Association of Aster Yellow Phytoplasma with Witches' Broom Disease of Ash (Fraxinus rhynchophylla Hence) in Korea

  • Typical phytoplasma whiches' broom symptoms were observed in Ash (Fraxinus rhynchophylla Hence) in Korea. The symptoms of the disease were showing abnormally small leaves, shorted internodes and proliferation of shoots. Examination of fluorescent and electron microscopy of leaf midribs revealed numerous phytoplasma bodies localized in the phloem tube cells. The phytoplasmas were detected in all the symptomatic samples by the amplification with phytoplasma specific primer pair P1/P7 consistently, and the expected size was 1.8 kb. However, the phytoplasma DNA was not detected in healthy seedlings. Based on sequence analysis of amplified region, this phytoplasma has close homologies with eqilodium phyllody, mulberry dwarf, and aster yellow phytoplasmas, 99.95%, 99.79% and 99.78%, respectively, This phylogetic analysis indicates that ash witches' broom phytoplasma should be classified in the aster yellow group 16SrVI and clearly distinct from the ash yellow group 16SrVII.

  • Testes from the allied rock wallaby(Petrogale assimilus) have been examined by the electron microscopy in thin sections in order to examine spermiogenesis and structure of spermatozoa. The spermiogenesis can be divided into nine stages: early Golgi, late Golgi, collapsing, nuclear protrusion, condensation and flattening, nuclear shaping, rotation, nuclear ring contraction, and maturation. The acrosome has been abruptly formed following the collapse of expanded acrosomal vesicle without the cap stage described in the eutheria. The flatly condensed nucleus rotates obliquely to the axis of the axial filament complex and the folded acrosome covers the anterior third of the dorsal nuclear surface forming a wide subacrosormal space as the nuclear ring has contracted. The Sertoli cell reaction and spur are prominent during the nuclear protrusion and rotation stages. A mature spermatozoon has S-shape head which has an extended part reaching to the vicinity of the middle piece.

  • New type of ordering along 1110] direction was found In BZT sintered at $1400^{\circ}C$ for 90 hours by using X-ray diffraction analysis and high resolution transmission electron microscopy. The wavelength of the modulated structure was 0.9 nm which is three times larger than the interplanar distance of (110) plane. New type of ordering is considered to be formed as a result of Zn vancacy ordering and new structural model was proposed. The computer simulated electron diffraction and the structural images are in good agreement with those obtained by experiments.

  • We report a fabrication of 6, 13-bis(triisopropylsilylethynyl) (TIPS) pentacene nanowires that made on Si substrates by liquid bridge-nanotransfer molding (LB-nTM) with polyurethane acrylate (PUA) mold. LB-nTM is based on the direct transfer of various materials from a stamp to a substrate via a liquid bridge between them. In liquid bridge-transfer process, the liquid layer serves as an adhesion layer to provide good conformal contact and form covalent bonding between the TIPS-pentacene nanowire and the Si substrate. The patterned TIPS-Pentacene nanowires have been investigated by Atomic force microscopy (AFM), Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM) and electrical properties.

열압착 공정을 통해 형성된 나노와이어와 금속전극간의 기계적/전기적 접촉특성 분석

  • 나노와이어는 센서, 메모리소자, 태양전지등과 같은 다양한 소자로 응용이 되고 있다. Bottom-up 방법으로 길러진 나노와이어들을 금속전극 위에 정렬 및 접합시킬 때, 나노와이어와 금속전극간의 기계적 접합강도와 안정적인 전기적 특성이 매우 중요하다. 본 연구에서는 열압착 공정과 솔더전극(Cr/Au/In/Au, Cr/Cu/In/Au)을 사용함으로써, 나노와이어를 금속전극에 압입시켜 강한 기계적 접합강도와 안정적인 전기적 특성을 얻을 수 있는 공정을 제안하였다. 나노와이어와 금속 전극간의 접합부 분석을 위해 scanning electron microscopy (SEM)와 transmission electron microscopy (TEM)을 이용하였으며, 기계적 특성은 lateral force microscopy (LFM), 전기적 특성은 semiconductor analyzer (Keithley 4200-SCS)를 사용하여 측정하였다. 접합강도 측정결과 lateral force가 나노와이어에 가해질 때 나노와이어가 파괴되는 힘에서도 나노와이어와 금속전극간의 접합부파괴가 일어나지 않았다. 또한 나노와이어와 금속전극간의 전기적 접촉특성은 안정적인 ohmic contact을 이루었다.

  • The compound eyes of the adult Drosophila melanogaster were. fixed in 1.25 per cent glutaraldehyde and 1 per cent osmium tetroxide buffered with sodium cacodylate at pH 7.2. Double fixed specimens were dehydrated using the alcohol series and embedded in Epon 812. They were sectioned with porter blum and JUM 5B ultra-microtome and then stained with lead hydrooxide and uranyl acetate. All thin sections were examined with Hitachi HS-7 or HU-11 electron microscope. The rhabdomere of the compound eye is composed of numerous microvilli packed, arranged, and projected from inner edge of each retinal cell. Each microvillus consisted of a centrum, about $82{\AA}$ in diameter, surrounded by the substances, about $105{\AA}$ in width, which were bounded with double membrane about $44{\AA}$ in thickness. In each inner edge of the microvilli, there was a cylinder, about $175{\AA}$ in diameter, in parallel with retinal cells, which contained a cylindrical axis about $583{\AA}$ in diameter. The surface of the outer edges .of .the microvilli was bounded with reticullar substances about $500{\AA}$ in thickness.

  • Midgut cells of the dead Dendrolimus spectabilis larvae by infection of D. spectabilis nuclear polyhedrosis virus (DsNPV) were observed by transmission electron microscopy. DsNPV replicated in the nucleus of the infected midgut cells and the virogenic stroma of DsNPV appeared in the nucleus, from which nucleocapsids were formed. Also the formation of polyhedral inclusion bodies were observed in the nucleus.

  • In performing in situ heating transmission electron microscopy (TEM) for materials characterizations, arising concerns such as specimen drifts and unintentional Cu contamination are discussed. In particular, we analysed the thermal and mechanical characteristics of in situ heating holders to estimate thermal drift phenomena. From the experimental results, we suggest an empirical model to describe the thermal drift behavior so that we can design an effective plan for in situ heating experiment. Practical approaches to minimize several hindrances arisen from the experiment are proposed. We believe that our experimental recommendations will be useful for a microscopist fascinated with the powerful potential of in situ heating TEM.

  • The endocrine cells in the midgut epithelium of the Japanese cockroach, Periplaneta japonica were observed by the light and electron microscopy. The midgut epithelium of the last instar larva and adult cockroach consisted of principal columnar cells, regenerative cells, and secretory granular cells. Midgut endocrine cells were positioned basally as a cone-shaped single cell in the epithelium or underneath the regenerative crypt cells. When midgut epithelium grows and the cell composing it transforms, between the endocrine cells and regenerative cells were made desmosome type junction and large vesicular shaped stretches of loose contact. The endocrine cells were characterized by a clear cytoplasm with abundant Golgi complex and numerous secretory granules. The secretory granules in the cell were spherical and electron dense with their diameter of $200{\sim}400nm$. The secretory granules have been observed as discharged by exocytosis on the basal and lateral side of the cell.

  • The distribution, ultrastructure, and cytochemical properties of microbodies (peroxisomes) from the main roots and mature leaves of Panax ginseng were studied by electron microscopy included the activity of catalase in 3, 3'-diaminobenzidine (DAB) medium at pH 9, using glutaraldehyde-fixed tissues. The microbodies, which are about $0.5{\sim}1.5{\mu}$ diameter, were described from mesophyll cells of mature leaves and storage cells and cambial cells of main roots. The microbodies of the ginseng include the coreless, homogeneous matrix, in which catalase activities are present.
