Total saponin from Korean red ginseng changed thermodynamic parameters of membranes from dipalmitoylphosphatidylcholine (DPPC) and ghost erythrocytes of human. In liposomes from DPPC, temperature of the main transition (Lb'-La) in liquid-crystalline phase increases by 0.2$^{\circ}C$ in average, but enthalpy does not change. Total saponin at a concentration of smaller than $10^5$% "stabilizes" the timid bilayers. At larger than 0.07 of saponin/DPPC ratio, saponin leads to an exclusion of the bound lipid molecules from the main phase transition into lamella liquid crystalline La-phase. Total saponin influences specifically all erythrocyte membrane transitions in a concentration-dependent manner, i.e. on the structures of all the main membrane skeleton proteins. A high structural specificity of saponin with membrane proteins, could be a base of specificity of physiological response of not only erythrocytes, but also other cells.her cells.
The superoxide dismutase (SOD)-mimetic activities of copper complexes of a series of salicylic acid (SA) analogs were tested and compared to the activity of bovine erythrocyte SOD using ferricytochrome c reduction assay. Stability constants of copper complexes were measured potentiometrically using SCOGS2 program. In the presence of 10 g/l albumin, all the copper complexes lost their SOD mimetic activities. Multiple regression analysis was employed for the statistical comparisons between the SOD mimetic activity and their physicochemical properties. Correlation exists for the SOD mimetic activity and steric parameter $(E_s)$ and/or electronic parameter $({\Sigma}{\sigma})$ in xanthine/xanthine oxidase (XOD) system, demonstrating that E, plays a key role in SOD activity whereas ${\Sigma}{\sigma}$ influences it to a lesser extent. The protective effect of copper complexes against membrane damage was measured by counting D-glucose released frm $EG_s$. D-glucose and XOD were entrapped within $EG_s$ and acetaldehyde was used as a substrate for XOD. In this membrane model system using $EG_s$, hydrophobic parameter $({\Sigma}{\pi})$ is of most importance, producing parabolic equation while $E_s$, and ${\Sigma}{\sigma}$ appear to playa minor role in protection against D-glucose release. In summary, to design an efficient SOD mimetic, stability, steric factor, lipophilicity and redox potential should be considered.
We present a novel cell deformability monitoring chip based on the digitally measured cell lysis rate which is dependent on the areal strain of the cell membrane. This method offers simple cell deformability monitoring by automated high-throughput testing system. We suggest the filter design considering the areal strain imposed on the cell membrane passing through the filter array having gradually increased orifice length. In the experiment using erythrocytes, we characterized the cell deformability in terms of average fracture areal strain which was $0.24{\pm}0.014\;and\;0.21{\pm}0.002$ for normal and chemically treated erythrocytes, respectively. We also verified that the areal strain of 0.15 effectively discriminates the deformability difference of normal and chemically treated erythrocytes, which can be applied to the clinical situation. We compared the lysis rates and their difference for the samples from different donors and found that the present chips can be commonly used without any calibration process. The experimental results demonstrate the simple structure and high performance of the present cell deformability monitoring chips, applicable to simple and cost-effective cell aging process monitoring.
In order to evaluate the anticancer activity of 1,4-naphthoquinone derivatives, several 1,4-naphthoquinone derivatives were newly synthesized and subjected to mouse leukemia p-388 cell line by MTT cytotoxicity assay. Among the several 1,4-naphtho-quinone derivatives, YC-001 has showed the most potent anticancer activity. To determine the safety of YC-001, hematotoxicity was tested. YC-001 induced hemolysis increased with both concentration and time dependent manner. The mechanism of hemolysis considered to be the generation of oxygen free radicals and lipid peroxydation of erythrocyte membrane which composed of phospholipids. Also methemoglobin, oxidized form of hemoglobin, was formed by YC-001.
This study was designed to elucidate the mechanism of nephrotoxicity caused by antitumor agent tetraphosphine platinum (II) complex (RC-1), which was synthesized as a tetraphosphine Pt (II) derivatives recently. Rats treated with RC-1 (20mg/kg/day) showed the increase of BUN value and malondialdehyde contents in kidney homogenate, compared to the control and which means the lipid peroxidation was a main cause of nephrotoxicity. In order to investigate the cytotoxic mechanism of RC-1, we also tested and revealed the generation of oxygen free radicals derived from neutrophil stimulated by RC-1 and interaction of the oxygen free radicals with the erythrocyte membrane. From the above results, we suggest that nephrotoxicity of general platinum (II) antitumor compounds as well as RC-1 were inhibited by radical scavengers.
Iron deficient anemia in piglets could be overcome by supplementary iron. Overloaded iron induced peroxidation of cell membrane and increased malonaldehyde (MDA). Antioxidant activity of vitamin C has been studied in iron-overloaded swine plasma. Erythrocyte fragility, MDA, glutathione, vitamin A, and vitamin E were measured in swine plasma with or without iron (0~1mg/dl) and vitamin C (0~10mg/dl). Erythrocyte fragility increased from 8% to 45% in iron group and reduced from 57% to 43% in vitamin C group with dose dependant response. MDA was $0.94{\pm}0.05$ and $1.86{\pm}0.10$ nmol/ml in piglet and pig, respectively, and significantly high in pig (p<0.05). Iron increased MDA from $1.86{\pm}0.10$ to $9.46{\pm}0.04$ nmol/ml in pig, but not in piglet (p<0.05). Vitamin C reduced MDA from $9.46{\pm}0.04$ to $4.80{\pm}0.10$ nmol/ml in pig. Iron increased glutathione from $90.12{\pm}0.10$ to $108.52{\pm}5.29$ nmol/dl in pig, and vitamin C reduced glutathione from $108.52{\pm}5.29$ to $93.52{\pm}2.44$ nmol/dl (p<0.05). Vitamin A and E were $24.86{\pm}2.70$ to $138.29{\pm}6.70{\mu}g/dl$, respectively in iron group, and $35.76{\pm}0.60$ to $177.21{\pm}2.95{\mu}g/dl$, respectively in supplementary vitamin C group (p<0.05). These data indicated an antioxidant activity of vitamin C in iron-overloaded swine plasma.
Objectives : JiaoTeng-Yuan(교등원) is said to be a prescription for preservation of health in $\ulcorner$HuaTuo.ZhongZangJing (화타.중장경)$\lrcorner$. It is known to have the effect of Bu-Shen (보신: strengthening kidney) and Yi-Shou (익수: prolonging the span of one's life). This study investigates whether JTY is effective on inhibition of oxidation stress. Methods : Sprague-Dawley rats (12-week-old, weight $300{\pm}20g$) were divided into 3 groups. The normal group (n=8) was injected with PBS (1ml/body, s.c) subcutaneously behind the neck. The control group (n=8) was injected with D-galactose (50mg/kg, 1ml PBS/body, s.c) to induce pathological animals. The JTY group was injected with the same treatment as the control group, and fed containing JTY (10%). All groups were treated 1 time per day for 6 weeks. After the rats were sacrificed anti-oxidant enzyme (SOD, CAT, G-px) activity, GSH quantity of RBC and tissue (heart, liver and kidney), and plasma Vit-C quantity were examined. Also, the MDA levels of liver and kidney, lipofuscin of heart and endurance of erythrocyte membrane were measured. Results : In the JTY group, RBC's SOD activity decline was halted by 21 % of the normal level, compared to the control group, G-px activity (unit/g of Hb) increased significantly, compared to the normal group, and the level of Vit-C in plasma increased by 16%. Heart's SOD activity was kept at the same level as that of the normal group; and CAT activity decline was halted by 26%. Kidney's CAT and G-px activities were kept at the same level as that shown in the normal group, implying the existence of a halting effect. Liver also showed a slight halting effect against the decline of anti-oxidant ability, but the effect was not significant (${\alpha}=0.05$). A comparison between the levels of peroxide in SD rats showed that the level of TBARS in plasma increased significantly in the control group and that it was normal in the JTY group. The livers in the JTY group, compared to those in the control group, showed 36% halting effect of the normal level while their kidney's indicated the level significantly lower than the normal level. Heart's lipofuscin increased significantly in the control group, but was alike in both the JTY and the normal groups. Endurance of erythrocyte membrane (%) decreased significantly in the control group while it was kept at the similar level in both the JTY and the normal groups, indicating the halting effect. Conclusions : This study suggests that JTY is effective to defend oxidation stress caused by D-galactose in the animals. It showed that the anti-oxidant ability was maintained and strengthened. On the other hand, it reduced the level of peroxide in animals. In sum, JTY appeared to have the equilibrium normal physiological function in SD rat.
This study was conducted to evaluate the effect of dietary antioxidant and energy density on performance and antioxidative status in transition cows. Forty cows were randomly allocated to 4 dietary treatments in a $2{\times}2$ factorial design. High or low energy density diets (1.43 or 1.28 Mcal $NE_L$/kg DM, respectively) were formulated with or without antioxidant (AOX, a dry granular blend of ethoxyquin and tertiary-butylhydroquinone; 0 or 5 g/cow per d). These diets were fed to cows for 21 days pre-partum. During the post-partum period, all cows were fed the same lactation diets, and AOX treatment followed as for the pre-partum period. Feeding a high energy diet depressed the DMI, milk yield, and 4% fat-corrected milk (FCM) of cows. However, AOX inclusion in the diet improved the milk and 4% FCM yields. There was an interaction of energy density by AOX on milk protein, milk fat and total solids contents. Feeding a high energy diet pre-partum increased plasma glucose and ${\beta}$-hydroxybutyrate, whereas dietary AOX decreased plasma ${\beta}$-hydroxybutyrate value during the transition period. There were also interactions between time and treatment for plasma glutathione peroxidase activity and malondialdehyde content during the study. Cows fed high energy diets pre-partum had higher plasma glutathione peroxidase activity 3 days prior to parturition, compared with those on low energy diets. Inclusion of AOX in diets decreased plasma glutathione peroxidase activity in cows 3 and 10 days pre-partum. Addition of AOX significantly decreased malondialdehyde values at calving. Energy density induced marginal changes in fatty acid composition in the erythrocyte membrane 3 days post-partum, while AOX only significantly increased cis-9, trans-11 conjugated linoleic acid composition. The increase in fluidity of the erythrocyte membrane was only observed in the high energy treatment. It is suggested that a diet containing high energy density pre-partum may negatively affect the anti-oxidative status, DMI and subsequent performance. Addition of AOX may improve the anti-oxidative status and reduce plasma ${\beta}$-hydroxybutyrate, eventually resulting in improved lactation performance; the response to AOX addition was more pronounced on the high energy diet.
Wistar rat에 $^{88}SrCl_2$를 꼬리 정맥에 주사하여 체내 기관과 혈액 내 분포, 잔존율을 조사하였고 착화제와 유기산을 투석하여 혈장 단백질에 결합하는 Sr 양의 변화를 측정하였다. 혈액내에서 Sr은 혈장에 60%, 세포에 40% 부착되어 이동하였다. 혈장에 존재하는 Sr 중 약 50%정도는 혈장 단백질과 결합한 상태였고, 세포에는 세포 표면에 가볍게 부착되어 있었다. Erythrocyte나 granulocyte보다 lymphocyte에 많은 양의 Sr이 부착되어 있었다. 투여후 초기 1시간 이내에 혈액 내에서 급격히 감소하여 뼈에 침착되었다. 이때 각 기관에서도 Sr의 잔존율은 24시간 이내에 크게 감소하였고, 뼈로 침착된 Sr은 24시간 이후에 서서히 감소하였다. 착화제 EDTA, EGTA 및 DTPA를 투여한 경우, 혈장 단백질에 결합하는 Sr의 양은 대조군의 57%에서 27-33%로 감소하였으며 citrate 및 oxalate의 투여시는 이 값이 19%와 40%로 각각 감소하였다.

To simplify the different biological investigation of the methanolic extract and solvent-solvent partitioning of Lablab purpures (L. purpures) bark. In-vitro anti-oxidant study was determined using total DPPH radical scavenging assay. In vitro antimicrobial study was measured by observing zone of inhibition. The cytotoxic activity was studied using brine shrimp lethality bioassay and thrombolytic activity by clot disruption method. The antioxidant potential was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Folin-Ciocalteau reagents using butylated hydroxytolune (BHT) and ascorbic acid as standards. The Aqueous soluble fraction revealed the highest free radical scavenging activity ($IC_{50}=48.76{\mu}g/mL$). The antimicrobial screening of the bark of L. purpures exhibited mild to moderate activity in test microorganisms. The CSF showed the maximum relative percentage inhibition against Salmonella parathyphi (34.2%) for bacteria and C. albicans (28.8%) for fungi whereas, lowest relative percentage inhibition against Sarcina lutea (22.0%) for bacteria and Aspergillus niger (24.4%) for fungi. In the brine shrimp lethality bioassay, The $LC_{50}$ values of Carbon tetrachloride and N-Hexane soluble fraction were found $92.18{\mu}g/mL$, and $68.95{\mu}g/mL$ respectively while the $LC_{50}$ values of standard Vincristine sulphate was $1.37{\mu}g/mL$. The methanolic extract and its organic soluble fractions of Lablab purpureus at concentration 2.0 mg/mL, significantly protected the lysis of erythrocyte membrane induced by hypotonic solution and heat as compared to the standard, acetyl salicylic acid (0.10 mg/mL). The MSF and AQSF produced 61.48 % and 53.75% inhibition of hemolysis of RBC caused by hypotonic solution respectively, whereas acetyl salicylic acid (0.10 mg/mL) showed 76.42%. Ethanol extract of L. purpures and all of its different partitions exhibited moderate thrombolytic activity of 37.25%-2.40%. Very good preliminary screening and simplified experiments were able to show the different biological activity of methanolic extract and its soluble fractions of L. purpures at a time.
