참기름 제조의 부산물인 참깨박의 활용도를 높이기 위하여 효소 처리에 의한 참깨박 불용성 단백질의 추출 조건을 조사하였다. 단백질 분해효소인 Alcalase, Flavourzyme, Neutrase, Protamex의 처리 결과를 대조구와 비교한 결과 Protamex가 고형분과 단백질 함량 증가에 효과적이었다. Protamex의 반응 조건(50 ℃, pH 6.0)에서 효소의 사용량은 탈지 참깨박의 1%, 효소반응시간은 3시간이 적당하였다. 효소 처리 효율을 향상시키기 위하여 참깨박을 열처리하면, 열처리 온도가 증가할수록 추출되는 단백질 함량은 증가하였으며 110이상에서는 미미하게 증가하였다. 세포벽 분해효소(Tunicase)와 단백질 분해효소의 병용처리가 단백질 가용화에 미치는 효과를 조사한 결과, 단백질 분해효소를 처리한 다음에 세포벽 분해효소를 처리하는 것이 가장 효과적이었다. 열처리(110 ℃, 10분) 후 Protamex와 Tunicase를 순차적인 처리로 단백질 함량이 열처리와 효소처리하지 않은 대조구의 약 3.6배(9.85→35.58 mg/mL), 열처리만 실시한 대조구의 약 2.2배(15.83→35.58 mg/mL) 증가하였다.
The influences of protease on the removal of various protein soils from cotton fabrics were studied. The human epidermal stratum corneum, hemoglobin and casein were used as protein soils. The soiled fabrics were denatured by steaming for 30 min. before washing and laundered using Terg-O-Tometer under washing conditions. The removal efficiency was evaluated by analysis of protein on the fabrics before and after washing by means of copper-Folin method. The relations between the removal and the characteristics of protease were discussed. Also the degradation of protein were examined by microscopy. The seperation of human epidermal stratum corneum after hydrolysis was examined by SDS-PAGE. The results obtained were as follow : 1. The protein from the soiled cotton fabric was removed effectively by adding protease. The removal of protein was increased in proportion to increasing of the enzyme concentration up to a certain point, but it began to decrease above the point. The removal effect was high in the order of casein>human epidermal stratum corneum>hemoglobin. Especially the protein was more effectively removed in ADS solution(pH 9.5) containing enzyme. 2. When protease was used with ADS. the removal of protein was efficiently showed in relatively short time(5~15min.) compared to using ADS only. It is due to the properties of this enzyme that reacts with very short time. 3. Even at low temperature the removal efficiency of enzyme was relatively higher compared with the activity of enzyme. The removal of protein soil was increased up to a maximum near $50^{\circ}C$, and then decreased. 4. The removal of protein by protease was improved with the increase of alkalinity in the pH range from 9.5 to 11.0 but it began to decrease above pH 11.0. 5. According to the increase of mechanical agitation, the removal effect was increased. But the removal efficiency of protease was more effective compared with the agitation in detergency. 6. According to the SDS-PAGE separation and micrograph it was confirmed that the human epidermal corneum was effectively hydrolysed by the enzyme added. So the fragments of protein were removed more efficiently by means of the interfacial reaction of AOS.
In this work, colorants extraction process from gromwell was studied for making powder form of colorants by solving the high viscosity problem of gromwell extracts. In order to do that, sugar extracted together with colorants must be pre-extracted. For sugar decomposition, gromwell roots were pretreated with various enzyme solutions. The total sugar content of pre-extract with enzyme solution was measured. Accordingly, the effects of enzyme type and pretreatment condition on sugar decomposition were investigated to find appropriate enzyme(amylase, hemicellulase, pectinase) and enzyme activity (100~1000unit), pre-extracted time(3~24hr). Color characteristics and dye uptake of dyed fabrics were evaluated. Gromwell colorants were assessed for their potential antimicrobial activities, which possibly expand their end use as functional pigments. The efficiency of removing sugar was increased in the order of hemicellulase, pectinase, amylase, $H_2O$. Gromwell colorants powder yield was in the range of 4.4% to 9.8% depending on pretreatment enzyme. Gromwell colorants produced RP color on the silk and wool fabrics with good dye uptake. Antimicrobial activity of gromwell colorants will greatly increase its potentiality for applying as functional natural colorants in the future.
Deinking is a series of unit operations designed to detach ink from cellulose fibers and separate the dispersed ink from the pulp slurry. Deinking chemicals are process aids that enable expensive mill equipment used in these unit operations to operate more efficiently - often much more efficiently. We propose the blended deinking agent with cellulolutic enzymes and synthetic collector in deinking pulp of conventional alkaline method. The deinking efficiency of old news print in alkaline pH was enhanced with enzyme treatments. The brightness of deinked pulp was increased with less residual ink particles and yield of enzymatic deinked pulp was improved compared to the deinked pulp of conventional alkaline method. Enzymes in biomass were use to Chemical Deinking for reduce environment pollution through surfactant and improve surfactants. examining into compatibility Enzymes and surfactants, these new materials are studied efficiency of deinking efficiency.
The effect of solubility and hygroscopicity of some tablet diluents on the disintegration of enzyme tablets was investigated. Tablets were prepared by direct compression method using sodium starch glycolate, crospovidone, croscarmellose sodium and low-substituted hydroxypropyl cellulose as super disintegrants. Lactose, dextrose, sucrose, sorbitol and calcium phosphate dibasic were selected as typical diluents in this study. They were different in solubility (sucrose, sorbitol>dextrose>dextrose>lactose>calcium phosphate dibasic) and hygroscopicity (sorbitol>sucrose>dextrose>caicium phosphate dibasic, lactose). The disintegrants accelerated differently the disintegration of the tablets prepared with different diluents in the decreasing order of calcium phosphate dibasic>lactose>dextrose>sucrose and sorbitol. These results indicate highly soluble and/or hygroscopic diluents decrease the efficiencies of super disintegrants in the enzyme tablets.
IPSKCDNA gene(1.8 kbp) encoding rat brain IP3K enzyme contained Not I restric site in open reading frame. The Not I sequence, GCGGCCGC, was converted to GCAGCCGC by site-directed mutagenesis. The mutated IP3KcDNA was digested with EcoR I and ligated with EcoR I-restricted psp72·Not2 vector. The resulting psp72 · Not2-IP3KCDNA was digested with the Not I restriction enzyme and then subcloned into the Not I -digested PZIP · NeoSV(X) mammalian expression vector. The PZIP · NeoSV(X) -IPSKCDNA was transfected into CCL39 hamster lung fibroblast cells. The efficiency of the expressed IPSKCDNA gene was significantly higher than expected generally, not only a mean 5-fold increase in the amount of enzyme, but also 16-fold increase in enzyme activity from tractsfected CCL39 cells by the method of Western blot using anti-lP3K antibodies. Both distribution of IPSK in various rat tissues and biochemical properties were discussed.
For the purpose of evaluating the cleansing efficiency against Candida albicans detected frequently in patients with denture stomatitis, two denture cleansers with or without enzymes were studied under the same conditions. The results were as fellows: 1. Enzyme-contain denture cleanser was showed more Candida albicans lytic ability than non-enzyme-contained denture cleanser. 2. It was observed that Candida albicans lytic activity in further diluted manufacturerers' recommended concentration was decreased. 3. In fungicidal test, the enzyme-contained denture cleanser sterilized Candida albicans, and the non-enzyme-contained denture cleanser did not sterilize Candida albicans. 4. Sterilizing time of Candid albicans was needed for at least 60 minutes in enzyme-contained denture cleanser solution which was diluted with manufacturerers' recommended concentrations., and was needed for more times with further diluted manufacturerers' recommended concentrations. 5. In vitro growth test of Candida albicans on acrylic resin surface, the only enzyme-contained denture cleanser inhibited growth of Candida albicans, and it was observed that inhibiton ability of growth of Candida albicans on arrylic resin surface was decreased in further diluted manufacturerers' recommended concentrations.
Antioxidant enzymes such as catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) are known to inhibit oxidative reactions by incativating compounds responsible for the formation of ree radicals. SOD transforms superoxide radical into hydrogen peroxide which is precursor to active free radicals. CAT reduces hydrogen peroxide to water. GSH-Px reduces hydroperoxides to corresponding alcohols. Antioxidant enzyme activities of muscle are different by animal species age, stress and exercise, muscle type and part, conditions of post mortem, storage and processing which are related to oxidative deterioration I muscle foods as well as oxidative defence in living systems. Antioxidant enzyme systems are enhanced rather than weakened in aging skeletal muscle. Red muscle contains higher antioxidant enzyme activity than white muscle. The antioxidant enzyme activities of poultry are higher in leg than in breast, and those of beef are higher in redder and more unstable muscles. It is clear that the effectiveness of the antioxidant enzyme in muscle foods seems to be influenced by meat processing operations. Both GSH-Px and CAT are inactivated by heat processing NaCl also influence the efficiency of the antioxident enzymes since its presence diminishes their catalyitc activity.
Ile-224 in I269S mutant horse liver alcohol dehydrogenase isoenzyme S (HLADH-S) was mutated to serine by site-directed mutagenesis in order to study the role of the residue in c oenzyme binding to the enzyme. The specific activity of the I269S and I224S mutant enzyme to ethanol was increased 6-fold and all Michaelis constants($K_a,\;K_b,\;K_p,\;and\;K_q$,/TEX>) were larger than those for the wild-type and I269S enzyme. The substitution decreased the afffinity to coenzymes and increased the specific activity of the enzyme. The mutant enzyme showed the highest catalytic efficiency for octanol among the primary alcohols. But it didn`t have activities on retinoids and 5${\beta}$-cholanic acid-3-one. From these results, it was confirmed that the hydrophobic interaction of Ile-224 residue with coenzyme was related to coenzyme affinity in ADH reaction. The substitution also affected the substrate affinities to the enzyme.
본 연구는 목재부후균 유래 $\beta$-글루코시다제, 엑소글루카나제 및 엔도글루카나제 유전자를 함유한 형질전환체 효모를 배양한 액상섬유소분해효소에 대하여 인공반추위 배양시험을 통해 완전혼합사료에 대한 적용 수준 조사와 더불어 한우 비육 거세우의 사양시험을 통해 2 종류의 액상섬유소분해효소를 완전혼합사료에 1.0% 수준으로 첨가 급여하여 가축의 성장, 영양소소화율에 미치는 영향을 알아보고자 실시하였다. 1. 인공반추위 배양시험을 통해 완전혼합사료에 대한 액상 섬유소분해효소 첨가에 의해 pH가 약간 증가하는 경향이었다. 휘발성지방산에서 acetate 비율이 배양 12와 24시간에 유의적으로(p<0.05) 높게 나타났으며 총휘발성지방산 농도 역시 배양 12와 24시간에 유의적으로(p<0.05) 증가시켰으며 1.0% 첨가구에서 효과적이었다. 건물소화율은 배양 24시간에 액상 섬유소분해효소 첨가구에서 유의적으로(p<0.05) 증가하였다. 2. 한우 거세우 사양시험에서는 액상 섬유소분해효소 급여에 의해 일당증체량과 사료효율이 개선되는 경향이었다. 영양소 소화율에서는 DM, 조섬유, NDF 및 ADF 소화율이 액상 섬유소분해효소 첨가에 의해 유의적으로(p<0.05) 증가하였다. 따라서 인공반추위 배양시험을 통해 액상 섬유소분해효소 1.0% 첨가가 비교적 반추위발효 양상을 개선시키었으며, 한우 사양시험에서도 2종류의 액상 섬유소분해효소 1.0% 급여가 일당증체량 및 사료효율 개선효과가 있었다.
