Up-regulation of Early Growth Response-1 Expression by Endoplasmic Reticulum Stress

  • Endoplasmic reticulum (ER) plays formation of disulfide bonds and proper folding of secretory proteins. Cellular responses to ER stress enhances the stress-activated kinase pathway and the induces a lot of immediate-early genes. Among of them, the early growth response-1 (Egr-1), a transcription factor, which plays an important role in cell growth, development, differentiation, apoptosis and various types of injury. For that reason, we have tested the expression of Egr-1 against ER stress inducible drugs (tunicamycin, DTT, A23187 and BFA) to understand what kind of aspect occurred by ER stresses.

A study on performance and smoke emission characteristics by blending low purity methanol in a DI diesel engine with the EGR rates of 0, 12.8 and 16.5%

  • The purpose of this study is to investigate experimentally the effect of low purity methanol (LPM) on performance and smoke emission characteristics by using a four-cycle, four-cylinder, water-cooled, direct injection diesel engine with EGR system. The experiments are performed by the change of engine load in the engine load ranges of 25 to 100% with an interval of 25% under the constant engine speed of 2000 rpm. The LPM in the fuel blends contained 24.88% water by volume. The blended fuel ratios of diesel oil to LPM are maintained at 100/0, 95/5, 90/10 and 85/15% on the volume basis. In this paper, EGR rates are varied in three conditions of 0, 12.8 and 16.5%. The result shows that the brake power of a blended fuel with 15% LPM is reduced more 11.1% than that of the neat diesel oil at the full load with the EGR rate of 16.5%. At this condition, also, the brake specific fuel consumption (BSFC) is increased by 3.2%, the exhaust gas temperature is decreased by 10.7%, the smoke opacity is decreased by 18.7% and the brake thermal efficiency is increased by 7.3%. The sharp reduction of smoke opacity for a blended fuel with the LPM content of 15% at the full load without EGR system is observed by 68.4% compared with that of the neat diesel oil due to the high oxygen content of LPM.

EFFECT OF VALVE TIMING AND LIFT ON FLOW AND MIXING CHARACTERISTICS OF A CAI ENGINE

  • To increase the reliability of auto-ignition in CAI engines, the thermodynamic properties of intake flow is often controlled using recycled exhaust gases, called internal EGR. Because of the internal EGR influence on the overall thermodynamic properties and mixing quality of the gases that affect the subsequent combustion behavior, optimizing the intake and exhaust valve timing for the EGR is important to achieve the reliable auto-ignition and high thermal efficiency. In the present study, fully 3D numerical simulations were carried out to predict the mixing characteristics and flow field inside the cylinder as a function of valve timing. The 3D unsteady Eulerian-Lagrangian two-phase model was used to account for the interaction between the intake air and remaining internal EGR during the under-lap operation while varying three major parameters: the intake valve(IV) and exhaust valve(EV) timings and intake valve lift(IVL). Computational results showed that the largest EVC retardation, as in A6, yielded the optimal mixing of both EGR and fuel. The IV timing had little effect on the mixing quality. However, the IV timing variation caused backflow from the cylinder to the intake port. With respect to reduction of heat loss due to backflow, the case in B6 was considered to present the optimal operating condition. With the variation of the intake valve lift, the A1 case yielded the minimum amount of backflow. The best mixing was delivered when the lift height was at a minimum of 2 mm.

배기가스 재순환을 적용한 희박-과농 연소시스템의 공해물질 배출특성 연구 (The Pollutant Emission Characteristics of Lean-Rich Combustion System with Exhaust Gas Recirculation)

  • In this study, the CH4/air lean-rich combustion system with exhaust gas recirculation (EGR) was investigated to explore the potential for lowering pollutant emissions. To achieve this purpose, experiments of lean-rich combustion system with EGR were conducted to measure the changes in the characteristics of the pollutant emission and flame shape with various equivalence ratios and EGR rates. Here, this study was applied to the fuel distribution ratio of 3:1 for the formation of the lean and rich flames. Additionally, the results were compared with $CH_4$/air lean premixed combustion system. The results show that flame shape of lean-rich combustion system was determined by lean and rich equivalence ratios (${\Phi}_L$ and ${\Phi}_R$) and stratified flame was formed with increasing ${\Phi}_R$. According to the pollutant emission characteristics based on experimental results, the NOx and CO emission index (EINOx and EICO) decreased with increasing EGR rate. Especially, in the range needed to form a stable flame, the reduction rates of EINOx and EICO were approximately 47% and 48% for an EGR rate of 25%, global equivalence ratio of 0.85 and ${\Phi}_L$ of 0.80 compared with lean premixed combustion system (${\Phi}$ = 0.78).

  • 점막 상피는 외부 인자를 감지하는 최전선의 인식부위로서 외부스트레스 자극을 하부의 반응 신호로 전달하는 주요 세포이다. 리보솜독성 반응을 유발하는 데옥시니발레놀 (DON) 및 그 관련 곰팡이 독소는 푸자륨 곰팡이 오염에 의한 식중독성 소화기 염증성 질환과의 연관성이 알려 져 있다. 본 연구의 목적은 DON이 상피세포 감지 신호 전달 분자로서 PKR과 EGR-1이 관련 되고 이들이 상피세포에서의 염증성 사이토가인 인터루킨 8의 생성에 관련 된다는 가정 하에서 연구를 수행 하였다. PKR 발현의 세포내 작용 억제는 DON에 의해 유도되는 인터루킨 8의 생성을 감소시켰다. 또한 DON에 의한 IL-8 전사 활성화는 PKR 억제에 의하여 장관 상피세포에서 감소하였다. PKR 저해제의 처리는 EGR-1 promoter 활성, mRNA, 단백질 유도 등을 감소를 유발하였으며 MAP kinase (ERK1/2, p38, JNK)는 변화가 적거나 오히려 PKR 저해제의 전처리에 의하여 항진 되었다. 결론적으로 DON에 의해 자극된 감지신호인 EGR-1은 자체적으로 또는 PKR 신호를 경유하여 인터루킨8의 생산을 항진하는데 주요한 기능을 하였다. 이를 통하여 향후 리보솜 독성 반응과 관련된 소화기 염증유발의 주요한 기전을 제공하고 있다.

  • Recently, the application of variable geometry turbocharger (VGT) to the high speed direct injection (HSDI) diesel engine has gained more and more interest in automotive industry. A steady state experimental investigation has been undertaken on a 1.5L HSDI diesel engine to verify the benefits of VGT comparing to the standard engine having a waste gate turbocharger (WGT). Specifically, part load performances (e.g., fuel economy and emission) have been investigated under various vane angles of the VGT. The results show that the real exhaust gas recirculation (EGR) rate as well as the pumping loss is very important to improve break specific fuel consumption (BSFC). It was previously known that the pumping loss only is a main parameter. In addition, the trade-off relationship between BSFC and NOx according to boost pressure, and the decreasing tendency of NOx with increasing real EGR rate have been verified. 1-D numerical analysis also has been performed, and the numerical results are in good agreement with experimental results.

온도와 시간을 주요 변수로 한 냉장 돈육에서의 native isolated Listeria monocytogenes에 대한 성장예측모델 (Predictive Growth Model of Native Isolated Listeria monocytogenes on raw pork as a Function of Temperature and Time)

  • 본 연구는 냉장돈육에서의 식중독 원인균이면서 냉장온도에서 성장이 가능한 병원성균인 L. monocytogenes에 대한 적절한 위생관리를 제시하기 위하여 포장돈육 작업장 원료돈육에서 분리된 야생균주 L. monocytogenes 이용하여 돈육포장공정 및 유통조건에서의 L. mnocytogenes에 대한 성장예측모델을 제시하고자 실시하였다. 성장실험은 온도 5, 10, 15, $20^{\circ}C$ 시간은 0, 1, 2, 3, 18, 48, 120시간에서 실시하였으며, 이를 바탕으로 온도별 Gompertz value인 A, C, B, M의 값과 Growth kinetic인 exponential growth rate(EGR), generation time(GT), lag phase duration(LPD), maximum population density(MPD)를 산출하였다. GT, LPD는 온도가 상승할수록 그 값이 점점 낮아지는 경향을 나타났으며, EGR의 경우는 반대로 온도가 높아질수록 점점 높아지는 경향을 나타냈다. Gompertz value중 B와 M 값을 이용하여 온도를 주요 control factor로 선정한 반응표면분석(Response surface analysis)을 실시하여 온도에 따른 다항식을 산출하였고 이 식을 Gompertz 식에 적용하여 온도와 시간에 따른 냉장돈육에서의 L. monocytogenes에 대한 성장정도를 예측할 수 있는 성장예측모델을 제시하였다. 개발된 성장예측모델에 대한 검증은 GT, LPD, EGR에 대한 실험값과 예측값의 비교를 통하여 실시하였으며, 그 결과 GT, LPD, EGR 모두 통계적으로 유의하게 나타났다(p<0.01). 따라서 이 모델은 risk assessment 중 exposure assessment를 위한 성장예측모델로 충분히 이용가능 한 것으로 보이며, 추후 냉장돈육 위성관리기준에 대한 과학적 근거자료로 활용될 수 있을 것으로 보인다.

고온 배기가스의 산소농도가 비예혼합화염의 NOx 발생에 미치는 영향 (Effects of Oxygen Concentration on the NOx Emission of Non-premixed Flame in Hot Exhaust Gas)

  • The present study examined the possibility of NOx reduction in the high temperature industrial furnaces, duct burner of gas turbine cogeneration and two-stage gas turbine combustor. The experimental study was carried out for the non-premixed flame of second stage combustor with the variations of oxygen concentration in the hot exhaust gas of first stage combustor. It also examined the flammability range, temperature and NOx, $CO_2$, $O_2$formation in the combustor with respect to oxygen concentration in which the fuel(natural gas) is supplying into the hot exhaust gas. The results show that the inner temperature of flame reaches 1,20$0^{\circ}C$ at EGR $O_2$23% and that 15ppm of NOx at EGR $O_2$15.5% increases up to 60ppm at EGR $O_2$23%. It is believed that Fenimore's prompt NOx mechanism is more influential on the NOx formation than Zeldovich's thermal NOx mechanism does.

  • HCCI engines fueled DME and natural gas have been studied on single-cylinder engine due to availability of reducing on $NO_X$ and PM simultaneously without deteriorating into high thermal efficiency, and thus it is clarified that higher maximum engine load is achieved as DME equivalence is smaller. In this study, combustion tests were accomplished on multi-cylinder engine for practical use of it. When minimum DME equivalence achieved maximum engine load on single-cylinder engine was applied to 4-cylinders engine, there was in unstable running condition that engine revolution fluctuated greatly and cyclically. It is the reason what misfire occurred intermittently with one the same as minimum DME equivalence on single-cylinder due to increase in energy for ignition at No. 1 cylinder with lower cylinder liner temperature. Maximum engine load was achieved by adopting EGR, though it decreased because of knocking at smaller engine load than single-cylinder due to increase in minimum DME equivalence.

  • Emission characteristics of regulated pollutants (CO, NOx, HC and PM) and hazardous air pollutants (HAPs) from diesel heavy duty trucks equipped with EGR+pDPF and SCR for Euro 5 emission standards were investigated using a chassis dynamometer. In the case of regulated pollutants, diesel heavy duty trucks with EGR+pDPF emitted 79% less CO than those with SCR. Also, those with the SCR emitted 36% less NOx than those with the EGR+pDPF. The results of VOCs have show that alkanes emissions for heavy duty trucks with the EGR+pDPF and the SCR have been higher than alkenes, cycloalkanes and aromatics. In the case of individual VOCs, the highest of propene emissions for 11.3~16.1% occupied. For aromatics group, benzene emissions are the highest percentage for 4.4~15.5%. In the future, the results of present study will provide basic data to set up HAPs emissions inventory for mobile source.
