성스테로이드 호르몬과 고수온 처리가 조피볼락, Sebastes schlegeli의 성분화에 미치는 영향 (Effects of Sex Steroid Hormones and High Temperature on Sex Differentiation in Black Rockfish, Sebastes schlegeli)

  • 조피볼락, S. schlegeli 치어에 $estradiol-l7{\beta}\;(E_2)$$17{\alpha}-methyltestosterone$ (MT) 그리고 고수온을 처리하여 성분화에 미치는 영향을 조사하였다. 실험어는 출산 후 56일부터 72일까지 $E_2\;20,\;40,\;60\;{mu}g/g\;diet$ 농도로 경구투여 하였고, MT는 $20, 50{\mu}g/g diet$ 농도로 경구투여 하였다. 고수온 처리는 대조구 그리고 호르몬 처리구와 비교하여 약 $10^{\circ}C$ 이상인 $27.0{\pm}0.5^{\circ}C$로 사육하였다. 출산 후 56일까지 성분화 과정에서, 생식소는 대부분 생식원세포로 구성되어 있어 성적으로 미분화 단계였다. 그러나 출산후 128일째에, 난소는 난소강파 난소박판으로 구성되어 있고, 난원세포와 주변인기난모세포는 난소박판에 분포하고 있다. 그리고 정소는 많은 정세관으로 이루어 졌고, 이들 세관내에는 정원세포들이 무리를 지어 분포하고 있긴 또한 색소포들이 정소의 수질층과 기부에 드문드문 분포하고 있다. 성비에 있어, 대조구와 비교하였을 때 모든 $E_2$, 처리구는 더 많은 암컷이 관찰되었고, 반면에, 모든 MT 처리구와 고수온 처리구에서는 더 많은 수컷이 관찰되었다. 이러한 결과들에서, 성스테로이드 호르몬의 농도와 종류 그리고 고수온 사육이 조피볼락의 성분화 과정에 있어서 성결정에 영향을 주고 있다고 볼 수 있다.

산수유 에탄올 추출물의 생리활성 (Biological Activities of Ethanol Extract from Corni fructus)

  • 산수유를 기능성식품 소재로서 이용하기 위하여 산수유 에탄올추출물의 생리활성을 조사하였다. 산수유 에탄올추출물의 수소공여능은 농도 의존적으로 증가되었으며, 300 및 $500{\mu}g/mL$ 농도에서 각각 64, 73%의 활성을 나타내었다. 대식세포 RAW264.7에 산수유 에탄올추출물을 100, 300 및 $500{\mu}g/mL$의 농도로 처리하였을 때 산수유추출물은 농도 의존적으로 NO(nitric oxide)의 생성을 유도하였다. 또한 산수유 에탄올추출물을 유방암세포에 다양한 농도로 처리하여 24, 48, 72시간 반응시킨 결과 농도 및 시간에 의존적으로 유방암세포의 증식을 억제하였으며, $500{\mu}g/mL$ 농도로 72 시간 처리 시 60% 이상의 높은 증식억제율을 보여주었다. 또한, MCF-7 유방암 세포에 bisphenol과 $17{\beta}$-estradiol과 같은 환경호르몬을 $0.1{\mu}M$의 농도로 처리하였을 때 세포의 증식이 유도되었으며, 이 세포에 산수유 에탄올추출물을 농도별로 72시간 처리하였을 때 유방암 세포의 증식이 억제되었다. 따라서 본 연구결과는 산수유의 기능성식품 소재로서의 활용 가능성을 시사한다.

소 수정란의 간이 동결기법 개발에 관한 연구 I. 내동제의 종류, 농도 및 동결방법이 체외발생율에 미치는 영향 (Studies on the Development of Easy Cryopreservation Technique of Bovine Embryos I. Effects of Kinds, Concentration and Freezing Method of Cryoprotectants on In Vitro Developmental Rates of Embryos)

  • The studies on the carried out to investigate the effective concentration of cryoprotectant agents and sucrose by one-step straw method of bovine embryos. The follicular oocytes were cultured in TCM-199 medium containing 10 IU/ml PMSG(Sigma, USA), 10 IU/ml hCG(Sigma, USA), 1$\mu\textrm{g}$/ml $\beta$-estradiol(Sigma, USA) and 10% FCS for 24~48 hrs in incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 12~18 hrs with motile capacitated sperm by preincubation of heparin. The bovine embryos following dehydration by cryoprotective agents and various concentrations of sucrose were directly plunged into liquid nitrogen and thawed in 3$0^{\circ}C$ water. Survival and in vitro developmental rate was defined as devellpmental rate on in vitro culture or FDA-test. The results are smmarized as followes : 1. The high in vitro developmental rates of bovine frozen embryos after rapidly thawed in freezing medium was attained 2.0M glycerol, 2.0M DMSO, 1M or 2.0M propanediol. 2. The high in vitro developmental rates of bovine frozen embryos after rapidly thawed in freezing medium was obtained single cryoprotectant(6.7~17.4%) than mixed cryoprotectants(6.7~16.7%). 3. In vitro developmental rate of bovine embryos after rapid frozen-thawing in the freezing medium added 0.25M and 0.50M sucrose were higher cleavage rate than those of sucrose concentration of 0.75M and 1.00M. 4. The freezing methods on in vitro developemental rates of bovine embryos was attained slow freezing method(9.70~15.6%) higher than rapid freezing method(9.4~13.3%).

Toxicity of Methylcyclohexane and Its Effect on the Reproductive System in SD Rats

  • Objectives: There is limited data regarding the toxicity of methylcyclohexane, despite its wide use in rubber adhesives, paint diluents, and cleansing agents. This study aimed to verify the toxicity and influence on the reproductive system of methylcyclohexane after its repeated injection in Sprague Dawley (SD) rats. Methods: Methylcyclohexane was injected subcutaneously into male and female SD rats once a day, five times a week, for 13 weeks at different doses (0, 10, 100, and 1,000 mg/kg/day) for each group. The toxicity of testing material was verified by observing the change in body and organ weight, hematological change, pathological findings, and effect on the reproductive system at each different concentration. Results: In the 1,000 mg/kg/day group, there were cases of animal deaths. In animals that survived, hematological changes, including a decrease in the red blood cell count, were observed. A considerable weight gain or loss and pathological abnormalities in the liver, kidney, and other organs were found. However, the 10 and 100 mg/kg/day groups did not cause deaths or other specific abnormalities. In terms of reproductive toxicity, there were changes in hormone levels, including a significant decrease in hormones such as estradiol and progesterone (p < 0.001) in male animals. Menstrual cycle change for female animals did not show concentration dependency. Conclusion: When injected repeatedly for 13 weeks, methylcyclohexane proved to be toxic for the liver, heart, and kidney at a high dose. The absolute toxic dose was 1,000 mg/kg/day, while the no observed adverse effect level was less than 100 mg/kg/day. The substance exerted little influence on the reproductive system.

Endocrine Profiles of Oestrous Cycle in Buffalo: A Meta-analysis

  • A meta-analysis was conducted to summarize the results of studies which have described the profiles of hormones during the oestrous cycle in buffalo using a fixed effect model and a random effect model. Plasma progesterone concentrations were lowest (0.30${\pm}$0.06 ng/ml) during the peri-oestrous phase and increased (p = 0.067) through the early luteal phase to a maximum concentration (1.94${\pm}$0.03 ng/ml) during the mid-luteal phase. Circulating plasma inhibin and estradiol concentrations were lowest (0.31${\pm}$0.01 and 11.04${\pm}$0.13 ng/ml) during the mid-luteal phase, increased through the late luteal phase to maximum concentrations (0.44${\pm}$0.02 and 22.48${\pm}$0.32 ng/ml) during the peri-oestrous phase. Plasma FSH concentrations were lowest during the early luteal phase and increased through the mid-luteal phase to a maximum concentration during the peri-oestrous phase. Peripheral prolactin concentrations were lowest during the late luteal phase and increased to a maximum concentration during the peri-oestrous phase which then declined (p = 0.716) during the early luteal phase. Peripheral plasma cortisol concentrations decreased from 2.68${\pm}$0.14 ng/ml during the early luteal phase to 1.43${\pm}$0.27 ng/ml during the mid-luteal phase (p<0.001) which then increased to 2.06${\pm}$0.17 ng/ml during the late luteal phase. Plasma $T_{5}$ concentrations decreased from the late luteal phase to the peri-oestrous phase (p<0.001) which then increased during the early luteal phase. $T_{4}$ concentrations increased from the late luteal phase to the peri-oestrous phase which then decreased during the early luteal phase.

Inhibition of $IL-1{\beta}$ and IL-6 in Osteoblast-Like Cell by Isoflavones Extracted from Sophorae fructus

  • Osteoporosis is recognized as one of the major hormonal deficiency diseases, especially in menopausal women and the elderly. When estrogen is reduced in the body, local factors such as IL-1 $\beta$ and IL-6, which are known to be related with bone resorption, are increased and promote osteoclastogenesis, which is responsible for bone resorption. In the present study, we investigated whether glucosidic isoflavones (Isocal, PIII) extracted from Sophorae fructus affect the proliferation of osteoblasts and prevent osteoclastogenesis in vitro by attenuating upstream cytokines such as IL-1$\beta$ and IL-6 in a human osteoblastic cell line (MG-63) and in a primary osteoblastic culture from SD rat femurs. Interestingly, IL-1$\beta$ and IL-6 mRNA were significantly suppressed in osteoblast-like cells treated with 17$\beta$-estradiol (E2) and PIII when compared to positive control (SDB), and this suppression was more effective at $10^{-8}$% than at the highest concentration of $10^{-4}$%. In addition, these were confirmed in protein levels using ELISA assay. In the cell line, the cells showed that E2 was the most effective in osteoblastic proliferation over the whole range of concentration ($10^{-4}%-10^{-12}$%), even though PIII also showed the second greatest effectiveness at $10^{-8}$%. Nitric oxide (NO) was significantly (p<0.05) upregulated in PIII and E2 over the concentration range $10^{-6}% to 10^{-8}$% when compared to SDB, without showing any dose dependency. In bone marrow primary culture, we found by TRAP assay that PIII effectively suppressed osteoclastogenesis next to E2 in comparison with SDB and culture media (control). In conclusion, these results suggest that local bone-resorbing cytokines can be regulated by PIII at lower concentrations and that, therefore, PIII may preferentially induce anti-osteoporosis response by attenuating osteoclastic differentiation and by upregulating NO.

Characteristics of New Estrogen Biosensor Employing Taste Principles

  • Measurement of estrogen concentration in bio-samples are very important for differential diagnosis of various disease or evaluation of health status. However, it is difficult to collect immediate data of estrogen concentration because they are measured by radioimmunoassay or chromatography which need time- and cost-consuming sample pre-treatment. This study was performed for development of new estrogen biosensor employing taste principles, and for evaluation of cross reactivity between various steroid hormones. Gene sequence of ligand binding domain of ${\alpha}$-human estrogen receptor (amino acid 302-553; hER-LBD) was cloned from human breast cancer cell line. The proteins of hER-LBD were produced by T7-E.coli expression system, and isolated by chromatography. hER-LBD were coated on the gold plated quartz crystal (AT-cut 9MHz), and resonance frequencies were measured by universal frequency counter. Estradiol, progesterone, testosterone, and aldosterone were used for cross reactivity of the hER-LBD. We also monitored influences of pH change in resonance frequency. The resonance frequencies of hER-LBD coated quartz crystal were decreased during increase of estrogen concentration from $15 \;{\mu}g/mL$ to $50\;{\mu}g/mL$. However, similar steroid hormones, progesterone and aldosterone, did not elicit the change in resonance frequency. Testosterone evoke weak change in resonance frequency. The new estrogen biosensor was more sensitive in pH 7.2 than in pH 7.6. These results suggest that hER-LBD coated quartz crystal biosensor is a probable estrogen biosensor.

돼지에 있어서 분만전후(分娩前後)의 혈청(血淸) 성(性) Hormone 수준(水準)에 관(關)한 연구(硏究) (Studies on the Sex Hormone Level is Serum of Puerperal Sow)

  • 돼지에 있어서 분만전후(分娩前後)의 혈청중(血淸中) peptide 및 steroid hormone 수준(水準)의 변화(變化)를 구명(究明)하기 위(爲)하여 8두(頭)의 임신돈(妊娠豚)을 대상(對象)으로 분만전(分娩前) 20일(日)부터 분만후(分娩後) 20일(日)까지 혈청중(血淸中)의 FSH, LH, prolactin, estradiol-$17{\beta}$, progesterone 및 cortisol의 농도변화(濃度變化)를 radioimmunoassay 방법(方法)에 의(依)하여 측정(測定)하였다. 혈청(血淸) FSH의 농도(濃度)는 분만전후(分娩前後)에 큰 변화(變化)가 없었으며, $8.1{\pm}1.8mIU/ml$에서 $9.0{\pm}2.6mIU/ml$의 사이에 있었다. LH농도(濃度)는 분만(分娩) 20일전(日前)에 $2.6{\pm}0.3mIU/ml$이던것이 분만일(分娩日)에는 $3.9{\pm}1.1mIU/ml$로 증가(增加)하였으며, 분만후(分娩後) 2일(日)에는 $3.2{\pm}0.9mIU/ml$로 감소(減少)되었고, 그 후(後)로는 거의 비슷한 수준(水準)을 유지(維持)하였다. Prolactin의 농도(濃度)는 분만일(分娩日)에 $68.5{\pm}9.5ng/ml$로 최고수준(最高水準)을 나타냈으며, 분만당일(分娩當日) 전후(前後)에 비교적(比較的) 높은 수준(水準)을 나타냈다. Estradiol-$17{\beta}$는 분만전(分娩前) 6일(日)에 $205.0{\pm}29.5pg/ml$로 증가(增加)하였고, 분만일(分娩日)에는 $425.0{\pm}35.6pg/ml$로 급증(急增)하였으나 분만후(分娩後) 2일(日)에는 $59.5{\pm}7.8pg/ml$로 다시 감소(感少)했다. Progesterone 농도(濃度)는 분만전(分娩前) 20일(日)부터 6일(日)까지는 $18.4{\pm}1.6{\sim}20.2{\pm}2.1ng/ml$사이에 있었으나, 분만전(分娩前) 2일(日)부터 감소(減少)하기 시작(始作)하여 분만일(分娩日)에는 $2.6{\pm}0.8ng/ml$까지 감소(減少)하였고, 분만후(分娩後) 2일(日)에는 $0.9{\pm}0.3ng/ml$에 도달(到達) 하였으며, 그 후(後)로는 거의 같은 수준(水準)을 유지(維持)하였다. Cortisol은 분만일(分娩日)에 $86.5{\pm}10.5ng/ml$로 최고수준(最高水準)을 나타냈다.

산란계의 육성기 제한급여가 체성장 특성 및 내분비 변화에 미치는 영향 미치는 영향 (Effects of Restricted Feeding during Growing Period on Growth and Endocrine Profile in Layers)

  • 본 연구는 산란계의 육성기 제한급여가 체성장 특성 및 내분비 변화에 미치는 영향을 구명하고자 1일령 갈색산란계 Isa-brown 1,080수를 공시하여 시험을 실시하였다. 육성기 제한급여 처리내용으로는 자유채식, 6~18주령까지 제한급여, 12~18주령까지 제한급여를 각각 실시하였으며 모든 사료영양소 함량 및 단계별 사료급여는 NRC(1994)에 준하여 실시하였다. 육성기간동안 체중과 사료섭취량은 6~18주령 제한급여 처리구가 가장 낮았으나(P<0.05) 20주령에서 각 처리구간 유의적 차이는 나타나지 않았다. 소화기관 및 실질장기 발달은 처리구간에 뚜렷한 차이는 없었으나 육성기간동안 체중이 낮았던 제한급여 처리구에서 다소 높은 비율을 보였다.체조성은 지방 함량에서 처리구간 유의성이 나타났는데 자유급여 처리구에 비해 6~18주령 제한급여 처리구에서 육성기간동안 유의적으로 낮게 나타났다(P<0.05). IGF-Ⅰ는 육성기간동안 높은 수준을 보였으나 번식기관이 발달하는 시기부터 급격히 하락하여 낮은 수준으로 유지되면서, 사료섭취량에 민감한 반응을 보였다. estradiol은 6주령 이후 전체적으로 증가하는 양상을 보였는데 16~17주령 사이에 증가하는 양상을 보였으며 종료 시까지 높은 수준을 유지하였는데 처리구간에 유의성은 나타나지 않았다.이상의 결과에서 6~18주 제한급여 처리구가 육성기간동안 체중, 증체량 및 사료섭취량은 자유급여 처리구에 비해 낮았으며(P<0.05), 지방함량 또한 적게 나타났다(P<0.05). 따라서 육성기 제한급여를 육성초기에 실시할 경우 체내 대사에 있어 큰 영향을 미치지 않았으며 estradiol 수준 역시 안정적으로 나타났다. 따라서, 본 연구 결과는 육성기 제한급여가 산란계 성장에 있어서 내분비와 체성장 조성에 영향하지 않으며 또한, 최근 곡물가격의 상승으로 인한 사료가격의 상승에 대한 문제를 해결 할 수 있는 해결방안으로 충분한 가치가 있을 것으로 사료된다. 하지만, 좀 더 개선되어져야 할 부분이 많은 만큼 향후 지속적으로 제한 급여 수준 및 기간 등에 관한 세밀한 연구가 이루어져야 할 것으로 사료된다.(색인어:산란계, 제한급여, Insulin-like growth factor-Ⅰ, 체성분)

E-screen Assay 및 상경적 결합반응을 이용한 Phthalate Esters의 내분비계 장애 작용 연구 (Study on Estrogenic Activities of Phthalate Esters Using E-screen Assay and Competitive Binding Assay)

  • Phthalate esters are used extensively as a plasticizer in the manufacture of plastic products such as PVC bags and medical devices. Recently, phthalate esters have been shown to induce endocrine system mediated responses. However. only a Jew studies have been conducted for estrogenic activity of phthalate esters. In this study estrogenic activities of seven phthalate esters. butyl benzyl phthalate (BBP), di(2-ethylhexyl) phthalate (DEHP), di-n-butylphthalate (DBP), diethylphthalate (DEP), di-n-pentylphthalate (DPP), di-n-propylphthalate (DPrP) and dicyclohexylphthalate (DCHP), were examined in vitro using E-screen assay and competitive binding assay. From the E-screen assay, BBP. DEHP. DBP and DEP showed weak estrogenic activity at the concentration of 5 $\mu\textrm{M}$. The relative proliferative effect (RPE) and the relative proliferative potency (RPP) were 50~70% and 0.01%. respectively, when compared with 500 pM of 17$\beta$-estradiol (E2). In competitive binding assay with the rat uterine estrogen receptor (ER), BBP and DEP showed weak binding potency [(l/$10^4$~1/$10^5$ of E2] while DEHP and DBP scarcely bound to ER. These results suggest that some phthalate esters have weak estrogenic activities in vitro.
