여러 가지 배양조건에서 돼지 난포란의 체외수정 및 체외발달에 관한 연구 (Studies on the In Vitro Fertilization and In Vitro Development of Porcine Embryos in Different Culture System)

  • 본 연구는 돼지 체외수정란의 생산에 있어서 체외배양체계를 확립하기 위해 정액의 차이와 보관시간에 따른 체외성숙난자의 체외수정 및 체외발달율을 조사하였고, NCSU23와 G1.3/G2.3 배양액으로 체외배양 시 분할율과 체외발달율을 조사하였고, 체외배양액에 GSH를 첨가 시 배발달율을 조사하였다. 본 연구에서 얻은 결과는 다음과 같다. 1. 액상정액과 동결정액을 사용하여 정액에 따른 체외수정율은 각각 46.2 및 39.7%로써 액상정액을 이용한 것이 수정율이 높았지만 유의적(P<0.05)인 차이는 없었다. 한편 배반포로의 발달율은 액상정액이 동결정액보다 유의적으로 높았다.(P< 0.05) 2. 정소상체미부 정액을 day 1, 2, 3 보관후 체외수정을 유도하였을 때 수정율은 각각 60.5, 61.0 및 56.8%로서 2일 동안 보관후 사용하였을 때가 높았지만 유의적(P<0.05)인 차이가 없었다. 또한 상실배와 배반포기까지의 배발달율에서도 2일 동안 보관 후 사용하였을 때가 높았지만 유의적(P<0.05)인 차이는 없었다. 3. NCSU-23와 G1.3/G2.3으로 나눠 배양 시 분할율과 상실배기까지의 배발달율은 각각 52.8, 62.1%와 16.0, 28.9%로서 G1.3/G2.3에서 유의적(P<0.05)인 차이로 높았다. 그러나 배반포기로의 배발달율을 각각 11.6와 4.7%로서 NCSU-23에서 유의적으로 높았다.(P<0.05) 4. NCSU-23을 기본 배양액으로 하여 1 mM GSH가 첨가된 군의 분할율은 62.3%로서 GSH가 첨가되지 않은 군 53.5%보다 유의적(P<0.05)인 차로 높았지만, 상실배나 배반포기까지의 배발달율은 GSH가 첨가되지 않은 군보다 높았지만 유의적(P<0.05)인 차는 존재하지 않았다.ne을 처리하는 것이 수태율을 향상시키는 것으로 나타났다.25^{\circ}C$에서 발현되었고 이 중 79.1%가 3일령, 4일령과 5일령에 집중적으로 나타났다. 계분의 경우 $25^{\circ}C$ 처리구에서 35.12%(56.95mg/kg), $37^{\circ}C$에서 45.89%(74.40mg/kg)로 나타나 주로 $25^{\circ}C$ 이상에서 발현한 것으로 특징지어졌다. 우분의 경우 $10^{\circ}C$ 처리구에서 28.21%(43.86mg/kg), $25^{\circ}C$에서 49.30% (76.66mg/kg)로 나타나 주로 $25^{\circ}C$ 이하에서 발현한 것으로 나타났다. 3. 배양온도에서 검지 된 뷰틸산의 량은 6일 동안 돈분에서 1,463.87mg/kg, 계분에서 96.72mg/kg, 우분에서 129.18mg/kg이 발현되었으며 돈분의 경우 93.31%(1,365.95mg/kg)가 $25^{\circ}C$에서 발현되었고 이 중 87.92%가 3일령, 4일령과 6일령에 집중적으로 나타났다. 계분의 경우 $37^{\circ}C$ 처리구에서 76.60%(74.09 mg/kg)로 발현되었고 이 중 88%가 1일령, 2일령과 5일령에 집중적으로 나타났다. 우분의 경우 61.55%(79.51mg/kg)가 $25^{\circ}C$에서 발현되었고 이 중 89.6%가 1일령, 3일령과 4일령에 집중적으로 나타났다. 4. 배양온도에서 검지된 이소밸릭산의 량은 6일 동안 돈분에서 6,885.99mg/kg, 계분에서 307.47mg/kg,

  • 야생의 우수리땃쥐, Crocidura lasiura의 부정소 미부의 정상과 비정상 형태의 정자를 광학 및 전자현미경으로 비교 관찰하였다. 정상 정자와 함께 다수의 형태 비정상 정자가 관찰되었다. 이것은 비정상 정자의 존재가 개체의 생식력 없음을 나타내는 절대적 지표가 아님을 의미한다. 하지만 비정상 정자의 존재는 개체의 생식력 없음과 연관되며 특히 두부의 형태의 이상은 생식력 없음과 밀접한 관계가 있다. 건강한 야생의 우수리땃쥐, C. lasiura의 부정소 미부에서 관찰된 정자의 비정상 두부의 형태는 다음과 같은 유형으로 나타났다. 1) 핵질의 응축이 이루어지지 않거나 부분적으로 이루어진 형태 2) 첨체의 분열 혹은 파손이 나타나는 형태 3) 첨체 혹은 원형질막이 부분적 혹은 완전히 접혀진 형태 4) 첨체가 핵과 분리되는 형태 5) 첨체가 불규칙적으로 응축하는 형태 6) 잘못된 위치에서 관찰되는 첨단체

수컷랫드에 있어서 새로운 안트라사이클린계 항암제 DA-125의 생식독성 연구 (Male Reproductive Toxicity of DA-125, a New Anthracycline Anticancer Agent, in Rats)

  • The toxicity of DA-125. a new anthracycline anticancer agent, on the male reproductive system was studied in Sprague-Dawley rats. Forty male rats were rando$m\ell$y assigned to Jour groups with ten rats in each group and given single intraveneous doses of DA-125 at dose levels of 0. 12.5. 25. and 50 mg/kg body weight. On day 56 after treatment the animals were allowed to mate. and their male reproductive Junctions and organs were examined in detail. Copulated females were sacrificed on day 20 of gestation for examination of embryo-fetal development. One out of ten rats in the 50 mg/kg group died on day 12 after treatment. Clinical signs such as emaciation. sedation, anorexia. swelling. dark material around eye. alopecia. and diarrhea were observed in the 25 and/or 50 mg/kg groups. Reduction in the body weight gain. decrease in the absolute weights of testes. epididymis and seminal vesicles. and/or decrease in the number of testicular sperm heads were also found. Although histopathological changes such as atrophy of seminiferous tubules. loss or decrease of spermatogenic cells. exfoliation of spermatogenic cells. vacuolization of Sertoli cells. decrease of sperm. and/or increase of necrotic spermatogenic cells in epididymal ducts were observed. no adverse effects on the motility and morphology of epididymal sperm. copulation index. fertility index. and embryo-fetal development were detected in the 25 and 50 mg/kg groups. There were no evidences of male reproductive toxicity in the 12.5 mg/kg group. These results show that single intravenouse doses of DA-125 produce significant dose-related testicular atrophy. histopathological changes. and oligozoospermia in rats and $LD_{10}$ for DA-125 appears to be 50 mg/kg body weight.

Effect of Sperm Heat Stress on Embryo Development in Cattle

  • Heat stress to bovine oocytes and embryos has suggested a potential role of retardation of their development. Limited study has reported on the effect of heat shock on sperm before using it for IVF. Caudal epididymal sperm cultured in 42$^{\circ}C$ incubator for 0.5, 1 and 2 h compared on sperm viability and oocyte development after its use for IVF to those of control. Oocytes were matured for 22 h and then inseminated with treated or control sperm for 16 h. Embryos were cultured in CRlaa medium, transferred to TCM199+10% FBS on day 4, and maintained on day 9. A higher proportion (84.1%, 0.5 h; 72%, 1 h: 65%, 2 h) in treated sperm was observed dead and abnormal pattern as 100% of consider as control. In control the rates of cleavage and development into blastocyst were 76% and 22%, respectively, and did not differ the rates between 1 h and 2 h of culture. Significant differences were appeared in the rates between treated for an hour and control (32% and 5% vs. 54% and 10%, respectively). Moreover increased time of culture is more retardation to be cleaved the oocytes. However, the rates of blastocyst from cleaved embryos in treated group similar to control (25% vs. 29%, respectively). The reason for this remains unclear, but male sperm, from preliminary experiment(data un-shown) for sexing of resulting embryos, would be more fragility on heat stress.

The effects of Commiphora mukul extract on spermatogenesis and testosterone levels in male diabetic rats

  • Objective: The strong antioxidant activity of Commiphora mukul prompted us to conduct the present study to explore whether treatment with C. mukul extract (CME) would have any protective influence on sperm parameters, testosterone levels, and plasma glucose levels in streptozotocin (STZ)-induced diabetic rats. Methods: Male Wistar rats were randomly divided into four groups: control, control animals treated with CME, diabetic animals, and diabetic animals treated with CME. CME extract (300 mg/kg) was administered for 60 days by daily gavage. Diabetes was induced by an intraperitoneal injection of 50 mg/kg STZ. The epididymal sperm count, weight, motility, morphology, viability, and serum testosterone and glucose levels were determined. Results: In the diabetic animals, CME decreased blood glucose levels (p< 0.05), increased the total sperm count (p< 0.05), and decreased the proportion of sperm with abnormal morphology (p< 0.05). Diabetes reduced sperm motility (p< 0.001), and CME supplementation partially reversed this effect of diabetes (p= 0.003). Furthermore, in diabetic animals, CME decreased the proportion of immotile sperm (p< 0.001). In rats, diabetes caused a significant decrease (p< 0.05) in serum testosterone levels (F[3, 28] = 3.283, p= 0.035), but treatment of diabetic animals with CME increased serum testosterone levels. Conclusion: The present study demonstrated that C. mukul possesses proandrogenic activity and exerts a beneficial effect on sperm parameters in diabetic rats.

Impact of glycosylation on the unimpaired functions of the sperm

  • One of the key factors of early development is the specification of competence between the oocyte and the sperm, which occurs during gametogenesis. However, the starting point, growth, and maturation for acquiring competence during spermatogenesis and oogenesis in mammals are very different. Spermatogenesis includes spermiogenesis, but such a metamorphosis is not observed during oogenesis. Glycosylation, a ubiquitous modification, is a preliminary requisite for distribution of the structural and functional components of spermatids for metamorphosis. In addition, glycosylation using epididymal or female genital secretory glycans is an important process for the sperm maturation, the acquisition of the potential for fertilization, and the acceleration of early embryo development. However, nonemzymatic unexpected covalent bonding of a carbohydrate and malglycosylation can result in falling fertility rates as shown in the diabetic male. So far, glycosylation during spermatogenesis and the dynamics of the plasma membrane in the process of capacitation and fertilization have been evaluated, and a powerful role of glycosylation in spermatogenesis and early development is also suggested by structural bioinformatics, functional genomics, and functional proteomics. Further understanding of glycosylation is needed to provide a better understanding of fertilization and embryo development and for the development of new diagnostic and therapeutic tools for infertility.

세포질내 정자주입을 시행한 인간 난자의 전핵 형성 및 배아 발생에 관한 연구 (Pronucleus Formation and Embryonic Development of the Human Oocytes Fertilized by Intracytoplasmic Sperm Injection)

  • Mammalian, including human, spermatozoa undergo morphological and physiological changes during sperm maturation. There were, these changes may affect the fertilization and embryonic development. In this study, we examined the pronucleus formation, pronucleus disappearance and embryonic development in the human oocytes fertilized by intracytoplasmic sperm injection (ICSI). The injected spermatozoa were grouped into ejaculated, epididymal and testicular by the collecting region. Among 363 metaphase II injected oocytes, 287(79.1%) oocytes were normally fertilized and displayed two pronuclei. There were no difference in the fertilization rates and in the pronucleus formation and pronucleus disappearance at 16, 20 and 24 hr after ICSI, among the each spermatozoa group. Also, at 64 hr, the appearance of embryonic development was similar. From these results it can be concluded that there was no difference of maturity among the sperm collected from ejaculated, epididymis and testis in the pronucleus formation and embryonic development. Therefore, testicular spermatozoa are successfully used with ICSI in IVF-ET program.

Spermatogenic index and hormonal profile in the rats received chromatographic fractions of ethanol extract of Crotalaria juncea L. seeds

  • The ethanol extract of the Crotalaria juncea seeds, which showed promising antispermatogenic and antiandrogenic activities in albino mice, was taken up further for the isolation of the active fractions present in it. Two fractions that were obtained from thin layer chromatography were subjected for testing to know their antispermatogenic and antiandrogenic activities. After preliminary trials the fraction I showed maximum antifertility activity at the dose level of 200 mg/kg body weight when administered orally to the rats for 50 days. The fraction I was found to affect spermatogenesis as well as the endocrine functions of the testis as indicated by gravimetric, histopathological and biochemical changes. Further this fraction has caused degenerative changes in the seminiferous tubules and Leydig cells of the testis. The accessory reproductive organs like epididymis, seminal vesicles, vas deferens, prostrate, Cowper's gland and Levator Ani muscle showed significant malfunction. Cauda epididymal sperm count and sperm motility were reduced significantly. The treatment has also resulted in increase in the cholesterol level and alkaline phosphatase activity, and decrease in protein, glycogen, sialic acid contents and acid phosphatase activity in testis. It is noteworthy that RIA studies have shown significant reduction in serum FSH, LH and testosterone. Scanning electron microscopic observations revealed abnormalities in sperm structure.

Capacitation-associated Changes in Protein-tyrosine-phosphorylation, Hyperactivation and Acrosome Reaction in Guinea Pig Sperm

  • The aim of this study was to evaluate the effects of $Ca^{2+}$, $HCO_3{^-}$ and BSA on the in vitro capacitation-associated protein tyrosine phosphorylation, hyperactivation and acrosome reaction in guinea pig sperm. Caudal epididymal sperm were incubated in four different groups: modified TALP (Tyrode's albumin lactate pyruvate) or TALP without one of the medium constituents ($Ca^{2+}$, $HCO_3{^-}$ and BSA). After incubation for the required time (0 h, 0.5 h, 1 h, 3 h, 5 h, and 7 h), sperm were removed for further experiment. The capacitation effect was assessed by CTC (Chlortetracycline) staining. Western blotting and indirect immunofluorescence were used to analyze the level and localization of tyrosine phosphorylation. The results showed that guinea pig sperm underwent a time-dependent increase in protein tyrosine phosphorylation during the in vitro capacitation and the percentage of protein tyrosine phosphorylated sperm increased from 36% to 92% from the beginning of incubation to 7 h incubation. Also, there was a shift in the site of phosphotyrosine-specific fluorescence from the head of sperm to both the head and the flagellum. Moreover, an absence of $Ca^{2+}$ or $HCO_3{^-}$ inhibited in vitro hyperactivation and acrosome reaction and decreased the phosphorylation of the proteins throughout the period of in vitro capacitation. However, an absence of BSA could not influence these processes if substituted by polyvinyl alcohol (PVA) in the medium.

Effect of Epididymal Fluid on In Vitro Maturation and Subsequent Sperm Penetration in Porcine Follicular Oocytes

  • The aim of this study is to investigate the effect of porcine epididymal fluid (pEF) on in vitro-maturation and subsequent fertilization of porcine follicular oocytes. Porcine cumulus-oocytes complexes retrieved from antral follicles were cultured in tissue culture medium (TCM)-l99 supplemented with pEF of different concentrations. At 48 h after culture, development of oocytes to germinal vesicle (GV) breakdown, metaphase I, anaphase-telophase I, and metaphase II were examined Significant (p<0.05) increase in the proportion of oocytes developed to MII stage was observed in oocytes cultured in pEF-containing TCM-l99 than in oocytes cultured in pEF-free TCM-199 (46.2% vs 16.7%), which was a dose-dependent manner. Subsequently, the proportion of monospermic fertilization were significantly (p<0.05) increased in oocytes cultured in the TCM supplemented with pEF than those cultured in pEF-free TCM-199 (51.0% vs 24.1%). In the second series of experiment, the percentage of MII oocytes was significantly (p<0.05) increased after exposure of oocytes to pEF during the first 22 h period of culture than after exposure of oocytes to pEF during the next 24 h of culture, while no significant difference in the percentage of monospermy was observed. The results of this study demonstrate that pEF contains at least enhancing component(s) for nuclear maturation.
