허혈후 칼슘 결핍 용액의 재관류가 적출 관류 기니픽 심근 세포에 미치는 영향에 관한 전자현미경적 관찰 (An Electron Microscopic Study on the Effect of Calcium-free Reperfusion in Isolated Perfused Guinea Pig Heart after Global Ischemia)

  • The effect of calcium-free reperfusion for 5, 10, and 15 minutes, respectively, followed by continuous reperfusion with normal Tyrode solution containing 1.0mM calcium chloride, after global ischemia in the isolated perfused guinea pig heart by Langendorff techniques was examined with transmission electron microscope. Compared to the nomal Tyrode solution-perfused control hearts, the 5 minute calcium-free-reperfused hearts showed loss or thickening of Z lines, focal sarcolemmal disruption, mitochondrial swelling, clumping of chroma-tin, intracellular fluid accumulation, and some separation of cell junctions, especially the fasciae adherentes. These changes became more severe in the hearts of 10 minute calcium-free reperfusion. Subsarcolemmal larger bleb and near complete separation of cell junctions were noticed. In the 15 minute calcium-free-reperfused hearts, irreversible ultrastructural changes including contraction bands, biazrre mitochondria, and sarcolemmal destruction were widely distributed. The severity of myocardial changes were in accordance with the duration of calcium-free reperfusion. These changes indicate that calcium-free reperfusion regardless of its duration could not salvage the post-ischemic myocardium probably due to development of calcium paradox.

성장기 흰쥐 시각피질의 신경연접에 대한 도은법 및 전자현미경적 연구 (Silver Impregnation and Electron Microscopic Studies on the Synapse in the Visual Cortex of Rat during Postnatal Development)

  • These studies were performed to observe the morphological changes of synapses in the visual cortex of rat during early postnatal development. Specimens of the visual cortex were taken from rats (Sprague Dawley) at 1, 3, 7, 14 and 21 days of age, and prepared for silver impregnation and electron microscopy. The number of synapse and the length of postsynaptic thickening were increased progressively with age, especially 14 and 21 days. The number of dendritic spine was increased conspicuously on postnatal days 14-21. And asymmetic, curved and axo-spinous synapses were increased markedly at the same ages. The present findings suggest that spurt of synaptogenesis in the rat visual cortex occurs during early postnatal development, especially in second to 3rd week period and asymmetric and/or curved axo-spinous synapse is a matured form of synapse with advanced age.

  • A Scanning Electron Microscopic Study of the root surface changes was carried out in advanced periodontal disease. The results obtained were summerized as follow; 1. The root surface of cementoenamel junction of healthy root showed smooth surface, while the root surface of diseased state showed covering of deposits which regareded as a course of calcification. 2. At the mid-portion of the root, the regular cemental projection were observed on the healthy root surface and on the root surface of diseased state, cemental projection showed relatively irregular pattern. 3. On the root surface which consist of wall of periodontal pocket, there were various deposits which could be considered as subgingival calculus, bacterial plaque and epithelial attachment remnant. 4. The bottom area of the pathologic pocket, tearing altered collagen fibers were seen on the root surface. 5. At the apical portion of root surface, calcified fibers ran parallel to the root surface in healthy tooth and the fiber bundles of periodontal ligament were seen in the diseased state.

  • This study was taken to clarify the histopathological changes of pigs naturally infected with hog cholera. Microscopic observations of the necrotic lesion and inclusion body in the lymphoid organs were carried out in the natural cases of hog cholera and experimental cases inoculated with ALD virus and isolated virus strains. Electron microscopic findings of the intranuclear inclusion bodies in the reticular cell of spleen and lymph node were also observed in the experimental cases. The results obtained are as follow, As the histological findings necrosis of lymphoid organs was observed mainly in the lymph follicle. The necrotic lymphoid organs were found to contain 35.0% in the natural and 37.5% in the experimental cases. Intranuclear inclusion bodies were found mainly in the reticular cells of lymphoid organ, the epithelium of bronchiole and alevolus, and the vascular endothelium of brain. These inclusion bodies were seen in 40.0% of the natural cases and all of the experiment. The inclusion body was appeared to compose of activated nucleoli and chromatin granules (interchromatin and perichromatin) by electron microscopy.

방사선조사가 백서 협점막에 미치는 영향에 관한 전자현미경적 연구 (AN ELECTRON MICROSCOPIC STUDY ON THE EFFECTS OF IRRADIATION ON THE BUCCAL MUCOSA OF RAT)

  • The author studied the acute reaction of cobalt-60 irradiation to buccal mucosa in rats and difference of the effects of single versus fractionated exposure. 195 Sprague Dowley strain rats, weighing about 120gm, were used in this experiment. 3 rats served as controls and the remaining 192 rats were divided into six groups of 32 rats each. Experimental group Ⅰ, Ⅱ, Ⅲ were received a single dose of 15Gy, 16.5Gy, 18Gy and group Ⅳ, Ⅴ, Ⅵ were received two equal sized fractionated dose of 9Gy, 9.75Gy, 10.5Gy at 4 hour intervals, respectively. The experimental groups were irradiated with cobalt-60 teletherapy unit, Picker model 4M 60 (Field size, 12x5 cm, SSD, 50㎝, Dose rate, 222cGy/min, Depth, 1㎝). The animals were sacrificed at 1, 2, 3, 6, 12 hours, 1, 3, 7 days after irradiation and the changes of the irradiated buccal mucosa were observed by electron and light microscopy. The results were as follows: 1. A single exposure was more damaging than fractionated exposure, and as the radiation dose increased, the changes of cell organelles became faster, but the healing of radiation-induced damage in fractionated exposure was faster than in single exposure. 2. The radiation-induced changes of the basal cells were the most prominent in 18Gy-single exposure group, and the least in 18Gy-fractionated exposure group. 3. Electron-microscopically, there appeared nuclear changes, swelling of mitochondria and rough endoplasmic reticulum, decrease of free ribosome, presence of vesicles, widening of intercellular space, and loss of basal lamina. The early remarkable changes were partly loss of nuclear membrane and swelling of mitochondria. 4. Light-microscopically, derangement and pyknosis of basal cells, hydropic changes of spinous cells, enlargement of granular cells, indistinctness of basement membrane, and proliferation of epithelium were observed.

Occurrence of Hydropericardium-Hepatitis Syndrome of Baeksemi and Broiler Raised in Korea

  • The occurrence of HHS was confirmed for the first time in Korea from chickens submitted for diagnosis to our laboratory from broiler and baeksemi farms. Clinical signs included depression, inappetence, ruffled feathers and a increase in mortality. At necropsy, severe hydropericardium and hepatic necrosis was founded characteristically and the most remarkable microscopic changes were seen in the liver. These included basophilic intranuclear inclusion bodies in the hepatocytes, massive hemorrhages and necrosis in the liver parenchyma. We could also identify fowl adeno-virus(FAV) by polymerase chain reaction(PCR) and electro-microscopic confirmation. Monosodium Glutamate가 초생추 대퇴골 근위골단에 미치는 영향에 관한 투과 및 주사전자현미경적 연구 (Ultrastructural Studies of Effect of Monosodium Glutamate on the Epiphyseal Plate of Femur in Young Chicken)

  • This study was carried out to investigate the ultrastructural changes of the proximal epiphyseal plate of the femur in young chickens that had been treated with monosodium glutamate(MSG). Eighty 1-day old broiler chickens(Hubbard strain) were divided into control and experimental groups. The experimental group received daily administration of MSG(3mg/g of body weight in 0.75% saline) per orally for 1, 3, 6, 9, 12, 15, 18 and 21 days, and were sacrificed with exanguination. The control group received an equal volume of 0.75% saline. For the transmission electron microscopy, the prehypertrophic cartilage zone of epiphyseal plate was cleaved, fixed with 2% glutaraldehyde(containing 0.2% ruthenium red), postfixed with 1 % osmium tetroxide, embedded in Epon 812, and stained with uranyl acetate and lead citrate. For the scanning electron microscopy, the calcified zone of epiphyseal plate was cleaved and coated with gold palladium. The results obtained were as follows; 1. On transmission electron microscopic examination, the sacculation decreased from 12 day to 21 day MSG administrated groups, and the vesiculation decreased in 18 and 21 day MSG administrated groups in rough endoplasmic reticulum of chondrocytes in prehypertrophic cartilage zone. The ruthenium red binding particles in pericellular rim, territorial matrix and interterritorial matrix increased from 9 day to 21 day MSG administrated groups, but the crystalloid materials decreased. 2. On scanning electron microscopic examination, the trabecular formation and calcospherites of calcification zone decreased in 18 and 21 day MSG administrated groups. The resorption cavities widened from 15 day to 21 day MSG administrated groups.

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.

가미계혈등탕(加味鷄血藤湯)이 고혈압(高血壓)에 미치는 영향(影響) (The Effect of Gamigehyuldeung-tang on Hypertension)

  • Objectives : This study was done to investigate the effect of Gamigehyuldeung-tang on hypertension. Methods : After administering Gamigehyuldeung-tang extract to SHR(Spontaneous Hypertensive Rats) for 5 weeks, changes in blood pressure, pulse rate, aldosterone and catecholamine levels in plasma were examined, and immunohistochemical changes and scanning electron microscopic changes were observed. Results : The following results were obtained; blood presure decreased significantly as well as levels of aldosterone, dopamine and epinephrine in SHR. But levels of norepinephrine were unaffected. No capillary vessel dilation was observed. A decrease in cell damage was seen in microscope investigation. Conclusions : These results support a role for Gamigehyuldeung-tang might be usefully applied in treatment of hypertension.
