Changes of Indicator Microorganisms and Pathogenic Bacteria in Spinach during Cook-Chill Process (시금치의 cook-chill 가공 중 오염지표균 및 병원성세균의 변화)

  • Spinach minimally processed using cook-chill and sous vide techniques was vacuum-packed in low gas permeable plastic film, pasteurized at $70^{\circ}C$ for 2 min, cooled rapidly at $3^{\circ}C$, and stored at 3 and $10^{\circ}C$. Contents of mesophilic bacteria, psychrophilic bacteria, anaerobic bacteria, spore-forming bacteria, total coliforms, yeast and molds, fecal Streptococcus, and Enterobacteriacea were measared to identify the degree of food contamination. Number of mesophilic bacteria, detected at $2.2{\times}10^8\;cfu/g$ in raw spinish, decreased to about $6.0{\times}10^3\;cfu/g$ after cook-chill process. During the storage at 3 or $10^{\circ}C$, levels of mesophilic, psychrophilic and anaerobic bacteria increased, whereas total coliforms, yeast and molds, fecal Streptococcus, and Enterobacteriacea were not detected. Twelve strains of Aeromonas hydphila, Escherichia coli O157:H7, Plesiomonas shigelloides, Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Yersinia enterocolitica, Bacillus cereus, Campylococcus spp., Clostridium perfringens, Listeria monocytogenes, and Staphylococcus aureus were examined for detecting the presence of pathogenic bacteria in spinach. B. cereus and C. perfringens were isolated from raw, washed, and cook-chilled spinach, whereas A. hydrophila was isolated only from washed spinach. S. aureus was isolated from raw and washed spinach, but not from cook-chilled spinach. Other pathogenic organisms were not detected in raw, washed, and cook-chilled spinach.

  • Frog culture industry is not yet familiar but has much potential. Generally, in farm, the population density is higher than that of in nature and frog farm is not the exception. But when population density is high, it can easily leads to stressful condition, poor sanitation. When a disease occur, it is a primary factor that makes the population more susceptible and the results more grave. Because of severe Rhabditoidea- helminth infection and subsequent bacterial septicemia, 50~70% of the total population had been died in a farm in Jeong-sun in Gangwon-do and Chungju in Chungcheongbuk-do from late June, 2012 to September, 2012. Diseased frogs showed ruptured lung, bloody ascites, liver discoloration, myocardium weakness, congested kidney, microcytic anemia and so on. Enterobacteriacea, Citrobacter.sp, Cupriavidus metallidurans, Acinetobacter.sp were isolated as major bacterium that had caused septicemia in frogs. Among isolated bacterium, Cupriavidus metallidurans, Ewingella americana, Shewanella aquimarina and Pseudoalteromonas sp. have not reported as potential pathogens in frogs before. It is a good example that severe helminth infection in frogs can lead to secondary infection of bacteria.

  • Seasonal distribution of various microorganisms was investigated in the effluent water in various sewage treatment plants from January to November in 2007. The target microorganisms were composed of total heterotrophic bacteria, Escherichia coli, coliform and enteric bacteria, Staphylococcus aureus, Yeast and Mold. The number of total bacteria and the indicator microorganisms, such as E. coli, coliform, and Enterobacteriacea, were increased during the summer season. S. aureus, however, was not influenced by the weather, showing higher number even in November and January in most sewage treatment plants investigated. Meanwhile, disinfection methods employed in various plants had no direct relationship with the growth of microorganisms, and small-scaled plants were found to be more effective than large-scaled ones in the sterilization of microorganisms.

Characterization of plasmid-mediated quinolone resistance genes in Enterobacteriaceae isolated from companion animals (반려동물 유래 장내세균에서 plasmid 매개 퀴놀론 내성 유전자의 특성)

  • The aim of this study was to investigate the prevalence and characterization of plasmid-mediated quinolone resistance (PMQR) gene in 79 Enterobacteriaceae isolated from dogs and cats. Of 79 isolates, PMQR genes were found in 10 (12.7%) isolates, including aac(6')-lb-cr, qnrB, qnrS and qnrA detected alone or in combination in 8 (10.1%), 4 (5.1%), 2 (2.5%) and 1 (1.3%) isolates, respectively. Interestingly, two qnrS genes were detected in nalidixic acid and ciprofloxacin susceptible isolates. Extended-spectrum ${\beta}$-lactamase (ESBL) was detected in 90% (9 isolates) of PMQR positives isolates. Among ESBL genes, CTX-M, TEM and SHV were detected in 9, 8 and 3 isolates, respectively. Almost all PMQR genes were detected in co-existence with ESBL genes. All PMQR positives isolates were multidrug resistance (i.e. resistant to five or more antibiotics). qepA, OXA and CMY-2 genes were not found. The six transconjugants were obtained by conjugation experiment. The aac(6')-lb-cr, qnrB and qnrS were co-transferred with CTX-M, TEM and/or SHV, whereas qnrA was not observed among transconugants. This is the first report of the presence of aac(6')-lb-cr and qnrA gene among Enterobacteriaceae isolates from dogs in Korea. The prudent use of antimicrobials and continuous monitoring for companion animals are required.
