AN IMMUNOHISTOCHEMICAL STUDY ON PROLIFERATION, DIFFERENTIATION, AND APOPTOSIS IN THE EPITHELIAL CELLS OF THE ODONTOGENIC CYSTS (치성낭종 상피세포의 증식, 분화 및 세포능동사망현상에 관한 면역조직화학적 연구)

  • The epithelium of odontogenic cyst seems to be in a specific status of cellular proliferation and cytodifferentiation. With the identification of various genes, which play essential roles in the specific stages of cellular proliferation and differentiation, the cellular conditions of odontogenic cyst epithelium need to be reevaluated. This study aimed to estimate the degree of proliferating, differentiating and apoptotic activities of odontogenic cyst epithelium using antisera of PCNA, Ki-67, MPM-2, transglutaminase C, heat shock protein 70 and $ApopTag$^{(R)}$. method in 19 cases of odontogenic cysts. Cellular changes of the cyst epithelium were measured by intensity of each immunohistochemical staining. Results were as follows: 1. The proliferating activity of the cyst epithelium was slightly lower than that of normal oral mucosal epithelium, with the use of primary antibodies against PCNA, Ki-67, and MPM-2. And the proliferating activity of the epithelium in OKC group was even higher than that of the epithelium in non-OKC group. 2. The odontogenic cysts showed weakly positive reaction with transglutaminase C, but strongly positive reaction with HSP 70. 3. Occasionally, only a few apoptotic cell was observed in the superficial keratin layer of OKC. 4. The hyperplastic cyst epithelium infiltrated with mild inflammatory cells showed diffusely positive reaction with different proliferating factors. From the above results, we presumed that the endogenous proliferating and differentiating activity of the cyst epithelium was slightly lower than that of normal oral mucosal epithelium, and also supposed that the cyst epithelium could be reactivated for the further proliferation by the exogenous factors, such as inflammatory reaction and any chemicophysical irritations.

Development on the esophagus of fetuses and neonates in Korean native goats (한국재래산양의 태아 및 신생아의 식도 발달에 관하여)

  • The development of esophagus in fetuses between 60, 90, 120 days of gestation and neonates of Korean native goats was investigated by light, scanning electron microscopy. The results were summarized as follows; 1. The esophageal wall appeared to be differentiated into the epithelium, lamina propria, tunica muscularis and tunica adventitia at 60 days of gestation. The esophageal epithelium was stratified cuboidal at 60 days, being transformed into stratified squamous epithelium at 90 days, and completely transformed into squamous epithelium an 120 days. 2. In scanning electron microscopy, the longitudinal furrows and ridges appeared on the epithelium of the esophagus of the fetuses at 60 days of gestation. The longitudinal furrow became deeper and microplicae appeared in 90-day-old fetuses. The transverse and longitudinal folds appeared on the epithelial surface of esophagus and the esophageal epithelium started to be keratinized in the neonates.

Histological Changes of Hepatocyte and Intestinal Epithelium during Starvation in Olive Flounder Paralichthys olivaceus (넙치 Paralichthys olivaceus 기아시 간세포 및 장상피세포의 조직학적 변화)

  • The effects of starvation on histological changes in hepatocytes and the intestinal epithelium in the olive flounder, Paralichthys olivaceus were examined. The starved group was not fed for 12 weeks, whereas the fed group was given food. Food deprivation resulted in a significant decrease in hepatocyte nucleus size and nuclear height in the intestinal epithelium (P<0.05). In the starved group, pronounced degenerative changes were seen in the histology of hepatocytes and the intestinal epithelium; the tissue morphology was irregular and the nuclei were stained basophillic, shrunk irregularly, and very compact. These results suggest that histological changes in hepatocytes and the intestinal epithelium can be used as an indicator of starvation in the olive flounder. In addition, the data are interpreted in detail and some biologically important relationships are discussed.

  • The endocrine cells in the midgut epithelium of the Japanese cockroach, Periplaneta japonica were observed by the light and electron microscopy. The midgut epithelium of the last instar larva and adult cockroach consisted of principal columnar cells, regenerative cells, and secretory granular cells. Midgut endocrine cells were positioned basally as a cone-shaped single cell in the epithelium or underneath the regenerative crypt cells. When midgut epithelium grows and the cell composing it transforms, between the endocrine cells and regenerative cells were made desmosome type junction and large vesicular shaped stretches of loose contact. The endocrine cells were characterized by a clear cytoplasm with abundant Golgi complex and numerous secretory granules. The secretory granules in the cell were spherical and electron dense with their diameter of $200{\sim}400nm$. The secretory granules have been observed as discharged by exocytosis on the basal and lateral side of the cell.

Oxygen matters: hypoxia as a pathogenic mechanism in rhinosinusitis

  • The airway epithelium is the first place, where a defense mechanism is initiated against environmental stimuli. Mucociliary transport (MCT), which is the defense mechanism of the airway and the role of airway epithelium as mechanical barriers are essential in innate immunity. To maintain normal physiologic function, normal oxygenation is critical for the production of energy for optimal cellular functions. Several pathologic conditions are associated with a decrease in oxygen tension in airway epithelium and chronic sinusitis is one of the airway diseases, which is associated with the hypoxic condition, a potent inflammatory stimulant. We have observed the overexpression of the hypoxia-inducible factor 1 (HIF-1), an essential factor for oxygen homeostasis, in the epithelium of sinus mucosa in sinusitis patients. In a series of previous reports, we have found hypoxia-induced mucus hyperproduction, especially by MUC5AC hyperproduction, disruption of epithelial barrier function by the production of VEGF, and down-regulation of junctional proteins such as ZO-1 and E-cadherin. Furthermore, hypoxia-induced inflammation by HMGB1 translocation into the cytoplasm results in the release of IL-8 through a ROS-dependent mechanism in upper airway epithelium. In this mini-review, we briefly introduce and summarize current progress in the pathogenesis of sinusitis related to hypoxia. The investigation of hypoxia-related pathophysiology in airway epithelium will suggest new insights on airway inflammatory diseases, such as rhinosinusitis for clinical application and drug development.

Postpartum Changes in the Uterus of Goats II. Electron Microscopic Observations in the Uterine and Vaginal Epithelium of Post-partum Korean Native Goats (산양에 있어서 분만후 자궁의 변화 II. 한국재래산양에 있어서 분만후 자궁 및 질상피세포의 전자현미경적 관찰)

  • Morphological changes in the uterine and vaginal epithelial cells of the Korean native goats were studied in fifteen primiparous goats slaughtered on the day of parturition and on days 1, 3, 10 and 21 postpartum. 15 uterus and vagina from goats were examined by scanning and transmission electron microscopy. The results obtained in this study were summarized as follows : 1. Transmission electron microscopically, long microvilli which sometimes ramified were found until 10 days postpartum, while short microvilli were found at 21 days. The high electron dense irregular-shaped mitochondria were found in the cytoplasm and the crystalline structure of the mitochondrial matrix was also found from 1 day to 10 days postpartum. Well-developed rough-endoplasmic reticulum (rER) with dilated cisternae which contained the proteins materials was observed at 21 days postpartum. These materials were fused each other and then large granules were found in the free surface of the cytoplasm. A few lipid droplets were generally appeared in the cytoplasm, while numerous droplets were found at 21 days postpartum. A moderate number of ribosomes, a few multivesicular bodies, vesicles, lysosomes and macrophages were found. The globule leucocytes were observed from 0 to 3 days postpartum by transmission electron microscopy. The short microvilli, high electron dense cytoplasm and severe indentation of the nuclear enbelope were found in the vaginal epithelium. Numerouos small vesicles and a few vacuoles were observed in the apical cytoplasmic portion of the epithelium. A few mitochondria were high electron dense and irregular in shape. A moderate amounts of microfilaments, loose intercellular space and dilated rER were also found at 21 days postpartum. 2. Scanning electron microscopically, the folds of the uterine mucosa were generally deep. The long microvilli of the epithelium were found until 3 days postpartum, while short microvili were found at 10 and 21 days postpartum. The distinct intercellular boundary was seen. The apporcine secretory profile of the epithelium observed at between 3 and 10 days postpartum and the cells were somewhat protruded into the lumen. The short microvilli were found on the surface of the protruded cells, while polygonal microridge profile of the epithelium and some dome-shaped epithelium were also observed at 21 days postpartum. The folds of the vaginal mucosa were deep and epithelium was polygonal in shape. The microvilli of the epithelium were long until 3 days postpartum, while they were short at 10 and 21 days. The polygonal epithelium was invaginated into the center of the cell surface until 10 days postpartum. The microridge and dome in shape of the epithelium were found at 10 days postpartum, while the polygonal and exfoliating epithelium were observed at 21 days.

The Papillar and Pelvic Epithelia of the Bat: A Scanning and Transmission Electron Microscopic Study (집박쥐 신유두(腎乳頭) 및 신우상피(腎盂上皮)에 미세구조(微細構造))

  • The anatomy of the renal papilla and pelvis of the bat(Pipistrellus abramus abramus Temminck) and their ultrastructure of the epithelium has been studied by means of light, scanning and transmission electron microscopy. The bats were captured at Ondal cave located in Yangchun-Myun, Tanyang-Kun, Chungchongpuk-Do, Korea in the active(July) and hibernating(January) phases. A conical renal papilla with several papillary foramina at the apex was curved toward the ureter. At the margin of the funnel-shaped renal pelvis, the fornix extended deeply into the kidney parenchyme was formed between the outer zone of the medulla and the cortex. The epithelium of the papilla covered the inner zone of the medulla. The epithelium of the fornix medially overlayed the outer zone of the medulla, and laterally the cortex. The surface epithelium of the papilla was simple columnar in shape with the scanty organelles and the short microvilli on their luminal surface. The fornix was Jined with $1{\sim}2$ cell layered squamous epithelium. On the medial surface of the fornix, two types of the superficial epithelial cells were identified; one had numerous short microvilli and the other had the microplicae and plaques on their luminal surface. The lateral surface of the fornix lined with the epithelial cells showed microridges and plaques on the luminal surface. The epithelial cells on the medial and lateral surfaces of the fornix were similar to their internal ultrastructures. The pelvic epithelium was a typical transitional type, which was composed of a layer of basal cells, one or two layers of intermediate cells, and a layer of superficial(facet) cells. The fusiform vesicles were observed in the cytoplasm of the intermediate and facet cells. There were no noticeable morphological changes in the epithelium of the papilla, fornix and pelvis except the medial epithelium of the fornix in the hibernating cycle. The microvillar cells were more numerous in the hibernating phase than in the active phase. The morphology, of the renal papilla and fornix of bats, indicates that a functional relationship may exist between the pelvic urine and the renal parenchyme.

Histochemical Study on the Uterine Development in Fetues and Neonates of Korean Native Goats (한국재래산양의 태아 및 신생아 자궁의 조직 발달에 관한 조직화학적 연구)

  • The present study was performed to investigate the patterns of a, pp.arance of lectin in uterus of fetuses of 90 and 120 days old and neonates of Korean native goat. The carbohydrate markers were used in histochemistry for the determination of the lectin by staining of avidin-biotin-per-oxidase complex(ABC), rincinus communis agglutinin(RCA-I), ulexeuropalus communis agglutinin(UEA) and wheat germ agglutinin(WGA). 1. The effects of this study were as follows; 1. The binding reactions for Con-A were weak on the mucosal epithelium of endometrium in 90 and 120 days old fetuses, and neonates and moderate at the free surface of mucosal epithelia. 2. The binding reactions for DBA was partially moderate on the mucosal epithelium of endometrium and partially strong at the free surface of mucosal epithelia in 120 days old fetuses. In neonates, the reactions were strong on the mucosal epithelium and gland primordium of endometrium, and the secretions at the free surface showed strong reactions for DBA. But, in 90 days old fetuses, the reaction was negative. 3. The binding reactions for RCA-I were moderate on the mucosal epithelium of endometrium and at the free surface of mucosal epithelia in 90 days old fetuses. In 120 days old fetuses, the reactions were weak on the mucosal epithelium of endometrium and moderate at the free surface of mucosal epithelia. In neonates, the reactions were moderate on the mucosal epithelium of endometrium and strong at the free surface of mucosal epithelia and also strong in the uterine gland. 4. The binding reactions for UEA were negative in 90 and 120 days old fetuses and neonates. 5. In 90 days old fetuses, the binding reactions for WGA were generally weak on the mucosal epithelium of endometrium, but several epithelial cells showed moderate reaction for WGA. In 120 days old fetuses and neonates, the reactions were moderate on the mucosal epithelium and blood vessels of endometrium and strong at the free surface of mucosal epithelia.

Short-Term Changes in Gut Microflora and Intestinal Epithelium in X-Ray Exposed Mice

  • Background: Gut microflora contributes to the nutritional metabolism of the host and to strengthen its immune system. However, if the intestinal barrier function of the living body is destroyed by radiation exposure, the intestinal bacteria harm the health of the host and cause sepsis. Therefore, this study aims to trace short-term radiation-induced changes in the mouse gut microflora-dominant bacterial genus, and analyze the degree of intestinal epithelial damage. Materials and Methods: Mice were irradiated with 0, 2, 4, 8 Gy X-rays, and the gut microflora and intestinal epithelial changes were analyzed 72 hours later. Five representative genera of Actinobacteria, Firmicutes, and Bacteroidetes were analyzed in fecal samples, and the intestine was pathologically analyzed by Hematoxylin-Eosin and Alcian blue staining. In addition, DNA fragmentation was evaluated by the TdT-mediated dUTP nick-end labeling (TUNEL) assay. Results and Discussion: The small intestine showed shortened villi and reduced number of goblet cells upon 8 Gy irradiation. The large intestine epithelium showed no significant morphological changes, but the number of goblet cells were reduced in a radiation dose-dependent manner. Moreover, the small intestinal epithelium of 8 Gy-irradiated mice showed significant DNA damaged, whereas the large intestine epithelium was damaged in a dose-dependent manner. Overall, the large intestine epithelium showed less recovery potential upon radiation exposure than the small intestinal epithelium. Analysis of the intestinal flora revealed fluctuations in lactic acid bacteria excretion after irradiation regardless of the morphological changes of intestinal epithelium. Altogether, it became clear that radiation exposure could cause an immediate change of their excretion. Conclusion: This study revealed changes in the intestinal epithelium and intestinal microbiota that may pave the way for the identification of novel biomarkers of radiation-induced gastrointestinal disorders and develop new therapeutic strategies to treat patients with acute radiation syndrome.

Histological and Histochemical Studies on the Epididymal Region and Deferent Ducts of the Drakes by the Age in Weeks (오리 부고환(副睾丸) 및 정관(精管)의 주령별(週齡別) 조직학적(組織學的) 및 조직화학적(組織化學的) 연구(硏究))

  • This study was made for the better information of the male reproductive system on the meat-type drake, Cherry Belly X White Golden. The epithelium of ductules of epididymal region and deferent duct were observed histologically and histochemically with the progress of their development. India-ink absorbability on the luminal epithelium was also investigated after the administration of India-ink. The results are as follows; 1. Rete testis and various round ductules in immature form appeared in epididymis within 6 weeks after hatching, and simple cuboidal and simple columnar epithelium were found in the epithelia of the ductules within 8 weeks after hatching. Larger ductules were found on epididymal surface which was in the developing stage near to the immature efferent ductule. From 10th to 20th week, various ductules appeared in epididymis, and developing form of efferent ductules were much more increased on epididymal surface. The luminal epithelium of the ductules were composed of ciliated simple columnar and pseudostratified ciliated columnar cells. At the same time, deferent duct appeared. From the 21th week, various ductules in epididymis became abruptly matured. Lumen of rete testis was lined by simple squamous or simple cuboidal epithelium, and that of efferent ductules, having many folds and being larger than any others were lined by pseudostratified ciliated columnar epithelium in which ciliated columnar cells, non-ciliated cells(clear cells) and basal cells were noted. Connecting tubules of star shaped lumen were composed of pseudostratified ciliated columnar epithelium in which ciliated columnar cells, nonciliated cells, and basal cells were observed. The luminal surface of epididymal ducts was smooth and has thick pseudostratified columnar epithelium which was composed of high columnar cells and basal cells. From 26th week after hatching, sperm pooling was started in various ductules. 2. From 4th to 10th week, simple cuboidal epithelium of deferent duct transformed to simple columnar epithelium with the progress of aging. At the basement of epithelium, clear round cells were noted. From 12th to 20th week, high columnar cells with enlongated nucleus were noted on the luminal border of deferent ducts, forming folds of pseuclostratified columnar epithelium. From 20th week, the deferent duct started to have septa in it's lumen and composed mainly of pseudostratified columnar epithelium, and round cells disappeared. From 20th week, the lumen diameter of deferent duct became wider with the progress of aging, but there was no difference among the values of lumen diameter in upper, middle, and lower part of deferent ducts. At 26th week, the pooling period of sperms in deferent ducts, the lumen diameter became rapidly widen, especially in the lower part of deferent ducts. Thickness of muscular layer of ductus deferens showed gradual growth within 24 weeks but did abrupt thickening from 26th week. 3. Saliva resistant PAS granules were dotted on the top of nucleus in efferent ductules epithelium but the amount of the granules were little in the connecting ductules's epithelium. The granules reactive to acid phosphatase were abundant in the some epithelial cells of efferent ductules and connecting ductules, especially above the nucleus of cells. The granules reactive to alkaline phosphatase were noted on the luminal border of efferent ductules. Parts of free border of efferent ductules and middle portion of deferent ducts were stained slightly by alcian blue technique. India ink granules were found mainly in the epithelium of efferent ductules but were few in that of connecting ductules.
