Essentiality of Histidine in Ruminant and Other Animals Including Human Beings

  • Concept and establishment of essential amino acids in animals and human beings rendered immeasurable contributions to animal production and human health. In ruminant animals, however, essential amino acids have never been completely established. The present review proposes a hypothesis that histidine may not be an essential amino acid for normal growing cattle (Japanese black) at least at the growing stage after about 450 kg of body weight on the basis of the experimental results of histidinol dehydrogenase activities in some tissues of the cattle together with hints from which the hypothesis was derived. At the same time, histidinol dehydrogenase activities in liver, kidney and muscle of swine, mouse, fowl and wild duck will be shown and the essentiality of histidine in these animals will be discussed. Finally, the essentiality of histidine for adult human will briefly be discussed.

INFLUENCE OF AMINO ACID SUPPLEMENTS TO A STRAW-MAIZE-BASED UREA DIET ON DUODENAL DIGESTA FLOW AND DIGESTION IN SHEEP

  • Amino acid (AA) substituted diets had no influence on rumen levels of total volatile fatty acids (VFA), ammonia and ${\alpha}$-amino-N, but tended to increase molar proportions of isovalerate and counts of total viable AA utilizing and celluloytic bacteria in the rumen as compared with the control urea diet. The AA diets did not affect daily flow to the duodenum of dry matter (DM), organic mater (OM) and acid detergent fibre (ADF), and rumen digestibility of these nutrients. However, the AA diets, in particular the 10 essential AA (EAA) diet improved total digestibility of DM, OM and ADF by decreasing faecal output of these fractions. Although N flow to the duodenum and N retention were not affected with the dietary treatments, duodenal bacterial flow appeared to increase by the AA diets when it was estimated by means of 2,6-diaminopimelic acid (DAP) and nucleic acid-purine bases (PB) as markers. The results suggest that AA supplements to a urea diet could improve feed utilization by stimulating microbial activity and proliferation in the rumen but and increased microbial activity per se is not necessarily associated with improvement of feed conversion.

Functional Characteristics of Whey Protein-Derived Peptides Produced Using Lactic Acid Bacteria Hydrolysis

  • Hydrolysis of whey-derived proteins using lactic acid bacteria (LAB) utilizes the mass culture method and fermentation of LAB to produce effective bioactive peptides. Whey protein has the biological potential of its precursors, but the active fragments may not be released depending on the hydrolysis method. As an alternative to these problems, the nutritional and bioactive functionality of the hydrolysis method have been reported to be improved using LAB for whey protein. Peptide fractions were obtained using a sample fast protein liquid chromatography device. Antioxidant activity was verified for each of the five fractions obtained. In vitro cell experiments showed no cytotoxicity and inhibited nitric oxide production. Cytokine (IL [interleukin]-1α, IL-6, tumor necrosis factor-α) production was significantly lower than that of lipopolysaccharides (+). As a result of checking the amino acid content ratio of the fractions selected through the AccQ-Tag system, 17 types of amino acids were identified, and the content of isoleucine, an essential amino acid, was the highest. These properties show their applicability for the production of functional products utilizing dietary supplements and milk. It can be presented as an efficient method in terms of product functionality in the production of uniform-quality whey-derived peptides.

  • We quantitated the major chemical components of ramie leaves (Boehmeria nivea L.) powder. The proximate compositions (all w/w) was 5.42% moisture, 28.15% crude protein, 6.95% crude fat, 15.27% crude ash, and 54.79% carbohydrate, respectively. The total, insoluble, and soluble dietary fiber contents were $39.66{\pm}1.84g/100g$, $20.32{\pm}2.02g/100g$, and $19.34{\pm}2.84g/100g$, respectively. The major free sugars were glucose, galactose and lactose. Seventeen amino acids were isolated. Essential amino acids constituted 44.65% of the total. When free amino acid levels were evaluated, 25 kinds of components were detected, of which 18.15% were essential. Only caproic acid methyl ester and pentadecanoic acid were detected when fatty acid examined.   • Rapeseeds (Brassica napus L.)were germinated at $25^{\circ}C$ for 60 hours under dark condition in order to investigate the compositional changes in amino acids and fatty acids. The weight loss was most significant after 30 hours of germination and root growth was observed from that time. Analysis showed that Glu, Lys and Asp were the major amino acids and Ile was the limiting one. Germination caused an increase in Lys, Thr, Val and Leu while Met and Phe decreased among essential amino acids, resulting Met to be the limiting amino acid. The major fatty acids were oleic (49.3%), linoleic (22.0%) and eicosenoic (10.5%) acids. The decrease in oleic and stearic acids was measured while linoleic, linolenic, eicosenoic and erucic acids showed initial decrease followed by gradual increase.

Molecular Studies on the Disease Resistance Gene, Proopiomelanocortin (POMC), from Flounder (Paralichthys olivaceus)

  • Proopiomelanocortin (POMC) plays an essential role in the disease resistance system and is the precursor protein of biologically active peptides such as adrenocorticotropin (ACTH), $\alpha-melanocyte-stimulating$ hormone $(\alpha- MSH)$, $(\beta-melanocyte-stimulation hormone\;(\beta- MSH)$ and $\beta-endorphin$. We have isolated and sequenced two different forms of POMC cDNA, POMC-I and POMC-II, from a pituitary cDNA library of flounder. POMC-I cDNA consisted of 956 bp corresponding to deduced amino acids of 216 residues and POMC-II cDNA was 982 bp in length corresponding to 194 amino acids, respectively. The results of deduced amino acids analysis of the clones showed high sequence homology with previously reported POMCs amino acid sequences from various species. The homology between flounder POMC-I and -II is$57\%$ identity. We also constructed a phylogenetic tree based on POMC amino acid sequences.

Investigations on Nutrient Intakes Among Korean Female College Students -Quality Evaluations for Fat and Protein Consumption- (우리나라 일부 여대생의 영양섭취실태에 관한 연구 -지방 및 단백질섭취의 질적 평가를 중심으로-)

  • This study was performed to investigate the adequacy of dietary fat and protein intakes among female college students. Daily intakes of energy, fat, protein, major amino acids and other nutrients were measured in 52 female college students. Daily energy intake was 75.8% of the recommended intake. Fat and protein consist 19.2% and 16.7% of the total calorie, respectively. The average protein consumption per day was 105% of the recommended intake. Essential amino acids intakes were more than the recommended amounts which appears in the 6th edition of Recommended Dietary Allowances for Koreans. However, when the intake of each essential amino acid was compared to the recommended amino acid requirement pattern, these subjects did not meet the estimated requirements. There was a highly significant correlation between daily protein intake and lipid intake implying the major sources of protein in the diet were also major sources of fat. Daily intakes of dietary fiber, vitamin C, iron, and phosphorous were above the recommended levels of intake. However, blood hemoglobin concentration was marginal indicating dietary iron consumption is not a good marker for iron status. Also, calcium intake was only 63.5% of the recommended intake. Therefore, these results imply that main problems for these subjects are low energy consumption, low calcium intake, and the quality of protein. However, as opposed to the hypothesis, the main energy sources were not the food items high in saturated fats such as instant foods, which should be emphasized further.

Stability of Antioxidant Properties and Essential Amino Acids in Squid Viscera Hydrolysate Produced using Subcritical Water

  • Subcritical water hydrolysis was carried out in a batch reactor to produce valuable materials, such as low-molecular weight (MW) peptides and essential amino acids with antioxidant properties, from heat-dried squid viscera. Hydrolysis of squid viscera was performed at 160 to $280^{\circ}C$ for 3 min. The yield was increased by increasing the temperature and pressure, while the protein content of squid viscera hydrolysate decreased with increasing temperature. Low-MW peptides were detected in all hydrolysates by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The highest yields of free and structural amino acids in heat-dried squid viscera hydrolysate were at $160^{\circ}C$ and were $411.95{\pm}1.15$ and $346.62{\pm}1.25$ mg/100 g, respectively. All essential amino acids were detected in viscera hydrolysates; leucine was the most abundant. Antioxidant activities of hydrolysates were highest at $220^{\circ}C$. Greater than $98{\pm}0.26%$ of the ABTS antioxidant activity was retained in hydrolysates after long-term heat treatment.

Comparisons of Physicochemical Composition of Korean and Chinese Crataegi Fructrus (한국산 산사와 중국산 산사의 이화학적 성분 비교)

  • This study was conducted to compare the major chemical components of Korea Crataegi fructrus (KCF) and Chinese Crataegi fructrus (CCF). Among the proximate compositions, the curde fat content of KCF was lower than that of CCF, whereas their crude protein, crude ash and carbohydrate contents were similar. CCF had a higher total free sugar content than KCF. The major free sugars of KCF and CCF were identified as fructose and glucose. The value of glutamic acid was greater in the amino acids of KCF and CCF, and KCF had higher total amino acids and essential amino acids contents than CCF. KCF also had a higher level unsaturated fatty acids than CCF. CCF had a higher organic acid content, but both KCF and CCF had high citric acid levels. and Chinese The vitamin C contents of KCF and CCF were 272.69 mg per 100 g and 262.38 mg per100 g, respectively. The mineral content of KCF was higher than that of CCF, in the following order : K > Ca > Mg > Fe. The results showed that KCF had higher total amino acid, essential amino acid, unsaturated fatty acid and mineral contents and CCF had higher free sugar and organic acid contents.

Effect of Amino Acids Supplemented to Culture Medium on Development of Porcine Embryos Culturde in Vitro (아미노산의 첨가가 돼지 체외수정란의 후기배의 발달에 미치는 영향)

  • The objective of this study was to investigative the effects of amino acids supplementation on maturation, fertilization and embryo development of pig oocytes. Essential amino acids (EA), non-essential amino acids (NA) or both amino acids (EA + NA) were supple-mented to North Carolina State University (NCSU) 23 medium containing porcine follicular fluid (pFF). When the amino acids were supplemented to the maturation medium, the maturation rates were higher (p<0.05) in the NA group than control ($83.3{\pm}0.04\%\;versus\;70.0{\pm}0.05\%$, but the subsequent cleavage rates and development to morula and blstocyst stage between aminoacid supplement groups and control were not different. The developmental rates to morula and blastocysts stage were not significantly different regardless of amino acid supplementation to culture medium. In addition, supplementation of amino acids did not significantly affect the rate of fertilization and polyspermy. When the amino acids were supplement to culture medium, the number of trophectodermal (TE) cells was significantly (p<0.05) higher in amino acid supplement group than that of control ($18.6{\pm}0.5\;versus\;16.1{\pm}0.6$), whereas the numbers of inner cell mass (ICM) cells were not different among the treaonent groups and control ($29.0{\pm}0.9\~31.5{\pm}1.2$). Total cell number was also significantly (p<0.05) higher in EANA group ($50.0{\pm}1.0$) than that of control group ($44.2{\pm}1.1$). These results indicate that the amino acid supplementation to maturation and culture medium may not significantly stimulate early embryo development, but may improve the TE cell number of blastocyst stage in the pig.
