Preparation and Characterization of Hyaluronic Acid Loaded PLGA Scaffold by Emulsion Freeze-Drying Method (히알루론산을 함유한 PLGA 지지체의 제조 및 특성결정)

  • Poly(lactide-co-glycolide)(PLGA) and hyaluronic acid (HA) has been widely used as biocompatible scaffold materials to regenerate tissue. In this present study, we fabricated microporous PLGA and HA loaded PLGA scaffolds by a emusion freeze-drying method. In order to confirm that the release profile of cytokine or water-soluble drugs, we manufactured the granulocyte macrophage colony stimulating factor(GM-CSF) loaded PLGA and HA-PLGA scaffold. All scaffolds were characterized using scanning electron microscope(SEM), mercury porosimeter and wettability measurement. Cell proliferation and viability were assessed by a 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide (MTT) test. The porosity of HA-PLGA scaffold was greater than 95% with the total pore area of $261\;m^2/g$. The HA-FLGA scaffold exhibited well interconnected pores to allow greater cell adhesion and prolixferation. It was proven by higher cell viability in the HA-PLGA scaffold than PLGA alone. This may be due to the enhanced natural properties and higher water retention capacity of HA.

Preparation of $ZrO_2/Al_2O_3-Mullite$ Composites Using the Silica Sol Infiltration Method (실리카 졸 침투법을 이용한 $ZrO_2/Al_2O_3-Mullite$ 복합체의 제조)

  • ZrO2/Al2O3-Mullite composites were prepared by infiltration of the silica sol to the porous ZrO2/Al2O3 bodies. The porous ZrO2/Al2O3 bodies for infiltration were fabricated using ZrO2 (20wt%)/Al2O3 composite powders synthesized by the emulsion-hot kerosene drying method. The preparation of silica sols was conducted by the hydrolysis-peptization of an alcoholic TEOS solution. When ZrO2/Al2O3-Mullite and ZrO2/Al2O3 composites were sintered at 1$650^{\circ}C$ for 4 hrs, both of them showed an excellent sinterability. As the amount of mullite added in the composites increased, the ratio of the tetragonal phase of zirconia to the monoclinic phase at the room temperature became higher. It was known that values of the fracture toughness of the ZrO2/Al2O3-Mullite composites were about 5.48 MPa.m1/2 much larger than that of the ZrO2/Al2O3 system.

Preparation of Cefaclor-Containing Gelatin Microcapsules and Their Drug Release Characteristics (수용성 약물인 세파클러를 함유하는 젤라틴 마이크로캅셀의 제조 및 약물 방출특성)

  • In order to formulate a controlled release system for oral drug delivery, the microcapsules were prepared in w/o emulsion containing cefaclor as a water-soluble model drug by th e method of interfacial polycondensation. Gelatin wis selected as a suitable polymer for interfacial polycondensation. Gelatin solution containing drug was emulsified in an organic phase under mechanical stirring. After emulsification, terephthaloyl chloride was added as cross linking agent, followed by mechanical stirring, washing and drying. Physical characteristics of microcapsules were investigated by optical microscopy, scanning electron microscopy and particle size analysis. Mean particle sizes of gelatin microcapsules were, in the range, of about 20~50 ${\mu}$m. The microcapsules were in good apperance with spherical shapes before washing, but were destroyed partially after washing and drying, even though some microcapsules were still maintained in their shapes. Contents of cefaclor in the microcapsules were calculated by UV spectrophotometry after 3 days extraction with pH 4 carbonate buffer solution. The effects of cross linking time. pH. concentration of cross-linking agent, and temperature on drug release kinetics have been discussed extensively.

  • Al2O3-20 wt% ZrO2 composite powders to be used as the starting materials of the Al2O3/ZrO2-Spinel composite system were prepared by the use of the emulsion-hot kerosene drying method. The crystalline phase of ZrO2 in the synthesized Al2O3-ZrO2 composite powders was 100% tetragonal but the small amount of t-ZrO2 was transformed into m-ZrO2 after crushing. The hardness, fracture toughness, and flexural strength of the composite, which was sintered at 1650$^{\circ}C$ for 4 hrs after calcining at 1100$^{\circ}C$ for 2 hrs and had the relative density of 99%, were 15.7 GPa, 4.97 MN/m3/2, and 390 MPa, respectively. The fracture form in the sintered composites was found to be the intergranular fracture.

  • Poly(${\varepsilon}-caprolacton$)/ethyl cellulose (PCL/EC) microcapsules containing pluronic F127 were prepared by a spray drying method. The aqueous phase, 20% of pluronic F127 was dissolved in distilled water, and the organic phase, 5% of PCL and EC were dissolved in dichloromethane. The microcapsules were obtained by spray drying the water-in-oil (W/O) emulsion. According to the data of scanning electron microscopy and particle analyzer, tens of micro size microcapsules were observed. On a differential scanning calorimeter, the phase transition temperatures of microcapsules were observed and they were found around those of pluronic F127 and poly(${\varepsilon}-caprolacton$), which were the main components of the microcapsules. At the range of $30{\sim}45^{\circ}C$, temperature-dependent release properties were investigated using fluorescein isothicyanate-dextran (FITC-dextran) and blue dextran as a model drug. When the temperature was increased, the degree of release of microcapsule was also increased. FITC-dextran, the relative low molecular weight, was more released than blue-dextran.

  • We developed the nerve growth factor (NGF) loaded poly (L - lactide) (PLA) scaffolds by means of emulsion freeze drying method to the possibility for the application of the nerve regeneration of spinal cord disease and the degeneration in Alzheimer's disease. The release amount of NGF from NGF loaded PLA scaffold were analyzed over a 4 week period in vitro at phosphate buffered saline (PBS), pH 7.4, at $37^{\circ}C$. It can be observed the open cell pore structure of porous scaffolds and can be easily controlled the pore structure by the controlling of formulation factors resulting in the controlling of the release rate and the release period. The stability of NGF during the preparation of PLA scaffold was evaluated by comparing the released amounts of total NGF, assayed NGF enzyme - linked immunosorbent assay (ELISA). Released NGF has been found to enhance the neurite sprouting and outgrowth from pheochromocytoma (PC-12) cells. These results suggest that the released NGF from NGF loaded PLA scaffold such as conduit type can be very useful for the nerve regeneration in the neural tissue engineering area.

  • The tendency of the miximum brightness of $Y_2$O$_3$:Eu phosphor with various activator concentration, by different surface treatment methods as well as different exciting energies were investigated. The surface treatment methods were the adsorption method used $\alpha$-Fe$_2$O$_3$powder prepared emulsion-drying process and the precipitation method used FeSO$_4$/ethanol. Eu concentration of maximum brightness of $Y_2$O$_3$:Eu phosphor prepared by solid-solid state was changed with various exciting energies. The concentrations were 0.02 mol at VUV(147 nm) as well as 400 V and 0.03 mol at 5 kV. The phosphor treated both by adsorption method and precipitation method showed decreasing luminescent intensity with increasing amount of $\alpha$-Fe$_2$O$_3$, and the methods are chosen by exciting energy. Adsorption method was effective in a low voltage and VUV(147nm) region, and precipitation method was effective in the high voltage region.

  • In the field of makeup cosmetics, especially, powder-based foundations such as two-way cake, pact and face powder, the quality of which is known to be strongly influenced by the properties of powder, surface treatment technology is widely used as a method to improve the various characteristics of powder texture, wear properties, dispersion ability and so on. The two-way cake or pressed-powder foundation is one of the familiar makeup products in Asian market for deep covering and finishing purpose. In spite of the relent progress in surface modification method such as composition of powders with different characteristics and application of a diversity of coating ingredient (metal soap, amino acid, silicone and fluorine), this product possess a technical difficulty to enhance both of the adhesion power and spreadability on the skin in addition to potential claim of consumer about heavy or thick feeling. This article is covering the preparation and coating method of nano-vesicle that mimic the double-layered lipid lamellar structure existing between the corneocytes of the stratum corneum in the skin for the purpose of improving both of two important physical characteristic of two-way cake, spreadability and adhering force to skin, and obtining better affinity to skin. Nano-vesicle was prepared using the high-pressure emulsifying process of lecithin, pseudo ceramide, butylene glycol and tocopheryl acetate. This nano-sized emulsion was added to powder-dispersed aqueous phase together with bivalent metal salt solution and then the filtering and drying procedure was followed to yield the nano-vesicle coated powder. The amount of nano-vesicle coated on the powder was able to regulated by the concentration of metal salt and this novel powder showed the lower friction coefficient, more uniform condition of application and higher adhesive powder comparing with the alkyl silane treated powder from the test result of spreadability and wear properties using friction meter and air jet method. Two-wav cake containing newly developed coated powder with nano-vesicle showed the similar advantages in the frictional and adhesive characteristics.
