 1, 2, 5, 7, 10, 13, 15, 18 and 19 days after onset of estrus, and 1, 30, 60, 90, 120 and 149 days of pregnancy, and -5, -2, -1, 0, +1, +2 and +5 days of periparturient period where minus figures denote the days before paturition. The progesterone, estradiol-17$\beta$ in the serum samples were assayed by radioimmunoassay methods. The results of this study are summarized as follow: 1. The progesterone levels during the estrous cycles reached a peak level of 0.98$\pm$0.60ng/ml at 13 days after onset of estrus and decreased thereafter and were lower than 0.09$\pm$0.02mg/ml on the first day of estrus. 2. The estradioe17$\beta$ levels during the estrous cycles showed a peak level of 15.97$\pm$1.72pg/ml at onest of estrus, and decreased (5.41$\pm$0.51pg/ml-9.09$\pm$1.82pg/ml) during luteal phase. 3. The progesterone levels during the gestation period increased from day 1 and peaked at 90 days after mating and then decreased until 149 days. The peak level was 6.27$\pm$0.23ng/ml at 90 days. 4. The estradiol-17$\beta$ levels during the gestation period showed gradual increase, which were 9.03$\pm$0.88, 32.96$\pm$2.85, 46.03$\pm$2.42, and 54.06$\pm$1.64pg/ml on 30, 60, 90 and 120 dyas after mating respectively. 5. The progesterone levels measured from 5 days before the parturition to 5 days after showed the highest level at the shart of measurement (4.46$\pm$0.31ng/ml) and decreased gradually and bottomed out at one day post-partum and thereafter (0.24$\pm$0.02-0.45$\pm$0.06ng/ml). 6. The estradiol-17$\beta$ levels measured during the same periparturient period as progesterone showed increase to reach the peak level at 1 day before parturition and decreased rapidly thereafter (-5 dyas 69.46$\pm$3.62, -2 days 107.07$\pm$1.91, -1 days 137.83$\pm$7.54, 0 days 50.06$\pm$6.71 and +1 to +5 days 3.21-4.72 pg/ml).

Efficiency of Equilume light mask on the resumption of early estrous cyclicity and ovulation in Thoroughbred mares

  • Equilume light masks had no impact on hastening the resumption of estrous cyclicity in mares maintained in outdoor pastures on the mainland of Korea due to the cold weather conditions. Jeju Island is a major horse-breeding site in Korea and is warmer than the mainland during the winter season. Therefore, the primary objective of this study was to explore the efficiency of the Equilume light mask on the resumption of seasonal estrous cycles in Thoroughbred mares on Jeju Island. A total of 20 nonpregnant mares were randomly divided into the Equilume light mask (n = 9) and stable lighting (n =11) groups. The experiment was performed at seven different horse-breeding farms located on Jeju Island from November 15, 2020, to February 15, 2021. The mares were exposed to the respective lights from 16:00 to 23:00. Follicle size and uterine edema were measured by ultrasound scanning. Body condition scores (BCS) were also monitored during the experiment. Statistical analysis was conducted using the SAS and SPSS software, and p-values of < 0.05 were considered statistically significant. Two of the nine (22.2%) mares in the Equilume light mask group and three of the 11 (27.28%) mares in the stable lighting group were still cycling in December and January, which were considered as all-year-round cycling mares. On February 15, there was no difference between groups in the resumption of early seasonal estrus cycle, which was determined by follicles > 25 mm in addition to uterine edema. All mares in the Equilume light mask group and five of the eight mares (62.5%) in the stable lighting group had resumed cycling. Interestingly, six of the seven mares (87.5%) in the Equilume light mask and four of eight mares (50%) in the stable lighting group had already ovulated on February 15 (p > 0.05), as determined by the presence of a recent corpus luteum. No difference was observed in BCS and uterine edema between groups (p > 0.05). In conclusion, the Equilume light mask can be an effective approach to induce early seasonal estrus cycles of mares in Jeju Island, and it also enhances the efficiency of farm management by reducing labor.

난소 내 황체조직에서 발정주기별 H-Ras, RLIP76, Angiogenic Receptors mRNA와 Protein의 발현 (Expression of H-ras, RLIP76 mRNA and Protein, and Angiogenic Receptors in Corpus Luteum Tissues during Estrous Cycles)

  • 황체는 발정주기에 따라 형성과 퇴행이 반복되는 일시적인 내분비기관이다. 본 연구에서는 황체와 종양의 혈관신생과정이 기능적과 구조적 기전이 유사하다는 가정하에 실시하였다. 먼저, 우리는 혈관신생관련 수용체인 VEGFR2와 Tie 2 mRNA와 단백질 발현을 검토하였다. 또한, RLIP76와 H-ras의 발현도 측정하였다. 그 결과, 초기와 중기황체에서 VEGFR2와 Tie 2 mRNA와 단백질의 발현되었으나, 후기황체에서는 발현이 감소하였다. H-ras의 경우, mRMA와 단백질 모두 초기와 중기황체에서 발현되었으나, 후기황체에서는 발현되지 않았다. RLIP76 mRNA은 모든 황체주기에서 발현되었고, 단백질은 초기황체에서 강하게 발현되었다. 이상의 결과를 토대로, RLIP76와 H-ras는 황체의 기능에 관여하고, 황체의 혈관신생과정 메커니즘에 중요한 역할을 할 것이다.

  • This experiment was examined the effect of splenectomy on the hematology in pregnant wistar rat. Only animals that had been shown regular 4-day estrous cycles for more than two cycles were used. The day after mating with the same male animal ws designated Day 0 of pregnancy. Spleen was removed from Day 0(early), 6(middle) and 13(late) of pregnant rat, respectively. Blood sample was collected at Day 1, 7, 14 and 21 of the pregnancy. 1. RBC was increased significantly(P<0.05) to the progress of pregnancy in control rat. The late splenectomized rats were decreased significantly(P<0.05) at Day 21 of pregnancy than control rats. 2. Hb was increased significantly (P<0.05) at 21th day of pregnancy in late splenectomized groups than others group. 3. In the late splenectomized rats, Ht was decreased significant (P<0.05) due to the progress of pregnancy and decreased significantly (P<0.05) at Day 21 of pregnancy in all splenectomized groups. 4. WBC was increased significantly (P<0.05) at Day 1 of pregnancy in splenectomized groups compared with control. 5. In differential leukocyte rate, the Basophils and Monocytes was not significantly changed. Neutrophils was increased significantly(P<0.05) at Day 14 and 21 than Day 1 and 7 of pregnancy in control. Lymphocytes was decreased significantly(P<0.05) in control due to progress of pregnancy. Neutrophils was increased and Lymphocytes was decreased significantly(P<0.05) in splenectomized groups compared with control.

비장적출이 임신 rat의 혈액성분 및 번식력에 미치는 영향 (Effect of Splenectomy on the Blood Component and Reproductive Performance accordingto Gestation in Rats)

The total serum protein was increased significantly (P<0.05) to the progress of pregnancy in control rat. All rats that splenectomized groups were decreased significantly(P<0.05) at Day 21 of pregnancy than control rats. 2. Albumin was not significantly changed. 3. Globulin was decreased significantly(P<0.05) between contol and middle treatment at Day 7, 14 of pregnancy. 4. Glucose was increased for the due to the pregress of pregnancy. It was no significance differences among the each groups. 5. Rate of abortion was increased in groups of early and middle of splenectomy compared with control. 6. Period of pregnancy was delayed in middle and late splenectomized gruops companed with control. 7. Litter size was lowed significantly (P<0.05) in early and middle splenectomized groups compared with control.

소동물의 자궁축증에 대한 침술요법의 개발 (Acupuncture Therapy for Pyometra in Small Animal)

  • 개에서 정상 발정주기 때와 자궁 축농증에 이환 되었을 경우 이에 대한 전침 자극의 효과를 알아보기 위하여 본 실험을 실시하였다. 정상 자궁 운동은 발정기 때에 최대이었으며 발정정지기 때에 운동성이 감소되거나 결여되었다. 발정정지기 때에 전침자윽을 하거나 Prostagrandin $F_2{\alpha}$를 투여하였을 경우에는 자궁의 운동성이 크게 증가하였다. 자궁경관이 열린 자궁축농증에 이환된 두 마리에서 전침자극을 3~4일간 실시하였다. 자궁의 직경은 감소되었고 질 분비물은 증가한 후 이후 감소하였다. 환축은 회복되는 경향을 보였으나 축주의 요구에 의하여 난소자궁절제술을 시행하였다. 본 실험의 결과를 통하여 전침자극은 자궁 축농증에 이환된 환축에서 축주가 번식용으로 계속 기르기를 원한다거나 또는 난소자궁절제술을 환축에 적용하는데 있어서 수술적인 위험이 높을 때에 적용 가능한 새로운 치료적 대안이 될 수 있음을 알 수 있었다.
