Effects of Feeding Betaine on Performance and Hormonal Secretion in Laying Hens

  • The effects of dietary betaine on performance, blood compositions, hepatic amino acid concentrations and hormonal secretions were examined in laying hens. Egg production was significantly higher in birds fed the 16.5 % protein diet compared to those fed 14.5 % protein diet(p<0.05), whereas dietary supplementation of betaine did not show any significant effect. The high level of protein and betaine supplementation significantly improved egg weight, egg mass and feed conversion(p<0.05), while eggshell breaking strength, eggshell thickness and Haugh unit were not influenced by betaine and dietary protein levels. Supplemental betaine did not affect serum total protein, albumin and BUN concentration. However, uric acid concentration significantly increased in 600 ppm betaine-fed groups(p<0.05). Concentrations of most hepatic amino acid were influenced by increased protein feeding and dietary betaine supplementation. Hormone studies recorded significantly higher serum and hepatocyte IGF-I concentration in 600 and 1,200 ppm betaine treatments(p<0.05) compared to those of control group. IGF-I mRNA gene expression of hepatocytes revealed statistically correlated increase in 600 and 1,200 ppm betaine-fed groups compared to the controls(p<0.05). Serum IGFBP-3 concentration was significantly elevated in 600 ppm betaine treatments. However, the secretion of IGFBP-1 in hepatocyte of laying hens fed with 600 and 1,200 ppm of betaine showed a significant decrease compared to the control group(p<0.05). Results of these study show that dietary betaine supplementation affects protein and hormone metabolism in laying hens.

Lymphopenia by Pure Zinc Deficiency : Role of Corticosterone

  • The effect of zinc deficiency on certain immunological parameters was investigated using intragastric tube feeding to obviate decreased food intake and altered eating pattern. Male, Fischer 344 rats were bilaterally adrenalectomized(ADX) or sham operated(SHAM). ADX rats received 0.9% NaCl in their drinking water and corticosterone injections at the dose of 1mg/kg of body weight three times per day. After recovery, one half of ADX and SHAM animals were tube-fed a purified, liquid diet containing either two ppm of zinc(zinc-deficient, force-fed ; ZDF) or 50ppm(zinc-replete, force-fed ; ZRF) for 19 days. They received identical amounts of diet based on the intake of ad libitum-fed, zinc-replete rats. Although they received identical amounts of food, ZDF rats grew at a slower rate compared to ZRF rats in both SHAM and ADX rats. Regardless of surgery, force-feeding rats the zinc-deficient diet resulted in a substantial decrease in serum zinc levels. The weights of the thymus, lymph node, and spleen were lower in SHAM-ZRF rats compared to SHAM-ARF rats. Marginal zinc deficiency caused lymphopenia in SHAM animals. However, these differences in lymphoid tissues and cells between SHAM-ZDF and SHAM-ZRF rats disappeared in ADX rats. These results indicate that the impaired growth of lymphoid tissues observed in zinc-deficient, sham-operated animals can be attributed to elevated serum corticosterone levels under the conditions of our experiments.

Effect of Iron Deficiency on the Capacity for Peroxisomal and Mitochondrial $\beta$-oxidation

  • In order to determine the effect of iron depletion and subsequent supplementation on the muscle capacity for peroxisomal (PO) and mitochondrial(MO) $\beta$-oxidation during high fat feeding, weanling rats were fed a 44% (HF) or 2.5% (LF) fat diet with (+Fe) or without (-Fe) iron for 6 or 9 weeks. After 1 week rats fed HF+Fe or HF-Fe had 50-100% more PO and MO in heart, soleus, psoas and gastrocnemius than did rats fed low fat, but after 3 weeks rats fed HF-Fe had lower muscle PO and MO. In muscles of iron depleted rats PO and MO were not increased by supplementation with iron for 3 weeks. After 6 weeks MO and PO in skeletal muscles of rats fed HF+Fe were lower than after 3 weeks. It is concluded that adequate iron is necessary for miaximum response of muscle PO and MO to high fat feeding. However, after 6 weeks both PO and MO have returned to levels similar to those of rats fed low fat diets, hence, the elevated catalase activities seen at this time do not reflect peroxisomal $\beta$-oxidation.

Pig Feeding under the Potato-green Forage Base System with or without Addition of Herbs versus a Concentrate Based System: Effect on Post-slaughter Performance and Pork Characteristics

  • This study examined carcass and meat quality parameters in growing/finishing pigs fed unconventionally versus the concentrate-based system. Ninety-six, 12 wk old pigs ($34{\pm}SD0.3kg$) were randomly divided into three groups, assigned to one of the three dietary treatments: standard complete concentrate mixture, conventional (C diet); unconventional, steamed potato-green forage-concentrate based diet (U diet), and unconventional basal diet+herbage mix (UH diet). Pigs fed U diet showed lower dressing percentage, meatiness, loin eye area, and weight of pork neck ($p{\leq}0.05$), but their carcasses were significantly ($p{\leq}0.05$) longer and had increased backfat depth ($p{\leq}0.05$). There was no impact of the diet on the meat content of dry matter, crude ash, acidity, and colour parameters of m. longissimus. Unconventional feeding significantly ($p{\leq}0.05$) elevated water the holding capacity of m. longissimus and slightly improved the sensory attributes analysis of meat. The addition of herbs resulted in increased loin eye area ($p{\leq}0.05$), decreased fat content ($p{\leq}0.05$) in m. longissimus, and tended to improve some sensory attributes of meat. There were significant gender differences in response to all diets. There were significant diet${\times}$sex interactions for some measured variables, but there were no clearly identifiable trends with regard to any specific carcass or meat parameters. Feeding unconventional diet to pigs may offer better culinary attributes of the meat, and improve some technologically important characteristics of pig carcass, but may negatively affect some carcass or meat parameters.

DMPT(dimethyl-$\beta$-propiothetin)첨가가 조피볼락의 화학적 성분에 미치는 영향 (Effects of Dimethyl-$\beta$-propiothetin (DMPT) on Chemical Components of Korean Rockfish, Sebastes schlegeli)

  • 수산물 중에 다량으로 분포하는 섭이촉진물질로 알려진 DMPT의 추출 이용을 위한 기초 자료를 얻 고자 DMPT를 3 mM, 5 mM, 10 mM로 첨가한 실험용 사료로 8 주간 사육한 조피볼락의 성장효과 및 어체 성분에 미치는 영향을 검토한 결과를 요약하면 다음과 같다. 1. DMPT를 첨가하지 않은 대조구의 체중증가율은 약 1.2 배였으나 10 mM을 첨가한 사료의 경우는 약 1.4배로 성장효과가 크게 증가하였고, 사료효율 및 사료 섭취량도 많아 DMPT가 사료 섭이를 촉진시키는 것으로 나타났다. 2. 일반성분 변화 를 보면 DMPT의 농도가 높을수록 수분 함량은 감소하고 조단백질과 조지방의 함량은 함께 증가하는 경향이었다. 3. 아미노산 함량의 변화에서는 대조구가 3286.3 mg%에 비하여 10 mM 첨가구에서는 3568.3 mg%로 높은 값을 나타내어 DMPT 첨가구가 높데 나타났으며, 구성아미노산 조성에 있어서는 서로 비슷한 경향이었다. 4. 지방산 함량의 변화를 살펴보면, 실험개시전과 대조구에 비하여 DMPT 첨 가구에서 높게 나타났으며, DMPT의 농도가 높을수록 지방산의 함량도 증가하여 지방의 함량 변화와 일치하였다.

Effect of Environmental Factors on Flavonol Glycoside Production and Phenylalanine Ammonia-lyase Activity in Cell Suspension Cultures of Ginkgo biloba

  • A study was carried out to elucidate the relation between the production of flavonol glycosides and the change of phenylalanine ammonia-lyase activity in cell suspension cultures of Ginkgo biloba by the unassisted and synergistic effects of various factors. The quercetin production showed a mixed-growth-associated pattern in cell suspension cultures. Fluorescent light and UV radiation increased phenylalanine ammonia-lyase (PAL) activity, and resulted in the increase of the production of quercetin and kaempferol ten- and four-fold, respectively, as compared to that obtained in the normal culture condition. The cell growth of Ginkgo biloba was enhanced .at higher temperatures whereas the quercetin production was at its maximum at low temperatures. Moreover, the quercetin production was increased by temperature change during the culture period. In particular, the quercetin production was at the highest level when the culture temperature was elevated from $10^{\circ}C\;to\;30^{\circ}C$. The addition of phenylalanine as a precursor in the culture medium stimulated an 8-fold increase in the production of quercetin; the addition of naringenin caused a l0-fold increase. The quercetin production was also greatly increased by feeding enzyme cofactors such as 2-ketoglutarate and ascorbic acid in the culture medium, but specific PAL activity was not increased except with phenylalanine feeding. The synergistic effect of UV radiation and naringenin feeding was observed, resulting in the increase of flavonol glycoside production at a rate higher than in any other case investigated.

Effects of Feeding Increasing Proportions of Corn Grain on Concentration of Lipopolysaccharide in the Rumen Fluid and the Subsequent Alterations in Immune Responses in Goats

  • This study was conducted to investigate the effects of feeding increasing proportions of corn grain on concentration of lipopolysaccharide (LPS) in the rumen fluid and the subsequent alterations in immune responses as reflected by plasma concentrations of serum amyloid A (SAA) and haptoglobin (Hp) in goats. Nine goats were assigned to three diets (0%, 25%, and 50% corn grain) in a $3{\times}3$ Latin square experimental design. The results showed that as the proportion of dietary corn increased, the ruminal pH decreased (p<0.001), and the concentrations of propionate (p<0.001), butyrate (p<0.001), lactic acid (p = 0.013) and total volatile fatty acid (p = 0.031) elevated and the ruminal LPS level increased (p<0.001). As the proportion of dietary corn increased, the concentration of SAA increased (p = 0.013). LPS was detectable in the blood of individual goats fed 25% and 50% corn. A real-time PCR analysis showed that the copy number of phylum Bacteroidetes (p<0.001) was reduced ($4.61{\times}10^9$ copies/mL to $1.48{\times}10^9$ copies/mL) by the increasing dietary corn, and a correlation analysis revealed a significant negative correlation between the number of Bacteroidetes and rumen LPS levels. Collectively, these results indicated that feeding goats high proportions (50%) of corn grain decreased the ruminal pH, increased LPS in the rumen fluid and tended to stimulate an inflammatory response.

Comparative Effects of Dietary Quercetin and Rutin in Rats Fed with the Lieber-DeCarli Ethanol Diet

  • Flavonoids including quercetin and rutin are a group of naturally occurring compounds widely distributed in plants, especially in buckwheat. Thus, cereal and the leaf of the plant have increasingly used as a source of nutritional and functional foods such as noodle, cake or soup in Korea, Japan and other countries. This study investigated comparative effects of dietary rutin rich in buckwheat and its aglycone, quercetin, on serum biomarkers and antioxidant parameters in rats treated with chronic ethanol. Rats were fed with the liquid diets prepared by the method of Lieber Decarli. Serum alanine transaminase (ALT) and aspartate transaminase (AST) activities increased significantly by alcohol feeding. Dietary flavonoids including rutin, quercetin and their mixtures (1/1, v/v) decreased significantly the activities of serum ALT whereas the feeding of quercetin decreased only the activity of serum AST. The concentration of serum malondialdehydes elevated by chronic alcohol feeding decreased markedly in all the experimental groups that were fed with the flavonoids; however, the combined administration of quercetin or rutin, but not that of rutin or quercetin alone decreased significantly the concentration of liver malondialdehydes to the normal range in rats fed without ethanol. Our results suggested that dietary combined mixture of rutin and quercetin might be effective in ameliorating adverse responses seen in rats exposed to ethanol chronically.

Effects of High Cholesterol Feeding on Regulation of Plasma Lipids and Reverse Cholesterol Transport in Rabbits

  • this study was conducted to examine the atherogenic effect of high cholesterol diet (experimental diet) that influences changes of lipoprotein cholesterol metabolism and arterial wall. Seven NewZealand white rabbits were fed control diet, an the other 7 rabbits 2% cholesterol diet for 10 weeks. Results obtained from this study are as follows: 1) High cholesterol diet resulted in a gradual increase of plasma total cholesterol level, reaching upto 1422 mg/dl at the seventh week. 2) CETP (cholesteryl ester transfer protein) activity was significantly higher in high cholesterol group (64.9% at the 7th week) than control group (49.3% at the 7th week) during most of the experimental period except the 6th week. 3) The cholesterol supplementation induced fatty liver and a decrease of hepatic HMG-CoA reductase activities (2.1 moles vs. 0.3nmoles) compared to control group. 4) Bands of apo B-100 and apo E in plasma lipoprotein were thicker in high cholesterol-fed animals tan control animals as visualized by SDS-PAGE. 5) Oxidizability of plasma lipoproteins measured in vitro was greater in high cholesterol group tan control group, but vitamin E level higher in control group. 6) he effect of cholesterol feeding for 10 weeks also led to early fatty streaks in aortic intima. High cholesterol feeding was atherogenic to rabbits, an this seems to be mediated through elevated CETP activities that regulate plasma HDL cholesterol level and decrease an efficiency of reverse cholesterol transport in lipoprotein cholesterol metabolism. The enhanced oxidizability of plasma lipoproteins and lowered vitamin E level may also contribute to the formation of faaty streaks in aorta of cholesterol-fed rabbits.

신생아의 조기수유에 관한 실험적 연구 (An Experimental Study on the Early Feeding of Infants)

  • The newborn human is the only mammalian whose mother does not have a food supply ready for it's offspring at birth. From two to four days usually elapse before the mother's supply If milk appears, and during this period, some kind of artificial feeding should be supplied to the infants. Because of this factor, there has been continued debate fog the past hundreds of years as to when the first feeding should be started Accordingly, many experiments were carried out by scholars and because of these, Pre-lateral feedings were believed to be necessary. Many types of pre-lateral feedings were tried and the conclusion was reached that glucose water was the best food for the first infants'feedings. Traditionally, This has been started 12 hours after birth. The causes for the 12 hours delay were thought to (1) provide rest for the infants: (2) prevent regurgitation ana vomiting which tended to be prevalent during this tine: (3) in cases of low weight infants, prevention of aspiration pneumonia. From recent studies of newborn physiology and as pediatric medicine has been rapidly advancing, many studies hare been carried out concerning the improvement of infant nutrition and the early feeding of infants has been emphasized. This author believes it would be very beneficial to try two different kinds of feedings for the infant. (1) experimental feedings ana (2) comparative feeding, and during this period to investigate and compare the infants blood sugar level, hematocrit, gamma globulin level weight changes and to observe the infant reaction ill order to search for a more desirable feeding program. This study was conducted from January to March 1974 with data related to 40 healthy newborn infants (male 21, female 19: weight, 2.79∼4.20㎏ : gestation, 39∼40 weeks) born at Ewha Womens University Hospital and the results obtained were as follows : 1. At time of birth the blood sugar level from the cord sample averaged 88.99㎎/100㎖, but the blood sugar level rapidly dropped after 2 to 3 hours and reached the lowest point after 10 to 11 hours (54.48㎎/100㎖) and rose again by the 24 hour time period (76.80㎎/100㎖). Changes in the blood sugar level of the experiments: groups and the compare-five group was not significantly different until the 6 to 7 hour period, but by the 10 to 11 hour period the blood sugar levels of the experimental group (49,10㎎/100㎖) and the comparative group (49.70㎎/100㎖) were lower than the remainder of the experimental groups. 9. There ware no significant weight changes between the two groups. Average weight at birth was 3.35㎏, but at the 24 hours period birth weight averaged 3.29㎏. (1.8% reduction of birth weight). It continually lowered until at 48 hours, average weight was 3.26㎏ (2.7% reduction from birth weight.) 3. Hematocrit readings showed no significant difference between the groups. Hematocrit, the average value at birth, was 28.07% and abruptly elevated to average 64.35% at the 2 to 3 hour period, then slowly lowered to an average of 59.67% at the 6 to 7 hour period, 55.10% at the 10 to 11 hour period, ana 53.70% at the 24 hour period. 4. At birth, average gamma globulin value averaged 1,39㎎/100㎖. and at the 24 hour period averaged 1,52㎎/100㎖ revealing no significant difference between the two feeding groups. 5. Such factors as voiding, passing of meconium, regurgitation and vomiting showed no significance between the two feeding groups. However, the number of infants voiding and passing meconium in the experimental groups during the first 12 hours was slightly greater. In general there was an increased tendency for regurgitation and. vomiting among a small group of the infants during the first 24 hours which thereafter decreased. 6. Fluid intake averaged 24.38cc at the first feeding and increased to average 30.48cc at the third feeding and further increased to 73. 00cc at the fifteenth feeding. Finally it was suggested that the most reasonable method of early feeding is to give less than 25cc of 5% glucose water and/or 8% powdered milk at 8 to 9 flours after birth in order to prevent hypoglycemia and dehydration.
