Determination of Ephedrine Hydrochloride and its Analogs by GC-FPD (GC-FPD를 이용한 염산에페드린 및 그 관련물질의 정량)

  • A gas chromatographic procedure is described for the quantitative analysis of ephedrine hydrochloride, pseudoephediiiie hydrochloride and norephedrine hydrochloride in pharmaceutical preparations. The procedure was based upon the determination of thioureas formed from amines and phenylisothiocyanate. The recoveries of ephedrine, pseudoephedrine and norephedrine from pharmaceutical preparations were 100.7(S.E. 0.9), 99.5(0.8), 99.1(0.8) and 99.1%(O.9), respectively. The high accuracy and precision of this assay were inherent in the flame photometric detectors normally used in gas chromatography.

Gas chromatographic determination of l-ephedrine and dl-methylephedrine (가스크로마토그라피에 의한 l-에페드린과 dl-메칠에페드린의 정량)

  • The determination method of l-ephedrine and dl-methylephedrine by gas chromatography was developed, using reactive alkaline precolumn packed with celite 545 containing KOH. Symmetrical peaks were achieved under the condition, inlet temperature, $180^{\circ}C$-$230^{\circ}C$; column temperature, $180^{\circ}C$- $160^{\circ}C$; carrier gas flow rate, 30ml/minute. The peaks of the salts coincided with those of bases. When this method was applied to preparations, using d-dimethylaminobenzaldehyde as inner standard, good results were obtained. The relative retention times of l-ephedrine and dl-methylephedrine to p-dimethyl aminobenzaldehyde were 0.50 and 0.65 respectively.

  • Pyrogallol is recently known to be a inhibitor of catechol-o-methyl transferase (COMT) and increase the action of epinephrine on the isolated rabbit atria. In this experiment, the author attempated to investigate the influence of pyrogallol on the effect of ephedrine and epinephrine on the blood pressure of the rabbit and isolated atria and excised intestine of the rabbit. The results obtained were summarized as follows; 1. Pyrogallol tends to increase the blood pressure and respiration of the rabbit. But it has no significant effect on the excised rabbit atria and intestine. 2. The effect of ephedrine on the blood pressure and respiration, isolated atria and excised intestine of the rabbit were not influenced by the pretreatment with pyrogallol. 3. The effect of epinephrine on the blood pressure and isolated atria of the rabbit is potentiated with pyrogallol pretreatment.

The Safety Guidelines for use of Ma-huang in Obesity Treatment (비만처방에서의 안전한 마황사용 지침)

  • Objectives : Ma-huang (Ephedra sinica) is frequently prescribed for obesity management in oriental medicine. The main component is ephedrine alkaloids which can have serious adverse side effects such as heart attack, stroke, sudden death. There are no scientific guidelines for Ma-huang usage in the safe treatment of obesity in oriental medicine. We reviewed published studies on its safety to make evidence based guidelines. Methods : We searched electronic databases up to May 2006. We limited evidence to controlled trials for efficacy or safety, case reports for safety, and studies for Ma-huang contents analysis. Results and Conclusions : In clinical trials for weight loss, Ma-huang and ephedrine promote modest short-term weight loss but have no serious adverse effects, have only a few adverse effects associated with increased risk of psychiatric, autonomic, gastrointestinal symptoms and heart palpitations. In case reports, there have been serious adverse effects including stroke, heart attack, and death using typical doses of ephedrine or no associated illness. There are factors related to serious adverse effects, such as overuse, lack of standardization, individual sensitivity, and interactions with other drugs. Studies relating to these factors should be analyzed for safe use of Ma-huang and ephedrine. After analyzing related studies, we suggest guidelines for Ma-huang usage. We propose that the dosage should be within 4.5-7.5g per day for up to 6 months for generally healthy individual. It's use is contraindicated in individuals with heart disease, thyroid disease, diabetes mellitus, hypertension, psychiatric disorders, glaucoma, urination disorders, enlarged prostate, persons using MAOIs, methyldopa and sympathomimetic agents.

  • Cholesterol esterase(pCEH, pancreas cholesterol ester hydrolase, E.C.3.1.1.13) which is secreted from pancreas has been known as an important lipase for cholesterol uptake. cholesteryl acyl esters from a diet must be hydrolyzed to free cholesterol and fatty acid by cholesterol esterase before the absorption in small intestine. For the development of inhibitory substances from natural source, we screened many extracts of oriental herbs for the inhibition of cholesterol esterase in vitro. The ethanol extract of Ephedra herba showed strong inhibitory activity. Solvent fractionation and silica gel column chromatography with the extract lead to the purification of the inhibitory principle in Ephedra herba. Crystallized inhibitor was identified as ( ) ephedrine by using UV, FT IR, 1H NMR, 13C NMR and GC/Mass. These results suggest that ( ) ephedrine can be used as a potential lead compound for the development of inhibitor for cholesterol uptake by cholesterol esterase inhibition.

Discrimination of the Origin of Ephedrine and Pseudoephedrine for Pharmaceutical Powder Materials by Carbon and Nitrogen Stable Isotope Ratio Analysis ($^{13}C$$^{15}N$ 안정성 동위원소 비율 분석에 의한 원료의약품에서 에페드린 및 슈도에페드린의 기원 판별)

  • The abuse of methamphetamine (MA) is one of the most serious drug abuses in Asia. And, the prevention of precursor production for abuse drug is one of the most effective drug control system. Isotope ratio analysis at natural abundance levels have been used to establish the environmental source or the geographic origin of various biological and nonbiological materials. Ephedrine, the precursor of MA, is produced by one of three methods; extraction from Ephedra plants, full chemical synthesis or via a semi-synthetic process involving the fermentation of sugar, followed by amination. We investigated the origin of ephedrine and pseudoephedrine based on the carbon and nitrogen values for nineteen pharmaceutical powder materials (PPMs) obtained from pharmaceutical company in Korea by stable isotope ratio mass spectrometry coupled to an elemental analyser (EA-IRMS). The carbon delta values for the ephedrine and pseudoephedrine were -24.21~-22.72 (mean=-23.72) $^{\cir}/_{\circ\circ}$ and -23.79~-22.71 (mean=-23.48) $^{\cir}/_{\circ\circ}$. The nitrogen delta values were 3.51~5.55 (4.43) $^{\cir}/_{\circ\circ}$ and 2.24~8.22 (5.42) $^{\cir}/_{\circ\circ}$. These results indicate that PPMs are semi-synthetic products. Therefore the origins of ephedrine(natural, semi-synthetic or synthetic) could be discriminated by using carbon and nitrogen stable isotope ratios. we are sure tat this stable isotope ratio analysis can discriminate the origins of precursors of methamphetamine.

HPLC Analysis of Marker Substances in Taeumin Gamijowi-tang (태음인(太陰人) 가미조위탕(加味調胃湯)의 HPLC 분석)

  • 1. Objectives This study was aimed to evaluate marker substances in Gamijowi-tang by high performance liquid chromatography(HPLC) 2. Methods The seperation was performed an AquasilC18(4.6X250mm)column by gradient elution with 0.1N H3PO4 in H2O - acetonitrile(0min 100:10,20min60:40, 40min 80:20, 60min 100:0) as the mobile phase at a flow-rate of 1.0 ml/min with detection at 190-800nm 3. Results Ephedrine and schizandrin were established as marker substances in Gamijowi-tang and its qualitative analysis was carried out. Analyzed was similarity of marker substances between in standard article and in Gamijowi-tang. The result of the analysis was that ephedrine and schizadrin showed 0.9999 of similarity, which meant they were really ephedrine and schizadrin. In addition, ephedrin content was 12.16mg, pseudoephedrine conrenr was 5.09mg, and schizandrin content was 26.6mg in 76g of Gamijowi-tang. 4. Conclusions Qualitative and quantitative analyses of ephedrine and schizandrin in Gamijowi-tang were conducted, and it was identified thar they could be used as marker substances in standardizing quality of Gamijowi-rang.

Effects of Methanol Extract of Ma-huang on Adipocyte of Epidiymal Fat in Rat Fed High Fat Diet (고지방 식이를 섭취한 흰쥐에서 마황의 메탄올 추출물이 체지방조직 세포에 미치는 영향)

  • Objectives : Ma-huang is a traditional Chinese medicinal herb, derived from Ephedra sinica Stapf and other Ephedra species, used to treat asthma, nose and lung congestion, and fever with anhidrosis. It contains 0.5-2.5% by weight of total alkaloids, of which ephedrine accounts for 30 to 90%. Recently, Ma-huang has been used as a source of ephedrine in many dietary supplements formulated for the treatment of obesity, since ephedrine has been found to be effective in inducing weight loss in the obese. In this study the effects of the methanol extract of Ma-huang on the adipocyte of epididymal and brown fat pads in rats fed a high fat diet for six weeks were studied. Methods : Male Sprague Dawley rats weighing an average 94g (4 weeks old) were fed either a regular diet (RE) or a high fat diet (HF), and the HF group was subdivided into a Ma-huang methanol extract (30mg/100g body weight) group (HF+MH). The weight of epididymal fat pad and brown adipose tissue were measured. The cell size and cell number per unit area of epididymal fat pad were investigated. Results : The yield weight of methanol extract of Ma-huang was 3.63mg per l00g of Ma-huang. The body weight gain of the HF group was similar with that of the HF+ MH but higher than that of the RE. The weights of the epididymal fat pads and brown adipose tissue of the RE group were lower than those of HF and HF+MH groups. The cell sizes and numbers per unit area of epididymal fat pads of the RE and HF+MH groups were larger than those of HF group. The cell numbers per unit area size of epididymal fat pads were the smallest in the RE group. Conclusions : It could be concluded that the Ma-huang extract has no effect on the epididymal fat pads in rats fed a high fat diet and the clinical application of Ma-huang for the treatment of obesity should be re-considered.
