Suppressive Effect of 4-Hydroxy-2-(4-Hydroxyphenethyl) Isoindoline-1,3-Dione on Ovalbumin-Induced Allergic Asthma

  • 4-Hydroxy-2-(4-hydroxyphenethyl)isoindoline-1,3-dione (PD1) is a synthetic phthalimide derivative of a marine compound. PD1 has peroxisome proliferator-activated receptor (PPAR) ${\gamma}$ agonistic and anti-inflammatory effects. This study aimed to investigate the effect of PD1 on allergic asthma using rat basophilic leukemia (RBL)-2H3 mast cells and an ovalbumin (OVA)-induced asthma mouse model. In vitro, PD1 suppressed ${\beta}$-hexosaminidase activity in RBL-2H3 cells. In the OVA-induced allergic asthma mouse model, increased inflammatory cells and elevated Th2 and Th1 cytokine levels were observed in bronchoalveolar lavage fluid (BALF) and lung tissue. PD1 administration decreased the numbers of inflammatory cells, especially eosinophils, and reduced the mRNA and protein levels of the Th2 cytokines including interleukin (IL)-4 and IL-13, in BALF and lung tissue. The severity of inflammation and mucin secretion in the lungs of PD1-treated mice was also less. These findings indicate that PD1 could be a potential compound for anti-allergic therapy.

The First Case of Capillaria hepatica Infection in a Nutria (Myocastor coypus) in Korea

  • This study reports the first case of Capillaria hepatica infection in a nutria in Korea. Ten nutrias, captured near the Nakdong River, were submitted to our laboratory for necropsy. White-yellowish nodules were found in the liver of 1 of the nutrias at necropsy. Histologically, the lesions were granulomatous, and infiltrations of lipid-laden macrophages, eosinophils, and several multinucleated giant cells were observed. The lesions consisted of numerous eggs and necrotic hepatocytes. The eggs were lemon-shaped and had polar plugs at the ends of both long sides. The eggs were morphologically identified as those of C. hepatica. Worldwide, C. hepatica infection in nutrias is very rare. Nutrias are a kind of livestock, as well as wildlife; therefore, an epidemiological study for parasitic infections needs to be conducted.

  • The immune response against Trichinella spiralis at the intestinal level depends on the $CD4^+$ T cells, which can both suppress or promote the inflammatory response through the synthesis of diverse cytokines. During the intestinal phase, the immune response is mixed (Th1/Th2) with the initial predominance of the Th1 response and the subsequent domination of Th2 response, which favor the development of intestinal pathology. In this context, the glucocorticoids (GC) are the pharmacotherapy for the intestinal inflammatory response in trichinellosis. However, its therapeutic use is limited, since studies have shown that treatment with GC suppresses the host immune system, favoring T. spiralis infection. In the search for novel pharmacological strategies that inhibit the Th1 immune response (proinflammatory) and assist the host against T. spiralis infection, recent studies showed that resiniferatoxin (RTX) had anti-inflammatory activity, which decreased the serum levels of IL-12, $INF-{\gamma}$, $IL-1{\beta}$, $TNF-{\alpha}$, NO, and $PGE_2$, as well the number of eosinophils in the blood, associated with decreased intestinal pathology and muscle parasite burden. These researches demonstrate that RTX is capable to inhibit the production of Th1 cytokines, contributing to the defense against T. spiralis infection, which places it as a new potential drug modulator of the immune response.

  • Eosinophilic gastroenteritis is a rare disease occurring especially in children, and shows various nonspecific presentations with infiltration of eosinophils in the gastrointestinal organs. The pathophysiology of eosinophilic gastroenteritis is not yet clearly known, but allergic reactions are suspected to be related with the disease. Eosinophilic gastroenteritis is categorized into the mucosal, muscularis and subserosal types based on which layer of the intestinal wall is involved. There are different clinical manifestations according to the involved layer. Most cases to date have responded well to steroid therapy. In this study, we diagnosed and treated a case of non-IgE-mediated, subserosal eosinophilic gastroenteritis in a child with abdominal pain, diarrhea and ascites.

The Reference Values on Hematologic Parameters in Clinically Normal Thoroughbred Neonatal Foals (건강한 Thoroughbred 신생망아지의 혈액상)

  • Next to performing an expert physical examination, a blood sample submitted for a complete blood count is the most basic tool available to owners or veterinary practitioners. Reference values of complete blood count were determined at 6 different ages in 114 Thoroughbred foals during the second month of life. Hematologic results were as follows: RBC 8.2-10.5 ($10^6/{\mu}l$), Hb 10.9-13.3 (g/dl), HCT 28.2-35.2 (%), MCV 30.7-35.8 (fL), MCH 11.9-13.5 (pg), MCHC 37.9-40.5 (g/dl), RDW 24.5-25.7 (%), PLT 146.3-256.4 ($10^3/{\mu}l$), MPV 6.7-8.3(fL), total WBC 8.1-12.5 ($10^3/{\mu}l$), basophils 0.0 ($10^3/{\mu}l$), eosinophils 0.0-0.3 ($10^3/{\mu}l$), neutrophils 2.4-8.6 ($10^3/{\mu}l$), lymphocytes 1.8-2.9 ($10^3/{\mu}l$) and monocytes 0.0-1.4 ($10^3/{\mu}l$). The results of this study serve as reference ranges for Thoroughbred neonatal foals populations and can be useful for health control, regular examination and pre-sale soundness examination.

Prevalence of Fascioliasis of Korean Native Cattle in Kangwon Province in Korea (강원도 사육 한우의 간질 감염실태)

  • A field survey of fascioliasis of Korean native cattle raising and raised in specialized commercial breeding farms and local farms in Kangwon province using both intradermal test and sedimentation technique for feces was carried out from November to December, 1996. Fecal samples were taken from fascioliasis positive cattle by the intradermal test for the fecal examination. Liver tissues were randomly collected from an abattoir for histopathological examination of liver fluke infection in cattle. The results are as follows. 1. By the intradermal test for a total of 211 cattle raising in both Wonju and Wheongsung, Kangwon province, 60 heads(28.4%) showed positive reaction. Among 60 positive cattle, eggs of Fasciola hepatica were found from 51 heads(85.0%) by sedimentation technique. 2. According to the cattle raising areas, the positive rates by the intradermal test were 26.7%(20 out of 75 heads) in Wonju and 29.4%(40 out of 136 heads) in Wheongsung. 3. According to the age of cattle examined, the positive rates by the intradermal test in 1~3, 4~6 and 7~10 years old were 11.7%(7 out of 100 heads), 68.3%(41 out of 93 heads) and 20.0%(12 out of 18 heads), respectively. 4. The overall infection rates of fluke larvae from the slaughtered cattle at an abattoir in Wonju was 24.7%(37 out of 150 heads). In histopathology, liver lesions were observed such as inflammation with infiltration of eosinophils, polymorphonuclear cells, mononuclear cells and multinucleated giant cells, proliferation of connective tissues, calcification and abscess formation.

Treatment of prednisolone for eosinophilic enteritis in a dog (개에서 prednisolone을 이용한 호산구성 장염 치료 증례)

  • A 7-year-old, castrated male Yorkshire terrier dog with chronic vomiting, and melena lasted for 3 weeks was referred to the Veterinary Medical Teaching Hospital of Chungnam National University. The complete blood counts and serum biochemistry revealed no remarkable findings. Gastrointestinal series revealed irregular margin in small intestinal lumen. After upper gastrointestinal series, erythema of the small intestinal lesions was found by exploratory laparotomy and full-thickness biopsy was performed. Histopathologic examination revealed infiltration of predominantly eosinophils and other inflammatory cells in small intestine. Based on these findings, the dog was diagnosed as eosinophilic enteritis. After 4 weeks medical treatment and hypoallergenic diet, the patient showed no vomiting, melena and other complications.

The Effects of Oyo-Tang(五拗湯) on the Respiratory Patterns and Tracheal Tissues in Allergic Asthma (오요탕(五拗湯)이 알레르기 천식(喘息)의 호흡(呼吸) 양상(樣相)과 기관(氣管) 조직(組織)에 미치는 영향(影響))

  • This study aimed to find out Oyo-Tang's effects on allergic asthma using the Egg-White Implant(EWI) method model recommended by Siqueira et al. Oyo-Tang has widely been prescribed and used in Oriental Medicine for controlling and curing of cough, nasal obstruction, pharyngeal pain and hoarseness. Previous studies have demonstrated that Oyo-Tang has analgesic, sedative, anticonvulsive, antitussive and antihistamine effects. But it was uncertain whether Oyo-Tang could inhibit the allergic reactions, to alleviate the symptoms of allergic asthma. With such aim. the respiratory patterns and eosinophil infiltration in the tracheal mucosa of rats with experimentally induced allergic asthma (using EWI method) were mainly observed. The results are as follows: 1. Oyo-Tang decreased the number of poor respiration with respect to control group (P<0.05) 2. Oyo-Tang decreased the number of eosinophils infiltrated in tracheal mucosa with respect control group (P<0.05). The above results demonstrate that Oyo-Tang has the inhibitory effects on the process allergic asthma and suggest that it could be used in relieving patients of the symptoms caused allergic asthma.

Effects of Benzene, Phenol and Hydroquinone on Proliferation, Differentiation and Migration of Human Eosinophilic EoL-1 Cells

  • Benzene is known as a ubiquitous air pollutant and has a carcinogenic influence on the human body. Benzene is also metabolized to other volatile organic compounds (VOCs) in the body such as phenol and hydroquinone (HQ). The metabolites are accumulated and further oxidized by myeloperoxidase in bone marrow. They act as toxic agents and cause a variety of diseases, including cancer, atopic dermatitis and asthma. In this study, we examined the effects of benzene and its metabolites on proliferation, differentiation and chemotaxis of EoL-1 cells, the human eosinophilic leukemia cell line. These chemicals had no effect on the proliferation of EoL-1 cells. Benzene decreased the differentiation of EoL-1 cells induced by butyric acid. HQ was induced the cell death during butyric acid-induced EoL-1 cell differentiation. In a chemotaxis experiment, benzene, phenol and HQ enhanced the cell migration induced by Lkn-1 but not by MCP-1, eotaxin, MIP-$1{\alpha}$ and RANTES. These findings provide the effect of VOCs on the regulation of eosinophil-involved immune response.

Increased B cell-activating factor (BAFF) level in the sputum of children with asthma

  • Purpose: B cell-activating factor (BAFF) is a tumor-necrosis factor (TNF) superfamily member best known for its role in the survival and maturation of B cells. BAFF activity is observed in naive cells as well as in effector/memory T cells. We aimed to explore whether BAFF in sputum is expressed at elevated levels in asthmatic airways and associated with eosinophilic inflammation, pulmonary function, and bronchial hyperresponsiveness in children. Methods: One hundred and fifty-four asthmatic children and 98 healthy children were enrolled in the study. Sputum supernatants were collected and sputum BAFF and eosinophil cationic protein (ECP) levels were measured. We performed pulmonary function tests and methacholine challenge tests, while measuring total eosinophil count, total serum IgE, and serum ECP in all subjects. Results: Asthmatic children had significantly higher levels of BAFF in induced sputum [26.50 (10.50-100.27) pg/mL] compared to healthy children [18.32 (7.68-44.63) pg/mL; $P$=0.011]. Sputum BAFF positively correlated with sputum eosinophils (${\gamma}$=0.406, $P$<0.001) and sputum ECP (${\gamma}$=0.789, $P$<0.001). Significant negative correlations were found between sputum BAFF and FEV1 (${\gamma}$=-0.291, $P$<0.001) or post-bronchodilator FEV1 (${\gamma}$=-0.334, $P$<0.001), whereas nonsignificant correlations were found between sputum BAFF and bronchial hyperresponsiveness, serum eosinophil count, and serum ECP. Conclusion: These findings suggest that BAFF may play a role in childhood asthma, and BAFF levels in sputum could be a supportive marker that represents airway inflammation, especially eosinophilic inflammation.
