체세포복제수정란 이식에 의한 젖소 수란우의 유도분만 (Induction of Parturition after Transfer of a Hanwoo-Somatic Cell Cloned Embryo in Holstein Cow)

  • 한우 태아 섬유아세포 유래 체세포복제수정란 이식으로 임신된 수란우가 분만예정일이 도래하였으나 분만징후가 없어 수정란이식 후 272일째에 dexamethasone 20 mg을 근육주사하고 24시간후 PG $F_{2a}$ 25 mg과 estradiol 20 mg을 근육주사하여 분만유도를 실시하였다. dexamethasone 투여후 48시간만 분만징후를 나타내었으며, dexamethasone 투여후 50시간째에 40 kg의 수송아지를 견인에 의해 정상적으로 분만시켰다. 그러나 수란우의 후산은 정상적으로 배출되지 않았다.다.

난포액내 정자유인물질의 분석 (Analysis of Sperm Chemoattractant in Follicular Fluid)

  • Among proteins separated from methanol extract of follicular fluid with superose column, the components inducing sperm swim-up separation through sucrose layer were analysed with superose column in Smart system and SDS-PAGE. And the results obtained were as follows; The fractions of retention volume (RV) 0.83ml and RV 1.36ml separated with superose column should stimulate sperm migration and movement. However, RV 0.83 fraction was consisted of complex materials containing RV 1.36 component. RV 1.36 fraction contained a BSA analogue of 67 kilodaltons (Kd) and showed identical peak pattern with BSA fraction V. In conclusion, the protein of 67 Kd in follicular fluid should stimulate sperm migration and movement.

Reproductive outcomes of retransferring retained embryos in blastocyst transfer cycles

  • Objective: To determine the incidence of embryo retention (ER) in the transfer catheter following embryo transfer (ET) in blastocyst transfer and investigate whether retransferring retained embryos has an impact on reproductive outcomes in patients undergoing in vitro fertilization-ET. Methods: We retrospectively analyzed the records of 1,131 blastocyst transfers, which comprised 223 single blastocyst transfer (SBT) and 908 double blastocyst transfer (DBT) cycles. Each SBT and DBT group was classified depending on whether ET was performed without retained embryos in the catheter during the first attempt (without-ER group) or whether any retained embryos were found following ET (ER group) for the purpose of comparing reproductive outcomes in a homogenous population. Results: The overall incidence of finding retained embryos was 2.8% (32/1,131). There were no retained embryos in SBT cycles. In DBT cycles, implantation rates (30.0% vs. 26.6%), positive ${\beta}-hCG$ rates (57.2% vs. 56.2%), clinical pregnancy rates (45.3% vs. 46.9%), and live birth rates (38.9% vs. 43.8%) were not significantly different between the without-ER and ER groups. There were no significant differences in the mean birth weight (g) $2,928.4{\pm}631.8$ vs. $2,948.7{\pm}497.8$ and the mean gestational age at birth ($269.3{\pm}17.2days$ vs. $264.2{\pm}25.7days$). A total of nine cases of congenital birth defects were found in this study population. Eight were observed in the without-ER group and one in the ER group. Conclusion: Our results suggest that retransfer of retained embryos does not have any adverse impact on reproductive outcomes in blastocyst transfer cycles. Furthermore, our results support finding that SBT might be advantageous for decreasing the incidence of retained embryos in catheters.

Reproductive Disorders that Limits the Reproductive Performances in Dairy Cows of Bangladesh

  • The reproductive disorders are the major causes of reproductive infertility in cows that affect the total annual calf crop, resulting in great economic loss in Bangladesh. The aim of the study was to find out the reproductive disorders (RD) in dairy cows that markedly influences the reproductive performances in aspect of Bangladesh. A total number of 1658 dairy cows were selected according to their body condition score (BCS) in different farms at the southern part of Bangladesh during the period of 2011 to 2012. The preliminary data (basic information) were collected directly from the dairy farmer's record books and asking questions according to a prescribed questionnaires as well as the diagnosis of RD was presumptively confirmed on the basis of history, clinical signs and examination of animals by ultrasonography and others necessary tools. There are thirteen major reproductive disorders were identified. Overall prevalence of reproductive disorders at that area were 23%, among of these anoestrus 5.1%, repeat breeder 3.7%, metritis 4.4%, poor heat detection 1.6%, ovarian cyst 0.36%, retain placenta 4.6%, dystocia 0.97% and pyometra 0.24%. It is indicated that anoestrus and retention of placenta after calving was most hazardous cause of infertility whereas the metritis and repeat breeder were the second line of consequence. RD had shown significantly higher incidence in low BCS (${\leq}2$) than that of fair (2.5) and very good (${\geq}3{\sim}3.5$). In conclusion, the highest RD especially anoestrus and retention of placenta is very alarming for reproductive loss which might be needed further research to identify the specific cause of these disorders for establishment a profitable dairying and dairy population.

돼지 난포액내 난구세포 난자복합체 팽창 억제 성분 (Cumulus Oocyte Complex Expansion Inhibiting Ingredient in Porcine Follicular Fluid)

  • The objective of this study was to identify a follicular fluid ingredient inhibiting the cumulus oocyte complex (COC) expansion. Thus, follicular fluid or liquid chromatographic fractions of follicular fluid was supplemented in COC culture medium. And COCs were incubated for 48 hours to investigate about cumulus expansion and also the first polar body extrusion. The results obtained were as follows; 1. The fluid of medium follicle significantly inhibited the COC expansion. 2. The fluid of large follicle inhibited the COC expansion. 3. Follicular fluid showed six major fractions at retention volumes (RVs) 1.83, 1.91, 2.15, 2.34, 2.53 and 2.74 ml after separation with Superose 12 column. Of the major fractions, fractions RV2.15, RV2.34, RV2.53 and RV2.74 inhibited both COC expansion and polar body extrusion. Especially, fractions of RV2.15 and RV2.53 significantly inhibited COC expansion, oocyte denudation and polar body extrusion. In conclusion, porcine follicular fluid contained a COC expansion inhibiting ingredient (CEI) that may be contained largely in fractions RV2.15 and RV2.53. And CEI may inhibit oocyte maturation by inhibition of oocyte denudation and extrusion of the first polar body.

생쥐 난자의 제2극체 방출을 유발하는 정자 성분 (Sperm Component Inducing 2nd Polar Body Extrusion in Mouse Oocytes)

  • This study was carried out to elucidate whether sperm contain a factor inducing second polar body extrusion and to search for an effective collection method of the sperm factor Thus, sperm extract, dialyzed sperm-extract or liquid chromatographic fractions of sperm extract was microinjected into ovulated oocytes. And the microinjected oocytes were incubated for 24 hours to investigate about the extrusion of second polar body. The results obtained were as follows; 1. Sperm extract significantly increased the second polar body extrusion. 2. Sperm extract showed five major fractions at retention volumes (RVs) 1.25, 1.37, 1.84, 2.10 and 2.67ml after separation with Superose 12 column. These sperm extract fractions did not significantly increase the second polar body extrusion. 3. Dialyzed sperm-extract significantly increased the second polar body extrusion 4. Dialyzed sperm-extract showed three maior fractions at RVs 1.88, 2.14 and 2.77ml after separation with Superose 12 column. Of these fractions, the fraction RV2.14 significantly increased the second polar body extrusion. In conclusion, sperm extract contained a factor inducing the second polar body extrusion and the factor was contained largely in fraction RV2.14 after dialysis and liquid chromatographic fractionation of sperm extract.

High Performance Liquid Chromatographic Analyses of Higenamine Enantiomers in Aconite Roots

  • The enantiomers of higenamine were directly separated by high performance liquid chromatography using a chiral stationary phase and detected by UV. The R- and S-isomers of higenamine were eluted at the retention time of 22 min and 27 min, respectively. Higenamine was determined to be present as R-(+)-enantiomer not only in the embryo of Nelumbo nucifera, from which the separation of R-(+)-higenamine was reported, but also in various Aconite roots, from which higenamine was separated as optically inert racemic mixtures.

In Vitro Differentiation-induced hES Cells Relieve Symptomatic Motor Behavior of PD Animal Model

  • Human embryonic stem (hES) cells can be induced to differentiate into tyrosine hydroxylase expressing (TH+) cells that may serve as an alternative for cell replacement therapy for Parkinson's disease (PD). To examine in vitro differentiation of hES (MB03, registered in NIH) cells into TH+ cells, hES cells were induced to differentiate according to the 4-/4+ protocol using retinoic acid (RA), ascorbic acid (AA), and/or lithium chloride (LiCl) followed by culture in N2 medium for 14 days, during which time the differentiation occurs. Immunocytochemical stainings of the cells revealed that approximately 21.1% of cells treated with RA plus AA expressed TH protein that is higher than the ratio of TH+ cells seen in any other treatment groups (RA, RA+LiCl or RA+AA+LiCl). In order to see the differentiation pattern in vivo and the ability of in vitro differentiation-induced cells in easing symptomatic motor function of PD animal model, cells (2 $\times$ 10$^{5}$ cells/2${mu}ell$) undergone 4-/4+ protocol using RA plus AA without any further treatment were transplanted into unilateral striatum of MPTP-lesioned PD animal model (C57BL/6). Following the surgery, motor behavior of the animals was examined by measuring the retention time on an accelerating rotar-rod far next 10 weeks. No significant differences in retention time of the animals were noticed until 2 weeks post-graft; however, it increased markedly at 6 weeks and 10 weeks time point after the surgery. Immunohistochemical studies confirmed that a reasonable number of TH+ cells were found at the graft site as well as other remote sites, showing the migrating nature of embryonic stem cells. These results suggest that in viかo differentiated hES cells relieve symptomatic motor behavior of PD animal model and should be considered as a promising alternative for the treatment of PD.

Chromatography용 Paper, $\mu$RPC Column 및 Superose Column을 이용한 정자의 이동을 자극하는 난포액 성분의 분리 (Separation of Follicular Fluid Components Stimulating Sperm Migration with Chromatographic Paper, $=mu$RPC and Superose Columns)

  • 난포액내 함유되어 있는 단백질성분 중에서 sucrose 층으로부터 정자의 swim-up 이동을 자 극하는 성분을 분리하기 위하여 paper chromatography (PC) 및 reverse phase column (RPC) 과 superose column (SC)를 이용한 액체 chromatography의 분리효과를 조사하였던 바 결과는 다음과 같다. 1. Chromatography용 paper로 분리한 각 band 의 성분은 첨가농도가 증가할수록 정자의 이동과 운동을 자극하였으며, 특히 band 1 성분은 정자의 이동을 유의하게 증가시켰다. 그러나, 동일 첨가수준에서 bands 성분의 정자 이동과 운동자극효과는 난포액의 효과에 비하여 유의하게 낮았다. 2. $\mu$RPC를 이용 2~5mm 난포로 부터 분리한 성분중 RT3.33, RT7.00, RTl3.87 및 RTl6.6A 성분은 정자의 이동을 자극하였으나, 자극효과는 매우 적었다. 3. $\mu$RPC를 이용 10mm 난포로 부터 분리한 성분은 정자의 이동과 활력을 자극하지 않았다. 4. SC를 이용 2-5mm 난포로 부터 분리한 성분 중 RVI.35 성분과 RV0.82 성분은 정자의 이동과 운동을 유의하게 자극하였다. 결론적으로 난포액내 정자의 이동과 운동을 자극하는 단백질 성분은 superose column을 이용하여 효과적으로 분리할 수 있으며, 분리된 RVI.35 성분과 RV0.82 성분은 정자의 swim-up 분리를 자극하였다.

Green Light-emitting Diodes Light Stimuli during Incubation Enhances Posthatch Growth without Disrupting Normal Eye Development of Broiler Embryos and Hatchlings

  • Monochromatic green light-emitting diodes (LED) light stimuli influences the posthatch growth performance of chicks. This study was undertaken with the following objectives: i) to examine whether the green LED light stimuli induces an overheating effect by determining weight loss rate of fertile eggs during incubation period; ii) to look for the development of eyes and other primary organs at different ages of embryos and newly hatched chicks. Arbor Acres fertile broiler eggs (n = 480) were randomly assigned to 3 incubation groups and exposed to continuous white light, green light, or a dark environment (control) from the first day to 19 d of incubation. The light sourced from LED lamps with the intensity of 30 lx at eggshell level. The results showed that either green or white light stimuli during incubation did not significantly affect the weight loss rate of fertile eggs, hatching time, hatchability, chick embryo, or body weight (BW), the weight percentage of heart, liver, and eyes, as well as obvious systematic abnormalities in eye weight, side-to-side, back-to-front, or corneal diameter from 15 d of embryogenesis to 6 d of posthatch (p>0.05). Compared with the dark condition, green light stimuli during incubation tended to increase feed intake (p = 0.080), improved the BW gain of chicks during 0 to 6 day posthatch (p<0.05), and increased the percentage of pectoral muscle to the BW on 3- and 6-day-old chicks. In addition, embryos or chicks in green light had lower weight percentage of yolk retention on 19 d of embryogenesis and 1 d of posthatch in comparison to those in dark or white group (p<0.05). These results suggest that providing 30 lx green LED light stimuli during incubation has no detrimental effect on the development of eyes, heart and liver of embryos and hatchlings, but does have potential benefits in terms of enhancement of the chick growth during the early posthatch stages. In addition, the fertile broiler eggs stimulated with 30 lx green LED light during incubation does not cause an overheating effect.
