Enrichment of Pork with Omega-3 Fatty Acids by Tuna Oil Supplements: Effects on Performance as well as Sensory, Nutritional and Processing Properties of Pork

  • The effects of tuna oil supplementation (0, 1, 2 and 3%) to pig diets on growth and carcass yield as well as meat quality were determined in 40 crossbred pigs. Animals were fattened from 30 to 90 kg of live-weight. Twenty-four hours after slaughter, following various early- and late-post mortem measurements, loin, backfat and belly were prepared from the carcasses. Bacon was produced from the belly part by curing and smoking. Neither performance (feed intake, daily gains, feed conversion efficiency) nor carcass quality (slaughter weight, dressing percentage, lean percentage, nutrient composition of the loin) were significantly affected by tuna oil supplementation. Tuna oil also had no clear effects on early- and late-post mortem meat quality traits, water-holding capacity and tenderness of the M. longissiumus dorsi (LD). Colour traits of LD and backfat, and backfat firmness were not significantly affected by tuna oil, either. However, there was a certain trend to elevated fat contents of LD (and bacon), but not of backfat, with increasing levels of tuna oil in feed. Pigs receiving elevated proportions of tuna oil expressed lower VLDL cholesterol and triglyceride concentrations in blood plasma, whereas the cholesterol content of LD, backfat and bacon did not reflect this trend. Effects of tuna oil on fatty acids in LD, backfat and bacon were often small in extent, except those concerning the long-chain polyunsaturated fatty acids. With 3% tuna oil in the diet, the contents of the particularly desired omega-3 fatty acids, C20:5 and C22:6, were 0.1 and 0.2 g/kg in LD. The corresponding values for backfat and bacon were 2.6 and 12.6 g/kg, and 1.3 and 9.2 g/kg, respectively. Tuna oil supplementation was associated with significant adverse effects on flavour and overall acceptance of bacon (not significant in LD although numerically the same trend was noted), but these effects on sensory ratings were limited in extent. Also shelf life of the products, determined as TBA value after different storage periods at $4^{\circ}C$ in LD, backfat and bacon, was significantly reduced. Overall, the present study suggests that omega-3 fatty acids may be enriched in pork by feeding tuna oil with few undesired side-effects, particularly those on sensory perception and shelf life, suggesting immediate consumption of the products is advisable. Most economically important traits (performance, slaughter and physical meat quality) remained unaffected.

Adriamycin을 투여한 흰쥐의 간 지질대사에 미치는 비타민 A와 E의 급여 효과 (Effect of Dietary Vitamin A and E on Hepatic Lipid Metabolism in Adriamycin-Treated Rats)

  • Adriamycin의 투여로 변화되는 생체내 지질과산화대사에 대하여 비타민 A및 E의 급여와 이들의 동시급여의 효과를 살펴보기 위해 이유한지 1주된 Sprague-Dawley 흰쥐를 6주간 사육하였다. 대조군을 제외한 모든 실험군에게 매주 일정시각에 ADR(2mg/kg/week)을 복강주사 한 후 비타민A와 E의 급여효과를 관찰하였으며 그 결과는 다음과 같다. 간 조직에서의 과산화지질 함량은 ADR투여로 현저한 증가로 보여주었고, 비타민 A 와 E급여가 이들의 생성을 저하시켰다. Catalase 활성도는 ADR의 투여로 증가된 반면 GSH-Px는 감소되는 경향이었다. 이에 비타민 A나 E의 급여로 ADR투여에 의한 효소 활성도의 변화를 완화시켰다. SOD의 활성은 ADR의 투여로 유의적인 변화는 나타나지 않았지만 비타민 A와 E동시 급여군에서 그 활성도가 현저히 유도외었다. 간 미토콘드리아의 지방산조정은 ADR투여로 큰 변화는 없었으나 비타민 E급여군이 비타민 A급여군 보다 방어효과가 더 좋았다. 또한 지질성분들의 변화는 ADR의 투여로 중성지질과 콜레스테롤 함량이 증가하였고 비타민 A의 급여에 의해 이들 함량이 유의적으로 감소되었다.

Coenzyme $Q_10$과 Vitamin E 첨가식이가 Adriamycin을 투여한 흰쥐의 체내 지질과산화에 미치는 영향 (Effects of Dietary Coenzyme $Q_10$ and Vitamin E on Lipid Peroxidation in Adriamycin-treated Rat)

  • 본 연구는 항암제로서의 효능을 가진 ADR을 투여하여 유도된 흰쥐의 체내 지질과산화에 대하여 vitamin E와 coenzyme $Q_{10}$ 의 항산화 효과를 구명하고자 실시되었다. 횐쥐의 성장상태는 ADR의 투여로 대조군에 비하여 저하되었으나 vitamin E나 coenzyme $Q_{10}$ 공급에 의하여 별다른 영향을 받지 못했다. 혈장과 심장 미토콘드리아 분획에서의 지질과산화물 함량은 ADR 단독 투여군에서 현저하게 증가되었으나 vitamin E나 coenzyme $Q_{10}$ 의 공급량에 비례하여 저하되었고 그 작용 능력은 vitamin E가 더 우수한 것으로 나타났다. 지질과산화물 대사효소 활성도에 있어서는 catalase 활성도가 전 처리군에서 유의적 차이를 보이지 알은 반면 GSH-Px 활성도는 ADR투여로 대조군에 비해 증가되었으나 vitamin E의 공급으로 대조군의 수준으로 저하되었다. Adriamycin에 의해 증가된 SOD 활성도는 vitamin E와 coenzyme $Q_{10}$ 공급에 의해서 모두 영향을 받았으나 coenzyme $Q_{10}$ 보다는 vitamin E에 의해 더 큰 효과를 나타내었다. 심장 미토콘드리아 분획의 지방산 조성은 ADR 단독 투여군에 있어서는 arachidonic acid가 다른 처리군들에 비해서 가장 높게 나타난 반면에 docosahexaenioc acid는 관찰되지 않았다. 그러나 높은 수준의 vitamin E나 coenzyme $Q_{10}$ 을 공급시켰을 때 arachidonic acid는 감소하는 대신 docosahexaenioc acid는 증가되었으며 높은 수준의 vitamin E가 coenzyme $Q_{10}$ 보다도 더 큰 보호작용을 나타내었다. 이상의 결과를 종합해서 볼 때 ADR 투여로 인해 심장에서 일어난 지질과산화 반응에 대해 vitamin E 나 coenzyme $Q_{10}$ 이 정상 수준에는 미치지 못하나 지질과산화 방어계 효소변화를 통해 보호작용을 나타낸다는 것을 알 수 있었다. 또한 심장에서 ADR에 의한 지질과산화 반응을 억제시키는 능력에 있어서는 농도에 비례하여 vitamin E가 coenzyme $Q_{10}$ 보다도 더 우수한 것으로 나타났다.

대추 메탄을 추출물이 사염화탄소투여에 의한 흰쥐의 간 세포독성에 미치는 영향 (Effect of Jujube Methanol Extract on the Hepatotoxicity in $CCl_4$-Treated Rats)

  • 대추 메탄을 추출물이 사염화탄소에 의한 휜쥐의 간손상에 미치는 영향을 알아보기 위하여 대조군, 대추메탄올 추출물 투여군, 사염화탄소 투여군 및 대추메탄올 추출물을 투여한 후 사염화탄소를 투여한 군으로 나눠, 2, 4주간 사육하여 혈청 중의 transaminase (GOT 및 GPT), 간 중의 lipid peroxide량, glutathione 함량, xanthine oxidase, SOD, catalase, GSH-Px 등의 활성도의 변화를 관찰하였다. 사염화탄소(50%, 1.0ml/kg) 투여군의 혈청 중 GOT및 GPT의 활성도, 간중TBA 반응성 산물량은 2, 4주에서 대조군에 비하여 현저히 증가되었으나, 대추 메탄올 추출물을 투여한 후 사염화탄소를 투여할 때 GOT의 경우 2, 4주에서, 간 중 TBA 반응성 산물량은 4주에서 사염화탄소 투여군에 비하여 유의성있는 감소효과를 보였다. 유리기 생성계 효소인 xanthine oxidase 활성도에서, 사염화탄소 투여군은 대조군에 비하여 현저한 증가를 보였으나, 대추 메탄올 추출물을 투여한 후 사염화탄소를 투여한 군에서는 감소되었다. 유리기 해독계 효소인 SOD, catalase 및 GSH-Px의 활성도도 사염화탄소 투여로 모두 증가되었으나, 대추 메탄올 추출물을 투여한 후 사염화탄소 투여로 SOD는 4주에서, GSH-Px는 2, 4주에서 대조군에 비하여 유의한 감소효과를 나타냈으며, catalase는 투여기간에 따라 감소는 되었으나 유의성은 보이지 않았다. 이상의 실험결과에서, 사염화탄소 투여로 혈청 및 간 중의 각종 효소 활성도 및 지질과산화물량이 증가되었는데 이는 사염화탄소가 간 중의 효소계 및 간세포의 microsome을 심하게 손상시켰음을 알 수 있으며, 대추 메탄올 추출물을 전처리하므로써 사염화탄소에 의하여 현저히 증가된 각종 효소활성도 및 지질과 산화물량이 감소되었는데, 이는 대추 메탄올 추출물이 간세포의 괴사와 효소의 유출을 저 해하고 간의 저항력 및 간기능을 유지시키므로써 간 보호작용을 할 수 있을 것으로 사료된다.

IRS-2 Partially Compensates for the Insulin Signal Defects in IRS-1-/- Mice Mediated by miR-33

  • Insulin signaling is coordinated by insulin receptor substrates (IRSs). Many insulin responses, especially for blood glucose metabolism, are mediated primarily through Irs-1 and Irs-2. Irs-1 knockout mice show growth retardation and insulin signaling defects, which can be compensated by other IRSs in vivo; however, the underlying mechanism is not clear. Here, we presented an Irs-1 truncated mutated mouse ($Irs-1^{-/-}$) with growth retardation and subcutaneous adipocyte atrophy. $Irs-1^{-/-}$ mice exhibited mild insulin resistance, as demonstrated by the insulin tolerance test. Phosphatidylinositol 3-kinase (PI3K) activity and phosphorylated Protein Kinase B (PKB/AKT) expression were elevated in liver, skeletal muscle, and subcutaneous adipocytes in Irs-1 deficiency. In addition, the expression of IRS-2 and its phosphorylated version were clearly elevated in liver and skeletal muscle. With miRNA microarray analysis, we found miR-33 was down-regulated in bone marrow stromal cells (BMSCs) of $Irs-1^{-/-}$ mice, while its target gene Irs-2 was up-regulated in vitro studies. In addition, miR-33 was down-regulated in the presence of Irs-1 and which was up-regulated in fasting status. What's more, miR-33 restored its expression in re-feeding status. Meanwhile, miR-33 levels decreased and Irs-2 levels increased in liver, skeletal muscle, and subcutaneous adipocytes of $Irs-1^{-/-}$ mice. In primary cultured liver cells transfected with an miR-33 inhibitor, the expression of IRS-2, PI3K, and phosphorylated-AKT (p-AKT) increased while the opposite results were observed in the presence of an miR-33 mimic. Therefore, decreased miR-33 levels can up-regulate IRS-2 expression, which appears to compensate for the defects of the insulin signaling pathway in Irs-1 deficient mice.

Effects of Herb-Mix Supplementation on the Growth Performance and Serum Growth Hormone in Weaned Pigs

  • Two hundred sixteen crossbred ($Landrace{\times}Yorkshire$) castrates with an average weight of $7.4{\pm}0.3kg$ were used in a $3{\times}3$ factorial treatment array. The treatments were three levels of Herb mixture (HM; 0, 0.40 and 0.80 g/kg BW/day) and three levels of dietary nutrient (17.30% CP, Level-1; 17.90% CP, Level-2; and 18.50% CP, Level-3). The influence of HM intake and nutrient level on growth performance and ADG in 0.40- and 0.80-HM pigs increased significantly (p<0.01) as nutritional level was elevated. Although very little enhancement of ADG was observed at Level-1, peak ADG occurred in 0.8-HM treated pigs at Level-3. Feeding of 0.80 g HM/kg/d to pigs consuming Level-1 diet resulted in a 8.7% increase in ADG compared with control pigs, whereas the increase in ADG as a result of 0.80-HM with Level-3 treatment was 39%. ADFI in Level-2 pigs improved linearly (p<0.01) as HM level was increased. Treatment with HM resulted in a 12.0% increase ranging 4.7 to 20% in the ADFI compared with respective controls. ADFI at all nutritional level was significantly higher in 0.80-HM pigs (p<0.02). F/G in Level-2 pigs improved significantly as HM was fed (p<0.01), and in HM-0.80 pigs was also significantly improved as nutritional level was increased (p<0.05). Pigs fed HM had higher bone mineral density (BMD) at Level-1, longer dorsal spine length (DSL) at level-2 (p<0.05) than pigs fed basal diets. Pigs fed HM tended to higher BMD and DSL than those fed basal diets. The level of GH secretion declined with age. There was no difference between treatments (p>0.05) in the serum growth hormone at the same age. The GH was higher in pigs fed HM than those fed basal diets and increased in all pigs after 2wks feeding. A positive effect of added Herb-Mix on growth performance in weaned pigs was demonstrated by measuring the serum growth hormone, bone mineral density and length of dorsal spine.

Hypotriglyceridemic effects of brown seaweed consumption via regulation of bile acid excretion and hepatic lipogenesis in high fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: The present study aimed to further investigate the potential health beneficial effects of long-term seaweed supplementation on lipid metabolism and hepatic functions in DIO mice. MATERIALS/METHODS: Four brown seaweeds (Undaria pinnatifida [UP], Laminaria japonica [LJ], Sargassum fulvellum [SF], or Hizikia fusiforme [HF]) were added to a high fat diet (HFD) at a 5% ratio and supplemented to C57BL/6N mice for 16 weeks. Triglycerides (TGs) and total cholesterol (TC) in the liver, feces, and plasma were measured. Fecal bile acid (BA) levels in feces were monitored. Hepatic insulin signaling- and lipogenesis-related proteins were evaluated by Western blot analysis. RESULTS: Fasting blood glucose levels were significantly reduced in the LJ, SF, and HF groups compared to the HFD group by the end of 16-week feeding period. Plasma TG levels and hepatic lipid accumulation were significantly reduced in all 4 seaweed supplemented groups, whereas plasma TC levels were only suppressed in the UP and HF groups compared to the HFD group. Fecal BA levels were significantly elevated by UP, LJ, and SF supplementation compared to HFD feeding only. Lastly, regarding hepatic insulin signaling-related proteins, phosphorylation of 5'-AMP-activated protein kinase was significantly up-regulated by all 4 types of seaweed, whereas phosphorylation of protein kinase B was up-regulated only in the SF and HF groups. Lipogenesis-related proteins in the liver were effectively down-regulated by HF supplementation in DIO mice. CONCLUSIONS: Brown seaweed consumption showed hypotriglyceridemic effects in the prolonged DIO mouse model. Specifically, combinatory regulation of BA excretion and lipogenesis-related proteins in the liver by seaweed supplementation contributed to the reduction of plasma and hepatic TG levels, which inhibited hyperglycemia in DIO mice. Thus, the discrepant and species-specific functions of brown seaweeds provide novel insights for the selection of future targets for therapeutic agents.

Feeding of Juvenile Purple Washington Clam, Saxidomus purpuratus (Sowerby): Effects of Algal Concentration and Temperature

  • To find the optimal rearing conditions for Saxidomus purpuratus juvenile, filtering activity was estimated as functions of algal concentration and temperature by measuring the rates of clearance (CR) and ingestion (IR), when S. purpuratus was feeding. The clams were fed on unialgal diet of Isochrysis galbana at 6 algal concentrations (4.6$\times$$10^4$~2.6$\times$$10^6$ cells/ml) and at 6 temperatures (5, 10, 15, 20, 25, and 30^{\circ}C ). Algal concentration significantly affected the CR and the IR at all temperatures. At lower algal concentrations, CR increased, but decreased beyond a particular concentration. The maximum CR ($CR_{max}$) at 5, 10, 15, 20, 25, and 30^{\circ}C were 0.30, 1.73, 5.95, 15.17, 21.12, and 0.33 $l/g/h$, respectively. Below the level of 5.6$\times$10$^{5}$ cells/ml, IR increased as algal concentration increased, but was saturated at higher concentrations. To maintain high growth rate of S. purpuratus, I. galbana should be supplied with more than 5.6$\times$10$^{5}$ cells/ml. The maximum IR ($IR_{max}$) at 5, 10, 15, 20, 25 and30^{\circ}C were $2.2$\times$10^8, $1.5\times$10^9, 3.4$\times$10^9, 4.9$\times$10^9, 5.3$\times$10^9, and 1.0$\times$10^8$ cells/g/h, respectively. As for temperature, both $CR_max$ and $IR_max$ increased remarkably with raising temperature from 5 to 25^{\circ}C, but rapidly decreased at 30^{\circ}C. Between 15 and 25^{\circ}C $CR_{max} and IR_{max}$ were higher and most stable, At this temperature range, the $Q_{10}/s for CR_{max} and IR_{max}$ were 3.5 and 1.6, respectively. Therefore the optimal thermal range for the juvenile is 15~$25^{\circ}C$. The annual variation in IR$_{max}$ predicted by natural seawater temperature shows that inactive period (with lower $IR_max$) lasts for 5 months (from December to April). To ensure higher growth of juvenile during this inactive period at hatcheries, rearing temperature should be elevated to $15^{\circ}C$.>.

GALK Hyperactivity로 인한 갈락토스혈증의 임상적 특성에 관한 연구 (Clinical and Laboratory Characteristics of Galactokinase Hyperactivity)

  • Purpose: Galactose is metabolized to galactose-1-phosphate by galactokinase (GALK), galactose-1-phosphate uridyltransferase (GALT) and UDP-galactose-4-epimerase (GALE), and galactosemia occurs when each enzyme is deficient. In Korea, unlike foreign countries, classic galactosemia is rare and transient galactosemia due to GALK hyperactivity is reported, but studies on frequency, clinical significance, and genetic variation are lacking. In this study, we analyzed the clinical characteristics of patients with galactosemia due to GALK hyperactivity. Methods: We investigated 85 patients who had an elevated galactose level in the neonatal screening test without deficiency of enzymes at Department of Pediatrics, Seoul & Bucheon Soonchunhyang University Hospital from January 2008 to June 2016. We investigated the level of galactose, galactose-1-phosphate, GALK and duration of galactose normalization, and analyzed the correlation between GALK elevation and galactose, galactose-1-phosphate and duration of galactose normalization. And the levels of galactose, galactose-1-phosphate, and duration of galactose normalization were compared between the galactose-free formula feeding group and non-feeding group. Results: Mean age of visit was $26.7{\pm}16.1days$. Duration of galactose normalization was $35.3{\pm}20.5days$. Mean galactose level was $18.5{\pm}7.3mg/dL$ in the neonatal screening and follow-up galactose level in serum was $2.3{\pm}5.4mg/dL$. The mean value of galactose-1-phosphate was $6.0{\pm}4.7mg/dL$ and the mean GALK level was $3.84{\pm}1.28{\mu}mol/Hr/gHb$. There was no significant correlation between GALK levels and galactose levels in the neonatal screening test (P=0.351), and we analyzed the correlation between GALK levels and follow-up galactose levels in serum, there was no significant correlation (P=0.101). There was a significant correlation between GALK levels and galactose-1-phosphate (P=0.015), and the correlation between GALK levels and duration of galactose normalization was not statistically significant (P=0.176). 49% of the patients were fed galactose-free formula, and 45% were not. Galactose and galactose-1-phosphate levels in the neonatal screening test were statistically significantly higher (P=0.004, 0.034) in using galactose-free formula group. Duration of galactose normalization was not related to the use of galactose-free formula (P=0.266, 0.249). Conclusion: Galactosemia due to GALK hyperactivity seems to be a temporary phenomenon and may not require galactose restriction. More research is needed on the role of the nuclear protein, racial traits and genetic variations in Korean patients.

젖소에서 유성분 분석을 통한 영양상태 평가 및 건강관리에 관한 연구 III. 고능력우 위주의 대규모 목장에서 우유중 단백질과 요소태질소 수준이 수태율에 미치는 영향 (Studies on health management and nutritional evaluation by milk components analysis in dairy cows III. Relationship between conception rates, and milk urea nitrogen and milk protein concentration in a large dairy herd of high yielding cows)

  • Milk urea nitrogen (MUN) determination is being used an indicator of the protein-energy balance in dairy herds. A faulty balance can be corrected to optimize milk production and animal health. This parameter is regarded as a potential tool to evaluate suboptimal feeding practices and reproductive disorders. Therefore, the purpose of this study was to investigate the response of milk composition by regular feeding analysis and to compared the relationship between MUN and milk protein(MP) and fertility at the insemination period in Holstein dairy cows. Total of 355 artificial insemination (AI) for 150 Holstein cows in the herd were used to examine the relationship between MUN and MP content and conception rate. The AI occured for the cows 50 to 150 day in milk, and MUN and MP concentration were determined using automated infrared procedures. The mean${\pm}$standard deviation of MUN and MP concentration in the herd were $15.6{\pm}2.1mg/dl$ and $3.23{\pm}0.38%$, respectively. MUN contents of bulk milk were increase by elevated crude protein intake. The conception rate was lower in the cows in which the level of MUN was lower than > 8.0mg/dl (10.0%) or > higher than 25mg/dl (15.4%) relative to the cows in MUN content of 12.0~17.9 mg/dl (36.7%) at the time of insemination. Also, lower MP than 3.0% or higher MP than 3.25% were associated with a lower conception rates. Consequently, MUN and MP analyses may be used serve as a monitoring tool of protein and energy nutritional balance to improve reproduction efficiency in Holstein dairy cows.
