Detection and Extraction Condition of Physiological Functional Compounds from Bran of Heugjinju rice (Oryza sativa L.) (흑진주벼 미강으로부터 생리기능성 물질의 탐색 및 추출조건)

  • Physiological functionalities in various extracts of Heugjinju rice bran were determined and its optimal extraction condition were also investigated Angiotensin-converting enzyme(ACE) inhibitory activity, fibrinolytic activity and tyrosinase inhibitory activity were higher in water extracts than those of 80% ethanol and methanol, hexane. Electron donating abilities were 97.8% in hexane extract and 83% in water extracts. ACE inhibitor was maximally extracted from Heugjinju rice bran when it was treated with 20 times of distilled water for 12 h at 20 $\^{C}$. Fibrinolytic compound was also maximally extracted by treatment of 10 times of distilled water for 18 h at 20 $\^{C}$. Electron donating compound and tyrosinase inhibitor were maximally extracted by treatment of 20 times of hexane and 10 times of distilled water at 20 $\^{C}$ for 18 h, respectively

Functional Properties of Tea-fungus Beverage (Tea-fungus 발효음료의 기능성)

  • To develope tea-fungus beverage(TB), media added various kinds of ingredients including black tea, persimmon leave tea, pine needle, mugwort mycelia and fruiting body of Cordyceps spp. were prepared fur fermentation. Tea-fungus beverage(TB) was prepared with tea-fungus by fermentation for 2 weeks at 30 $^{\circ}C$. Functional properties of antibacterial activity, electron donating ability and nitrite scavenging ability were investigated in tea-fungus beverage(TB) and 5 times diluted tea-fungus beverage(DTB). Antibacterial activity against pathogenic E. coli and S. aureus was excellent in TB added pine needle, mycelia and fruiting body of Cordyceps japonioa while no activity in TB added persimmon leave tea. Electron donating ability of TB were ranged from 41% to 87% in TB and were ranged from 11% to 63% in DFB, high ability was in TB and DTB added pine needle and persimmon leave tea, while low ability in FB and DTB added mycelia and fruiting body of C. militaris. Nitrite scavenging ability was 63% in TB added black tea and 44% in TB added persimmon leave tea. Other ingredients added TB had weak nitrite scavenging ability.

Analysis of Chemical Compositions and Electron-Donating Ability of 4 Korean Wild Sannamuls (산나물류의 식품 화학적 성분과 전자 공여능)

  • The composition analysis and electron donating activity of 4 kinds of Korean wild sannamuls[Cacalia firma Komar. (CFK), Aster scaber Thunb. (AST), Ligularia fischeri (Ledeb.) Turcz. (LFT), Synurus palmatopinnatifidus (Makino) Kitam., SDN)] were investigated. The Korean edible mountainous vegetable was named a sannamul or chwinamul in Korea, generally. Most of them, the characterization of the each vegetable was reported, individually without comparative research. This study was compared the proximate compositions, mineral, dietary fiber and electron donating abilities on 2,2-diphenyl-1-picrylhydrazyl (DPPH) of sannamuls. In the chemical analysis of sannamuls, CFK was shown higher value in ash ($5.4{\pm}0.18%$), crude fat ($2.5 {\pm}0.14%$), carbohydrate ($8.1{\pm}0.21%$), potassium ($475.45{\pm}0.34mg$) and magnesium ($64.70{\pm}0.48mg$) than others. Each mineral of the 3 other samples was a similar value but phosphorus in LFT was contained $62.91{\pm}0.43%$. The compositions of total dietary fiber of the samples were listed CFK LFT SDN AST and the $RC_{50}$ Value on DPPH radicals, LFK CFT AST SDN. These results have susggest that CFK and LFT may be useful for a funtional material to food development.

Antioxidative Activity of the Durian (Durio zibethinus) Extract (두리안 추출물의 항산화 활성)

  • The purpose of this study was to examined the antioxidant activities by water and 70% ethanol extract from durian (Durio zibethinus.) seed, sarcocarp and peel. Durian extract were studied for reducing sugar content, polyphenol content, superoxide dismutase (SOD) like activity, electron donating ability, nitrite scavenging ability, flavonoid content, hydroxy radical scavenging activity. Reducing sugar content were increased peel > sarcocarp > seed. Total polyphenol, flavonoid content, DPPH radical scavenging ability and SOD like activity were increased seed > peel > sarcocarp. Total polyphenol content was relatively high as $21.90{\pm}0.50mg/g$ in the ethanol extract of the seed. DPPH radical scavenging ability was relatively high as $62.08{\pm}2.63%$ in the water extract of the seed. Nitrite scavenging ability was no significant difference. Hydroxy radical scavenging activity was increased seed > peel > sarcocarp, was relatively high as $58.27{\pm}1.13%$ in the water extract of the seed.

Antioxidative Properties and Whitening Effects of the Polygoni Multiflori Radix, Polygonati Rhizoma and Ephedrae Herba (하수오, 황정 및 마황의 항산화성 및 미백효과)

  • In order to fine antioxidant and whitening agent source from nature, the comparisons of antioxidative activity and tyrosinase inhibitory activity were carried out for various ethanol extract on Polygoni Multiflori Radix, Polygonati Rhizoma and Ephedrae Herba. Comparing for three ethanol extracts, the highest electron donating ability was found at Polygonati Rhizoma (86.6%), but, the highest SOD-like ability, at the Ephedrae Herba (47.8%). Xanthine oxidase experiment exhibited 95.7% of hindrance effect in Ephedrae Herba, and 84.0% in Polygonati Rhizoma. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 6.5% in the Polygoni Multiflori Radix, 32.6% in the Polygonati Rhizoma, 64.0% in the Ephedrae Herba. Based on these results, we suggest that the ethanol extracts of Polygoni Multiflori Radix, Polygonati Rhizoma and Ephedrae Herba can be used as food and cosmetic ingredients.

Functional Composition and Antioxidant Activity of Eel Teriyaki Sauce with Bokbunja (Rubus coreanus Miquel) (복분자 첨가 장어 데리야끼 소스의 기능성 성분 및 항산화 활성)

  • Antioxidant activity of factory eel teriyaki sauce with bokbunja was investigated by functional analysis. Sugar content analysis of eel teriyaki sauce-free with bokbunja showed fructose and glucose but not maltose. Anthocyanin content of eel teriyaki sauce with bokbunja juice concentrate was 40%. DPPH radical scavenging activity and total polyphenol content of bokbunja added eel teriyaki sauce increased with increasing amount of bokbunja and were 80% higher than control sample. Total polyphenol content and DPPH radical scavenging activity were closely related to electron donating ability. The preferred concentrations of eel teriyaki sauce for commercial production were 60% for bokbunja, 40% for bokbunja concentrate, 20% for bokbunja powder eel.

Antioxidative Property and Whitening Effect of the Pueraria Radix, Poria Cocos and Coptidis Rhizoma (갈근, 복령 및 황련의 항산화성 및 미백효과)

  • In order to fine antioxidant and whitening agent source from nature, the comparisons of antioxidative activity and tyrosinase inhibitory activity were carried out for various ethanol extract on Pueraria Radix, Poria Cocos, and Coptidis Rhizoma. Comparing for three ethanol extracts, the highest electron donating ability was found at Poria Cocos (81.2%), but, the highest SOD-like ability, at the Coptidis Rhizoma(58%). Xanthine oxidase experiment exhibited 23.3% of hindrance effect in Pueraria Radix, and 79.3% in Poria Cocos. To evaluate at the whitening effect, tyrosinase inhibitory activity was conducted. Tyrosinase inhibitory activity was detected at 17.9% in the Puerariae Radix, 5.2% in the Poria Cocos and 83.3% in the Coptidis Rhizoma. From these results, we suggest that the ethanol extracts from Pueraria Radix, Poria Cocos, and Coptidis Rhizoma can be used for cosmetic ingredients.

Study on the Antioxidant and Anticancer Effects of Extract of Stamens of Nelumbo nucifera and Kaempferol (연수 추출물과 Kaempferol의 항산화 및 항암작용에 관한 연구)

  • Objectives : The present study was performed to clarify the antioxidant and anticancer effects of extract of stamens of Nelumbo nucifera and Kaempferol. Methods : Antioxidant effect was measured by DPPH-radical scavenging activity for electron donating ability (EDA), superoxide dismutase (SOD)-like activity for SOD and lipid peroxidation. Anticancer effect was assessed by MTT absorbance for cytotoxicity. Results : Vitamin E, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera increased SOD-like activity and DPPH-radical scavenging activity dose-dependently. On the contrary, lipid peroxidation was time-dependently decreased. Furthermore, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera significantly decreased the growth rate of C6 glioma cells. Conclusions : These results suggest that ethyl acetate extract of stamens of Nelumbo nucifera may be a putative antioxidant or anticancer substance.

Screening of Hyaluronidase Inhibitory and Free Radical Scavenging Activity in vitro of Traditional Herbal Medicine Extracts (생약재 추출물의 hyaluronidase 저해 및 라디칼 소거 활성 검색)

  • For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined their hyaluronidase inhibitory effect and radical scavenging activity in vitro. From the result of the hyaluronidase inhibitory activity using a Morgan-Elson assay, Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix, Acanthopanacis Cortex, Chaenomelis Fructus, Amomum xanthioides Wallich and Moutan Radicis Cortex showed more than 50% hyaluronidase inhibitory effects at the concentration of 10 mg/mL. In the various solvent fractions (n-hexane, chloroform, ethyl acetate, water) prepared from ethanolic extracts, the ethyl acetate fraction of all extracts tested showed strong activity. Antioxidative activity was evaluated by assaying electron-donating ability to DPPH free radical and scavenging of hydroxyl radical (ㆍOH) generated through Fenton reaction, respectively. Rubus coreanus Miq, Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed high activity more than 90%, yet similar activity to $\alpha$-tocopherol and BHA at the concentration of 1 mg/mL in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical were investigated using a 2-deoxyribose oxidation method and tested all extracts showed significant radical scavenging activity. The experiment was also performed to examine whether herbal medicines having significant lipid peroxidation inhibitory activity, Schizandrae-Fructus is the strongest inhibitory activity in both linoleic acid and liposome peroxidation.

The Antioxidant and Nitrite Scavenging Activity of Wild Grape(Vitis coignetiea) Wine (머루와인의 항산화성 및 아질산염 소거능)

  • Total anthocyanin and color intensity contents of wild grape wine were $4.3{\pm}0.3%$ and $10.2{\pm}0.8%$, respectively. The contents of total phenols and flavonoid in wild grape wine were $18.8{\pm}3.9$ mg/100 g, $0.5{\pm}0.2$ mg/100 g, respectively. Total mineral content in wild grape wine was $22.6{\pm}0.2$ mg/100 g and the 10 minerals of the potassium content ($5.3{\pm}0.2$ mg/100 g) was the highest. Electron donating abilities of wild grape juice at concentration of 1,000 ${\mu}L/mL$ were $90.4{\pm}1.8%$. Reducing power of wild grape wine at concentration of 1,000 ${\mu}L/mL$ were 0.932. The electron donating abilities and reducing power were increased significantly by increased the sample concentration in the reaction mixture. The nitrite scavenging ability was dependent on pH of reaction mixture and sample concentration. The nitrite scavenging ability of wild grape win was $76.3{\pm}1.3%$ at concentration of 1,000 ${\mu}L/mL$ under pH 1.2.
